A mouse macrophage-like cell line (MMC-1) and its biological characteristics--biological characterization.

The biological characteristics of mouse macrophage-like cell line (MMC-1) are described. This cell line has been cultured in vitro for more than 20 months. MMC-1 cells are adherent to typical morphology of macrophages, but bigger in size than those of the uncultivated. They are actively phagocytic with abundant Fc receptors. The karyotypes are mostly pantaploidy. This cell line provides a useful tool for the biological and immunological study of macrophage.

IME -- an inhibitor of interferon activity isolated from mouse embryonic tissues. II. Biological characteristics.

The biological properties and mode of action of IME were studied. Extracts of homogenized and lyophilized mouse embryos served as source of IME, which was found to be localized mainly in cells of mouse amnionic membranes and embryonic tissues. Growth of embryos was accompanied by falling activity of the inhibitor. IME activity was manifested only if it acted on cells in established antiviral state, and only in systems with efficient protein synthesis in all stages. IME stimultes cell metabolism. The findings from this study constitute material for further research on factors antagonistic to interferon.

[Treatment based on biological characteristics of lung cancer: biological differences between small cell and non-small cell lung cancer and gene therapy].

Whether chemotherapy or surgical resection is primarily selected for lung cancer therapy is determined based on the pathological diagnosis of small cell or non-small cell lung cancer. In future, however, each standard therapy should be established against the respective subgroups of lung cancer, which will be more precisely defined depending on the biological or the molecular basis, like malignant lymphoma. Much evidence is increasing to help understand the mechanisms mediating differences in clinical behavior and neuroendocrine features between small cell and non-small cell lung cancer. Here we showed the experimental models of tissue-specific gene therapy targeting CEA- or Myc-overexpressing lung cancer cell lines.

[Biological characteristics and prognosis of childhood malignant lymphoma].

The biological characteristics and prognosis of childhood malignant lymphoma were reviewed. A national survey of 568 cases of childhood malignant lymphoma in Japan revealed that 505 cases (88.9%) were non-Hodgkin's lymphoma (NHL), and 63 cases (11.1%) were Hodgkin's lymphoma (HL). The biological characteristics of NHL in Japan were a male predominance, an average age of 7.8 years, and frequent occurrence in the head and neck (38.2%). Histopathologically, only 3.2% of NHL was nodular type and 96.8% was diffuse type (lymphoblastic 28.4%, histiocytic 19.5%, medium-size cell 18.6%, Burkitt's 14.6%, and miscellaneous 15.8%.) The characteristics of the 63 cases of HL were a male predominance, an average age of 9.3 years, 73% of primary lesions in the cervical nodes, and 46.6% of mixed cellularity type. The survival rate at 7 years estimated by the Kaplan -Meier method was 41.1% for patients with NHL, and 84.9% for patients with HL. Leukemic conversion and CNS involvement occurred in 27.9% and 22.9% of patients with NHL, respectively. The current multi-institutional treatment study for NHL conducted by the Children's Cancer and Leukemia Study Group (CCLSG) has improved the prognosis of childhood NHL to an 84.6% induction rate with initial therapy and a relapse-free survival rate of 55.6% at four years. The most important principle for the management of childhood malignant lymphoma is the recognition of the systemic nature of the disease.

[Biological characteristics of the spirulines].

The biological characteristics of Spirulina are reported in this study. The cellular structure and photosynthetic membrane structure of Spirulina (Cyanoschizophyta) were viewed by electron microscopy after freeze-etching. Special characteristics of the alga, its ecological behavior in natural carbonated water and its energetic behaviou in the laboratory cultures are also reported.

[Adoptive immunotherapy in experimental brain tumor in rats--induction of LAK cells and their biological characteristic].

We have investigated the biological characteristic of Lymphokine Activated Killer (LAK) cells induced from spleen cells (SPC) of Fischer rats. Supernatant of 48 hour culture medium (RPMI-1640, 10% FBS, 2-ME: 5 X 10(-5) M, HEPES: 10 mM) of SPC (1 X 10(6)/ml) from Wistar rats in the presence of Con A (5 micrograms/ml) was used as Interleukin 2 (IL-2). LAK cells were generated by co-cultivation of SPC (4 X 10(6)/ml) from Fischer rats with peak reactivity on the 2 nd or 3 rd day of culture. Lytic activity was observed against not only syngeneic tumor cells (T9, RSV induced brain tumor), but also allogenic (KMT-17) and xenogenic (K 562, Raji cell, G 361, YAC-1) tumor cells, while no lytic activity was observed against normal brain cells. Cell depletion test, dye exclution test and immunofluorescence method using monoclonal Antibodies (mAbs) revealed that LAK cells partially belonged to the population of activated T cell group (W 3/13, OX 4 positive) but the precursor cells did not react with any mAbs used (W 3/13, W 3/25, OX 4, OX 8). On the basis of these results, more effective and useful administration of LAK cells is now under investigations by using the rats with brain tumors.

Differences in biological characteristics of two strains of the cestode Hymenolepis diminuta.

Biological characteristics of infectivity, growth rate and fecundity of Hymenolepis diminuta isolated from wild Rattus rattus in Japan were compared with parasites of Texas origin maintained for several generations in this and many other laboratories in laboratory bred Rattus norvegicus. The timing of development and maturation was similar in parasites from both sources, but the mean parasite dry weight was less and the mean egg production lower in Japanese parasites in both single and multiple infections. The differences persisted over 10 weeks in single infections, and were unaffected by rat strain. In all experiments there was much greater variation and heterogeneity in the biological characteristics of the Japanese parasites. It was concluded that the Texas parasites were better adapted to R. norvegicus and in the course of adaptation had become more homogeneous, and that the difference between the parasite strains had a complex genetic basis. Some Japanese parasites of the early isolated generations failed to produce normal eggs, but the proportion of these decreased in later generations. However, selected individual Japanese parasites whose egg production was equal to that of parasites of Texas origin did not produce offspring of larger mean size or higher mean fecundity, and heterogeneity of these characteristics was maintained. In the intermediate host, the mean number of cysticercoids of the Japanese isolate per beetle was higher than that of the Texas strain in Tribolium confusum but lower in Tenebrio molitor. The greater heterogeneity exhibited by Japanese parasites and the genetic basis of the strain difference are discussed with particular reference to geographical isolation, differences in definitive hosts and co-evolution of wild and laboratory host and parasite populations.

[Establishment and biological characteristics of an immortalized human ovarian sarcomatoid carcinoma cell line BUPH:OVSC-2].

BACKGROUND & OBJECTIVE: Cell immortalization technique has been an important method to establish cell lines with expected phenotype. However, there was few report about the establishment of immortalized human ovarian carcinoma cell lines. The purpose of this study was to establish an immortalized human ovarian sarcomatoid carcinoma cell line, and to investigate its biological characteristics. METHODS: A specimen derived from metastatic tumor of a human ovarian sarcomatoid carcinoma in abdominal wall was obtained and cultured in vitro. The 10th passage was transfected with SV40 T gene, colonies were isolated by G418 selection and expanded to immortalized cell lines. The morphology of the cells was observed by microscope and electromicroscope, Growth curve, karyotype analysis, culturing in soft agar, nude mice transplantation, immunohistochemistry, etc. were used to investigate its biologic characteristics. The biologic characteristics of BUPH: OVSC-2 were compared with its original cells. RESULTS: After transfection and selection, an immortalized ovarian carcinoma cell line BUPH: OVSC-2 was established. The cell line has been maintained for over 90 passages. The H&E stain finding of transplanted tumor showed that the cells were morphologically sarcomatoid. However, the transmission electron microscopic observation exposed its epithelial origin. The cells grew exuberantly and had high malignant characteristics. Comparative studies revealed that the biological properties of BUPH: OVSC-2 and its original cells were identical. CONCLUSIONS: BUPH: OVSC-2 was demonstrated as an immortalized human ovarian sarcomatoid carcinoma cell line with high malignancy. It retains similar properties of their parental cells and could be a useful model for the study of human ovarian sarcomatoid carcinoma.

[Culture of the transformed human embryonic tendon cells and its biological characteristics in vitro].

OBJECTIVE: This paper was to study the biological characteristics of the transformed human embryonic tendon cells, the relation between cell growth and culture conditions, and to compare these features with that of human embryonic tendon cells. METHODS: The pts A58H plasmid had successfully used to transform a tendon cell line from human embryo in our past work. The human embryonic tendon cells and the transformed human embryonic tendon cells were cultured in vitro. In different culture conditions, the growth curve were drawn respectively. Population dependence and proliferation capability of the cells were investigated through plate cloning test and soft agar culture. The collagen secreted by cells was identified by immunohistochemical method. RESULTS: In routine culture condition, the growth properties of the human embryonic tendon cell and transformed cells were almost identical. The growth properties of the transformed cells were not changed when the cells were frozen storage. There were changes of growth characteristics of the transformed cells when the culture temperature was changed. The transformed cells could subcultured continually and permanently. The proliferation capability of the transformed cells were stronger than that of the human embryonic tendon cells. Moreover, the growth of the transformed cells was serum-dependent, and the phenomenon of contact inhibition was observed. The transformed cells were not able to grow on soft agar culture. They had the capacity of secreting collagen type I. CONCLUSION: The transformed human embryonic tendon cells could be subcultured continually and permanently, and their growth could be controlled by changing their culture conditions and they had no malignant tendency in biological characteristics. They could be taken as an ideal experimental material for tendon engineering.

[The phenoloxidase test and its application to identify Cryptococcus neoformans with various biological characteristics].

Cryptococcus neoformans strains with various biological characteristics were examinated the phenoloxidase activity on the caffeic acid cornmeal agar (CACA). Other medically important yeasts were also included in this research. Firstly, thirteen reference strains of Cryptococcus neoformans were confirmly found positive phenoloxidase production on the CACA medium. Then, in total 150 isolates of yeasts, forty three Cryptococcus neoformans strains were phenoloxidase positive, other than that 107 other yeast strains were all negative. It is suggested that Cryptococcus neoformans strains with different biological features specifically and uniquely produce phenoloxidase, thus the phenoloxidase test can be applied as a useful tool to identify this pathogenic yeast. Our results also show that the culture on the CACA medium is an effective method to test the phenoloxidase activity within 72 hours. It is further approved that the urease test will not be able to be a screening test for Cryptococcus neoformans in the clinical laboratory, but it is still valuable to identify urease negative Cryptococcus neoformans when the test is used in combination with the phenoloxidase test.

Biological characteristics of breast cancer in obesity.

Biological characteristics of breast cancer in obese patients were investigated in comparison with non-obese patients. 134 patients with breast cancer were divided into three categories; thin, normal and obese based upon the height and weight. Five-year survival rate was 55.6% in the obese patients, whereas it was 79.9% in the non-obese patients including thin and normal subjects. A cause for unfavorable prognosis in the obese patients with breast cancer was clinically and histologically investigated. It was found that in the obese patients primary tumor, regional lymph node engorgement and clinical stage were relatively advanced, scirrhous carcinoma was predominant ( 46%), tumor margin was more infiltrattive (gamma type 64%), the incidence of vascular involvement was high (73%), lymph node metastasis was also frequent (58%) and nuclear grade was low.

Lewis lung carcinoma and two transplantable sarcomas in mice as experimental therapy models: biological characteristics and cell population kinetics.

The biological characteristics of three transplantable tumours: two sarcomas (SaL, MCA) and Lewis lung carcinoma (LLC) have been studied. We investigated histology, DNA ploidy and cell kinetics parameters of the tumours. All examined tumours were aneuploid and rapidly proliferating, with the hyperdiploid fraction greater than 60%. The SaL tumour was found to have the lowest mean aneuploid bromodeoxyuridine labelling index (LI) equal to 21%, while the highest LI of 35.8% was measured for the LLC tumour. The mean S-phase time was short, lasting 8.5 - 10.9 hrs. The potential doubling time (T(pot)) assessed by BrdUrd staining and flow cytometry were as follows: 37.1 hours for SaL, 22.7 hours for MCA and 21.4 hours for LLC tumour. The MCA had the shortest volume doubling time (T(d)) equal to 1.7 days and the longest one, equal to 4.7 days, was found for the LLC. The lowest cell loss was found in the MCA tumour (44%), while the highest in the LLC tumour (81%). As all the examined tumours proliferate rapidly, there is a capacity for accelerated repopulation and therefore the tumours seem to be good models for experimental radiotherapy.

Effects of soybean (Glycine max) germination on biologically active components, nutritional values of seeds, and biological characteristics in rats.

We briefly reviewed the effects of soybean germination on biologically active components, nutritive value of seed and biological characteristics in rats. The purpose of this review is to evaluate the effects of soybean germination on nutritional values of seeds and the potential importance for the use of germinated soybeans, from a contemporary conception, in food preparation as well as on soybean possible influence in optimal health. Germination induced a substantial increase in the content of saponin, oestrogenic compounds and almost all phytosterols, particularly beta-sitosterol of seeds. Lecithin content increased slightly and gradually during germination process. Lipase and alpha-galactosidase activities increased whereas lipoxygenase activities reduced after a short period of germination (< or = 72 h). Therefore, the substantial odour and flavour scores of germinated soybean flour were improved. Germinated seeds were also beneficial to heat penetration, their thermolabile antinutritional factors were easier to inhibit than those of dry beans, also the seeds did not require a long cooking time to be palatable. Duration of the germination process greatly influenced the nutritional value and palatability of seeds and biological characteristics in rats. In rat bioassay, one-day germination of soybeans induced a significant increase of daily body weight gain, daily protein intake in rats and protein efficiency ratio (PER) of seed meal. Palatability of seeds was also improved whereas a 5-day germination resulted in a decrease of PER of seed meal (less than the value of unheated seeds) and induced thyroid enlargement in rats. A vapour thermal treatment (100 degrees C, 20 min) eliminated thyroid-active agents and improved PER of seed meal, food intake and final weight of rats. Well prepared germinated soybeans can be used as a good alternate to animal proteins for more balanced nutritional diet. Development of food products from germinated soybean may be another way to further increase the versatility and utility of soybeans for both developing and industrialized countries, as germination induced the modification of certain specific biologically active components, palatability and nutritive value of seeds.

Biologic characteristics of cultured human vitiligo melanocytes.

BACKGROUND: Vitiligo is a pigmentary disorder of unknown cause characterized by depigmented patches due to destruction of melanocytes. Recently, the inherent cellular defect theory has been discussed. To investigate the biologic characteristics of cultured melanocytes from normal and vitiligo subjects, this study had the purpose to examine the functional and ultrastructural characteristics of these melanocytes and to observe the morphologic and functional changes of melanocytes in response to ultraviolet B irradiation. METHODS: Melanocytes were isolated and cultured from foreskin and arm skin of normal and vitiligo subjects. The DNA synthesis, tyrosinase activity assay, transmission and scanning electron microscopic examination, and the effects of ultraviolet B(UVB)-irradiation on cultured melanocytes were studied. RESULTS: Vitiligo melanocytes showed no significant differences in DNA synthesis and tyrosinase activity compared with normal melanocytes, but the vitiligo melanocytes contained dilated and/or circular rough endoplasmic reticulum (RER) on transmission electron microscopic examination. Exposure of the cultured melanocytes to UVB resulted in increased protein synthesis and tyrosinase activity. Morphologic alterations and changes in DNA synthesis were also noted. Compared with normal melanocytes, the responses of vitiligo melanocyte to UVB showed no significant difference. CONCLUSIONS: Normal and vitiligo melanocytes showed similar biologic characteristics except in the changes of RERs in the vitiligo melanocytes. The ultrastructural aberrations in vitiligo subjects do not seem to be directly related to the biologic characteristics and the responses to UVB irradiation in vitiligo melanocytes.

[Biological characteristics of different forest soils in Nanjing-Zhenjiang mountan area].

Studies on the biological characteristics, including soil microbe, soil enzyme activity, soil nutritient content, and litter decomposition of different forest soils in Nanjing-Zhenjiang mountain area showed that the amounts of microbes and the activities of six enzymes in forest soils changed regularly in different forests during different months. The contents of nutritional elements varied regularly with forest growth bio-cycles, and were inerrelated prominently with the amounts of soil microbes and the activities of soil enzymes. There existed temporal-spatial differences in the decomposition rate of litters and the reverted velocity of nutrients among different forests. The comparisons of various biological characteristics among secondary Quercus variabilis forest, Phllostachys pubescens forest, and Cunninghamia lanceolata forest indicated that secondary Quercus variabilis forest had the most abundant nutrients in soil, and possessed the strongest ability of self-fertilization. Therefore, to construct coniferous forests with broadleaf trees in this area could avoid or abate the decline of soil fertility.
