Occurrence of four types of growth hormone-related dwarfism in Israeli communities.

Data are presented on 121 dwarfed patients belonging to 98 Israeli families with 4 types of dwarfism related to deficiency or in activity of human growth hormone (hGH): isolated growth hormone deficiency (IGHD), partial hGH deficiency (pIGHD), multiple pituitary hormone deficiencies (MPHD) and Laron-type dwarfism (LTD). These series are believed to comprise most of the dwarfs of these types in Israel; however, their distribution among the various ethnic communities varies greatly: LTD (15 families) is confined to a few communities of Oriental Jews; only 3 of 30 families with IGHD, but 7 of 10 families with pIGHD, are Ashkenazi, whereas the clinic distribution of 42 families with MPHD corresponds roughly to that of the general population. These data seem to reflect the role of genetic factors in the etiology of the various types of dwarfism: the stronger the genetic component, the greater is its deviation from random occurrence among the various communities.

[Cardiovascular reactivity to arachidonic acid, prostaglandin E2, prostacyclin and bradykinin in carrageenan treated rats].

In the rat, during carrageenan-induced peritonitis, the hypotensive activity of arachidonic acid, which depends on PG biosynthesis, is increased during the first hours, and thereafter is not modified. The vasodilator action of PGE2 is reduced during the first day while the actions of PGI2 and bradykinin are not modified. The concentrations of plasma albumin and haptoglobin, two inhibitors of PG biosynthesis are reduced during the first hours. Thereafter the concentration of haptoglobin is increased by 100% while the concentration of albumin remains diminished. During this peritonitis, no plasmatic inhibitory influence on PG synthesis is seen. The anti-inflammatory action of carrageenans does not depend on PG synthesis inhibition.

[Radioimmunologic determination of human kininogen of high molecular weight].

HHW-kininogen has been purified from human plasma. We have then studied the optimal conditions for labeling this protein with 125I. This molecule has been used for the preparation of a radioimmunoassay. We have studied the analytical performances of our dosage : sensitivity, precision, accuracy and specificity.

Polymorphisms of red-green vision in some populations of Southern Africa.

Some 5,000 schoolboys of the Khoikhoi, Negro, "Coloured," and Malay populations were screened with the Ishihara plates, and those with defective red-green vision were diagnosed with an anomaloscope. The findings are presented in terms of the six protan and deutan mutant alleles, a few large population-samples (e.g., Nama and Zulu) being characterized by absence of the allele for protanopia. The overall frequencies of mutants range from less than 1% to over 4%. No correspondence was found between these data and linguistic affinities of eight Bantu-speaking groups, nor between the frequencies of colorblindness and previously estimated proportions of San genes in these eight populations; on the other hand, a north-south cline of increasing frequences of mutants and of dichromacies among the Bantu-speakers was noted. The overall frequency of defective red-green vision among Cape Coloureds, 3.3%, is compatible with previously estimated racial composition of this population. The Malay sample is characterized by the highest frequency of protan mutants (2%), a 1:1 protan-deutan ratio, and an overall frequency of 4% of red-green defects. The study illustrates the potential value of anomaloscopic characterization of colorblindness in attempts to evaluate human evolutionary processes.

Opposite effects of the synthetic adjuvant N-acetyl-muramyl-L-alanyl-D-isoglutamine on the immune response in mice depending on experimental conditions.

The influence of N-acetyl-muramyl-L-alanyl-D-isoglutamine (muramyl dipeptide, MDP) on the plaque-forming cell (PFC) response of mice to sheep red blood cells (SRBC) was studied. The effect of MDP depends on the relative doses of antigen and adjuvant and on the timing of adjuvant addition. For a given concentration of antigen, when SRBC and MDP were simultaneously injected in the i.v. route, MDP led to an enhancement of the response; in contrast, when antigen was injected i.p., a depression of the PFC response was observed. In vitro, the same duality of effect was obtained, for the same spleen cell concentration; addition of MDP led to a stimulation of the response when cultures were set in petri dishes and to an important decrease in the response when cultures were set up in plastic tubes. This inhibitory effect of MDP did not appear when it was added one night after the initiation of the cultures. These contrasting results confirm the adjuvant properties of MDP, but they show that under certain conditions, MDP can stimulate suppressive processes.

[Radioimmunoassay of low molecular weight human kininogen].

A radioimmunoassay for low molecular weight (LMW) human Kininogen has been carried out. The first step was to prepare LMW Kininogen from human plasma. The proposed method allowed to get chemically pure and biologically active LMW Kininogen. This preparation was used to induce antibody. Optimal conditions for labelling and incubation were determined. This method may be applied to the assay of Kininogen in human plasma.

Dislodgment of pacemaker electrode simulating focal motor seizure.

The unusual finding of dislocation of the electrode of a pacemaker in a patient caused repetitive twitching of the right leg. Initial confusion with focal motor seizure resulted in the administration of antiepileptic medication. Malposition of the electrode was confirmed by appropriate roentgenographic studies, and subsequent repositioning terminated the episode.

Primary retinal dysplasia transmitted as X-chromosome-linked recessive disorder.

The familial occurrence of retinal dysplasia in five affected male children suggested X-chromosome-linked recessive inheritance. The clinical features were childhood onset, severe visual impairment, head posture, nystagmus, and strabismus. The ophthalmoscopic findings varied in shape and extension; they ranged from retinal folds to dysplastic tissue covering the posterior pole or gliosis with tumor-like protrusion in the vitreous. The marked variability of the retinal findings was paralleled by the visual acuity, which ranged from some vision to blindness. Electroretinograms coordinated well with ophthalmoscopic observations. Of the five mothers, who are the presumed heterozygous carriers, two showed retinal changes.

Kinetics of the humoral and cellular immune response of guinea pigs after injection of the synthetic adjuvant N-acetylmuramyl L-alanyl-D-isoglutamine: comparison with Freund complete adjuvant.

We studied the time course of humoral and cellular immunity of guinea pigs injected with the synthetic adjuvant N-acetylmuramyl L-alanyl-D-isoglutamine (MDP Pasteur, 10 microgram) in Freund incomplete adjuvant; the kinetics were compared with those obtained with Freund complete adjuvant (50 microgram of whole Mycobacterium butyricum). The antibody response to ovalbumin was faster and higher with MDP, but dropped sooner to a low level; the secondary response was, however, again higher for MDP than for Freund complete adjuvant. Cellular immunity, as measured by delayed hypersensitivity, and migration inhibition factor production werepositive for both adjuvants. The same response was followed in animals injected with MDPA, the nonamidated analog of MDP; the same kinetics as for Freund incomplete adjuvant were obtained for the primary response, but the secondary response was stronger and gave a positive delayed hypersensitivity reaction.
