On the metabolic detoxication of thymol in rabbit and man.

The metabolic detoxication of thymol was investigated in rabbit and man. Thymol glucuronide which the aglycone is intact, was isolated from thymol medicated rabbit urine and identified as a acetyl derivative of methyl glucuronate. The hydroxylated product of thymol, thymohydroquinone, was recognized in a small amount in thymol medicated human urine. It was presumed that thymohydroquinone is excreted as ethereal sulfuric acid conjugate in man.

The thymol turbidity test and determination of serum cholesterol : two tests to abandon in the routine evaluation of liver function.

The thymol turbidity test (Macglan's test) and the determination of serum cholesterol are still performed routinely as liver function tests in many laboratories. In this report, we have employed quantitative methods in order to evaluate the real usefulness of these two parameters. In a first step, the value of the thymol test and of cholesterol determination for the discrimination of the 13 most frequent hepato-biliary diseases was studied by analysis of variance, and compared to that of the following tests : serum bilirubin, serum glutamic pyruvic transaminase, serum alkaline phosphatase serum protein electrophoresis and prothrombin time. It was found that of all these parameters, the thymol test and cholesterol measurement had the lowest discriminatory powers. In a second step, the consequences of the suppression of the two tests were examined by linear discrimination analysis, which was done for all the possible pairs of diseases. It appeared that in each case, the loss of information due to the elimination of the thymol test or of cholesterol determination was nil or negligible. We conclude therefore that the thymol turbidity test and determination of serum cholesterol should be abandoned as routine tests of liver function.

Cholesterol

On the overshoot of calcium accumulation in fragmented sarcoplasmic reticulum induced by thymol.

The mechanism was studied of the overshoot of calcium accumulation in fragmented sarcoplasmic reticulum (FSR) which is observed when the active transport of calcium into FSR is performed in the presence of thymol; the amount of calcium in FSR increases steeply during the first minute of the reaction and then decreases markedly. In contrast to this behavior, the amount of calcium in FSR increases monotonically and then reaches saturation in the absence of thymol. It is shown that the amount of calcium accumulated in FSR is determined by the balance between the rapid influx and efflux of calcium, and that both processes are depressed by thymol. The overshoot of calcium accumulation can be explained as follows: thymol so markedly depresses the efflux of calcium uncoupled with Ca2+-ATPase activity that the amount of calcium is increased in FSR in spite of partial deactivation of Ca2+-ATPase. However, the state of the FSR membrane is rapidly changed when the concentration of accumulated calcium exceeds a certain critical value. Concomitant with this change, the calcium permeability of the membrane is increased, leading to a decrease in the amount of accumulated calcium. The effects of magnesium and temperature on the overshoot of calcium accumulation can be accounted for by this proposed mechanism.

Role of IgG fractions with high isoelectric points in the thymol turbidity test in syphilis. Evidence for an increase in basic IgG in early syphilis.

It is demonstrated that IgG fractions with isoelectric points above 7.6, isolated from pooled syphilitic sera, are able to elevate the thymol turbidity. The effect increases with increasing isoelectric points of the isolated fraction. IgG from individual syphilitic patients exerts a stronger effect on the thymol turbidity than normal IgG. It is concluded that elevated amounts of these basic immunoglobulins are present in the sera of syphilitic subjects.

445 workers occupationally exposed to subtoxic diphenylformamide concentration were examined. No significant differences were found in results of thymol test, concentration of total protein, bilirubin, and activity of basic phosphatase in blood serum, compared with the control group. A higher activity of asparagine--and--alanine aminotransferas and gamma--glutamylotranspeptidase was found; it was, however, statistically insignificant.

Adult

Serum alkaline phosphatase measured by using thymol blue monophosphate and a bichromatic analyzer.

Thymol blue monophosphate is evaluated as a substrate for use in kinetic bichromatic analysis of serum alkaline phosphatase activity. The effect of abnormally high concentrations of hemoglobin, bilirubin, or chylomicrons is negligible. The expected range for ambulatory adults established by assaying sera from 206 blood donors, was 12-44 U/lliter at 37 degrees C. Comparison of the method with a commonly used p-nitrophenyl phosphate method yielded a correlation coefficient, r, of 0.9949. Advantages of the method are the reduced cost per test and the exceptionally stable substrate.
