Arthrography with Amipaque (metrizamide) and other contrast media. A roentgenographic and histologic evaluation in rabbits.

Arthrography of the knee joint was performed in 30 rabbits for comparison of Amipaque (metrizamide) with Urografin 60% (meglumine-sodium diatrizoate), Isopaque Amin (meglumine-calcium metrizoate), Conray Meglumin 282 (meglumine iothalamate) and Dimer-X (meglumine iocarmate). Films taken at regular intervals show longer contrast duration, and less hydrarthrosis after injection of Amipaque. Minimal focal inflammatory reaction in the synovial membrane is found in many cases when the animals are sacrificed 1 day or 7 days after the injections. No significant difference can be found between the contrast media, and similar changes are also seen after injection of physiologic saline.

Histologic effects of Amipaque (metrizamide) and various contrast media on mouse peritoneum.

The histologic effects of Amipaque and various commonly used contrast media on mouse peritoneum were studied by intraperitoneal injections of Amipaque 370 mg I/ml and 170 mg I/ml, Conray 300 mg I/ml, Conray Meglumine 282 mg I/ml, Urografin 60% and 76%, Gastrografin and barium sulfate. Amipaque and the other water-soluble media cause no deleterious effects, whereas barium sulfate evokes granulomatous inflammatory reaction. Amipaque is suitable for gastrointestinal studies when leakage into the peritoneal cavity is expected.

Fine needle aspiration cytology as a diagnostic approach to lymphangioleiomyomatosis. A case report.

BACKGROUND: Lymphangioleiomyomatosis is a rare condition in women of childbearing age that affects the lymphatic system, giving rise to blockage of the thoracic duct with chylothorax and chyloascites. The diagnosis is often delayed because the characteristic muscular proliferation may be missed by the pathologist. CASE: A 29-year-old female presented with lymphangioleiomyomatosis and bilaterally dermoid ovarian cysts. She had enlarged paraaortal and supraclavicular "lymph nodes" and pleural chylothorax on the left side. Fine needle aspiration cytology from the supraclavicular "lymph node" yielded clear, watery fluid containing groups of relatively large monomorphic spindle cells. The final diagnosis was possible after histopathologic examination of an excised paratubar lymph node. CONCLUSION: Lymphangioleiomyomatosis can be suggested by means of ultrasonography and fine needle aspiration cytology.

Molecular characterization of cellular proteins from colorectal tumors.

AIMS AND BACKGROUND: Recent evidence has suggested that progressive stages of colorectal tumorigenesis can be defined by a sequence of genetic events characterized by altered expression of certain genes and the appearance of cancer-specific proteins. Although the significance of these events is still not clear, expression of cancer-specific protein components may be directly involved in the neoplastic transformation. The purpose of the present study was to compare molecular characteristics of cellular proteins from human colorectal tumors and normal colonic mucosa. METHODS: Normal mucosa and colorectal tumors from 18 patients were fractionated by a differential centrifugation scheme into four cellular fractions, i.e., nuclear, mitochondrial (10P), microsomal (100P) and cytosolic (100S). The proteins of these fractions from normal and tumorigenic mucosa were analyzed by one-dimensional polyacrylamide gel electrophoresis followed by Coomassie brilliant blue R-250 and silver nitrate staining. Nuclear proteins from normal and neoplastic tissues which had revealed the most significant diversities were further characterized by two-dimensional gel electrophoresis. Electrophoretically cancer-specific nuclear proteins in the molecular mass zone 35-40 kDa were used as immunogen to produce rabbit polyclonal antibodies. RESULTS: Electrophoretic analysis by one-dimensional gel electrophoresis showed clear differences in molecular characteristics of cellular proteins between normal and tumorigenic mucosa, especially among nuclear fractions. The latter were also confirmed by their two-dimensional electrophoresis results. Rabbit antibodies raised against electrophoretically specific nuclear proteins characterized by molecular mass of 35-40 kDa cross-reacted with 36 kDa polypeptide in 15 of 18 (83.3%) studied nuclear fractions of colorectal tumors but not with any normal mucosa. In some cases, nuclear cancer-associated components of 38 and 40 kDa were also recognized by these antibodies. CONCLUSIONS: During colorectal carcinogenesis, specific expression of several nuclear proteins takes place. One of them, the polypeptide of 36 kDa not found in normal colonic epithelium, was shared by over 83% of the studied carcinomas despite variations in detailed cancer properties. This particular nuclear protein may be considered as a potential marker for the colon malignancy.

Microscopic polyangiitis diagnosed at hysterectomy.

A 78 year old female developed polyneuropathy, weight loss, malaise, and joint pain. Necrotizing vasculitis was diagnosed at hysterectomy, and later renal biopsy demonstrated focal segmental necrotizing glomerulonephritis. The pathological findings together with the presence of pANCA was consistent with a diagnosis of microscopic polyangiitis (MPA). This is the first clinical description of MPA with involvement of the uterus.

[Comparative molecular-genetic mapping of genomes of rye (Secale cereale L.) and other cereals].

The genetic map of rye consisting of 149 RFLP, 20 isozyme and 12 microsatellite markers was developed. Using the collection of cross-hybridizing probes, the presence of multiple translocations in rye genome with respect to wheat and barley genomes was shown. However, within large regions of genome a strict collinearity of marker order was observed that allow us to use the method of comparative mapping for an introduction of new genes. In the developed genetic map 18 morphological and breeding-valuable genes mapped in different rye populations were integrated. The comparative analysis of homeological loci in genomes of Triticeae species as well as in genomes of rice and maize was carried out. The genes controlling a number of morphological traits, plant height, photoperiodic response and winter/spring growth habit were shown to be conserve among cereals and to form clear homoeologous rows.

p53 protein expression in squamocellular carcinomas of the lip.

Twenty-six cases of squamocellular carcinomas of the lip were immunostained for p53 protein. Nuclear accumulation of p53 protein was observed in 88% of the 26 carcinomas, in 64% of adjacent "normal" epithelium and in all four cases with adjacent atypical squamous epithelium. 93% of the specimens (13/14) with p53 protein accumulation in "normal" epithelium demonstrated evidence of solar elastosis. UV radiation may induce p53 gene mutation as an early event in the neoplastic transformation.
