Modifications of the digestive tract in rats submitted to an orbital flight aboard the Soviet satellite Cosmos 1129.

Investigations carried out on the digestive tract of rats submitted to an 18 1/2-day orbital flight aboard the Soviet biosatellite Cosmos 1129 revealed that flight produces a reduction of the glycoproteins (GP) of the submaxillary glands, gastric mucosa and the goblet cells of the small and large intestine, the last being the most nonreactive, as well as an augmented staining for leucine-aminopeptidase (LAP) and acid phosphatase (AP). In lamina propria an accumulation of eosinophils was noticed. These changes were more intense than those produced in the groups of rats maintained "synchronous" under environmental conditions similar to flight. On the basis of previous data obtained by various authors and by ourselves on hypokinetic rats from the biosatellite Cosmos 936, it can be concluded that these reactions have a nonspecific, stress character. A stress of restraint applied daily for 2 1/2 hours during a period of 6 days to the animals immediately after landing produced modifications similar to those following flight but more intense, thus underlying the hypothesis of a stress reaction.

[Normal values of the spontaneous cytochemical NBT test in adults].

The description is given of a working method adapted after Park, Fikrig and Smithwick, for testing the capacity of neutrophils to reduce tetrazolium nitroblue salts spontaneously. In contrast to the original technique, the present method uses : 10-20 units indigenous heparin/ml blood, leukocyte concentrate, other reaction times at 37 degrees C, contrast staining with a week Giemsa solution. Applied to young healthy subjects, the mean proportion of NBT-positive neutrophils was of 4.3 +/- 3.09 and the absolute number approximately less than 700/ml blood. The results obtained were consistent with the published data.

The digestive tract after 7 days of restraining conditions in normal and adrenalectomized rats.

Hypokinesia produced specific reactions in tissues related to motion (bones, skeletal muscles and myocardium) and nonspecific in others, as that of the digestive tract after 7 days of hypokinesia. A decreased content of glycoproteins from mucous salivary glands, stomach, small intestine and colon, and an increased intestinal content of leucine-aminopeptidase and acid and alkaline phosphatase were found as a result of histochemical reactions. An important plasmatic corticosterone increase was also found. The same histochemical modifications appeared in adrenalectomized rats in which the plasma corticosterone was substantially diminished. Therefore these glycoproteins and enzymatic secretions are not dependent on the glucocorticoid response.

Digestive tract of rats after hypokinesia and hypergravitation.

We found in rats submitted to a 7 days interval of hypokinesia a decreased glycoprotein content in salivary mucous glands, gastric and intestinal mucosa, an increased glycoprotein content and an increased content of leucine-aminopeptidase, acid and alkaline phosphatase. Similar changes were found after a short interval of centrifugation (+6 Gz, 10 min). These nonspecific responses were more accentuated if the hypergravitation was performed after 7 days of hypokinesia. We reproduced this way this events that occurred in space flight and deacceleration before landing.

Experimental researches regarding the metabolic changes determined by the prolonged exposure to light or by keeping in darkness.

The authors conducted complex investigations in the domain of the intermediary metabolism under experimental conditions on 24 dogs grouped into three lots: lot A, twelve dogs, exposed for six months to the action of continuous light; lot B, six dogs, maintained in darkness; lot C, six dogs, maintained for the same period in the animal house, getting the same food as the other two lots, which are the control lot. We determined some biochemical, hematological, enzymatical parameters, before the beginning of the experiment, and after 24 hours, 48 hours, 7, 14 and 21 days, one, two, three, six months. Then the animals were sacrificed, and fragments from the aorta, coronary vessels, myocardium, brain, adrenals, pancreas and liver were examined both on the optical and on the electronic microscope and we analysed some fats from the vascular tissues. The net decrease of proteinemia with lot A was registered, concomitantly with a net decrease of serumalbumins, of the alpha 1-globulins and of the gamma-globulins, and with a rise of the alpha 2- and beta-globulins. The values of glycemia increased with lot A, and less with lot B. The thrombocytes decreased obviously with lot A, and less with lot B. GPT decreased with lot A, and GOT increased. The serum Zn decreased slowly with lot A, and more irregularly but more obviously with lot B. The serum Cu increased with lot A, and decreased with lot B. Alterations on the level of arterioles from the brain, aorta, myocardium and pancreas were evidenced, as well as electronmicroscopical alterations of the arteriolar walls and of the cellular organelles. The lipids from the brain, aorta and myocardium increased with both of the experimental lots and so did the cerebrosides, lecithins and the sphyngomyelin.

Histoenzymology of muscular arterioles.

The authors investigated the dehydrogenase histochemistry of arterioles on 22 muscular biopsies from 14 male and 8 female patients with different clinical forms of atherosclerosis, and in 5 controls. There was a diminution with age of all the enzymes studied. In 3 of 6 cases with pathological lesions (thickening of endothelium, fragmentation of the internal elastic lamina, thrombosis) there was a regional diminution of NADH-diaphorase and NADPH-diaphorase activities parallel with an increase of the reaction for lactic dehydrogenase and glutamate dehydrogenase in the muscular cells and endothelium.
