Thymic hormones and syngeneic T-lymphocytes are not required for leukopoiesis in an in vivo culture system for mouse bone marrow cells.

In order to explore the regulatory influence of thymic hormones and T cells on leukopoiesis , bone marrow cells from normal and athymic nude mice were cultured in peritoneal diffusion chambers (DC) that were implanted intraperitoneally into either normal or nude mice. T-cell-deficient nu/nu-C3H marrow cells formed slightly above normal numbers and nu/nu-BALB/c formed slightly below normal numbers of progenitor cells for granulocytes and/or macrophages in four-day DC cultures. Seven-day leukopoiesis in DC, as estimated by 3H-thymidine incorporation and differential cell counts, was not detectably affected by T-cell absence from nu/nu-BALB/c marrow. Leukopoiesis , including formation of progenitor cells, was not significantly different in normal and T-cell-deficient DC hosts. Normal thymocytes added to the T-cell-deficient bone marrow inocula decreased the generation of progenitor cells. These results indicate that (a) expansion of a proliferating granulocyte-macrophage progenitor cell population takes place in in vivo DC cultures without the presence of thymic hormones and T-lymphocytes, and that (b) T-lymphocytes may either stimulate or inhibit progenitor cell growth, and the balance between stimulation and inhibition apparently varies between mouse strains.

[Stimulating and protective effect of recombinant interleukin-1-beta (beta-leukin) on leukopoiesis in chemotherapy of malignant tumors].

Clinical studies were carried out in a group of 71 cancer patients to evaluate the effects of intravenous infusion of human recombinant interleukin-1-beta (betaleukin) (I and II phases). It was shown that daily single doses of 10-20 ng/kg body, for 5 days, markedly stimulated leukopoiesis (granulocytopoiesis) in toxic (grade II-IV) leukopenia which hampered combination chemotherapy. A protective effect on leukopoiesis (granulocy-topoiesis) was recorded in cases of moderate leukopenia when chemotherapy was continued. Such side-effects as stage I-III hyperthermia (13.5%) or their combination (50.0%) were effectively managed with paracetamol and antihistamines administration without causing suspension of the drug.

Periodic oscillations in leukopoiesis models with two delays.

The term leukopoiesis describes processes leading to the production and regulation of white blood cells. It is based on stem cells differentiation and may exhibit abnormalities resulting in severe diseases, such as cyclical neutropenia and leukemias. We consider a nonlinear system of two equations, describing the evolution of a stem cell population and the resulting white blood cell population. Two delays appear in this model to describe the cell cycle duration of the stem cell population and the time required to produce white blood cells. We establish sufficient conditions for the asymptotic stability of the unique nontrivial positive steady state of the model by analysing roots of a second degree exponential polynomial characteristic equation with delay-dependent coefficients. We also prove the existence of a Hopf bifurcation which leads to periodic solutions. Numerical simulations of the model with parameter values reported in the literature demonstrate that periodic oscillations (with short and long periods) agree with observations of cyclical neutropenia in patients.

Anomalous leukopoiesis in two patients with Crohn's disease.

Crohn's disease is a major form of chronic inflammatory bowel disease in the western world. The molecular genetic basis of Crohn's disease is unknown. In this study, we present evidence for anomalous leukopoiesis-namely, the generation of a leukocyte subset characterized by aberrant expression of gammadelta T cell receptor (gammadeltaTCR) with or without CD19 on a myeloid background-in two patients with Crohn's disease. The aberrant cells of patient 1 have the surface phenotype gammadeltaTCR + CD19 - CD14 + CD64 +. The aberrant cells of patient 2 have the surface phenotype gammadeltaTCR + CD19 + CD14 - CD64 + CD16 + CD13 + CD33 +. The results presented here are significant both in light of recent speculation that a critical defect in Crohn's disease may be at the level of hematopoiesis and because the CD19 gene lies within the region on chromosome 16 that corresponds with the Crohn's disease susceptibility locus IBD1.

Increased bone marrow production of granulocytes and mononuclear phagocytes induced by mycobacterial adjuvants: improved recovery of leukopoiesis in mice after cyclophosphamide treatment.

The effects of complete Freund adjuvant (CFA) or Mycobacterium bovis BCG on leukopoiesis and on leukopoietic recovery from cyclophosphamide treatment in mice was studied. CFA injected subcutaneously or intraperitoneally resulted in increased blood granulocyte and monocyte counts, increased numbers of bone marrow granulocyte and mononuclear phagocyte progenitors, and increased hematopoietic colony-stimulating factor in the serum. Furthermore, the quantitative cellular response within 24 h to an induced sterile intraperitoneal inflammation (thioglycolate) was augmented by subcutaneous CFA. In mice given CFA subcutaneously, blood granulocyte counts, as well as the peritoneal granulocyte and macrophage response to intraperitoneal thioglycolate, recovered more quickly than did those of the controls after a 250-mg/kg dose of cyclophosphamide. CFA-treated mice consistently maintained blood granulocyte and monocyte counts 1.3-to 4-fold higher than those of the controls for 2 weeks while receiving 75 mg of cyclophosphamide per kg every other day. Mice pretreated with CFA intraperitoneally had higher numbers of bone marrow colony-forming units in culture and higher levels of serum colony-stimulating factor after 250-mg/kg injections of cyclophosphamide. Similarly, BCG resulted in increased bone marrow colony-forming units in culture, increased serum colony-stimulating factor, and a faster return of the peritoneal inflammatory response after cyclophosphamide injection. These results show that mycobacterial adjuvants accelerate recovery of leukopoietic functions after cyclophosphamide treatment and suggest a mechanism whereby such adjuvants afford nonspecific protection against infection in immunosuppressed mice.

[The effect of growth hormone on leukopoiesis: in vivo and in vitro studies].

Growth hormone (GH), which has been extracted from the pituitary gland since early times, has become easily available by the advance of genetic engineering in recent year. Its clinical application to treatment in various fields, involving obesity, wounds, fractures, gastric ulcers and so on, is being increasingly discussed. The presence or absence of the effect of GH on leukopoiesis was studied in vivo and in vitro experiments. In the in vivo experiment, GH was administered to rats whose bone marrow production had been suppressed by the injection of mitomycin C, and time-course changes in the peripheral blood leukocyte count in these rats were compared with those in rats given physiological saline solution alone (control group). The in vitro experiment was performed by colony assay of mouse marrow cells. Insulin growth factor-1 (IGF-1) was also studied in the in vitro experiment. The in vivo experiment revealed that GH promoted recovery of leukocytes from the nadir, and in the in vitro experiments GH and IGF-1 were demonstrated to increase the number of colonies in the presence of granulocyte macrophage colony stimulating factor (GM-CSF). GH was considered to exert effects on myeloid progenitor cells and the hemopoietic microenvironment simultaneously, resulting in an increase in leukocytes.

[Effect of autologous blood irradiated in isolation on the restoration of leukopoiesis in an experimental model of prolonged leukopenia].

Reinfusion of irradiated (220 Gy) isolated blood (IIB) was shown to accelerate leukopoiesis restoration in conditions of myelodepression induced by the injection of cyclophosphane. Restoration of leucocyte count in the peripheral blood was preceded by the increase in DNA synthesis and bone marrow cellularity. Reinfusion of IIB also promoted a more rapid restoration of cellularity of lymphoid organs with cAMP predominating therein. The comparison of the processes under study in time permits to assume that the stimulatory effect of IIB is related to activation of proliferation and stimulation of cell maturing in bone marrow and lymphoid organs.

[Comparison of oral administration and intramuscular injection of thymus preparations on the leukopoiesis test in rats and some of the preventive possibilities emerging from the results].

Considering the fact that all immunologic processes consume energy, we have been attempting for more than a decade to combine immunostimulating measures with such for increasing the energetic state. This basic resulted in designing several modern variants of the Oxygen Multistep Immunostimulation. In order to make this method broadly applicable to public health care, its protocol had to be simplified in such a way that it can easily be applied also to outpatients at a reasonable expenditure of time. Such a program is described here among other variants. Its concept is based on measurements of leukopoiesis in rats after administration of thymus extract. The test system used revealed that oral administration of the thymus extract taken from specified dragées was of at least the same effect as intramuscular injection. The simplified variant of the Oxygen Multistep Immunostimulation which can be practised ambulantly within a short time, consists of the administration of thymus dragées and the 15-min-oxygen multistep quick procedure of the Oxygen Multistep Therapy. It is demonstrated by comparison of photographs of a case report the precancerous skin lesions can be unprimed by this simple treatment within 10 months. The finding indicates that along these lines a permanently effective cancer prevention might become practicable. Furthermore, it is supposed that the simple treatment schedule described should be of benefit in preventive medicine and gerontology as well, in order to fight illness in general and to reduce the disposition to diseases having a bad prognosis particularly in old age.

[Effect of gold therapy in rheumatoid arthritis on leukopoiesis and granulocyte phagocytosis].

In 12 patients with rheumatoid arthritis the investigations of leukopoiesis and granulocytic phagocytosis were carried out before and after a gold therapy. A marked reduction of granulocytic phagocytosis could be observed here which decreased after the gold therapy, yet remained below the values of a normal collective. Partly contrary informations are discussed. The disturbance of the phagocytosis capacity of granulocytes may possibly be due to a rheumatic factor.

Effects of piracetam on erythropoiesis and leukopoiesis in rats.

Erythro- and leukopoiesis in rats after 10-day treatment with piracetam (2 x 200 mg/kg b.w.) were studied. Increase of reticulocytes and of 59-iron incorporation in newly formed erythrocytes accompanied by an increase of the index of maturation of erythroblasts in bone marrow were found suggesting a piracetam-induced stimulation of erythropoiesis. Piracetam also increased the number of small lymphocytes and the maturation of granulocytes in bone marrow. It is supposed that piracetam exerting a positive effect on the metabolic processes of erythroid, myeloid and lymphoid precursors stimulates their proliferation and differentiation.
