[Evoked visual potentials in tapeto-retinal degeneration].

A study of the behaviour of V.E.P. latencies and amplitudes in 247 eyes suffering from different clinical forms of chorio-retinal degenerations is presented. In 147 eyes the E.R.G. and the V.E.R. were simultaneously recorded, and this group was used for studying the relation ship between E.R.G., V.E.P. and visual acuity. The authors are of the opinion that the V.E.P., in that it investigates particularly the macular region, provides a clinical test of real interest in the prognosis of visual acuity in a known clinical context.

Circulating miRNAs and inflammatory markers - Associations between miRNAs and cytokine levels point to miRNA-mediated sCD40L release from platelets.

MicroRNAs (miRNAs) are gaining increasing attention, particularly because of their involvement in immune-related signaling pathways. We investigated the association between 179 plasma-circulating miRNAs (Plasma Focus microRNA PCR Panel) and 47 cytokines ("MILLIPLEX® panel) in 692 participants of the population-based SHIP-TREND cohort (age range 21-79) and two additional cohorts to present a comprehensive map of miRNA-cytokine relations. Multivariate linear regression models identified Bonferroni-corrected significant associations between miRNAs and cytokines for EGF (pro-epidermal growth factor), PDGF-AA, PDGF-AB/BB (platelet-derived growth factor subunit A and B), VEGF-A (vascular endothelia growth factor A), and sCD40L (soluble CD40 ligand) with sCD40L showing the most robust pattern. These models were adjusted for age, sex, platelet count, BMI, smoking, and technical parameters. In the follow-up sample (N = 191, 7 years after initial sampling), we confirmed that the observed associations were stable over time and replicated our findings in an independent clinical cohort (N = 74). Furthermore, the causal mediation results provide evidence for the involvement of platelet activity in the regulation of sCD40L mediated by five miRNAs in the range of 25 %-69 % of the effect being mediated (strongest mediation for hsa-miR-223-3p). Our study highlights a strong and stable miRNA-mediated modulation of sCD40L, at the stage of platelet activation with potential subsequent effects on the interaction of immune cells and haemostasis pointing to a complex regulatory mechanism. Future research is needed to determine the clinical relevance of our observations in the context of vascular thrombosis, immunological disorders, and neurodegeneration.

[Composition of the trace contaminants in the atmosphere of the Soiuz-22 space ship].

The Soyuz-22 space cabin atmosphere was studied for volatile organic trace contaminants. By gas chromatography the following constituents were identified: methane, ethane, heptane, methanol, ethanol, n-propanol, acetaldehyde, acetone, ethyl benzene. Except for acetone, concentrations of the above compounds were close to the values determined in the mock-up experiments.

[Index of fibrinolysis with new fibrin plate (author's transl)].

Plasminogen-free fibrin plate (fP) which was made from treated commerical bovine fibrinogen with Lysine-Sepharose was developed in our laboratory. This new fibrin plate showed the following specificities. a) This new fibrin plate did not show any lysis with high amount of streptokinase and Urokinase (10,000 u/ml and 500 u/ml). b) The concentrations of its substrate was the same as standard plate (SP) and its substrate was not denatured compared with heated plate (HP). c) The activity of plasmin can be measured quantitatively on fP and linear correlation between plasmin units and lysis area was showen. d) This procedure of new fibrin plate was easy and simple and could be applicable to the materials of other species, i.e., human, rabbit and porcine. With the use of two kinds of bovine fibrin plate (SP and fP), activation of fibrinolysis of human plasma, euglobulin and plasminogen induced by SK and UK was investigated and each correlation ship between sample and activator was studied statistically. From these results, "Index of fibrinolysis" meaning of fibrinolytic components such as plasmin, plasminogen, activator, proactivator, anti-activator and anti-plasmin were indicated. Indeed, these index of fibrinolysis were calculated from the lysis area of plasma+SK, Eug.+SK and Eug.+UK by each formula and index obtained from some physiological and pathological condition showed us many new information about fibrinolysis.

SHIP-1 Differentially Regulates IgE-Induced IL-10 and Antiviral Responses in Human Monocytes.

IgE-mediated stimulation of monocytes regulates multiple cellular functions including cellular maturation, cytokine release, antiviral responses, and T-cell differentiation. Expression of the high-affinity IgE receptor, FcεRI, is closely linked to serum IgE levels and atopic disease. The signaling molecules regulating FcεRI effector functions have been well studied in mast cells and basophils; however, less is known about the signaling and regulatory mechanisms in monocytes. This study sought to identify regulators of IgE-mediated cytokine release in human monocytes. SHIP-1 was identified as a negative regulator of IgE-induced IL-10 production. It was also determined that IgE-mediated stimulation and SHIP-1 inhibition decreased antiviral IP-10 production after liposomal poly(I:C) stimulation, indicating differential regulation by SHIP-1 in IgE-driven and antiviral response pathways. SHIP-1 and NF-κB were activated following IgE-mediated stimulation of monocytes, and NF-κB activation was related to both SHIP-1 and FcεRIα cellular expression levels. To our knowledge, this is the first study to identify a role for SHIP-1 in regulating IgE-mediated and antiviral responses in human monocytes. Given the importance of monocytes in inflammation and immune responses, a better understanding of the signaling and regulatory mechanisms downstream of the FcεRI receptor could lead to new therapeutic targets in allergic disease.

Investigation of factors of probable significance in the pathogenesis of pneumonic pasteurellosis in cattle.

Six groups of ten beef calves six to eight months of age were shipped from western Canada and observed untreated for one week after arrival. The following parameters were measured daily: body temperature, plasma fibrinogen, nasal bacterial mean colony counts of Pasteurella hemolytica and Pasteurella multocida, total and differential leukoyte counts, packed cell volumes and the following, twice during the week: serum and nasal antibody titres to P. hemolytica and parainfluenza-3 virus. The lungs from 44 of the calves were obtained at post mortem and given a numerical score based on the degree of pneumonia present. Animals were designated SICK and WELL according to body temperature and plasma fibrinogen. The SICK animals had higher nasal mean colony counts of P. hemolytica than the WELL animals. The SICK animals had lower levels of serum antibody to P. hemolytica than the WELL on day 1 but had a greater rise in titre over the week than did the WELL animals. Both groups were similar with regard to serum antibody to parainfluenza-3 virus and there was little change in these titres. The SICK animals had a much greater degree of pneumonia than the WELL. The values of some of the parameters were combined with the data of previously studied animals in order to provide a comparison of SICK and WELL with larger numbers of animals.
