Inhibitory mechanisms influencing complex cell orientation selectivity and their modification at high resting discharge levels.

1. These experiments have investigated the contribution made by GABA-mediated inhibitory processes to the orientation tuning of complex cells in the cat's striate cortex. The GABA antagonist bicuculline has been ionophoretically applied to individual complex cells and the modifications produced in their orientation tuning documented. 2. In terms of the type of change produced in orientation tuning by the application of bicuculline, it seems that there are two categories of complex cells. 3. In one of these categories the orientation selectivity was eliminated during bicuculline application. The excitatory input to these cells would therefore appear to be non-orientation specific. Their orientation selectivity is presumably generated by a GABA-mediated inhibitory input. 4. In the other category of complex cells, although the orientation selectivity was decreased during bicuculline application, the cells retained a preference for a range of orientations that was generally centred around the original optimal orientation. It is suggested that for these cells the inhibitory input enhances the orientation tuning of an excitatory input that is already broadly orientation tuned. 5. Comparison of normal orientation tuning curves with those observed during the application of bicuculline provides a basis for estimating the orientation tuning of the GABA-mediated inhibitory input. In all cases, it is clear that at normal resting discharge levels, orientations either side of the optimal, and not those centred on the optimal, generate the most powerful inhibitory input. 6. These results would seem to be best explained by inhibitory interconnexions between cortical columns sensitive to different orientations. This type of lateral interaction between columns may serve to enhance the contrast in the orientation domain for the cortical representation of a specific stimulus orientation. 7. Increasing the resting discharge level of a complex cell, without blocking the action of GABA appeared to increase the gain of the inhibitory mechanisms acting on the cell. The normal excitatory responses to optimal or near optimal orientations were greatly reduced, or replaced by inhibitory responses, and non-optimal orientations produced only inhibitory responses. These inhibitory effects were blocked on the context of other observations in the literature. It is tentatively suggested that the interneurones providing the inhibitory drive to complex cells receive an input from recurrent collaterals of the recipient complex cells. Their other inputs would derive from neighbouring colums and from the afferent input to the parent column. The inputs from neighbouring columns would mediate the lateral inhibitory interactions in the orientation domain, and the recurrent collateral feed-back the decreased responsiveness at high resting discharge levels.

Hodgkin's disease with lymphocytic predominance, nodular type (nodular paragranuloma) and progressively transformed germinal centres--a cytohistological study.

The histology, cytology, and enzyme cytochemistry of a nodular variant of Hodgkin's disease with lymphocytic predominance, called 'nodular paragranuloma', are presented. The histological features of nodular paragranuloma are compared with those of progressively transformed germinal centres, which are enlarged follicles showing a predominance of small lymphocytes and some residual germinal centre cells. Progressively transformed germinal centres are sometimes found in nonspecific lymphadenitis (reactive hyperplasia). The histological similarity and the association between lymph nodes with nodular paragranuloma and lymph nodes with progressively transformed germinal centres in the same patient at different moments or at the same time, suggest that progressively transformed germinal centres are the origin of nodular paragranuloma. Hence, it must be concluded that nodular paragranuloma takes place in B-cell areas of the lymph node, unlike the other, or at least most of the other, types of Hodgkin's disease.

An economical, semi-automatic system for measuring cellular tree structures in three dimensions, with special emphasis on Golgi-impregnated neurons.

The hardware and software of a semi-automatic system are described for the measurement of cellular tree structures in thick microscopical sections such as the process of Golgi-impregnated neurons. With this system, the morphologist--viewing the neuron directly through the microscope--measures the tree structures using three cartesian coordinates of characteristic points, e.g. centre of cell, origin of processes, bifurcation points, end points, deflection points, etc. The system requires an on-line computer, but does not demand a dedicated computer since a joy-stick generator usually drives the microscopical stage during cell tracking. A digital computer is utilized for acquisition of data, and, when needed, to return the stage to a required position. The microscope stage is coupled to three stepping motors with 0.5 micron as the smallest step. The stepping motor for movement of the stage parallel to the optical axis (z-direction) is coupled directly to a spindle mechanism, replacing the original cog wheel mechanism. This construction shows no measurable backlash. The practical advantages of this new measuring system in relation to the existing digital systems currently in use, its accuracy and its costs, are discussed.

Inhibitory epilepsy.

Unlike post-epileptic paralysis, inhibitory epilepsy is rare and not well known. Differentiation between the two phenomena is usually difficult because both may have associated seizures. A satisfactory physiological explanation has yet to be advanced to account for the paraplysis. The present is a report of such a case together with the results of electroencephalographic, electromyographic and nerve conduction studies both during and after an attack of paralysis. The results suggest that inhibitory impulses originating from higher centres acting on the upper motor neurone itself leaving the anterior horn cell and possibly the upper motor neurone input to the gamma efferents switched off, are probably responsible for the paralysis.

Native edaphoclimatic regions shape soil communities of crop wild progenitors.

Unveiling the soil biological communities ecologically associated with crop wild progenitors (CWPs) in their habitats of origin is essential for advancing productive and sustainable agriculture. A field survey was conducted to investigate the edaphoclimatic conditions and soil bacterial, fungal, protist, and invertebrate communities of 125 populations of direct progenitors of major crops for world agriculture. The wild populations clustered into four ecoregions shaped by two edaphoclimatic dimensions: one summarizing variations in soil sand contents and nutrients concentrations, and the other featuring changes in aridity, soil pH, and carbon storage potential. We identified a common soil core community across CWPs that varied significantly along deserts to tropical seasonal forests and savannas. The assembly of the soil core community was driven by varying environmental preferences amongst soil biodiversity kingdoms, reflecting potential shifts in their functional profiles. The tropical ecoregion exhibited higher proportion of acidophilic bacteria, fungal, and protist parasites, whilst desert ecosystems harboured greater abundances of saprophytic fungi and heterotrophic protists. Moreover, CWPs displayed unique microhabitats that incorporate variability into the soil community assembly. Our work reveals the biogeography of soil communities associated with CWPs, the first step towards the development of microbial rewilding initiatives.

Selective N-actylation of gentamicin antibiotics-synthesis of 1-N-acyl derivatives.

Treatment of acid addition salts of aminoglycoside-aminocyclitol antibiotics of the gentamicin-sisomicin class, with one equivalent of triethylamine and an acylating agent results in selective formation of 1-N-acyl derivatives. This is in contrast to acylation of the antibiotic free bases which results in preferential acylation of other basic centres in the molecule. Origins of the observed selectivity are discussed. In vitro antibacterial activities of several new 1-N-acy1 derivatives of gentamicin, sisomicin and verdamicin are reported.

In an earlier paper, Caelli and Finlay demonstrated that the perception of apparent motion was contingent on critical phase relationships between inducing sources, and their spatio-temporal frequency ranges. In their original stimulus red-green centre-surround sources were used and phase-specific inhibition was found at 60 degrees--90 degreee phase angle differences between the complementarily coloured elements. In this experiment we show that, when centre-surround sources are replaced by vertically-horizontally displaced complementary (or identically coloured) sources, symmetrical phase inhibition effects occur with both motions.

High-voltage electron microscopy reveals new components in the axonemes and centroplast of the centrohelidian Raphidiophrys amibigua.

A high-voltage electron microscope was used to study the structure of the centroplast and axonemes in the centrohelidian Heliozoan Raphidiophrys ambigua. In Raphidiophrys the microtubules (Mts) of each axoneme are arranged so that they describe the apices of an array of hexagonally shaped prisms and all axonemes originate from a centroplast located near the centre of the cell. The centroplast was found to consist of 3 components: (1) a tripartite central disk, (2) a sphere of electron-opaque material, approximately 0.75 micron in diameter, which immediately surrounds the central disk, and (3) a slightly less electron-opaque centroplast shell, approximately 2-3 micron in diameter which consists primarily of hitherto undescribed components associated with the proximal 1-2 micron of each axonemal base. The new axonemal components described in this study include: (1) a 20-nm rod positioned in the centre of each hexagonal prism of Mts comprising the axonemal pattern, and (2) an electron-opaque linkage material which connects each Mt of the axonemal pattern to its 4 nearest neighbours. Both of these structures are associated with the base of each axoneme within the centroplast shell, both form crossbridges with axonemal Mts, and both terminate at the periphery of the centroplast shell once the complete axonemal Mt pattern is established. It is suggested that these structures are involved in forming and maintaining the Mt pattern seen in the axopodia of Raphidiophrys.

Detection of mitogen-activated T and non-T lymphocytes by virus plaque assay. Virus plaque assay on the cells fractionated by unit gravity sedimentation.

Virus plaque assay (VPA) was utilized for the quantitative evaluation of activated lymphocytes. We examined what types of cells, especially which of activated T and non-T lymphocytes, were detected as infective centres after infection with vesicular stomatitis virus. Marked increases in DNA synthesis and in virus-plaque forming cells (V-PFC) were observed not only during the activation of T lymphocytes with Con A, but also, though to a lesser extent, during the activation with lipopolysaccharide (LPS) of non-T lymphocyte preparations of nude spleen from which theta-positive lymphocytes and macrophages were completely depleted. The latter observation was further confirmed by the VPA on the populations enriched in LPS-activated non-T lymphocytes fractionated by the unit gravity sedimentation method. Fast sedimenting cells were found to be more active in DNA synthesis and contained more infective centres after infection than those sedimenting slowly and original unfractionated cells. Both the capacity for DNA synthesis and virus-replication were considered to be general properties accompanying lymphocyte activation.

Characterization of free and tightly bound lipoprotein in intima by thin layer isoelectric focusing.

Thin layer isoelectric focusing followed by second dimension quantitative immunoelectrophoresis has been used to characterize the free and tightly bound lipoprotein (lp) fractions in human aortic intima. In 3.3% acrylamide gels plasma low density lipoprotein (LDL) focuses rapidly, but very low density lipoprotein (VLDL) fails to enter the gell and remains at the origin (point of application). Samples of normal intima and lesions were placed directly on the gel and focused for 10--12 h; in normal intima and gelatinous lesions which had not accumulated cholesterol 60--70% of the free LP focused with plasma LDL, but with increasing accumulation of cholesterol the proportion focusing with LDL decreased and the component remaining at the origin increased. In 9 of the 39 samples examined one or more components with pI acid to LDL were present, and in one sample the whole peak focused at a more acid pI. In 4 of 5 lipid-rich centres of gelatinous plaques all the free LP remained at the origin, and the bound LP released by incubation with plasmin remained at the origin in all samples in 3.3% gels although it would focus in 2.6% gels. With antiserum to apo-C this large LP did not behave like VLDL, and it appears to be an aggregated LDL. An increased proportion of aggregated LDL was associated with (a) accumulation of cholesterol, (b) topographical location towards the centre of plaques, and (c) partial destruction of endogenous LP during incubation of fresh samples of tissue.

[Autologous osteochondral graft: a method for resurfacing osteonecrosis of the medial femoral condyle in the elderly (author's transl)].

The author describes an original technique for resurfacing osteonecrotic medial femoral condyles. The centre and posterior part of the condyle is displaced anteriorly into the weight-bearing area. The indications for this technique are cases in which there is no deformity in the frontal plane and when the tibial articular cartilage is normal. Two cases were so treated. After a follow up of three and five years the results were very good.

Comparison of radioactive peptides obtained from specifically labelled human renal and placental alkaline phosphatases.

Peptides with different chromatographic and electrophoretic properties were obtained from human placental and renal alkaline phosphatases by tryptic digestion of the enzymes labelled with radioactive orthophosphate at their active centres. These results provide structural evidence for the distinct genetic origins of the two isoenzymes that had previously been inferred from their different properties and from the observed phenotypic variation of placental phosphatase.

[Participation of gonads in pulmonary silicosis].

A survey is given on the participation of endocrine glands in the pulmonary silicosis. At the same time a new clinical and experimental contribution to the question of the appearance of a hypogonadotropic hypogonadism is given. The frequency of this hypogonadism increases with the severity of the pulmonary silicosis. The immunological opinions as well as the neuroergonal ones concerning the pathogenesis of pulmonary silicosis gave the possibility of a better explanation of the mechanisms, by means of which also other organs may be attacked in pulmonary silicosis. The neuroendocrine system gives a defensive answer to the aggressive silicotic pulmonary process, which in general slowly progresses, but which may also be certain hormones from the group of the glucocorticosteroids in the adrenal cortex. The intervention into the axis hypothalamus-hypophysis-adrenal cortex has been provided in all defence processes of the organism against every aggression. By the functional or organic damage of the neuroendocrine system, by the immediate toxic effect of silicium dioxide or the monosilicium acid as well as by the effect of the secondary hypoxia in pulmonary silicosis an insufficiency in the stimulation and excretion of certain hormones develops. The origin of this insufficiency is found in the hypothalamo-hypophyseal centres, and the effects extend to all endocrine glands which depend on the system hypothalamus-hypophysis. This mechanism is made responsible for the appearance of a hypogonadotropic insufficiency of gonads in the course of a pulmonary silicosis and is proved clinically and experimentally in the cases investigated by the authors

Critical bandwidth in loudness summation in sensorineural hearing loss.

Critical band estimation was performed in thirty-three patients with sensorineural hearing loss by the method of loudness summation, using noise bands centred around 1 kHZ. In 17 patients the hearing loss was of hereditary origin, in 16 patients of various, mostly unknown, origin. The normal loudness difference between broad band noise and narrow band noise decreased with increasing hearing loss, most pronounced in recruiting ears. Judged individually, 1/3 of the patients appeared to have a widened critical band, but several biases appeared to be responsible for these results. The pooled data indicated a normal critical band, both in patients with hearing losses less than 50 dB HL and in patients with hearing losses greater than or equal to 50 dB HL. Based on this finding, the validity of the critical band as a measure of the frequency selectivity of the ear is discussed and a theory is proposed for the anatomical and functional correlate of the critical band in loudness summation.

Next-generation sequencing in head and neck sarcoma: a single-centre institutional experience and review of the literature.

Head and neck sarcomas (HNS) are rare, heterogeneous malignancies representing less than 1% of head and neck cancers. Their complex anatomy and overlapping morphologies pose significant challenges for traditional diagnosis. We aimed to evaluate the clinical utility of next-generation sequencing (NGS) within a tertiary referral centre and synthesise these findings with current global molecular standards. We conducted a retrospective review of an original, previously unpublished, cohort of 12 patients with histologically verified HNS treated at University College London Hospital (UCLH) between 2023 and 2024. Molecular profiling included targeted DNA (RMH200) and RNA-fusion panels. This was supplemented by a qualitative synthesis of 16 key studies (2010-2026) identified through a systematic search strategy. In the institutional cohort, NGS provided definitive diagnostic or therapeutic clarification in 66% of cases (8/12). Key findings included the identification of pathognomonic fusions (such as EWSR1::FLI1, PAX3::MAML3), a novel MAMLD1::VGLL3 fusion, and actionable variants such as BRAF V600E and MYOD1. Furthermore, the formal exclusion of Neurotrophic tropomyosin receptor kinase (NTRK) fusions in some cases allowed for therapeutic streamlining. Literature synthesis aligned these results and emphasised the need for NGS for more accurate diagnosis and adequate treatment. NGS is a clinical necessity in the management of ultra-rare HNS. By transitioning from traditional morphology to high-resolution molecular interrogation, clinicians can resolve diagnostic ambiguity and identify targeted therapeutic pathways. Integration with emerging 2026 standards, including epigenetic classification and liquid biopsy monitoring, represents the future of precision surgery in head and neck sarcoma.

[Light and electron microscopic studies to the development of muscle cysts of Sarcocystis suicanis in experimentally infected pigs (author's transl)].

Six young pigs were experimentally infected with 100000 sporocysts of Sarcocystis suicanis and killed on the 32nd, 47th, 55th, 64th, 81th and 90th day p.i. The ultrastructure of sarcocysts in the muscle cells was studied. Light microscopical preliminary examination on the 32nd day p.i. showed cysts which lay in muscle fibres and were surrounded by a fine membran. On the 55th day p.i. the cyst wall showed radial light and dark stripes of 2.6 micrometer thickness, which reached a thickness of 3.2 micrometer on the 90th day p.i. The electron microscopic study of the cyst wall on the 32nd day p.i. showed a unit membran the numerous small outward projections of which are underlined by osmiophilic material that is absent in the inwardly directed invaginations. During the growth of the cyst, this primary cyst wall became regularly folded, forming many protrusions on the 55th day p.i. These appeared palisade-like and contained 57--62 filamentous elements in a single layer under the primary wall at a distance of 10 nm. In the centre of the protrusions there were 5--8 groups, comprising 2--5 filaments each. They have their origin in the ground-substance. As these filaments maintain their arrangement also in old cysts, they can be regarded as characteristic of Sarcocystis suicanis. The groundsubstance is homogenous; on the 32nd day p.i. it contained only metrocytes, and developed septa towards the interior of the cyst on the 47th day p.i. The parasites were reproduced by endodyogeny and appeared very similar to the structure of all cyst forming coccidians. Some old cysts degenerated fattily and the host cells were destroyed. The cysts of Sarcocystis suicanis with complete development of the protrusions of the primary wall can thus be differentiated from cysts of Sarcocystis suihominis with certainty.

[Measurement of polymerization heat of the tissue adhesive Fimomed (alpha-n-butylcyanoacrylate) reduced in the GDR].

The authors describe a method for measurement of temperature that originates from the polymerisation of the adhesive Fimomed (alpha-n-butylcyanoacrylate). The intravascular application of the adhesive is taken as a basis of the measurement. This kind of application is applied by the authors for the closure of arterio venous fistulae. The measurement of the temperature with this method did not show both in the centre of polymerisation and in the surroundings any doubtful value if not more than 3 ml adhesive was applied. However the measurement of the temperature which originates from the polymerisation is recommended in all cases before any plane sticking is applied.

Aspects of the patient intake process in a psychotherapy clinic.

Recent interest in psychotherapy services in the National Health Service has raised questions regarding the availability of existing psychotherapy facilities. In attempting to answer some of these questions, a study was made of aspects of the referral and assessment segments of the intake process of 180 patients referred in 1975 to the Paddington Centre for Psychotherapy Adult Out-patient Department. Of this group, 137 were referred for individual psychoanalytically oriented psychotherapy, 82 of whom were offered assessment interviews, 45 offered treatment and 40 actually began treatment during the study period. An additional seven patients originally assessed for group therapy were offered and began individual treatment. Some characteristics of the referral population, the referring agencies, the effects of a waiting list, and the phenomenon of dropouts are discussed and compared with findings from other Centres.
