[Serological examinations on natural infections with mouse pathogens in inbred mouse strains: difference in antibody detection among the strains (author's transl)].

Serological surveys on several infections were performed on the inbred mouse strains maintained at the Central Institute for Experimental Animals. In the first survey, 11 strains of mouse, which were 8 weeks of age or older and were kept in separate cages in the same animal room, were tested for antibodies to Salmonella enteritidis, Corynebacterium kutscheri, Tyzzer's organisms, Mycoplasma pulmonis, mouse hepatitis virus (MHV), Sendai virus (HVJ), pneumonia virus of mice (PVM) and minute virus of mice (MVM). Positive results were obtained in MHV, HVJ, PVM and MVM. Positive rates for these viruses except for MVM were different among mouse strains. In the second survey, 5 strains of mouse kept together in the same cage for 4 weeks after weaning were examined for MHV and HVJ antibodies. Positive rates to MHV were different among mouse strains as observed in the first survey. For HVJ antibody, no difference was demonstrated in positive rates unlike in the first survey, but the titers varied between the strains. These results suggest the difference in antibody response to natural infections dependent on mouse strains.

The highly inbred STU mouse strain (up to 100 inbred generations) was developed from a German white mouse colony. Its properties are described with special emphasis on the occurrence of tumors, occurrence of C-type oncornaviruses and on its immunological reactivity. The predominant tumor is a fibrosarcoma (approximately 23% incidence).

Natural resistance to Salmonella typhimurium in different inbred mouse strains.

The mechanisms of natural resistance to intravenous challenge with Salmonella typhimurium C5 are complex. LD50 determinations showed inbred mouse strains of low, intermediate and high natural resistance, with BALB/c and B10 strains the most susceptible, A/J the most resistant. Delayed (footpad) hypersensitivity was not by itself a measure of natural resistance. Resistant mouse strains sensitized either s.c. or i.v. with an attenuated salmonella strain showed positive 48 h footpad reactions when tested 8 days later with a salmonella extract, but three very susceptible strains also showed positive reactions. Determinations of the in vivo net growth rate of salmonellae in the liver and spleen during the first phase of the infection (up to day 4) arrange the different mouse strains into two categories of fast and slow net growth rate. All fast net growth rate strains are susceptible, but not all slow net growth rate strains are resistant. Besides slow net growth rate, resistance requires the participation of other factors appearing in the second phase of the infection (towards the end of the first week) probably involving the cellular immune response, which halts further bacterial growth. Not all slow net growth rate strains are equally capable of suppressing bacterial growth in this second phase. The host mechanism determining slow net growth rate is inherited as a dominant trait, and appears to be operating before the main cellular immune response. The influence of this mechanism on net growth rate is reflected in the time to death following a given dose of salmonellae. The present results suggest that overall resistance to salmonellae is polygenic, but that the mechanism responsible for the differences in early net growth rate is less complex.

Genetic relation of life span to metabolic rate for inbred mouse strains and their hybrids.

Average life spans were estimated for the male progeny from 21 of the 25 possible matings of 5 inbred mouse strains. Oxygen consumption was measured in an open system over a 48-hour interval. Resting metabolism, Mre, and average metabolism, Mav, were determined at 6-8 months of age, and at 24-34 months. Body weight, W, was determined at the time metabolism was measured. Life span, L, is negatively correlated with Mre and Mav, and positively correlated with W at both ages of measurement. This is in accord with the metabolic wear factor that had previously been established among 85 different species of mammals. A new metabolism variable, the energy partition coefficient, defined as the ratio of average to resting metabolic rate, Mav/Mre, has a parabolic relation to body weight, i.e., is maximal at an intermediate body size. The squared body weight deviation in turn has a negative correlation with life span. The correlation of L with Mav/Mre is positive, as expected, but not significant. These data suggest the existence of a longevity factor dependent on the partition of energy between the phasic metabolism of activity and the continuous maintenance metabolism.

Effect of 3-methylcholanthrene treatment on phenacetin O-dealkylation in several inbred mouse strains.

An increase in the metabolism of phenacetin to N-acetyl-p-aminophenol is correlated with benzo[a]pyrene hydroxylase induction by 3-methylcholanthrene among several inbred mouse strains. While the magnitude of induction of phenacetin O-dealkylation is considerably less than that of benzo[a]pyrene hydroxylation, the data indicate that in mice, the metabolism of these two substrates is under similar regulatory control.

Nocardia infections in congenitally athymic (nude) mice and in other inbred mouse strains.

The mortality rate and histopathological features of Nocardia asteroides and Nocardia brasiliensis infections in congenitally athymic (nude) mice of ICR and C3H/eB origins were quite different from what we found for Swiss white mice and other inbred mouse strains (namely, C57/BL/6J, New Zealand Black, BALB/c, CBA/LAC, and C3H/eB). The immunocompetent littermates of the congenitally athymic mice occupied an intermediate position between their athymic siblings and Swiss white mice in terms of their responses to both these organisms. Macrophage ingestion and destruction of N. brasiliensis, as demonstrated by electron microscopy, was found to occur. The T-lymphocyte appears to be an essential component in normal mouse resistance to infection by both N. asteroides and N. brasiliensis.

Sensitivity to intravenous injections of histamine and serotonin in inbred mouse strains.

A large panel of inbred strains of mice and some of their Fl hybrids were tested on their sensitivity to the hemoconcentrating effects of intravenous injections of histamine alone, serotonin alone, or histamine-serotonin mixtures. Major differences in susceptibility were observed. Crosses and backcrosses were made between susceptible (SJL/J) and less susceptible (RF/J, C57BL/6J) strains; it was found that the sensitivity to vasoactive amines is, at least partially, inheritable.

Correlation between DNA repair of embryonic fibroblasts and different life span of 3 inbred mouse strains.

Primary mouse fibroblast cultures were established from 10 day old embryos of 3 inbred strains with a genetically determined different life expectancy. The capacity for unscheduled DNA synthesis following u.v. irradiation was studied in these cells at various passage levels. The mouse fibroblasts show considerable repair synthesis corresponding to the duration of exposure time. The capacity for induction of unscheduled DNA synthesis was different in the cells of each strain and correlated to the natural life span of the animal; in each case, however, the ability to perform repair synthesis was subjected to an age-associated decline.

The mouse fibroblasts show considerable repair synthesis corresponding to the duration of exposure time. The capacity for induction of unscheduled DNA synthesis was different in the cells of each strain and correlated to the natural life span of the animal. In each case, however, the ability to perform repair synthesis was subjected to an age-associated decline, although semiconservative DNA synthesis and proliferative potential of the cells was not changed until the cultures entered phase III passages.

Intrauterine transmission of Sendai virus in inbred mouse strains.

A study of Sendai virus infection in adult mice (2 to 3 months of age) showed that the inbred strains were more susceptible to infection than randomly bred Swiss white mice and that virus could be isolated from inbred strains for as long as 21 days postinfection. For this reason, these mouse strains (C57B1/6J [black] and C57Br [brown]) were selected for the study of intrauterine transmission of virus. The major effect of infection was a decreased weight of both embryos at 16 days of gestation and newborn mice. Virus was isolated from 17 to 20% of the embryos and at least 20 to 30% of the newborns from intravenously infected mothers. Fluorescent-antibody studies showed that the virus was widely distributed in the tissues of both embryos and newborns, including the central nervous system.

Developmental changes in serum luteinizing hormone, follicle stimulating hormone and androgen levels in males of two inbred mouse strains.

A comprehensive developmental study of serum LH, FSH and androgen concentrations was carried out in male mice of two inbred strains. Gonadotropic hormone (GTH) levels rose 10-15 days prior to the pubertal increase in serum androgen with FSH preceding LH in this regard. In both strains GTH titer rose to a peak at 30 or 35 days and then steadily declined to adult levels which were strain-specific. Serum androgen was detected at low, relatively steady levels until the pubertal increase between 30 and 50 days postpartum. The results are interpreted as supporting the hypothesis that a shift in feedback sensitivity, occurring at about 20 days of age, may be involved in the onset of puberty in the male mouse.

Effects of Aspiculuris tetraptera dn Syphacia obvelata on exploratory behavior of an inbred mouse strain.

Sixty C57BL/6 male mice were treated with an anthelmintic for 2 weeks to free them of pinworms. They were then divided into six groups. Two groups were infected with Aspiculuris tetraptera, two groups with Syphacia obvelata, and two groups were left uninfected as controls. At intervals of 2 and 4 weeks after infection, the mice were tested for exploratory activity in a barriered field apparatus. Following the second test, each mouse was necropsied to determine presence of nematodes and to estimate worm burdens. Statistical analysis showed significant depression of exploratory activity in mice harboring S obvelata. No significant depression was found in those harboring A tetraptera.

Butanediols: selection, open field activity, and NAD reduction by liver extracts in inbred mouse strains.

Mice from the high-ethanol preferring C57BL strain and low-ethanol preferring DBA strain were tested for their preference for butanediols. The C57BL strain showed a significantly higher preference for a 10% (v/v) solution of 1,3-butanediol than the DBA strain. The C57BL strain also showed a significantly greater consumption of 1,2- and 2,3-butanediol, but the separation between strains was much smaller than with 1,3-butanediol. Both strains uniformly avoided 1,4-butanediol. Tolerance for 1,3-butanediol was tested in an open-field monitor at 3 doses. At the lowest dose the DBA strain was hyperactive and the C57BL were unaffected. At the highest dose both strains were equally depressed. The specific activity of NAD reduction on incubation of liver extracts with 1,3-butanediol and ethanol as substrates was higher with both compounds in extracts from the C57BL strain.
