Suppression of immune responses to sheep red cell antigens in rats preimmunized with heterophilic antigen: cellular events.

Depression of humoral immune response to sheep red cells (SRC) has been observed in rats previously immunized with heterophilic (Forssman) antigen. Analogous phenomena were observed by assay of splenic plaque-forming cells obtained from rats immunized in vivo with heterophilic antigen (HA) and challenged with SRC. Isophilic immune responses were almost completely suppressed. Kinetics of the primary and secondary immune responses were analyzed. Intervals of two to four days between administration of HA and SRC yielded maximal suppression. Primary immune responses to SRC were determined in cultures of splenic cell suspensions derived from rats which had been either injected in vivo with HA or received no treatment. Under these conditions, too, administration of HA interfered with the antibody responses to SRC. Both adherent and nonadherent cells caused inhibition, with this property appearing earlier in adherent than in nonadherent cells. A hypothesis, capable of explaining the observations made in this study and compatible with similar phenomena described in other systems, has been proposed according to which after immunization with HA, SRC are diverted to HA-stimulated cells which are not equipped to deal with the antigen complex represented by SRC.

Identification of terminal N-acetylglucosamine residues of highly branched asparagine-linked oligosaccharides as immunoreactive domains of a chicken heterophile antigenic determinant.

Chicken heterophile antigenic determinant (CHAD-1) has been previously found in medullary lymphocytes of the bursa and thymus as well as in some non-lymphoid cells by the immunoperoxidase method, using rabbit antiserum to a complete Freund's adjuvant (CFA) as the first antibody. In this work we demonstrated that absorption of anti-CFA serum with highly purified preparations of hen egg white glycoproteins (ovomucoid, ovoinhibitor, ovalbumin) or chicken orosomucoid completely blocked immunoperoxidase staining for CHAD-1. Treatment of these glycoproteins with beta-N-acetylglucosaminidase suppressed their capacity to inhibit this staining. Absorption of anti-CFA serum with asparagine-linked glycopeptides which have the mannose alpha 1,3 arm disubstituted by GlcNAc residues and which have another GlcNAc residue linked beta 1,4 to the beta-linked mannose of the core also inhibited staining for CHAD-1. These data indicated that highly branched asparagine-linked oligosaccharides with terminal GlcNAc residues beta-linked to mannose represent immunoreactive domains of CHAD-1.

Hanganutziu-Deicher heterophile antigen in human retinoblastoma cells.

We investigated the possible existence of Hanganutziu-Deicher antigen (HD antigen) on retinoblastoma cells by means of indirect membrane immunofluorescence and a complement-dependent antibody-mediated cytotoxicity test. We prepared antiserum in the chicken by immunization with purified equine hematoside highly specific to HD antigen. Three kinds of retinoblastoma cell lines, Y-79, WERI-Rb 1, and TOTL-1, showed positive membrane staining. Y-79 and TOTL-1 cells were also confirmed by the cytotoxicity test to possess HD antigen. Retinoblastoma cells freshly isolated from two patients had a positive membrane immunofluorescence. These results indicated that HD antigen was expressed in human retinoblastoma cells in vitro as well as in vivo.

Heterophile antigens in serous cystadenocarcinoma of the human ovary.

As the aberrance of carbohydrate moieties, the appearance of heterophile antigens in human ovarian cancer tissue was studied using various heterophile antigens and antibodies together with antisera raised in rabbits by immunization with extracts of human ovarian cancer tissues. In the immunodiffusion test, rabbit antisera reacted with a heat-stable, perchloric-acid extractable antigen that was different from CEA and CA125 and possessed Hanganutziu-Deicher (HD)-type specificity in all 4 specimens of serous cystadenocarcinoma of the ovary. Furthermore, a pathologic serum with strong HD-antibody complex formed a precipitin line with one of them. In the cancer tissue, we also immunochemically demonstrated the presence of rat-erythrocyte antigens. However, other types of heterophile antigens, such as Paul-Bunnell and Forssman, were not detected. These results suggest that HD-type antigens appear in serous cystadenocarcinoma of the human ovary, one in broad distribution and some in specific cases.

Isolation and partial characterization of the heterophile antigen of infectious mononucleosis from bovine erythrocytes.

The heterophile antigen (Paul-Bunnell antigen, PBA) of infectious mononucleosis was isolated by extraction of an aqueous suspension of bovine erythrocyte stromata with chloroform-methanol (2:1). The upper aqueous layer contained gangliosides, PBA, and a high-molecular-weight glycoprotein. PBA and gangliosides were separated from the high-molecular-weight glycoprotein by extraction of lyophilized upper layer with chloroform-methanol solvents. Separation of PBA from gangliosides was carried out by chromatography on DEAE-cellulose with chloroform-methanol solvents. PBA appeared to be a minor glycoprotein component of the erythrocyte membrane and had both hydrophobic and hydrophilic properties. It was soluble in either organic or aqueous solvents. On SDS-polyacrylamide gel electrophoresis, it migrated as a single component that stained for protein with Coomassie blue, for carbohydrate with periodic acid-Schiff reagent, and for lipid with oil red 0; it had an apparent molecular weight of 26,000. It was composed of 62% protein with major amino acids; glutamic acid, proline, glycine, isoleucine, leucine, and threonine (158, 116, 98, 90, 85, and 82 residues per 1,000 residues, respectively). Carbohydrate content was 9.2% with major sugar constituents: sialic acid, galactosamine, and galactose. Serologic activity of PBA was destroyed by pronase but not by trypsin.

A new heterophile antigen: man distinguished from higher apes and other mammals.

Human cell membranes acquired an antigenic substance from the bovine serum in culture medium. The antigen was named the heterologous membrane antigen (HM Ag). Cross-reactive antigen was found in sera of all mammals tested, except for man. The distribution of the HM Ag serologically distinguished man from other mammals.

Heterophile antigen detectable by infectious mononucleosis sera on bone marrow cells.

By means of mixed agglutination test and indirect immunofluorescence test, reactions of infectious mononucleosis sera with bone marrow cells were investigated. Positive reactions were observed with bovine, murine and rat bone marrow cells, but negative results were obtained with cells of monkeys and guinea pigs. Absorption studies demonstrated that the observed reactions are due to interaction of the Paul-Bunnell antigen on bone marrow cells with its corresponding antibodies in infectious mononucleosis sera. Whereas all cells of bovine bone marrow gave positive results, only 50% of murine or rat cells were positive and these cells were shown to be neutrophilic granulocytes. Evidence was also presented that the Paul-Bunnell antigen and Fc receptor are present, apart from each other, on murine neutrophilic granulocytes.

Implications of heterophile antigens in immunological infertility in males.

Immunological study was conducted on serum and hydrocel fluids from males belonging to an equatorial African population heavily contaiminated by Filaria and Schistosoma. Anti-spermatozoa antibodies were detected by agglutination in 31 of 64 sera and 24 of 60 fluids, and by cytotoxicity in 19 of 64 sera and 8 of 60 fluids. Antinuclear antibodies were detected in 21 of 64 sera and 8 of 60 fluids. However, the titers of these antibodies were low, as were the frequency and titers of other tissues antibodies. The appearance of antispermatozoa antibodies is also correlated with extensive skin burns and infection by Pseudomonas aeruginosa or severe Candidiasis.
