Cycloartane triterpene glycosides from the roots of Astragalus brachypterus and Astragalus microcephalus.

Three new cycloartane-type triterpene glycosides, brachyosides A (1), B (3), and C (2), from the roots of Astragalus brachypterus and one new glycoside, cyclocephaloside II (4), from the roots of Astragalusmicrocephalus have been isolated together with five known saponins, astragalosides I, II, and IV, cyclocanthoside E, and cycloastragenol. The structures of the new compounds were established as 3-O-[beta-D-xylopyranosyl(1-->3)-beta-D-xylopyranosyl-6-O-beta-D-gluc opyranosyl-3beta,6alpha,16beta,24(S),25-pentahydrox ycycloartane (1), 3-O-beta-D-xylopyranosyl-6-O-beta-D-glucopyranosyl-24-O-beta-D-glucop yranosyl-3beta,6alpha,16beta,24(S),25-pentahydroxyc ycloartane (2), 20(R),24(S)-epoxy-6-O-beta-D-glucopyranosyl-3beta,6alpha,16beta , 25-tetrahydroxycycloartane (3), and 20(R), 24(S)-epoxy-3-O-(4'-O-acetyl)-beta-D-xylopyranosyl-6-O-beta-D-glucopy ranosyl-3beta,6alpha,16beta,25-tetrahydroxycycloart ane (4). For the structure elucidations, 1D- and 2D-NMR experiments and FABMS were used.

Effects of feeding Oxytropis and Astragalus pollen to mice and Astragalus seeds to rats.

Pollens collected from Astragalus lentiginosus and Oxytropis sericea were fed to mice. At the end of the feeding period, uterine weights were taken to determine estrogenic activity, and tissue was collected for microscopic examination. The pollen from A lentiginosus, but not from O sericea, induced lesions similar to those of locoweed poisoning. The pollen from neither plant was estrogenic. Seeds from A lentiginosus produced toxicosis when fed to rats.

[Clinical study of Astragalus injection plus ligustrazine in protecting myocardial ischemia reperfusion injury].

OBJECTIVE: To investigate the mechanism of Astragalus injection plus ligustrazine in preventing the occurrence of myocardial ischemia reperfusion injury (MIRI) during open heart surgery of cardiopulmonary bypass, and the treating principle of MIRI in TCM. METHODS: Twenty-four patients suffering from either valvular heart diseases or congenital ventricular septal defect were randomly divided into three treated groups (Astragalus, ligustrazine, Astragalus plus ligustrazine) and the control group, 6 in each group. Blood samples were taken via subclavian central vein at the time before anesthesia (T1), 10 minutes of occlusion of aorta (T2), 10 minutes (T3) and 30 minutes (T4) after the release, and the end of operation (about 180 minutes after release, T5) respectively; EKG was observed, and the levels of asparate aminotransferase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), MB isoenzyme of CK (CK-MB), malondialdehyde (MDA) and the activity of superoxide dismutase (SOD), nitric oxide (NO), nitric oxide synthetase (NOS) were determined. RESULTS: The treated groups could reduce the levels of AST, LDH, CK, CK-MB, MDA, SOD compared with the control group, particularly Astragalus plus ligustrazine, there had significant difference (P < 0.05, P < 0.01). In NO activity improvement, Astragalus plus ligustrazine won the best effect, Astragalus group the next. CONCLUSIONS: The mechanism of MIRI is Qi deficiency and blood stasis in TCM, its treating principles should be promoting Qi and removing the blood stasis. According to the authors' study, Astragalus plus ligustrazine could effectively protect against MIRI, which is better than using the 2 medicines separately.

[Characterization and recognition key components in Astragalus membranaceus].

AIM: To separate, identify and quantify multi-components in complex traditional Chinese medicine (TCM) Astragalus membranaceus by unified analysis for systematic study of its relative contents. METHODS: The analytical method was reverse-phase HPLC/photodiode array detector. Based on the comparison of UV spectra, software of UV database to assist recognition has been established. Key components have been found and recognized in two reference Astragalus membranaceus and three samples from different sources. With the unified analytical project, relative peak heights of key components were recognized and contrasted. RESULTS: The water-soluble components can be separated by solvent system A, a gradient program with mobile phase of methanol-water. The liposoluble components can be separated with the solvent system B, a gradient program of mobile phase of 2-propanol-acetonitrile-water. UV detection wavelength was set at 202 nm and the flow rate was 1 mL.min-1 on a Hypersil ODS column. Twenty-nine components in the four Astragalus membranaceus were recognized using characteristic parameters of chromatography and UV spectroscopy. All of the 29 key components in two reference samples and three samples from different sources were shown in their chromatograms to be the same. The concentrations of some the key components in different samples were similar while those of the others were different. The chromatographic peaks of the reference Astragalus membranaceus and two samples were matched with each other. Many different fingerprints were found and differences were shown on their chromatograms. Not only there were differences among three samples from different source, but also there were differences in the two reference samples. CONCLUSION: To find out the key components and recognize them are the basis of quantitative recognition and are very useful when standards are not available. The method of multi-component quantitative recognition in Astragalus membranaceus was developed. The method is convenient and reproducible. It can be widely used for the quality control of the Astragalus membranaceus.

[Adjustment effect of Radix Astragalus and Radix Angelicae sinensis on TNF-alpha and bFGF on renal injury induced by ischemia reperfusion in rabbit].

OBJECTIVE: To study the mechanism of Astragalus membeanaceus and Angelica sinensis inrenal ischemia reperfusion injury of rabbits. METHOD: Thirty-four heathy mature Japanese big-ear rabbits were randomized into control group, IR model group, Astragalus Membeanaceus + IR groupand Danggui + IR group. Renal ischemia lasted for 1 hour and then the renal reperfusion lasted for 48 hours. At the end, kidneys were observed with Electron Microscopy and the level of Cr in serum and the level of TNF-alpha and bFGF in kidneys were measured. RESULT: The changes of kidneys in IR model group were significant, but they were slight in Astragalus Membeanaceus + IR group and Danggui + IR group. The level of Cr in serum and the level of TNF-alpha in kidneys of IR model group were higher and yet the level of bFGF decreased obviously. The level of Cr and TNF-alpha in Astragalus Membeanaceus + IR group and Danggui + IR group was lower than that in IR model group and the level of bFGF was higher than that in IR model group. CONCLUSION: The mechanism of Astragalus Membeanaceus and Danggui in remedying renal IR injury may be relative to their regulation of TNF-alpha, bFGF and other cytokines.

[The effects of astragalus and shenmai injections on macrophage function in burned mice].

OBJECTIVE: To explore the dynamic postburn change in macrophage function in burned mice within 120 hrs after injury, and to investigate the effects of astragalus and shenmai injections on the macrophage function and surrvival rate of burned mice. METHODS: The mice were divided into 13 groups according to postburn time and handling methods, i,e, normal control (A), burn control (B), normal mice with astragalus (NA), normal mice with shenmai (NS), burned mice with astragal (BA), burned mice with shenmai (BS) 2 postburn hour (2 PBH), 6 PBH, 12 PBH, 24 PBH, 48 PBH, 72 PBH, 120 PBH groups. The changes in the various macrophage functions at different postburn time points and after the use of astragalus and shenmai injections were determined by means of phagocytic and RT-PCR methods. RESULTS: (1) Within 120 PBHs, the phagocytic function of murine macrophages decreased evidently. The ACP activity decreased obviously. The expression of IL-15 mRNA fluctuated and that of TNF mRNA enhanced significantly. (2) Five days after the application of astragalus in dose of 2 500 mg . kg(-1) .d(-1), the phagocytic function of macrophages and ACP activity increased markedly (P < 0.01). The expressions of IL-15 and TNF mRNAs were not influenced. The survival rate of mice was not increased. (3) Five days after the application of shenmai injection in dose of 2.5 ml . kg(-1) . d(-1), the phagocytic function of macrophages and ACP activity increased significantly (P < 0.01), while the expression of IL-15 mRNA exhibited no change. But the expression of TNFalpha mRNA decreased obviously (P < 0.01). Moreover, the survival rate of burned mice was evidently raised (P < 0.05). CONCLUSION: Peritoneal administration of shenmai injection at early postburn stage could significantly improve the macrophage function of burned mice, and it increase the survival rate of mice.

[Treatment of leucopenia with pure Astragalus preparation--an analysis of 115 leucopenic cases].

This article reports the effects of pure Astragalus preparation (PAP) in treating 115 cases of leucopenia. These cases were randomized and divided into two groups. Group I (58 cases) was treated by more concentrated PAP (every 10 ml equalled to 15 grams of Astragalus), Group II (57 cases) was treated by less concentrated PAP (every 10 ml equalled to 5 grams of Astragalus). The patients took the PAP twice a day, 10 ml each time. The course of treatment was 8 weeks for both groups. The results showed that effectiveness in Group I was 82.76%, while in Group II 47.37%, with a total effective rate of 65.22%, they were statistically different (P < 0.01). According to the comparison of average WBC counts after treatment, Group I was significantly higher than that of Group II (P < 0.05). There was an obvious rise of the WBC counts in both groups after treatment (P < 0.001). The results were dose-dependent. The author holds that Astragalus is an effective drug in treating leucopenia, and increasing the dosage could enhance its effectiveness.

Characterization of rhizobia isolated from legume species within the genera Astragalus and Lespedeza grown in the Loess Plateau of China and description of Rhizobium loessense sp. nov.

Twenty-nine rhizobial isolates from root nodules of Astragalus and Lespedeza spp. growing in the Loess Plateau of China were characterized by numerical taxonomy, RFLP and sequencing of PCR-amplified 16S rRNA genes, measurement of DNA G+C content, DNA-DNA relatedness and cross-nodulation with selected legume species. Based on the results of numerical taxonomy, the isolates formed two clusters (1 and 2) with some single isolates at a similarity level of 82 %. Cluster 1 contained six isolates from Astragalus and Lespedeza spp. Cluster 2 consisted of nine isolates from Astragalus spp. DNA relatedness was greater than 80 % among isolates within cluster 2. Phylogenetic analysis based on 16S rRNA gene sequences showed that CCBAU 7190B(T), representing cluster 2, was closely related to Rhizobium galegae and Rhizobium huautlense. DNA-DNA relatedness between CCBAU 7190B(T) and reference strains of R. galegae, R. huautlense and other related species ranged from 0 to 48.6 %. The cluster 2 isolates could also be differentiated phenotypically from related species. Based on these data, a novel species, Rhizobium loessense sp. nov., is proposed for cluster 2, with the type strain CCBAU 7190B(T) (=AS1.3401(T)=LMG 21975(T)).

RAPD analysis of Astragalus medicines marketed in Taiwan.

The genetic variability of Astragalus medicine materials sold by twenty randomly selected stores in Taiwan was investigated using RAPD analysis in order to obtain available primers which could clearly differentiate among them. Total DNA isolated from the rhizomes of the samples were used as templates, and sixty 10 mer arbitrary primers were used in the analysis. The aim of the present study is to construct an identification model of molecular biotechniques applicable to Chinese herbal medicines in RAPD analysis. Three of the primers, OPT-03, OPT-13, and OPT-17, revealed polymorphic RAPD fingerprints among the samples of Astragalus membranaceus, and between Astragalus membranaceus and Hedysarum polybotrys samples. SSCP analysis was also conducted on PCR products from the ITS-1 region of ribosomal DNA in order to differentiate the two species.

Complete chloroplast genomes of endemic Astragalus and Oxytropis species from Uzbekistan.

Chloroplast genomes provide important insights into plant phylogeny, genome evolution, and molecular marker development. In this study, we sequenced, assembled, and analyzed the complete chloroplast genomes of two endemic species from Uzbekistan, Astragalus nuratensis and Oxytropis pseudorosea. Genome skimming generated high-quality paired-end reads, enabling the recovery of complete plastomes with mean sequencing depths of 638&#xd7; and 1,725&#xd7;, respectively. The chloroplast genomes were 122,316&#xa0;bp in A. nuratensis and 122,708&#xa0;bp in O. pseudorosea. Both genomes encoded 110 unique genes, including 76 protein-coding genes, 30 transfer RNA genes, and 4 ribosomal RNA genes. Consistent with members of the inverted repeat-lacking clade of Fabaceae, both species lacked the typical inverted repeat regions, resulting in a single-copy genome structure. Phylogenetic analysis based on 119 complete chloroplast genomes resolved major lineages within Astragalus and related genera with strong support. Astragalus nuratensis was placed within the Phaca clade, while Oxytropis pseudorosea formed part of a distinct Oxytropis lineage. These results provide new genomic resources for understanding evolutionary relationships and plastome evolution in Central Asian legumes.

[The effect of herbal medicine including astragalus membranaceus (fisch) bge, codonpsis pilosula and glycyrrhiza uralensis fisch on airway responsiveness].

OBJECTIVE: To study the effect of astragalus membranceus(fisch) bge, codonpsis pilosula and glycyrrhiza uralensis fisch on airway responsiveness. METHOD: FVC, FEV1 and PEF of twenty-eight asthmatic patients were measured before and six weeks after treated with Astragalus membranceus(fisch) bge, codonpsis pilosual and glycyrrhiza uralensis fisch; methacholine challenge test with 0.033 mumol and 1.98 mumol. RESULT: After 28 asthmatic patients received the treatment of those herbs, the results showed that FVC value increased obviously (t = 2.217, P < 0.05) and the value of FEV1 and PEF was all higher compared with before treatment of the herbs (P < 0.01). CONCLUSION: The herbal medicine including astragalus memberanaceus (fisch) bge, codonpsis pilosual and glycyrrhiza uralensis fisch can decrease the airway responsiveness of the patients with asthma.

[Therapeutic effect of Astragalus and Angelica on renal injury induced by ischemia/reperfusion in rats].

OBJECTIVE: To observe the effect of Astragalus and Angelica on acute renal injury. METHODS: Using the ischemia/reperfusion model established by blocking blood flow through clamping of bilateral renal artery for 45 min, the changes of glomerular filtration rate (GFR), renal plasma flow (RPF), fractional excretion of sodium (FENa), urinary volume (UV) and mean arterial pressure (MAP) as well as the morphological change of kidney before and after ischemia/reperfusion were observed. RESULTS: Astragalus and Angelica could promote recovery of RPF and GFR after ischemia/reperfusion, prevent the oliguria or shorten the oliguria period, reduce the increment of FENa and improve the histomorphological injury of kidney. CONCLUSION: Astragalus and Angelica have certain effect in protecting kidney from acute renal injury.

[Clinical study on effect of Astragalus in efficacy enhancing and toxicity reducing of chemotherapy in patients of malignant tumor].

OBJECTIVE: To observe the effect of Astragalus membranaceus efficacy enhancing and toxicity reducing on chemotherapy in patients of malignant tumor. METHODS: One hundred and twenty tumor patients were randomly divided into the treated group and the control group. Both groups were treated with chemotherapy, but to the treated group, Astragalus injection was given additionally by intravenous dripping, 20 ml in 250 ml of normal saline once per day for 21 days as one course and 4 courses were given successively. RESULTS: Compared with the control group, the treated group showed a lower progressive incidence, lesser decrease of peripheral WBC and platelet count (P < 0.05), accompanied with CD8 significantly lowered (P < 0.05), CD4/CD8 ratio significantly increased (P < 0.01), IgG and IgM levels raised (P < 0.05) and Karnofsky scores elevated more than those in the control group. IgA level was unchanged in both groups. CONCLUSION: Astragalus injection supplemented with chemotherapy could inhibit the development of tumor, decrease the toxic-adverse effect of chemotherapy, elevate the immune function of organism and improve the quality of life in patients.

[Experimental study on effect of Astragalus extractum on canine isolated kidney during hypothermia perfusion and preservation].

OBJECTIVE: To investigate the effect of Astragalus Extractum on canine isolated kidney during hypothermia perfusion and preservation. METHODS: Isolated kidneys in the control group were hypothermia perfused and preserved using conventional hypertonic adenine citrate solution (HC-A), and for those in the experimental group, using HC-A plus Astragalus extract instead. The changes of renal tissue construction were observed with light microscopy and electron microscopy. Moreover, the kidney transplantation model of dog was established to determine the changes of biochemical parameters before and after transplantation. Data were analysed synthetically. RESULTS: The ultrastructural injury in preserved kidney of the experimental group was significantly milder than that in the control group. Parameters determined in the early stage of transplantation showed that the blood creatinine level was significantly lower and the endogenous creatinine clearing value was higher in the experimental group than that in the control group, the difference was significant (P < 0.05). CONCLUSION: When Astragalus extractum is used in preserving kidney with hypothermia perfusion, it shows definite protective effect on the ischemic reperfusion injured kidney.

Numerical analysis of Astragalus cicer microsymbionts.

Thirty-seven rhizobium strains, isolated from root nodules of Astragalus cicer (L.) (cicer milkvetch) deriving from different geographic regions, were compared with the representative strains of the known rhizobial species and genera by numerical analysis of phenotypic characteristics. Our results indicated that Astragalus cicer rhizobia were related to the bacteria of Mesorhizobium species and formed two major phena. One phenon, localized on Mesorhizobium loti branch, contained strains from Poland. Another cluster, placed in the vicinity of M. tianshanense, M. mediterraneum, M. ciceri, and M. huakuii, comprised cicer milkvetch nodule isolates from Canada, Ukraine, and one strain from Poland. The relationship of Astragalus cicer microsymbionts to bacteria of the Mesorhizobium species was also supported by phage typing.

The sterols of Astragalus species from Iran: GLC separation and quantification.

This paper presents the content and composition of free sterols in a number of Astragalus species from Iran. Free sterols of roots and seeds were analysed quantitatively and qualitatively by gas-liquid chromatography. Four major 3-beta-hydroxysterols (cholesterol, campesterol, stigmasterol and beta-sitosterol) were identified in roots of 18 species and in seeds of five species of Astragalus. beta-Sitosterol was the main free sterol in both roots and seeds. The cholesterol content in seeds was higher than that in roots. Trace amounts of cholesterol were detected in roots of only four species. beta-Sitosterol accounted for 34.4-69.2 and 76.8-81.8% of the total lipid content in roots and seeds, respectively. The beta-sitosterol content in roots and seeds of Astragalus species varied from 6.3 to 35.7 and 93.3 to 164.5mg/100g dry weight, respectively. Total free sterol content in seeds varied from 112.09 to 204.04mg/100g dry weight, significantly higher than those in roots (7.5 to 43.4mg/100g dry weight). The chemotaxonomic values of free sterols and total lipid profiles of some of the representative species are discussed.
