Detection and analysis of tumor fluorescence using a two-photon optical fiber probe.

The utility of a two-photon optical fiber fluorescence probe (TPOFF) for sensing and quantifying tumor fluorescent signals was tested in vivo. Xenograft tumors were developed in athymic mice using MCA207 cells expressing green fluorescent protein (GFP). The TPOFF probe was able to detect ex vivo fluorescence from excised tumors containing as little as 0.3% GFP-expressing cells. TPOFF results were similar to both flow-cytometric analysis of tumor cells after isolation and suspension, and fluorescence determined by microscope images of cryosectioned tumors. TPOFF was then used to measure GFP fluorescence from tumors in live mice. The fiber probe detected fluorescently-labeled Herceptin antibody targeted to HER2-expressing tumors in severe combined immunodeficient mice. Dendrimer nanoparticles targeted by folic acid and having 6-TAMRA as a fluorescent probe were also used to label KB cell tumors in vivo. The fiber probe documented a fourfold increase in tumor fluorescence in animals that received the targeted dendrimer. These results suggest TPOFF can be used as a minimally invasive system for identifying tumor markers and monitoring drug therapy.

Optical circulators in two-dimensional magneto-optical photonic crystals.

We propose an optical circulator formed of a magneto-optical cavity in a 2D photonic crystal. With spatially engineered magnetic domain structures, the cavity can be designed to support a pair of counterrotating states at different frequencies. By coupling the cavity to three waveguides, and by proper matching of the frequency split of the cavity modes with the coupling strength between the cavity and the waveguide, ideal three-port circulators with complete isolation and transmission can be created. We present a guideline for domain design needed to maximize the modal coupling and the operational bandwidth for any given magneto-optical constant.

High-contrast images of semiconductor sites via one-photon optical beam-induced current imaging and confocal reflectance microscopy.

We demonstrate a computationally efficient procedure for determining only the semiconductor sites in a confocal reflectance image of an integrated circuit. It utilizes a one-photon optical beam-induced current (1P-OBIC) and confocal reflectance images that are generated from the same focused excitation beam. A 1P-OBIC image is a two-dimensional map of the currents induced by the beam as it is scanned across the circuit surface. A 1P-OBIC is produced by an illuminated semiconductor material if the excitation photon energy exceeds the bandgap. The 1P-OBIC image has no vertical resolution because the 1P-OBIC is linear with the excitation beam intensity. The exclusive high-contrast image of semiconductor sites is generated by the product of the 1P-OBIC image and the confocal image. High-contrast images of the metal sites are also obtained by the product of the complementary OBIC image and the same confocal image.

A three-dimensional optical photonic crystal with designed point defects.

Photonic crystals offer unprecedented opportunities for miniaturization and integration of optical devices. They also exhibit a variety of new physical phenomena, including suppression or enhancement of spontaneous emission, low-threshold lasing, and quantum information processing. Various techniques for the fabrication of three-dimensional (3D) photonic crystals--such as silicon micromachining, wafer fusion bonding, holographic lithography, self-assembly, angled-etching, micromanipulation, glancing-angle deposition and auto-cloning--have been proposed and demonstrated with different levels of success. However, a critical step towards the fabrication of functional 3D devices, that is, the incorporation of microcavities or waveguides in a controllable way, has not been achieved at optical wavelengths. Here we present the fabrication of 3D photonic crystals that are particularly suited for optical device integration using a lithographic layer-by-layer approach. Point-defect microcavities are introduced during the fabrication process and optical measurements show they have resonant signatures around telecommunications wavelengths (1.3-1.5 microm). Measurements of reflectance and transmittance at near-infrared are in good agreement with numerical simulations.

Analysis and optimization of a diffuse photon optical tomography of turbid media

In a numerical study, we investigate a diffuse-photon computed tomography of a turbid medium. Using a perturbation approach, we relate through a matrix K a bulk heterogeneous distribution of the optical absorption coefficient &mgr;(a) that characterizes the heterogeneity in an otherwise homogeneous turbid medium to the diffuse photon flux that emerges from its surface. By studying the condition number (N(C)) of the matrix K as a function of illumination-detection schemes and choices of reconstruction grids, we explore strategies that optimize the fidelity and spatial resolution of the computed tomography.

Large optical photon sieve.

A photon sieve with 10(7) holes has been constructed for operation at optical wavelengths. Details of the design, fabrication, and performance of this device are presented. The 1 m focal-length, 0.1 m diameter element is diffraction limited over a significant bandwidth and has a moderate field of view.

Coherent optical photons from shock waves in crystals.

We predict that coherent electromagnetic radiation in the 1-100 THz frequency range can be generated in crystalline materials when subject to a shock wave or soliton-like propagating excitation. To our knowledge, this phenomenon represents a fundamentally new form of coherent optical radiation source that is distinct from lasers and free-electron lasers. The radiation is generated by the synchronized motion of large numbers of atoms when a shock wave propagates through a crystal. General analytical theory and NaCl molecular dynamics simulations demonstrate coherence lengths on the order of mm (around 20 THz) and potentially greater. The emission frequencies are determined by the shock speed and the lattice constants of the crystal and can potentially be used to determine atomic-scale properties of the shocked material.

Optics of photonic nanojets.

The detailed optics of photonic nanojets generated by normal plane-wave incidence on dielectric cylinders is discussed. These nanojets have a subwavelength beam waist and propagate with little divergence for several wavelengths. A physical explanation for this peculiar behavior is presented. Characteristic dimensions of the nanojets for a large range of physical parameters are calculated.

Binary projective measurement via linear optics and photon counting.

We investigate the implementation of binary projective measurements with linear optics. This problem can be viewed as a single-shot discrimination of two orthogonal pure quantum states. We show that any two orthogonal states can be perfectly discriminated using only linear optics, photon counting, coherent ancillary states, and feedforward. The statement holds in the asymptotic limit of a large number of these physical resources.

1.5 microm photon-counting optical time-domain reflectometry with a single-photon detector based on upconversion in a periodically poled lithium niobate waveguide.

Optical time-domain reflectometry (OTDR) is one of the most powerful tools in the characterization of optical fiber links. We demonstrate a photon-counting OTDR system at 1.5 microm with a single-photon detector, which combines frequency upconversion in a periodically poled lithium niobate waveguide and a silicon avalanche photodiode. The system exhibits high sensitivity, good spatial resolution, and short measurement time.

Ultralow loss, high Q, four port resonant couplers for quantum optics and photonics.

We demonstrate a low-loss, optical four port resonant coupler (add-drop geometry), using ultrahigh Q (>10(8)) toroidal microcavities. Different regimes of operation are investigated by variation of coupling between resonator and fiber taper waveguides. As a result, waveguide-to-waveguide power transfer efficiency of 93% (0.3 dB loss) and nonresonant insertion loss of 0.02% (<0.001 dB) for narrow bandwidth (57 MHz) four port couplers are achieved in this work. The combination of low-loss, fiber compatibility, and wafer-scale design would be suitable for a variety of applications ranging from quantum optics to photonic networks.

Discrete solitons in photorefractive optically induced photonic lattices.

We demonstrate that optical discrete solitons are possible in appropriately oriented biased photorefractive crystals. This can be accomplished in optically induced periodic waveguide lattices that are created via plane-wave interference. Our method paves the way towards the observation of entirely new families of discrete solitons. These include, for example, discrete solitons in two-dimensional self-focusing and defocusing lattices of different group symmetries, incoherently coupled vector discrete solitons, discrete soliton states in optical diatomic chains, as well as their associated collision properties and interactions. We also present results concerning transport anomalies of discrete solitons that depend on their initial momentum within the Brillouin zone.

All-optical tunable photonic bandgap microcavities with a femtosecond time response.

An all-optical tunable photonic bandgap microcavity made from a two-dimensional polystyrene photonic crystal is fabricated by focused ion beam etching. The pump and probe scheme is adopted to measure tunability based on the femtosecond optical Kerr effect. An ultrafast response time of less than 120 fs is achieved for the tunable photonic bandgap microcavity. The microcavity resonant wavelength shifts 3.1 nm under excitation of 9.4 GW/cm2 pump intensity, which is in agreement with the theoretical prediction.

781 Mbit/s photon-counting optical communications using a superconducting nanowire detector.

We demonstrate 1550 nm photon-counting optical communications with a NbN-nanowire superconducting single-photon detector. Source data are encoded with a rate-1/2 forward-error correcting code and transmitted by use of 32-ary pulse-position modulation at 5 and 10 GHz slot rates. Error-free performance is obtained with -0.5 detected photon per source bit at a source data rate of 781 Mbits/s. To the best of our knowledge, this is the highest reported data rate for a photon-counting receiver.
