[Animal experimental investigations for determining the duration of the "mechanical-effective period" of muscle action potentials and of the "electro-mechanical latency period" of "fast fibres" of external ocular muscles (author's transl)].

A report is given of an animal experimental study for determination of the duration of the "mechanical effective period" of the muscle fiber membrane action potential and the "electromechanical latency period" of "fast fibers" of the external ocular muscles in rabbits in vivo. The studies were carried out with an internationally standardized glass microelectrode technique. The statistical values are based on 250 tests employing the superposition technique. The mean "mechanical effective period" of the muscular fiber membrane action potential was determined with SE=0.52 ms, SD 0.08 ms. The confidence thresholds were 99% statistically reliable with 0.43 ms and 0.55 ms. According to these studies, the duration of the "mechanical effective period" of the "fast fibers" of the external ocular muscles was only one-third off the "mechanical effective period" of the skeletal muscles. The mean duration of the "electromechanical fast-fiber latency period" was calculated with SE=1.97 ms, SD=0.30 ms. The confidence threshold was 99% statistically reliable with 1.86 ms and 2.08 ms. The "electromechanical latency period" of the "fast fibers" of the external ocular muscles was therefore significantly less than the skeletal muscles, lying under the 50% limit of the "electromechanical latency period" of the analogous skeletal musculature. These studies permit determination of the most important electrophysiological parameters which define a threshold value of maximal speed in vision direction changes.

Action Potentials

Induction of suppressor cells by sodium periodate: the opposite expression of periodate-induced lymphocyte activation in spleen cells of normal and nude mice.

Normal mouse spleen cells treated with sodium periodate for 10 min at 4 degrees are stimulated to undergo blastogenesis and to incorporate thymidine. In contrast, periodate treatment does not stimulate DNA synthesis in nude mice spleen cells. The effect of such treatment on the antibody response in vitro induced by sheep red blood cells (SRBC) and by trinitrophenyl polyacrylamide (TNP-PAA), a T-independent antigen has been evaluated. Periodic-induced proliferation is accompanied by a marked inhibition of the immune response to both of these antigens in normal mouse spleen cells. Mild oxidation of nude cells with periodate markedly enhances their anti-TNP response elicited by TNP-PAA. The data suggest that periodate-induced immune suppression is associated with T lymphocytes. Furthermore, periodate-treated cells are capable of suppressing the in vitro antibody response of untreated normal cells.

[Electroencephalographic periodicity with asynchronous variations in period duration in both hemispheres following acute cerebral anoxia].

The periodicity of paroxysmal wave complexes in EEG after acute cerebral anoxia with special reference to the dynamics of period duration (alteration of Zeitmass) is described in case 20 hours before isoelectric tracing. The completely asynchronous feature of paroxysmal events within certain limits of period duration in both hemispheres is a rare condition. Late and pure cortical electrogenesis is suggested. Short and long EEG- periods in similar and different pathological processes and functional states are compared with respect to topography of lesions. Subcortical interactions and extraneuronal influences upon periodicty are discussed. The parameters "complex wave and periodicity" in EEG and the dynamics respectively are a matter of serial investigations for better understanding pathophysiology and prognostic value.

Comparison of autoregression and fast Fourier transform techniques for power spectral analysis of heart period variability of persons with sudden cardiac arrest before and after therapy to increase heart period variability.

Clinical studies have used two types of analysis of the power spectral estimates of heart period variability: autoregressive and fast Fourier transform techniques. Controversy exists regarding which method is the most valid. The specific aims of this study are: (1) to describe the power spectra of heart period variability before and after an intervention designed to increase heart period variability in persons after sudden cardiac arrest; (2) to compare the integral of power spectral density between autoregressive and fast Fourier transform techniques within low and high-frequency bands; (3) to compare the magnitude of the spectral peak values determined by autoregressive and fast Fourier transform techniques approaches within low and high-frequency bands; and (4) to compare the aforementioned parameters using 4-minute, 1-hour, and 24-hour blocks of heart period data. Results indicated high correlations between spectral estimations by autoregressive and fast Fourier transform techniques using integrals or peak values within either the low or high-frequency ranges. The autoregressive technique demonstrated better resolution of sharp peaks than the fast Fourier transform technique, and makes a smoother, more interpretable curve. Lastly, people can cognitively change their heart rate variability.

[Substitution of protein animal feed through lysine-supplemented high-protein wheat during the breeding and laying periods of hens. 5. Influence of graded lysine supplements fed during the laying period on the production characteristics of hens].

A large-scale trial was performed with 1500 laying hens which were kept under conditions of industrial egg production. The trial was carried out to investigate whether supplements of L-lysine would increase the food value of high-protein wheat and in which way a proportional decrease of dietary protein by 50% would affect the egg production of the hens. Additionally, studies were made to investigate whether the egg production of the hens was influenced by changes in the plane of feeding of the young hens during their rearing period. Both the young hens and the laying hens received isonitrogenous and isocaloric rations containing 4 graded levels of lysine. All of the 16 groups of the hens received a different diet. This was achieved by suitable combinations within the feeding regime. The control animals received, at all periods of age, food prepared on the basis of government-controlled standard rations. No significant differences in the level of food consumption were observed between the experimental groups whereas the control birds consumed more food (124 g per bird/day) than the experimental hens (120 g per bird/day; alpha less than 0.05). The levels of production were in all groups associated with the lysine content of the ration. The lysine-deficient birds produced the smallest eggs, layed the smallest number of eggs and consumed the largest quantity of food per 100 g of egg weight. The egg production of the hens in groups 11 and 12 (receiving a fish meal ration during the rearing period and a fish meal or wheat ration + 0.15% L-lysine during the laying period) was 70% of that of the birds in the control groups. The results of the present trial suggest that lysine-supplemented high-protein wheat rations for laying hens could be used but care should be taken that the plane of feeding for the young hens will not be too much restricted.

Quantitative determination of immunoglobulins, lysozyme, and certain electrolytes in breast milk during the entire period of lactation, during a 24-hour period, and in milk from the individual mammary gland.

During a period commencing at birth and lasting for up to 27 months 193 milk samples have been collected from 29 mothers. The IgA globulin content was high immediately after birth, averaging 2.7 arb.U, decreasing to 0.3 arb.U within the first 2 to 3 weeks after birth, then remaining almost constant for the rest of the lactational period. In the case of IgG globulin, similar results were obtained, but the quantity was much smaller. IgM globulin was demonstrated in small quantities during the first 3 weeks of lactation. The lysozyme content varied considerably during the whole lactational period. Individual variations were found for all the immunoglobulins, while the concentration in the individual woman varied only slightly from day to day following in other respects the pattern described above. In 19 mothers IgA, IgG, IgM, lysozyme and electrolyte content were determined in serum and in milk from the right and the left breast on the same day. No difference in content was found between milk from the left and the right mammary gland. A positive correlation was found between the concentrations of IgA and sodium chloride in milk, between those of IgG in milk and serum, and between those of lysozyme in milk and serum. No variations were registered during the individual breast feeding, nor for the 24-hour period as a whole.

Spatial periodicities of periodic complex cells in the visual cortex cluster at one-half octave intervals.

Within individual penetrations in the visual cortex, spatial periodicities of periodic complex cells differ by either one-half or one octave. When data are pooled from neurons subserving the central visual area in many cats, the results indicate that spatial periodicities cluster at one-half octave intervals over a 2 1/2-octave range (0.95 to 5.4 cyc/deg). Thus a relatively small number of such channels spaced at regular intervals along a logarithmic scale within each orientation column may suffice for this stage of spatial processing.

Effects on radioimmunoassay of digoxin of varying incubation periods for antigen---antibody reaction and varying periods of absorption by dextran-coated charcoal.

In radioimmunoassay of digoxin by the dextran-coated charcoal method, duration of the incubation period for the antigen-antibody reaction and time allowed for dextran-coated charcoal to absorb the free antigen are critical factors. We compared effects of prolonged periods of absorption on results obtained with five commercial kits. In all cases, the digoxin concentrations increased linearly with absorption time, but the increase varied among kits. Kallestad and New England Nuclear 3h-kits and the Squibb 125I-kit showed less increase (i.e., gave relatively consistent results after various periods of absorption); the Schwarz/Mann 125i-kit and Clinical Assays 3h-kit showed greater increases. Precautionary steps are outlined for radioimmunoassays by this technique.

Motor reactions during the antenatal period correlated with the periodic change in the activity of the cardiovascular system.

A connection between the periodic appearance of vagal bradycardia and of intrauterine large-amplitude respiratory movements, as well as the appearance of generalized jerky-type and more sustained extensor motor reactions, has been established in experiments on dog fetuses connected with the mother through the umbilical cord. Brief vagal bradycardia also arises immediately after completion of these reactions and is an analogue of the vagal phase in adults after motor activity. These correlated phenomena are caused by periodic reversible mild hypoxemia which through the chemoreceptors of the sinocarotid zone stimulates the centers of vagal innervation of the heart and the centers of innervation of skeletal muscles. These phenomena disappear when the mother inhales a gas mixture containing less than 14-15% oxygen or greater than 50-60% oxygen, as well as when the mother is under deep anesthesia.

[Substitution of protein feed through lysine-supplemented high-protein wheat during the breeding and laying period of hens. 1. Problems, analytical methods used and results obtained in the chick period].

Different parameters were investigated in a trial carried out for the 8-week chick period with an experimental group receiving a protein feed in which 7% of the feed, relative to that of the control group, was replaced by protein-rich wheat. Values for consumption were higher in the controls than in the experimental chicks because of the lower energy content of the ration. Statistically well-established data were obtained indicating that the amounts of crude protein, lysine and sulfur-containing amino acids consumed per unit of weight gain were lower in the experimental group than in the control group. Total body analyses showed that the bodies of the control birds contained more crude protein (alpha less than 0.05) and less fat (alpha less than 0.001). The rate of mortality during the 8-week chick period was 1.1% in the control group and 0.5% in the experimental group.

[The substitution of lysine supplemented high protein wheat for protein feeds during the rearing and laying periods of hens. 8. Nitrogen excretion and true amino acid digestibility as an effect of various lysine levels and successive laying periods].

Digestibility trials were performed with laying hens receiving 3 types of rations containing equal proportions of the same dietary components. The percentage of high-protein wheat in these rations was 74%. Ration I was supplemented with 0.12% lysine while 0.06% lysine was added to ration II; ration III remained unsupplemented. The crude protein content of rations 1 to 3 was 16.1%, 16.5% and 16.6%, respectively. The corresponding lysine percentages were 0.66, 0.62 and 0.56. Studies were made to investigate the influence of different lysine levels on the rate of N excretion and various amino acid data. These studies were carried out during two successive laying periods. Only random differences between the data for urinary N excretion and N balances per kg liveweight were obtained in the two laying periods and for the different lysine levels. The true lysine digestibility was significantly lower in the poor-lysine ration as compared with the ration with 0.66% lysine. Moreover, the absorptive capacity for thio-amino acids is influenced by the lysine level of the ration. The proportion of lysine contained in faecal crude protein increased with the decreasing lysine content of the ration. Similarly, the total number of amino acids present in faecal crude protein increased with the declining supply of lysine. These findings suggest that relatively more NPN compounds are excreted in the faeces if adequate amounts of amino acids are supplied rather than when one amino acid is deficient.

[The pre-ejection-period of the humen fetal heart: significance of base-line alterations in perinatal period].

The Pre-ejection period of the cardiac cycle, fetal heart rate and uterine contractions were monitored in a series of 115 unselected fetus sub partu. A new one-line technique permits a continuous registration of PEP patterns simultaneously with the CTG. Any change in base-line of the PEP war related to heart rate, acid-base-status and gestational age. The average value of PEP in fetus was 72,93 +/- 7,9 msec. The base-line of PEP increased paralelly with gestational age of fetus and decreased rapidly beyond normal values after the 41. week of gestation. Within normal values alterations in base-line of the PEP were negativly correlated to alterations in base-line of the heart rate. Abnormal high and low base-lines of the PEP were observed in fetal acidosis, hypoxemia, and in uteroplacental insufficiency. The prolonging or shortening of the PEP base-line seems to signalize an acute injury of the utero-feto-placental unit in perinatal period.

[Active period of genes during Drosophila ontogenesis. I. Temperature-sensitive period for the spineless effect in aristapedia mutants].

The temperature-sensitive period (TSP) was determined for the spineless effect (bristle shortening) in SSa40a mutant of Drosophila melanogaster. The length of front orbital and front vertical bristles was measured at different intervals after the temperature shift from 29 to 17 degrees and vice versa. The TSP was found to last from 7 to 14 hrs during pupal development. This corresponds to the activity of the spineless-aristapedia locus during the period of isolation of bristle initial cells. The discordance between the TSP of spineless effect and that of homoeotic effect of the same gene suggests that the locus SSa40a is activated twice during development.

Alleles

Responses of visual cortical cells to periodic and non-periodic stimuli.

1. The activity of neurones of the visual cortex (area 17) has been recorded in anaesthetized cats in response to gratings of different profile and to single light and dark bars. 2. At very low spatial frequencies, outside the frequency response range to sinusoidal gratings, the response to square-wave drifting gratings is obtainable from a combination of the response to the single bars of the grating presented in isolation. At higher spatial frequencies this is no longer true. 3. At very low spatial frequencies the responses to square-wave gratings and to missing-fundamental gratings (obtained by subtraction from the square-wave grating of its fundamental gratings (obtained by subtraction from the square-wave grating of its fundamental harmonic) are very similar. 4. At spatial frequencies near the peak of the spatial frequency tuning curve of the cell, the responses to square-wave grating and to sinusoidal gratings are very similar. At these spatial frequencies the response to the missing-fundamental grating is practically zero. 5. At spatial frequencies lower than that of best sensitivity for the cell, the response to square-wave gratings is correlated with the 1st and 3rd harmonic of the stimulus. 6. We conclude that at very low spatial frequencies of the grating the response of cortical cells is correlated with the light or dark edges (or light or dark bars) of the stimulus, because the edges contain high frequencies within the range of sensitivity of the cells. At higher spatial frequencies the results are interpreted best by assuming that cortical cells respond to the harmonics of the visual periodic stimulus. 7. When a background of dynamic visual noise is added to increase the spontaneous discharge of simple cells, their response to visual stimuli becomes linear or quasi-linear. The stimuli could be either single bars or gratings.

Action Potentials

Periodic remission in Cushing's disease with paradoxical dexamethasone response: an expression of periodic hormonogenesis.

A patient with Cushing's disease due to a chromophobe adenoma was studied for 243 days before pituitary surgery and evidence for periodicity in cortisol steroid production was found with cycles occurring every 85.8 days (peak-to-peak length), associated with laboratory remissions and paradoxical response to dexamethasone. The autonomy of ACTH secretion was suggested by the nonresponsiveness to repeated lysine-vasopressin stimulation tests and lack of increase in urinary 170HCS following metyrapone. A distinct response of the hyperplastic glands (as demonstrated by percutaneous adrenal venography) was obtained on several B1-24 corticotropin stimulation. The patient's hypercortisolism disappeared following removal of the chromophobe adenoma through transphenoidal hypophysectomy.

Periodate oxidation analysis of carbohydrates. XIII. Simultaneous gas chromatographic determination of the aldehydes in the periodate oxidation products of non-dialyzable urinary carbohydrate materials as diethyl dithioacetals.

The aldehydes in the periodate oxidation products of non-dialyzable urinary carbohydrate materials were determined simultaneously by gas chromatography of their diethyl dithioacetal derivatives. The yields of aldehydes for normal male urine varied considerably among subjects, but their molar ratios were almost constant. The average values for glyceraldehyde/glyoxal and lactaldehyde/glyoxal molar ratios were 0.41 and 0.27, respectively. The average amount of L-fucose at the non-reducing terminals of carbohydrate chains, as estimated from the yield of lactaldehyde, was about 5 mg/day.

1530 laying hybrid hens kept under conditions of large-scale production were used to test whether varying lysine levels given to young hens would later on influence the production of the birds during their laying period. Thus, young hens received isonitrogenous and isocaloric diets with varying lysine content (group 1-4: 0.59%, 0.61%, 0.54%, 0.46%) and varying sources of lysine (group 1: wheat/fish meal; group 2--4: wheat/synthetic L-lysine). The ration used for the controls was prepared on the basis of nationally prescribed formulas.

[Hyperglycemia and the glucose tolerance test in the acute period of myocardial infarct and in the remote post-infarct period].

The blood sugar content was determined in 142 patients with myocardial infarction on the 1--3rd, 7-10th, 28--30th days of the disease by the Hagedorn-Jensen method. The standard glucose tolerance test was conducted in 64 patients on the 30th day of the disease and repeated in 20 patients 18 months after infarction. It was concluded that transient hyperglycemia developed in 47.8% of patients with myocardial infarction in the acute period, predominantly in those with a sugar curve of the diabetes-decipiens type. Carbohydrate tolerance was reduced in two thirds of the patients. In some of them this disorder was attended with clinical signs of diabetes and a severe course of myocardial infarction. In patients with diminished carbohydrate tolerance the sugar curve remained abnormal 18 months after the disease. Obvious diabetes developed in 6 out of 20 patients examined. Among relatives of patients with disturbed carbohydrate metabolism diabetes mellitus was encountered more frequently and the incidence of ischemic heart disease and hypertension was higher.
