Skin reactivity and antibody response following vaccination against smallpox.

Revaccination between 2 and 5 years after the last vaccination induced higher complement-fixation titres than revaccination 7-63 years after the last vaccination. The highest CF titres were reached during the 3rd to the 5th week after vaccination.Fifty-two serum samples, taken 137 or more days after a successful vaccination, all showed CF titres lower than 1/2.In a group of successfully vaccinated persons with no post-vaccinal complications, 65 out of 66 had a successful ;take' after one or two subsequent vaccinations.Sera of persons receiving primary vaccination showed significantly lower CF titres than sera of revaccinated persons. Cases of post-vaccinal encephalitis showed greater antibody response than uncomplicated successfully vaccinated cases in both the primary vaccination and revaccination groups. The antibody response in revaccinated persons with post-vaccinal encephalitis was greater than that in the group of successfully revaccinated cases without encephalitis.A group of 26 naturally hyporeactive persons completely failed to ;take' even after vaccination repeated between 3 and 10 times (average 5 times per person). The frequency of seropositives in this group, and the height of their CF titres, were significantly lower than in the group of successfully vaccinated persons. These findings support the view that untreated persons who repeatedly fail to ;take' with the vaccine must not be considered immune. In vaccinated persons treated with immunoglobulin (with or without simultaneous treatment with Marboran) antibody production was apparently diminished. Treatment of patients with Marboran significantly lowers the capacity of the vaccine to ;take'.

Microimmiscibility and three-dimensional dynamic structures of phosphatidylcholine-cholesterol membranes: translational diffusion of a copper complex in the membrane.

Saturated and unsaturated phosphatidylcholine (PC)-cholesterol membranes have been studied, with a special attention paid to fluid-phase immiscibility in cis-unsaturated phosphatidylcholine (PC)-cholesterol membranes as previously proposed and to the three-dimensional structure of the membrane. The investigation was carried out with dual probes: a membrane-soluble, square-planar copper complex, (3-ethoxy-2-oxobutyraldehyde bis(N4,N4-dimethylthiosemicarbazonato]copper(II) (CuKTSM2), and one of several nitroxide radical lipid-type spin-labels. Bimolecular collision rates between metal ion and spin-label were determined by measuring the nitroxide spin-lattice relaxation times (T1's) in the presence and absence of CuKTSM2 by use of saturation-recovery ESR techniques, and from these measured rates, translational diffusion coefficients of CuKTSM2 were estimated. Profiles of the collision rate across the membrane bilayer were obtained with Tempocholine phosphatidic acid ester, 5-doxylstearic acid, 16-doxylstearic acid, and cholesterol-type spin-labels as a function of cholesterol mole fraction, length and unsaturation of acyl chains, and temperature. In the liquid-crystalline phase of saturated PC membranes, incorporation of cholesterol decreases the collision rate at all depths in the membrane, and the effect of cholesterol is smallest in the middle of the bilayer. In trans-unsaturated PC membranes, a cholesterol-induced decrease of the collision rate was also observed, except in the head-group regions. In cis-unsaturated PC membranes, virtually no effect of cholesterol was observed on the collision rate, either with phospholipid-type spin-labels or with cholesterol-type spin-labels. This result is in clear contrast with our previous observation, in which the effect of cholesterol in cis-unsaturated PC membranes is small on the alkyl-chain motion of phospholipid-type spin-labels but large on the wobbling rotational diffusion of cholesterol-type spin-labels [Pasenkiewicz-Gierula, M., Subczynski, W. K., & Kusumi, A. (1990) Biochemistry 29, 4059-4069]. A model is proposed to explain these results in which the fluid-phase immiscibility is prevalent in cis-unsaturated PC-cholesterol membranes, but where cholesterol-rich (cholesterol oligomeric) domains are small (several lipids) and/or of short lifetime (10(-9) s to less than 10(-7) s). It is suggested that this microimmiscibility arises from the structural nonconformability between the rigid cholesterol ring structure and the rigid bend at the cis double bonds in PC alkyl chains.(ABSTRACT TRUNCATED AT 400 WORDS)

Conversion of azomethine moiety to carboxamido group at cobalt(III) center in model complexes of Co-containing nitrile hydratase.

The Co(III) complex of the Schiff base ligand N-2-mercaptophenyl-2'-pyridylmethyl-enimine (PyASH), namely, [Co(PyAS)(2)]Cl (1), has been synthesized via an improved method and its structure has been determined by X-ray crystallography. The two deprotonated ligands are arranged in mer configuration around the Co(III) center and the overall coordination geometry is octahedral. The coordinated azomethine function in 1 is rapidly converted into carboxamido group upon reaction with OH(-). The product is the bis carboxamido complex (Et(4)N)[Co(PyPepS)(2)] (2), reported by us previously. Reaction of H(2)O(2) with 1 in DMF affords [Co(PyASO(2))(PyPepSO(2))] (3), a species with mixed imine and carboxamido-N donor centers as well as S-bound sulfinates. Further reaction with H(2)O(2) in the presence of NaClO(4) converts 3 into the previously reported bis carboxamido/sulfinato complex Na[Co(PyPepSO(2))(2)] (4). The reaction conditions for the various transformation reactions for complexes 1-4 and the structure of 3 are also reported. The mechanism of the -CH=NR + [O] --> -C(=O)NHR transformation has been discussed. The reactions reported here provide convenient alternate routes for the syntheses of Co(III) complexes with coordinated carboxamide, thiolate, and/or sulfinate donors as models for the Co-site in the Co-containing nitrile hydratase(s).

Separation of semiconducting from metallic carbon nanotubes by selective functionalization with azomethine ylides.

A mild and efficient method for the functionalization of SWNTs by cycloaddition of azomethine ylides derived from trialkylamine-N-oxides is described. Selective reaction of semiconducting carbon nanotubes was achieved by preorganizing the starting N-oxides on the nanotube surface prior to generating the reactive ylides. Separation of met-SWNTs from functionalized sem-SWNTs was successfully accomplished by inducing solubilization of sem-SWNTs in the presence of lignoceric acid.

Organophosphorus derivatives containing piperazine dithiosemicarbazones as chemotherapeutants against fungal pathogens of sugarcane.

Five novel organophosphorus derivatives have been synthesized by the reactions of O,O-diethylchlorophosphate with piperazine dithiosemicarbazones. The derivatives have been characterized on the basis of analyses and spectral (IR, 1H NMR) data. Fungicidal activities of these derivatives against Colletotrichum falcatum, Fusarium oxysporum, and Curvularia pallescence have been evaluated. The screening results have been correlated with the structural features of the tested compounds. Organophosphorus derivatives containing 1,4-bis(4-chlorobenzaldehyde)piperazine dithiosemicarbazone and 1,4-bis(4-methoxybenzaldehyde)piperazine dithiosemicarbazone proved to be more active than some prevalent commercial synthetic fungicides.
