(Benzoxazole-2-thiolato-S)(benzoxazole-2-thione-S)bis(triphenylphosph ine)copper(I).

The Cu atom in [Cu(C(7)H(4)NOS)(C(7)H(5)NOS)(C(18)H(15)P)(2)] is tetrahedrally coordinated by a P(2)S(2) donor set. One S atom is derived from neutral benzoxazole-2-thione and the other from the deprotonated form of the ligand. The two sulfur ligands are connected via an N-H.N interaction, leading to the formation of an eight-membered ring.

2-(2,1-benzoxazol-3-yl)-3,5-dimethoxyphenol and 3-phenyl-2,1-benzoxazole.

The title compounds, C(15)H(13)NO(4), (I), and C(13)H(9)NO, (II), are produced, along with the corresponding anilines, by the reduction of the appropriate o-nitrobenzophenones. In (I), the planar benzisoxazole and phenol fragments are tilted relative to one another by a rotation of 53.02 (14) degrees about the bond joining them, and the molecules are linked into chains by phenol O--H...N and phenyl C--H...O(oxazole) hydrogen bonds. The cell of (II) (space group I2/c) contains eight molecules in general positions, four more in the 2b sites, with twofold axial symmetry that induces a degree of disorder, and a further four as centrosymmetric pairs of complete molecules, each with an occupancy of one-half. The relative tilt of the planar fragments varies slightly from one molecule to another but is much less than that in (I), ranging from 8.8 (8) to 12.58 (15) degrees.

3-[4-(4-fluorophenyl)piperazin-1-ylmethyl]-5-methyl-1,3-benzoxazol-2(3H)-one and 3-[4-(2-fluorophenyl)piperazin-1-ylmethyl]-5-methyl-1,3-benzoxazol-2(3H)-one.

The title compounds, both C(19)H(20)FN(3)O(2), contain essentially planar benzoxazolinone ring systems, within which the C-N bond distances and angles do not differ significantly between the two compounds. In both cases, the piperazine ring adopts an almost perfect chair conformation and the benzoxazolinone ring system lies nearly perpendicular to it. The structures contain intermolecular C-H.O contacts, and the interactions between the benzoxazolinone and fluorophenylpiperazine portions of the molecules are segregated.

Synthesis and evaluation of anticancer benzoxazoles and benzimidazoles related to UK-1.

UK-1 is a structurally unique bis(benzoxazole) natural product isolated from a strain of Streptomyces. UK-1 has been reported to possess anticancer activity but no activity against bacteria, yeast, or fungi. Previous work has also demonstrated the ability of UK-1 to bind a variety of di- and tri-valent metal ions, particularly Mg(2+) ions, and to form complexes with double-stranded DNA in the presence of Mg(2+) ions. Here we report the activity of UK-1 against a wide range of human cancer cell lines. UK-1 displays a wide spectrum of potent anticancer activity against leukemia, lymphoma, and certain solid tumor-derived cell lines, with IC(50) values as low as 20 nM, but is inactive against Staphylococcus aureus, a methicillin-resistant strain of S. aureus, or Pseudomonas aeruginosa. A series of analogues of the bis(benzoxazole) natural product UK-1 in which the carbomethoxy-substituted benzoxazole ring of the natural product was modified were prepared and evaluated for their anticancer and antibacterial properties. An analogue of UK-1 in which the carbomethoxy-substituted benzoxazole ring was replaced with a carbomethoxy-substituted benzimidazole ring was inactive against human cancer cell lines and the two strains of S. aureus. In contrast, a simplified analogue in which the carbomethoxy-substituted benzoxazole ring was replaced with a carbomethoxy group was almost as active as UK-1 against the four cancer cell lines examined but lacked activity against S. aureus. Metal ion binding studies of these analogues demonstrate that they both bind Zn(2+) and Ca(2+) ions about as well as UK-1. The non-cytotoxic benzimidazole UK-1 analogue binds Mg(2+) ions 50-fold weaker than UK-1, whereas the simple benzoxazole analogue binds Mg(2+) ions nearly as well as UK-1. These results support a role of Mg(2+) ion binding in the selective cytotoxicity of UK-1 and provide a minimal pharmacophore for the selective cytotoxic activity of the natural product.

Catalytic turnover of substrate benzylamines by the quinone-dependent plasma amine oxidase leads to H2O2-dependent inactivation: evidence for generation of a cofactor-derived benzoxazole.

Incubation of bovine plasma amine oxidase (BPAO) with benzylamine and various p-substituted analogues results in a time-dependent inactivation that is attributable to buildup of the H(2)O(2)-turnover product on the basis of protection afforded by coincubation with catalase. The mechanism of inactivation is distinct from that effected by H(2)O(2) itself, which requires higher concentrations. Solution studies using models for the 2,4,5-trihydroxyphenylalanine quinone (TPQ) cofactor reveal a loss of catalytic activity arising from oxidation of the dihydrobenzoxazole tautomer of the product Schiff base, that competes with hydrolytic release of benzaldehyde product. The resulting stable benzoxazole exhibits a characteristic absorption depending on the nature of the benzylamine p-substituent. For benzylamine itself, the model benzoxazole absorbs at 313 nm, in an area of strong absorption by the enzyme, whereas for 4-nitrobenzylamine, the absorption of the model benzoxazole is sufficiently red-shifted (at 365 nm) to be discerned above the background enzyme absorption. Inactivation of BPAO by 4-nitrobenzylamine is accompanied by loss of the resting TPQ anion absorption at 480 nm concomitant with generation of a new absorption near 360 nm. Resonance Raman spectra of the inactivated enzyme show a close correspondence with those for the model 4-nitrobenzylamine-derived benzoxazole. Substrate-dependent inactivation is also observed for the other two mammalian enzymes examined, equine plasma amine oxidase and human kidney amine oxidase. Catalase provides complete protection in these instances as well. Benzoxazole formation may constitute a common mechanism of inactivation of quinone-dependent amine oxidases by normal substrates in vitro if the product H(2)O(2) is permitted to accumulate. More importantly, the results suggest that the benzoxazole inactivation pathway may be important physiologically and may have influenced the distribution of amine oxidases and catalase in cells.

Synthesis of some novel benzoxazole derivatives as anticancer, anti-HIV-1 and antimicrobial agents.

In an effort to establish new candidates with improved antineoplastic, anti-HIV-1 and antimicrobial activities we report here the synthesis and in vitro biological evaluation of various series of 2-substituted benzoxazoles: 2-[(Arylhydrazono, arylidene, cycloalkylidene and N-substituted thiocarbamoyl)cyanomethyl]-benzoxazoles(2-4 and 7, respectively); 2-[(4- or 5-oxothiazoliden-2-yliden)benzoxazoles (5 and 6) and 2-(4-amino-3-substituted-2-thioxo-2,3-dihydrothiazol-5-yl)benzoxazoles (8), together with the synthesis of some substituted 3H-pyrido[2,1-b]benzoxazoles (9-11). The absolute configuration of compound 3b was determined by X-ray crystallography. The results of the in vitro anticancer screening revealed that some of the tested compounds exhibited broad spectrum antitumor activity. The most active compounds are 2a, 3b, 8a and 8d, their GI50 MG-MID values: 37.7, 19.1, 20.0 and 15.8 microM; TGI MG-MID values: 75.9, 53.7, 53.7, and 58.9 microM; and LC50 MG-MID values: 97.7, 93.3, 89.1 and 93.3 microM, respectively. The in vitro microbiological data showed that compound 7c was the most active against Staphylococcus aureus (minimal inhibitory concentration (MIC)<12.5 microg ml(-1). While compounds 5, 8a, and 8d were the most active against Bacillus subtilis (MIC values<12.5 microg ml(-1)). On the other hand, compounds 5 and 7c were the most active against Escherichia coli (MIC<25 microg ml(-1)), their activity is about half the activity of ampicillin and streptomycin . In addition, compound 4b and 7c were the most active against Pseudomonas aeruginosa (MIC<25, 50 microg ml(-1)). Compound 4b was two times as active as ampicillin and streptomycin while compound 7c was active as both. The results of antimycotic activity indicated that, Compound 7c showed mild activity against Candida albicans when compared with clotrimazole (MIC<100 microg ml(-1)). In vitro HIV-1 testing revealed that compound 7a displayed moderate anti-HIV-1 activity (maximum % cell protection, 36.6 at 2 x 10(-5) microM).

Synthesis and antimicrobial activity of new 2-[p-substituted-benzyl]-5-[substituted-carbonylamino]benzoxazoles.

A series of 23 new 2-[p-substituted-benzyl]-5-[p-substituted-phenyl/benzyl-carbonylamino]benzoxazole derivatives has been synthesized by reacting 5-amino-2-[p-substituted-benzyl]benzoxazoles with the appropriate carboxylic acid chlorides. The structures of the synthesized compounds were confirmed by IR and (1)H-NMR spectral data. Antimicrobial activities of the compounds were investigated using the twofold serial dilution technique against two gram-positive and two gram-negative bacteria and three Candida species in comparison with standard drugs. Microbiological results indicated that the newly synthesized 2-[p-substituted-benzyl]-5-[p-substituted-phenyl/benzyl-carbonylamino]benzoxazole derivatives (3-25) possessed a broad spectrum of activity, showing MIC values of 6.25-200 microg/mL against the gram-positive and gram-negative microorganisms tested. Moreover, they showed significant antifungal activity with MIC values of 3.12-100 microg/mL against the Candida species tested. Especially, with a MIC value of 3.12 microg/mL, 2-benzyl-5-[p-bromobenzyl-carbonylamino]benzoxazole 9 displayed the same activity against C. glabrata as the standard drug myconazol.

Synthesis and antimicrobial activity of some 5-[2-(morpholin-4-yl)acetamido] and/or 5-[2-(4-substituted piperazin-1-yl)acetamido]-2-(p-substituted phenyl)benzoxazoles.

In this study, a series of twelve novel 5-[2-(morpholin-4-yl)acetamido] and/or 5-[2-(4-substituted pip-erazine-1-yl)acetamido]-2-(p-substituted phenyl]benzoxazole derivatives have been synthesized and their structures were confirmed by IR, (1)H NMR, and mass spectral data. These compounds were prepared by reacting 5-(2-chloroacetamido)-2-(4-p-substituted-phenyl)benzoxazoles, which were obtained by using 5-amino-2-[p-substituted-phenyl]benzoxazoles with chloroacetyl chloride, in the presence of morpholine or 1-substituted piperazines. All synthesized compounds 3-14 were tested by using the method of twofold serial dilution technique for in vitro activities against certain strains of Gram-positive, Gram-negative bacteria as well as the yeasts Candida albicans, Candida krusei, and Candida glabrata in comparison with standard drugs. Microbiological results showed that the newly synthesized compounds possessed a broad spectrum of activity, showing MIC values of 3.12-50 mug/mL against the Candida species.

Synthesis and microbiological activity of some N-(2-hydroxy-4-substitutedphenyl)benzamides, phenylacetamides and furamides as the possible metabolites of antimicrobial active benzoxazoles.

The synthesis of some N-(2-hydroxy-4-substitutedphenyl)benzamides, phenylacetamides and furamides as the possible metabolites of benzoxazoles (II1-15) was performed in order to determine their in vitro antimicrobial activity against three Gram-positive bacteria, two Gram-negative bacteria and the fungus Candida albicans and their activities were compared with several control drugs. The compounds II11, II12, and II13 were found active at a MIC value of 12.5 microg/ml against the Gram-negative microorganism Pseudomonas aeruginosa. Most of the compounds show antibacterial activity at MIC a value of 25 microg/ml against the Gram-positive bacteria Staphylococcus aureus. For the antifungal activity against C. albicans, compound II10 was found more active than the other derivatives. The antimicrobial activity of some of these benzamides, phenylacetamides (II1 and II10) which are the possible metabolites of benzoxazoles, was also compared to their corresponding cyclic analogues III-IV. Compound II10 possesses two dilutions better antifungal activity than its cyclic analogue, benzoxazole IV, against C. albicans.

Photophysics and spectroscopic properties of 3-benzoxazol-2-yl-chromen-2-one.

The photophysics of 3-benzoxazol-2-yl-chromen-2-one was studied in different solvents. High molar absorptivities, between 14,800 and 22,900 dm3/mol cm, were observed for the absorption peak related to the S0 --> S1 transition which suggests a pi --> pi* character. This compound presents a limited solvatochromism, attributed to the benzoxazole group, and high fluorescence quantum yields, phi(f). The fluorescence quantum yield is lowered with the increase of solvent polarity, favouring the participation of internal conversion as deactivation path of the S1 state. The Stokes shift shows that the excited state is stabilised with increasing solvent polarity. The dipole moment was estimated by ab initio calculations as being between 5.28 and 5.62 Debye for S1, and 4.75 Debye for S0. Phosphorescence was not observed. A small but not negligible quantum yield of singlet oxygen generation (phidelta = 0.15) was measured in chloroform. The geometric parameters obtained by semi-empirical calculation (PM3) are in good agreement with crystallographic data, showing a r.m.s. deviation of 0.153 A for the superposition of both structures. The predicted structure is all planar, while the crystallographic data reveal a dihedral angle of 6.5 degrees, between the coumarin and benzoxazole rings. The theoretical description of the electronic spectra, obtained from a PM3 CI calculation, shows excellent agreement with the experimental data. Deviations lower than 2% are observed in the predicted absorption maxima, with best results when solvation is considered. For electronic states calculation, ZINDO/S gave a better prediction of excited state energies, with a deviation lower than 7% for the S1 energy. The most probable sequence for the first four excited states is: Ti(n pi*) < T2(pi pi*) < S1(pi pi*) < S2(n pi*).

Benzoxazole derivatives as novel 5-HT3 receptor partial agonists in the gut.

A series of benzoxazoles with a nitrogen-containing heterocyclic substituent at the 2-position was prepared and evaluated for 5-HT3 partial agonist activity on isolated guinea pig ileum. The nature of the substituent at the 5-position of the benzoxazole ring affected the potency for the 5-HT3 receptor, and the 5-chloro derivatives showed increased potency and lowered intrinsic activity. 5-Chloro-7-methyl-2-(4-methyl-1-homopiperazinyl)benzoxazole (6v) exhibited a high binding affinity in the same range as that of the 5-HT3 antagonist granisetron, and its intrinsic activity was 12% of that of 5-HT. Compound 6v inhibited 5-HT-evoked diarrhea but did not prolong the transition time of glass beads in the normal distal colon even at a dose of 100 times the ED50 for diarrhea inhibition in mice. Compounds of this type are expected to be effective for the treatment of irritable bowel syndrome without the side effect of constipation.

Parallel synthesis of a library of benzoxazoles and benzothiazoles using ligand-accelerated copper-catalyzed cyclizations of ortho-halobenzanilides.

A general method for the formation of benzoxazoles via a copper-catalyzed cyclization of ortho-haloanilides is reported. This approach complements the more commonly used strategies for benzoxazole formation which require 2-aminophenols as substrates. The reaction involves an intramolecular C-O cross-coupling of the ortho-haloanilides and is believed to proceed via an oxidative insertion/reductive elimination pathway through a Cu(I)/Cu(III) manifold. The reaction is also applicable to the formation of benzothiazoles. A variety of ligands including 1,10-phenanthroline and N,N'-dimethylethylenediamine were shown to provide ligand acceleration/stabilization in the reaction. Optimal conditions for cyclization used a catalyst combination of CuI and 1,10-phenanthroline (10 mol %). The method was amenable to a parallel-synthesis approach, as demonstrated by the synthesis of a library of benzoxazoles and benzothiazoles substituted at various positions in the ring. Most examples utilized the cyclization of ortho-bromoanilides, but ortho-iodoanilides and ortho-chloroanilides also undergo a reaction under these conditions. The rate of reaction of the ortho-haloanilides follows the order I > Br > Cl, consistent with oxidative addition being the rate-determining step.

Novel benzoxazole 2,4-thiazolidinediones as potent hypoglycemic agents. Synthesis and structure-activity relationships.

A new series of benzoxazole 2,4-thiazolidinediones was synthesized and evaluated for hypoglycemic activity in genetically obese and diabetic yellow KK mice. 2-Arylmethyl- and 2-(heteroarylmethyl)benzoxazole derivatives showed far more potent activity than known 2,4-thiazolidinedione derivatives such as ciglitazone, troglitazone and pioglitazone. A facile synthesis of benzoxazole 2,4-thiazolidinediones was also established using aminophenol 2,4-thiazolidinedione (11) as a key intermediate. Details of synthesis and structure-activity relationships for this series are described.

Kinetics of some benzothiazoles, benzoxazoles, and quinolines as substrates and inhibitors of rabbit liver aldehyde oxidase.

1. Twelve oxygen and sulphur azaheterocycles were studied as potential substrates of rabbit liver aldehyde oxidase. Only benzoxazole and 1,2-benzisoxazole were found to be substrates. 2. Nine of the compounds inhibited the oxidation of quinazoline by aldehyde oxidase and in all cases mixed inhibition kinetics were observed. 3. pi-Excessive heterocycles consisting of a single 5- or 6-membered ring (thiazole, oxazole) were neither substrates or inhibitors. Addition of a carbocyclic ring (benzothiazole, benzoxazole, 1,2-benzisoxazole) allowed binding to the enzyme as a substrate and/or inhibitor. 4. The mixed inhibition exhibited by the pi-excessive azaheterocycles benzothiazole, 1,2-benzisoxazole, and 2-substituted benzoxazoles was characterised by a Ki/KI ratio greater than 1.0, where Ki is the inhibitor constant for binding to the free enzyme and KI is the inhibitor constant for binding to the ES complex. In contrast, five pi-deficient methyl-substituted quinolines, which are known substrates for aldehyde oxidase, exhibited a Ki/KI ratio of less than 1.0. 5. The pi-excessive heterocycles 2,3-benzthiophene and 2,3-benzfuran, which do not contain a nitrogen atom, exhibited weak inhibition with a very high Ki/KI ratio. 6. The results of the study indicated that whilst thiazoles and oxazoles are unlikely to be extensively metabolized by aldehyde oxidase, they may inhibit the metabolism of substrates of the enzyme.

Synthesis and antimicrobial activities of 2-[(alpha-methylbenzylidene)-hydrazino]benzoxazoles.

New 2-[(alpha-methylbenzylidene)hydrazino]benzoxazole derivatives have been synthesized by reacting ortho or para substituted acetophenones with 2-hydrazinobenzoxazole in ethanol. The structures of the synthesized compounds were confirmed by microanalysis, IR and NMR spectral data. Antimicrobial activities of the compounds were investigated by the microdilution susceptibility test in Mueller-Hinton broth and Sabouraud liquid medium. Test organisms: Staphylococcus aureus ATCC 29213 and Enterococcus faecalis ATCC 29212 as Gram (+) bacteria, Escherichia coli ATCC 25922 and Pseudomonas aeruginosa, ATCC 27853 as Gram (-) bacteria, and Candida albicans, Candida stellatoidea, Candida parapsilosis and Candida pseudotropicalis as yeasts. Among the compounds tested 2-[(alpha-methyl-4-chlorobenzylidene)hydrazino]benzoxazole (compounds 4) and 2-[(alpha-methyl-4-nitrobenzylidene)hydrazino]benzoxazole (compound 8) showed the most favorable activity.

Thermal reactions of benzoxazole. Single pulse shock tube experiments and quantum chemical calculations.

The thermal decomposition of benzoxazole diluted in argon was studied behind reflected shock waves in a 2 in. i.d. single-pulse shock tube over the temperature range 1000-1350 K and at overall densities of approximately 3 x 10(-5) mol/cm(3). Two major products, o-hydroxybenzonitrile at high concentration and cyclopentadiene carbonitrile (accompanied by carbon monoxide) at much lower concentration, and four minor fragmentation products resulting from the decomposition were found in the postshock samples. They were, in order of decreasing abundance, benzonitrile, acetylene, HCN, and CH=C-CN and comprised of only a few percent of the overall product distribution. Quantum chemical calculations were carried out to determine the sequence of the unimolecular reactions that led to the formation of o-hydroxybenzonitrile and cyclopentadiene carbonitrile, the major products of the thermal reactions of benzoxazole. A potential energy surface leading directly from benzoxazole to cyclopentadiene carbonitrile could not be found, and it was shown that the latter is formed from the product o-hydroxybenzonitrile. In order that cyclopentadiene carbonitrile be produced, CO elimination and ring contraction from a six- to a five-membered ring must take place. A surface where CO elimination occurs prior to ring contraction was found to have very high barriers compared to the ones where ring contraction occurs prior to CO elimination and was not considered in our discussion. Rates for all the steps on the various surfaces were evaluated, kinetic schemes containing these steps were constructed, and multiwell calculations were performed to evaluate the mole percent of the two major products as a function of temperature. The agreement between the experimental results and these calculations, as shown graphically, is very good.

Synthesis and antimicrobial activity of some 2-[p-substituted-phenyl]benzoxazol-5-yl-arylcarboxyamides.

New 2-[p-substituted-phenyl ]benzoxazol-5-yl-arylcarboxyamides derivatives have been synthesized by reacting 5-amino-2-[p-substituted-phenyl ]benzoxazoles with substituted-arylcarboxylic acid chlorides. The structures of the synthesized compounds were confirmed by IR and (1)H NMR spectral data. Antimicrobial activities of the compounds were investigated using the two-fold serial dilution technique against different Gram-positive and Gram-negative bacteria and the yeast C. albicans in comparison with standard drugs. Microbiological results indicated that the synthesized compounds possess a broad spectrum of activity, having an MIC value of 25-200 microg/mL at molar concentration values of 3.45 x 10(-5) and 5.74 x 10(-4) against the tested microorganisms.
