Antiallergic agent from natural sources. Structures and inhibitory effect of histamine release of naphthopyrone glycosides from seeds of Cassia obtusifolia L.

Two new naphthopyrones, cassiasides B2 (1) and C2 (2), were isolated from the seeds (Cassiae Semen) of Cassia obtusifolia L. The structures of the two new compounds 1 and 2 were established as rubrofusarin 6-O-beta-D- glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl-(1-->3)-O-beta-D- glucopyranosyl-(1-->6)-O-beta-D-glucopyranoside and toralactone 9-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl- (1-->3)-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranoside, respectively, on the basis of spectral and chemical evidence. Compound 2 was found to inhibit the histamine release from rat peritoneal exudate mast cells induced by antigen-antibody reaction.

Phenolic constituents of Cassia seeds and antibacterial effect of some naphthalenes and anthraquinones on methicillin-resistant Staphylococcus aureus.

Thirteen phenolic glycosides including six new compounds were isolated from seeds of Cassia tora (Leguminosae). The structures of the new compounds, rubrofusarin triglucoside (7), nor-rubrofusarin gentiobioside (9), demethylflavasperone gentiobioside (10), torachrysone gentiobioside (11), torachrysone tetraglucoside (12) and torachrysone apioglucoside (13), were elucidated on the basis of spectroscopic and chemical evidence. The effects of the phenolic glycosides, their aglycones and several other compounds structurally related to them on Escherichia coli K12, Pseudomonas aeruginosa PAO1 and some strains of Staphylococcus aureus were then examined. Among them, torachrysone (15), toralactone (16), aloe-emodin (18), rhein (19) and emodin (20) showed noticeable antibacterial effects on four strains of methicillin-resistant Staphylococcus aureus with a minimum inhibitory concentration of 2-64 micrograms/ml. On the other hand, the phenolic compounds tested did not show strong antibacterial effects on E. coli and P. aeruginosa.

Allixin induction and accumulation by light irradiation.

Allixin, a phytoalexin isolated from garlic, was induced by irradiating fresh garlic cloves with sunlight or UV light. Induced allixin was analyzed by HPLC, and the accumulated amounts of allixin were 3.1-6.3 microg/g under experimental conditions.

Inhibitory effects of 1,3-selenazol-4-one derivatives on mushroom tyrosinase.

This study reports depigmenting potency of 1,3-selenazol-4-one derivatives, which would be based upon the finding of direct inhibition to mushroom tyrosinase. 1,3-Selenazol-4-one derivatives exhibited inhibitory effect on dopa oxidase activity of mushroom tyrosinase. In this study, inhibitory effects of six kinds of 1,3-selenazol-4-one derivatives (A, B, C, D, E and F) on mushroom tyrosinase were investigated. Compounds at a concentration of 500 microM exhibited 33.4-62.1% of inhibition on dopa oxidase activity of mushroom tyrosinase. Their inhibitory effects were higher than that of kojic acid (31.7%), a well known tyrosinase inhibitor. 2-(4-Methylphenyl)-1,3-selenazol-4-one (A) exhibited the strongest inhibitory effect among them dose-dependently and in competitive inhibition manner.

Pharmacokinetic study of allixin, a phytoalexin produced by garlic.

The pharmacokinetic behavior of allixin (3-hydroxy-5-methoxy-6-methyl-2-penthyl-4H-pyran-4-one) was investigated in an experimental animal, mice. Allixin was administered using an inclusion compound because the solubility of allixin in aqueous solution is very low. The allixin content in serum and in the organs of administered animals was analyzed by liquid chromatography (LC)-MS. Most of the administered allixin disappeared within 2 h, and the bioavailability of allixin was estimated to be 31% by obtained area under the blood concentration-time curve (AUC). The metabolites of allixin were studied using the metabolic enzyme fraction of liver and liver homogenate. Several new peaks corresponding to allixin metabolites were observed in the HPLC chromatoprofile. The chemical structure of the metabolites was investigated using LC-MS and NMR. Three of them were identified as allixin metabolites having a hydroxylated pentyl group.

Allixin accumulation with long-term storage of garlic.

Extremely high accumulation of allxiin, a phytoalexin derived from garlic, was observed in necrotic tissue areas after long-term storage. The allixin produced recrystallized on the surface of the garlic clove. The amount of allixin produced in raw garlic with necrotic tissue areas was 1400 ng/mg wet garlic, which exceeds the minimum exhibitory concentration of allixin. After approximately 2 years of storage, amount of allixin accumulated reached slightly less than 1% of the dry weight of garlic cloves.

Development of 5-[(3-aminopropyl)phosphinooxy]-2-(hydroxymethyl)-4H-pyran-4-one as a novel whitening agent.

A stable derivative of kojic acid, 5-[(3-aminopropyl)phosphinooxy]-2-(hydroxymethyl)-4H-pyran-4-one (Kojyl-APPA), was synthesized in good yield. The effects of Kojyl-APPA on tyrosinase activity and melanin synthesis were investigated. Kojyl-APPA showed tyrosinase inhibition effect (30%) in situ, but not in vitro. Kojyl-APPA inhibited tyrosinase activity significantly at 24 h after treatment in normal human melanocytes. It means that Kojyl-APPA is not a direct inhibitor of tyrosinase itself, but it is converted to a potential inhibitor kojic acid enzymatically in cells. In addition, Kojyl-APPA decreased melanin content to 75% of control in melanoma cells and decreased neomelanin synthesis to 43% of control in normal human melanocytes. Its permeation through skin increased by about 8 times as compared with kojic acid.

A new naphthopyrone from the root of Pleuropterus ciliinervis.

A new naphthopyrone, pleuropyrone A (1), together with (-)-lyoniresinol 3a-O-beta-D-glucopyranoside (2) and (+)-lyoniresinol 3a-O-beta-D-glucopyranoside (3) was isolated from the roots of Pleuropterus ciliinervis. The structure of pleuropyrone A (1) was determined to be 2,5-dimethyl-8-hydroxynaphthopyrone 10-O-beta-D-glucopyranoside by spectroscopic methods including 2D-NMR. Compounds 2 and 3 showed moderate antioxidant activity.

Microbial metabolism. Part 5. Dihydrokawain.

Preparative scale fermentation of (6S)-dihydrokawain (1) with Rhizopus arrhizus (ATCC 11145) gave 3'-hydroxydihydrokawain (2) and (8S)-hydroxydihydrokawain (3). Structure elucidation of the metabolites was based on spectroscopic data. The C-8 absolute configuration of (3) was assessed via its Mosher's esters.

Construction of tricyclic enone, a common precursor for aphidicolane and stemodane B/C/D-ring system.

Synthesis of a tricyclic enone (B/C/D ring system), a common key precursor for the aphidicolane- and stemodane-type diterpene, is described. The key reaction for the construction of the quaternary carbon center is allylation of epoxide at the more substituted carbon with an organotitanium reagent. Asymmetric reduction with DIP-Cl followed by stereoselective cyclization of spirocyclic ketone and the functional group modification gave the desired tricyclic enone in good yield.

Ca2+-independent phospholipase A2 inhibitor impairs spatial memory of mice.

Pharmacological blockade of Ca2+-independent phospholipase A2 (PLA2) is reported to disintegrate hippocampal synaptic plasticity, which is thought to be the cellular mechanism underlying learning and memory. Therefore, we investigated the effect of the Ca2+-independent PLA2 inhibitor bromoenol lactone (BEL) on spontaneous alteration behaviors of mice. When 3 nmol BEL was intracerebroventricularly injected 30 min prior to the test, the mice showed a poor alternation ratio, compared with control animals. The data suggest that Ca2+-independent PLA2 activity is required for spatial memory.
