Identification and characterization of the dihydropyridine-binding subunit of the skeletal muscle dihydropyridine receptor.

Photoaffinity labeling of isolated triads and purified dihydropyridine receptor with [3H]azidopine and (+)-[3H]PN200-110 has been used to identify and characterize the dihydropyridine-binding subunit of the 1,4-dihydropyridine receptor of rabbit skeletal muscle. The 1,4-dihydropyridine receptor purified from rabbit skeletal muscle triads contains four protein subunits of 175,000, 170,000, 52,000, and 32,000 Da (Leung, A., Imagawa, T., and Campbell, K. P. (1987) J. Biol. Chem. 262, 7943-7946). Photoaffinity labeling of isolated triads with [3H]azidopine resulted in specific and covalent incorporation of [3H]azidopine into only the 170,000-Da subunit of the dihydropyridine receptor and not into the 175,000-Da glycoprotein subunit of the receptor. The [3H]azidopine-labeled 170,000-Da subunit was separated from the 175,000-Da glycoprotein subunit by sequential elution from a wheat germ agglutinin-Sepharose column with 1% sodium dodecyl sulfate followed by 200 mM N-acetylglucosamine. Photoaffinity labeling of purified dihydropyridine receptor with [3H]azidopine or (+)-[3H]PN200-110 also resulted in the specific and covalent incorporation of either ligand into only the 170,000-Da subunit. Therefore, our results show that the dihydropyridine-binding subunit of the skeletal muscle 1,4-dihydropyridine receptor is the 170,000-Da subunit and not the 175,000-Da glycoprotein subunit.

Competitive binding experiments reveal differential interactions for dihydropyridine calcium channel activators and antagonists at dihydropyridine receptors on mouse brain membranes.

The binding of the dihydropyridine (+/-)-202-791 and its corresponding calcium channel activating and calcium channel antagonist enantiomers ((+)-S-202-791 and (-)-R-202-791, respectively) to dihydropyridine receptors on mouse brain membranes was studied through competition for [3H]nitrendipine binding and 3H-labelled (+/-)-BAY K8644 ((+/-)-[3H]BAY K8644). Direct binding studies with (+/-)-[3H]BAY K8644 and [3H]nitrendipine revealed high affinity binding to a homogeneous set of dihydropyridine calcium channel activator and antagonist receptors on mouse brain membranes, (+/-)-[3H]BAY K8644 binding to approximately one half as many receptors as did [3H]nitrendipine. Competition binding studies revealed a significant discrimination of both high and low affinity receptors for (-)-R-202-791 and a homogeneous set of receptors for (+)-S-202-791 regardless of whether (+/-)-[3H]BAY K8644 or [3H]nitrendipine was the competing radioligand. Molar ratios (1:1, 5:1, 10:1) of (+)-S-202-792 to (-)-R-202-791 inhibited [3H]nitrendipine binding with displacement binding isotherms substantially different from those predicted on the basis of the binding properties of the individual enantiomers. These data suggest that dihydropyridine calcium channel antagonists and activators bind to different allosterically linked receptors or domains of the dihydropyridine protein associated with the voltage-dependent calcium channels. Furthermore, these results support the concept of multiple binding sites for dihydropyridine ligands.

Crystal structure of the dihydropyridine Ca2+ antagonist felodipine. Dihydropyridine binding prerequisites assessed from crystallographic data.

The molecular structure of the dihydropyridine Ca2+ antagonist felodipine (ethyl methyl 1,4-dihydro-2,6-dimethyl-4-(2,3-dichlorophenyl)-3,5-pyridinedicarboxy late) has been determined by X-ray crystallographic methods. The dihydropyridine ring in this potent smooth muscle relaxant is among the flattest found in such structures. This is in qualitative agreement with previous investigations of dihydropyridine Ca2+ antagonists; deviations from planarity in the dihydropyridine ring are generally smallest in the most active compounds. Hydrogen-bonding patterns observed in the crystal lattices of several dihydropyridine Ca2+ antagonists are compared. Antiperiplanar carbonyl groups are partly shielded from forming hydrogen bonds in compounds with relatively bulky ortho phenyl substituents. Conformational prerequisites for a favorable hydrogen-bonding geometry toward a receptor site may thus involve synperiplanar carbonyl groups.

Crystal structures and pharmacologic activities of 1,4-dihydropyridine calcium channel antagonists of the isobutyl methyl 2,6-dimethyl-4-(substituted phenyl)-1,4-dihydropyridine-3,5-dicarboxylate (nisoldipine) series.

A series of isobutyl methyl 2,6-dimethyl-4-(X-substituted phenyl)-1,4-dihydropyridine-3,5-dicarboxylates (X = H, 2-NO2, 3-NO2, 3-CN, 3-MeO, 4-F, 2-CF3, 3-CF3, and 4-Cl) related to and including nisoldipine (X = 2-NO2) has been synthesized, their solid-state structures determined by X-ray analysis (X = H, 2-NO2, 3-NO2, 3-CN, 3-MeO, and 4-F), and their pharmacologic activities determined, as the racemic compounds, against [3H]nitrendipine binding and K+-depolarization-induced tension responses in intestinal smooth muscle as measures of Ca2+ channel antagonist activity. Comparisons of structure are presented to previously analyzed 1,4-dihydropyridines. The degree of 1,4-dihydropyridine ring puckering is dependent on the nature and position of the phenyl ring substituent and the adopted interring conformation. Different ester substituents affect 1,4-dihydropyridine ring puckering to a small extent in most cases. Pharmacologic and radioligand binding activities for the nine compounds studied show a parallel dependence on phenyl ring substituent, but the compounds are approximately 10-fold more active in the radioligand binding assay than in the pharmacologic assay. Consistent with a previous report for the nifedipine series (Fossheim et al. J. Med. Chem. 1982, 25, 126), pharmacologic activity increases with increasing 1,4-dihydropyridine ring planarity.

[Efficacy of and tolerance to combined therapy with dihydropyridine and non-dihydropyridine Ca antagonists in patients with moderate arterial hypertension].

The authors of the article studied efficacy of combined therapy with dihydropyridine and non-dihydropyridine Ca antagonists, its influence on structural and functional condition of the heart in 53 patients (28 men and 25 women) with moderate arterial hypertension (AH), and their tolerance to the therapy. Before and during the treatment the patients underwent 24-hour arterial pressure (AP) monitoring and Doppler echoCG. Due to combined therapy with isoptin SR and corinfar retard complete hypotensive effect (AP < 140/90 mmHg) was achieved in 83% of cases, and partial effect (diastolic pressure lowered by 10 mmHg)--in 17%. The therapy significantly reduced left ventricular mass index (14.6% on the average; p < 0.01), and improved diastolic function: E/A increased by 10.3% (p < 0.05), and isovolumetric relaxation time decreased by 13.6% (p < 0.01). Combined therapy also resulted in a 1.5 to 4 time reduction in the frequency of side effects of isoptin SR and corinfar retard due to reduction in their doses and/or mutual neutralization of their side effects. The paper demonstrates high antihypertensive efficacy of and good tolerance to the combination of dihydropyridines and non-dihydropyridines when they are administered for prolonged therapy in patients with moderate AH.

Voltage-sensitive calcium flux into bovine chromaffin cells occurs through dihydropyridine-sensitive and dihydropyridine- and omega-conotoxin-insensitive pathways.

The fluorescent Ca2+ indicator FURA-2 was used to characterize the depolarization-related intracellular Ca2+ signalling process in bovine adrenal chromaffin cells. Depolarization with high K+ (10-65 mM) gave rise to a very rapid increase in intracellular free Ca2+ concentration, which subsequently decayed slowly towards a "plateau". The size of this initial increase varied sigmoidally with the calculated membrane potential, the relationship being described well by a Boltzmann distribution function for a transition between two states (transition potential, -23 mV). A dihydropyridine calcium channel agonist [(+)202-791, 1 microM] raised intracellular free Ca2+ concentration further in the presence of 30 mM K+, and it enhanced the initial intracellular Ca2+ response to depolarization. Voltage-sensitive calcium channels in chromaffin cells are believed to include the L-type. Several dihydropyridine calcium channel antagonists [(-)202-791, nifedipine, nitrendipine; 1-5 microM], known to be active on L-type channels, caused only modest inhibition of K+ -induced increase in intracellular free Ca2+ concentration: c. 50% (at 30 mM K+) and 25% (at 40-70 mM K+). In addition, omega-conotoxin GVIA (1-10 microM), a blocker of neuronal N- and L-type calcium channels, reduced the initial increase in intracellular free Ca2+ concentration only slightly at 55 mM K+. Further, the dihydropyridine-insensitive component of the intracellular Ca2+ signal was also insensitive to omega-conotoxin, which was otherwise quite active in a central nervous rat in vivo preparation Gd3+ (40 microM), a potent calcium antagonist in the chromaffin cell, blocked the intracellular Ca2+ response to depolarization. When added at different times after K+ stimulation, however, Gd3+ reduced intracellular free Ca2+ concentration to control levels along a slow time course of several minutes. Similar results were obtained when EGTA was added to reduce extracellular Ca2+ concentration to sub-nanomolar levels, in the presence of high K+. We conclude that bovine chromaffin cells are equipped with at least two different classes of voltage-dependent calcium channels, only one of which is likely to be the L-type channel. We also propose that depolarization, in addition to stimulating Ca2+ influx, may also lead to enhancement of Ca2+ release from an intracellular store.

Frequency- and potential-dependency of the negative inotropic action of various dihydropyridine and non-dihydropyridine calcium antagonists.

Transmembrane voltage and beat frequency are important determinants of the action of several organic calcium antagonists. This is well-known for the cationic amphiphilic calcium antagonists. We intended to assess the functional impact of these phenomena in cardiac muscle with special regard to dihydropyridines. Therefore, concentration-response curves were constructed in isolated guinea-pig left atria for the negative inotropic effect of various compounds. The dihydropyridines nifedipine, racemic nitrendipine, nisoldipine, and felodipine, and the enantiomers of isradipine were investigated at different stimulation frequencies (1 Hz, 2.5 Hz, 4.5 Hz), and at different extracellular K+ concentrations (2.7 mM, 5.4 mM, 10.8 mM). These drugs were compared with the cationic amphiphilic compounds gallopamil, verapamil and diltiazem. The potency of some dihydropyridines, particularly nitrendipine, could be modulated to a remarkable extent, covering several orders of magnitude. The potential-dependency of the drugs depended on stimulus frequency and ranged from less than a half to two orders of magnitude. At 2.5 Hz, the rank order of extent of potential-dependency was gallopamil greater than nitrendipine greater than diltiazem greater than verapamil = (+)-isradipine greater than (-)-isradipine greater than or equal to nisoldipine greater than or equal to felodipine = nifedipine. Based on data obtained from binding studies in intact atria and from patch-clamp measurements of calcium current blockade, a mathematical model was used which describes the observed potency changes.(ABSTRACT TRUNCATED AT 250 WORDS)

Novel 2-amino-1,4-dihydropyridine calcium antagonists. II. Synthesis and antihypertensive effects of 2-amino-1,4-dihydropyridine derivatives having N,N-dialkylaminoalkoxycarbonyl groups at 3- and/or 5-position.

Novel 2-amino-1,4-dihydropyridine derivatives I, which contain N,N,-dialkylaminoalkoxycarbonyl groups at the 3- and/or 5-position, were synthesized and their antihypertensive effects were evaluated in spontaneously hypertensive rats. Remarkably prolonged duration of antihypertensive action was observed when a tertiary amino group was introduced into either the 3- and/or 5-ester side-chain of the 1,4-dihydropyridine ring. In particular, the compounds containing cyclic amino moieties at the 3-position showed greater potency than those with acyclic amino moieties. Chemical modification studies indicated that the two ester side-chains of 1,4-dihydropyridine at the 3- and 5-position might function in a different manner in relation to the antihypertensive activities. 3-(1-Benzhydrylazetidinited potent and long-lasting antihypertensive effects with gradual onset of action, and is a promising candidate as an antihypertensive drug.

Influence of pHo on calcium channel block by amlodipine, a charged dihydropyridine compound. Implications for location of the dihydropyridine receptor.

We have investigated the modulation of L-type calcium channel currents in isolated ventricular cells by the dihydropyridine derivative amlodipine, a weak base with a pKa of 8.6. Under conditions that favor neutral drug molecules, amlodipine block resembles other, previously described, neutral dihydropyridine derivatives: block is more pronounced at depolarized voltages, repetitive pulsing is not needed to promote block, and recovery is complete at hyperpolarized voltages. When the drug is ionized, depolarized voltages still enhance block, however, the time course is slow and speeded by repetitive pulses that open channels. Recovery from block by ionized drug molecules is very slow and incomplete, but can be rapidly modified by changes in external hydrogen ion concentration. We conclude from these observations that the degree of ionization of the drug molecule can affect access to the dihydropyridine receptor and that external protons can interact with the drug-receptor complex even if channels are blocked and closed. These observations place limitations on the location of this receptor in the ventricular cell membrane.

Quantitative analysis of the dihydropyridines, 3-(2-furoyl)-5-methoxycarbonyl-2,6-dimethyl-4-(2-nitrophenyl)- 1,4-dihydropyridine and nifedipine, by high-performance liquid chromatography with electrochemical detection.

An analytical method based on solvent extraction and reversed-phase high-performance liquid chromatographic separation with electrochemical detection has been developed for the dihydropyridines, 3-(2-furoyl)-5-methoxycarbonyl-2,6-dimethyl-4-(2-nitrophenyl) -1,4-dihydropyridine (MDL 72.567) and nifedipine. The analysis includes an internal standard of similar light sensitivity to correct for possible photodegradation during the procedure. The specificity of electrochemical detection precludes interference from oxidized metabolites. Total analysis time was 35 min per sample, and the detection limit for quantification was 1-2 ng/ml. Linear regression analysis gave calibration curves with coefficients or correlation of 0.9992 for MDL 72.567 (1-100 ng) and 0.997 for nifedipine (1-50 ng). Assays for within-run and day-to-day reproducibility gave coefficients of variation of 3.9% and 6.0%, respectively, at concentrations of 50 ng/ml. The method has been applied to the analysis of plasma levels of nifedipine and MDL 72.567 in dogs.

[Regulation of hormone biosynthesis in guinea pig adrenal glands by potassium ions as affected by dihydropyridines. 1. Analysis of changes in steroidogenesis evoked by dihydropyridines].

Influence of Ca2+ channel modulators BAY K 8644, nitrendipine and newly synthesized derivative of 1,4-dihydropyridine: 4-(3', 4'-dimethoxyphenyl)-2,6-dimethyl-3,5- diethoxy-carbonyl-1,4-dihydropyridine-(DHP-51) on aldosterone production by adrenocortical slices depends on K+ content in the incubation medium. The modulators only slightly influence the hormone output at low K+ level in the medium. Intensive synthesis of aldosterone at high level of potassium in the medium was prevented in the presence of DHP-51 and low concentration of BAY K 8644. DHP-51 inhibited [3H]-cholesterol incorporation into all main corticosteroids in the high-potassium medium.

Solubilization and affinity labeling of a dihydropyridine binding site from skeletal muscle: effects of temperature and diltiazem on [3H]dihydropyridine binding to transverse tubules.

Effects of temperature and d-cis-diltiazem (DTZ) on [3H]nitrendipine (NTD) and [3H]nimodipine (NIM) binding to skeletal muscle t-tubular membranes were studied. A decrease in temperature from 37 degrees C to 10 degrees C decreased KD and increased Bmax slightly. DTZ increased binding by increasing Bmax under all conditions and also decreased KD for NTD at 37 degrees C. The binding protein labeled with [3H]isothiocyanate dihydropyridine revealed a molecular weight of 36,000. The binding site for NTD was solubilized by deoxycholate and dihydropyridine binding was still stimulated by DTZ in the solubilized form.

Reconsideration of vascular selectivity of dihydropyridine calcium antagonists: comparison of cardiovascular profile of mepirodipine, a novel dihydropyridine consisting of a single stereoisomer with (+)-(S)-(S) conformation, with those of nifedipine and nicardipine.

The coronary vasodilator effect and negative chronotropic, inotropic and dromotropic effects of mepirodipine, the single stereoisomer of a novel dihydropyridine Ca antagonist, were compared with those of nifedipine and nicardipine using canine isolated, blood-perfused heart preparations. Drugs were injected into each nutrient artery. In the sinoatrial node preparation, the dose producing a 15% decrease in sinoatrial rate was 1.3, 6.5 and 2.5 micrograms for mepirodipine, nifedipine and nicardipine, respectively. In the papillary muscle preparation, the dose causing a 50% decrease in developed tension was 44, 6.5 and 54 micrograms for mepirodipine, nifedipine and nicardipine, respectively. The dose causing a 50% increase in blood flow through the anterior septal artery was 0.26, 0.18 and 2.0 micrograms for mepirodipine, nifedipine and nicardipine, respectively, while the time required for return to half maximum at the above dose was 13.1, 1.8 and 4.1 min, respectively. In the atrioventricular node preparation, the dose producing a 50% increase in AH interval was 1.6, 2.4 and 3.7 micrograms for mepirodipine, nifedipine and nicardipine, respectively. These results indicate that mepirodipine is a potent and long-lasting dihydropyridine Ca antagonist, whose vascular selectivity is highest against cardiac contractility, but less selective against sinoatrial node automaticity and atrioventricular nodal conduction, compared with nifedipine.

Vasodilator profile of a new 1,4-dihydropyridine derivative, 2,6-dimethyl-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methylamino)]-ethyl ester 5-methyl ester hydrochloride (YC-93).

A new 1,4-dihydropyridine derivative, 2,6-dimethyl-4-(3-nitrophenyl)-4,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methylamino)]-ethyl ester 5-methyl ester hydrochloride (YC-93), when i.v. injected into anesthetized dogs, exhibited not only a greated vasodilation in both cerebral and coronary than in femoral vessels, but also about 100 to 300 times higher potency as well as longer durability than any of reference drugs such as isoxsupurine, papaverine and cinnarizine. YC-93 was also effective in vasodilation by i.m. and i.d. administration. When administered into vertebral and coronary arteries, YC-93 caused vasodilation at the doses that did not affect systemic blood pressure. YC-93 did not potentiate the vasodilator effect of adenosine, and vasodilation by YC-93 was influenced by neither propranolol, atropine, diphenhydramine nor aminophylline. Acute toxicity (LD50) of YC-93 was almost the same as that of papaverine in mice and rats. Thus, YC-93 is a potent bu low-toxic vasodilator agent acting preferentially and perhaps directly on cerebral and coronary vascular beds and is well absorbed from gastrointestinal tract into blood stream.

Pharmacological studies of a newly synthetized 1,4-dihydropyridine derivative, 2,6-dimethyl-3,5-dimethoxycarbonyl-4- (o-difluoromethoxyphenyl)-1,4-dihydropyridine (PP-1466).

Hemodynamic effects of a new 1,4-dihydropyridine derivative, 2,6-dimethyl-3,5-dimethoxycarbonyl-4-(o-difluoromethoxyphenyl++ +)-1,4-dihydropyridine (PP-1466), were examined by measuring systemic blood pressure, various arterial blood flow such as femoral, vertebral, renal and superior mesenteric artery, heart rate and respiratory rate in anesthetized dogs. Cardiohemodynamic effects were examined by measuring cardiac output, coronary blood flow, maximal rate of rise of left ventricular pressure (dp/dt max.) and right atrial pressure. The changes in myocardial oxygen consumption associated with the parameters mentioned above were also examined by measuring coronary sinus outflow and oxygen content in blood samples. The results obtained are as follows: The intraarterial administration of PP-1466 to the femoral artery caused vasodilating response. The vasodilating effect of PP-1466 was approximately 200 times more potent than that of papaverine and comparable to that of nifedipine or nicardipine (YC-93). The effective dose to induce vasodilation on femoral, vertebral and superior mesenteric artery by the systemic use of PP-1466 was more than 0.3 microgram/kg i.v. and more than 0.03 mg/kg i.d. The vasodilation in vertebral artery was most prominent. After intravenous administration of PP-1466, renal blood flow decreased probably due to the fall in systemic blood pressure, whereas the flow progressively increased after intraduodenal administration. The effective dose to induce a fall in systemic blood pressure by PP-1466 was the range from 1 to 3 micrograms/kg i.v. and more than 0.1 mg/kg i.d., respectively. Diastolic hypotension was remarkable. Heart rate and respiratory rate were increased at the same time.(ABSTRACT TRUNCATED AT 250 WORDS)

Purification of the cardiac 1,4-dihydropyridine receptor using immunoaffinity chromatography with a monoclonal antibody against the alpha 2 delta subunit of the skeletal muscle dihydropyridine receptor.

The 1,4-dihydropyridine receptor associated with L-type Ca2+ channels was purified about 1700-fold from porcine cardiac sarcolemmal membranes using a simple and rapid (ca. 8 h) two-step procedure: wheat germ agglutinin affinity chromatography followed by immunoaffinity chromatography with a monoclonal antibody (MCC-1) against the alpha 2 delta subunit of the skeletal muscle Ca2+ channel with a glycine elution buffer (pH 3). Gel electrophoresis of this purified sample under non-reducing conditions revealed a major polypeptide band with molecular weight of 190 kDa, which was separated under reducing conditions to a 155 kDa band and 2-3 bands with M(r) about 20 kDa, corresponding to alpha 2 and delta subunits, respectively. The peptide band corresponding to the alpha 1 subunit was not detected in this gel electrophoresis. However, the alpha 1 subunit without bound alpha 2 delta was selectively eluted from MCC-1 Sepharose with 1% Triton X-100. A 190 kDa band corresponding to the alpha 1 subunit was visualized by fluorography and by silver staining in the fraction eluted with Triton X-100. Electrophoretically, the amount of alpha 1 was smaller than that of the alpha 2 subunit in the purified sample obtained here.
