Primary structure of a precursor for the delta-subunit of sweet potato mitochondrial F1-ATPase deduced from full length cDNA.

A cDNA library constructed from poly(A)-rich RNA of the sweet potato tuberous root using a newly developed plasmid vector carrying tac-SP6 promoters was used to identify full length cDNAs for the nuclear-encoded delta-subunit of mitochondrial F1-ATPase by oligonucleotide-hybridization selection. Selected clones contained cDNA insert which carry the entire coding capacity for the pre-delta-subunit, since the RNA transcribed in vitro from SP6 promoter on the vector directed the synthesis of pre-delta-subunit polypeptide in a wheat germ in vitro translation assay. The nucleotide sequence of one of these cDNAs indicates that it can code for the pre-delta-subunit of 244 amino acids of which 199 amino acids encode the mature subunit. The amino acid sequence of the mature delta-subunit shows similarities of about 18-25% amino acid positional identity with the delta-subunits of bacterial F1-ATPases, about 26% with the delta-subunit of chloroplast CF1-ATPase, and about 32-37% with oligomycin sensitivity conferring proteins of animal and fungal mitochondria. The N-terminal presequence of the precursor composed of maximum of 45 amino acids does not show any obvious sequence homology with either the transit peptide of the nuclear-encoded pre-delta-subunit of chloroplast CF1 or the presequence of the nuclear-encoded pre-oligomycin sensitivity conferring proteins. At least two types of the delta-subunit cDNAs with very similar structures were identified from the library, and the presence of multiple copies of the delta-subunit gene in the hexaploid genome of the sweet potato is also suggested by genomic Southern blot hybridization.

[Functional properties of the pOV13 plasmid as a vector for DNA cloning in a broad spectrum of gram negative bacteria].

The birepliconed plasmid pOV13 possesses all the properties of a vector for DNA cloning in a broad host range of bacterial cells. pOV13 is transfered by transformation and stably inherited by Escherichia coli, Brucella, Pseudomonas cells determining the resistance to streptomycin, tetrocycline and kanamycin in these bacteria. The plasmid pOV13 is a multicopy plasmid optimal in replication capacity (23kb). The plasmid carries single sites for some restriction endonucleases that are used for DNA cloning, including some restriction sites in antibiotic resistance genes. The examples of DNA cloning with the selection of recombinant clones by the insertional inactivation of kanamycin or tetracycline resistance and expression of the cloned DNAs are presented.

[Adaptation of pathogens of purulent infections to surface-active antiseptics].

A comparative study was carried out of adaptative capacities of microorganisms to antiseptic agents from the group of cationic surface-active substances (SAS) and antibiotics. It was established that antiseptic agents from SAS group may induce the appearance of resistant to them populations of microorganisms. The resistance acquired to one of the surface-active antiseptics spreads to other agents of this group. At the same time, cationic SAS reduce the frequency and rate of appearance of antibiotico-resistant populations of bacteria during combined use with antibiotics.

[Disability in the population of the elderly: results of a field study].

The concept of disability has gained importance in geriatric research and practice. There is now insight into the impairing function not only of illness but also of its consequences for an autonomous and satisfactory way of life. The aim of the study reported here was to elucidate the relationships of old age disabilities and other potentially important factors such as physical and psychiatric impairments as well as demographic characteristics. A representative sample of old people in an urban community (Mannheim, F.R.G.) was the target population of this study. The results demonstrate--above all--a significant correlation between reduction in motor skill capacity and disability in carrying out activities of daily living. Psychiatric impairment--in particular organic psychiatric disorders--play a role as well. Old people with both physical and psychiatric impairments are the most disabled of the sample and must be considered a high-risk group for the loss of independent living.

[Adaptive reactions of the thyroid system in patients with ischemic heart disease to graded physical exertion].

Data reflecting thyroid response to rationed exercise (bicycle ergometry) in coronary patients are presented with respect to physical stress tolerance and clinical form of coronary disease. The results were compared with those obtained in healthy untrained male subjects. Exercise was carried on until submaximum capacity was reached, or clinical and/or electrocardiographic signs of coronary insufficiency appeared. Radioimmune assays of blood thyrotropin and total triiodothyronine (T3) and thyroxin (T4) were made in the course of exercise. Patients with better functional capacity were shown to develop the relatively-low T3 syndrome. A direct correlation was demonstrated between the T4/T3 index and the threshold effort. Patients with low stress tolerance showed increased T3 secretion and depressed T4/T3 at the peak of exercise.

[The potassium metabolism of typical northern Algerian soils. I. Global evaluation of the potassium supply of soils].

In samples from 87 and 39 profiles, respectively, of typical Northern Algerian soils the following parameters were determined of the K content: total K, exchangeable and water soluble K, K saturation degree, and potassium replenishment capacity. The latter was carried out by four methods (N HCl 50 degrees C, HNO3 boiling, 0.05 and 0.2 N NaTPB). The results obtained were used to check the widespread view that Algerian soils are well supplied with K. The following grouping is suggested from the results of the investigation: --soils with high to very high values for all forms of K (50% of the samples), --soils with lower and medium values for exchangeable K, which require K fertilization actually or in the near future if K uptake by crops increases (30 to 35% of the samples), --soils with TPB values indicating a high binding strength of Ki in which a very limited replenishment power can be expected despite partly high or very high values of acid soluble K (more than 25% of the samples). These results indicate that it is necessary to differentiate the assessment of the K content of Algerian soils.

[Antigenic determinant study of the hemagglutinin in the composition of influenza viruses A subtype H1N1 by an indirect solid-phase radioimmunological analytical method].

A comparative study of antigenic determinants of influenza A virus, subtype H1N1, hemagglutinins isolated in 1947-1953 and in 1977-1979 was carried out. The competitive capacity of 8 virus strains for antibody of 4 hyperimmune antisera to A/Fm/1/47, A/Moscow/Pan/52, A/USSR/090/77, and A/Brazil/11/78 viruses was studied by solid-phase radioimmunoassay (SPRIA) in two modifications: in the homologous and heterologous systems. The analysis of competition in the heterologous system permits one to study isolated subpopulations of antibody homologous to viruses fixed on the solid phase. The combination of both modifications allows investigations of a wider spectrum of antibodies than those possible in the homologous system alone. The results of the study showed the antigenic determinants of hemagglutinin exposed on the surface of particles of different influenza A virus strains to differ in their capacity to stimulate antibody synthesis. The analysis of the time course of changes in the antigenic properties of hemagglutinin regions of similar structure in various viral strains may facilitate prognosing of evolution of virus strains of the same subtype.

[Immunochemical method of determining the biosynthesis of hypophyseal hormones. Effect of liberins and cyclic AMP on the biosynthesis of luteinizing hormone, prolactin and growth hormone in the rat pituitary].

A simple procedure is developed, which enables to estimate sequentially the biosynthesis of luteinizing hormone (LH), prolactin (LTH) and somatotropic hormone (STH) in rat hypophysis using solid-phase immunosorbents prepared on the basis of antisera to the hormones studied, copolymerized with chlorcarbonic acid isobutyl ester. The immunosorbents exhibited high specificity and pronounced immunosorption capacity. The analysis was carried out as follows: incubation of hypophyses with 14C-lysine in vitro, chromatography of the hypophysis extracts on columns with Sephadex G-25 and immunosorption of hormones. Within 4 hrs of incubation the biosynthetic processes in hypophysis were distinctly increased, reaching maximal values between 4 hrs and 6 hrs of incubation. Biosynthesis of LH and LTH in adult females exceeded 2-fold the STH biosynthesis and in 23 days old female rats the STH biosynthesis was more than 1.5- and 3-times higher than that LH and LTH synthesis. Luliberin at a dose 1.5 X 10(-6) M/hypophysis stimulated biosynthesis of LH, inhibited LTH biosynthesis but did not affect the STH synthesis in vitro. Thyroliberin (3.0 X 10(-6) M/hypophysis) stimulated LTH and STH biosynthesis and did not affect the LH synthesis. Dibutyryl-cAMP (0.5 X 10(-3) M/hypophysis) stimulated biosynthesis of all these hormones.

Cyanide-resistant respiration in Streptomyces citreofluorescens.

The capacity of Streptomycetes to carry out cyanide-resistant respiration was investigated. With the model strain, Streptomyces citreofluorescens, it was shown that deprivation of glucose followed by transition from exponential to stationary growth was coupled with declining sensitivity of cellular respiration to cyanide ions. Cyanide-resistant oxidase is located within the cytoplasmic membrane. The occurrence of the cyanide-resistant oxidase did not correspond to qualitative changes of cytochrome spectrum. Cytochrome d is involved neither in cyanide-sensitive nor in cyanide-resistant respiratory chain.

Blood doping and related issues: a brief review.

The intent of blood doping is to increase maximal aerobic power by increasing the capacity of blood to carry oxygen. This manipulation gained notoriety in the sports world because of rumors of blood doping by competitors in endurance events. Researchers also have become interested in induced erythrocythemia because its study provides insight into the limiting factor(s) of maximal aerobic power (Vo2max). It is concluded in this review that to increase Vo2max, it is necessary to elevate significantly the hemoglobin concentration by infusing at least 900 ml of blood. The use of inadequate reinfusion volumes, premature reinfusion of the blood following withdrawal, and storage of the blood by refrigeration rather than by freezing are major reasons for the contradictory findings from earlier studies of blood doping. Changes in blood volume and 2,3-diphosphoglycerate following blood doping are transient and, other than during the first 24 h post infusion, appear to be inconsequential. In addition, this review also examines related issues such as attendant hemodynamic and lactate changes, the need of controls, and ethical considerations in blood doping.

[Preliminary results of otoacoustic emission measurement and standard values in young subjects with normal hearing. Part II. Measurement of effects of acoustic distortion].

The paper discusses the results of DPOAE measurement in 44 young subjects with normal hearing capacity. The measurements were carried out separately for each ear (88 measurements altogether). Otoacoustic responses to continuous two-tone stimulation (f1 and f2) with an intensity range of 46-71 dB SPL were recorded. Mean DPOAE values from 2f1-f2 measurement for the right ear in males and females varied from 2.6 +/- 5.6 to1 5.4 +/- 6.9 (within the range 0.7-6.0 kHz). For the left ear the respective values were 3.9 +/- 4.7 to 19.1 +/- 4.0. In most of subjects an increase in DPOAE values for frequencies higher than 3 kHz was found, which is characteristic of middle ear emission. The statistical analysis of DPOAE measurements in males and females did not reveal significant differences except for a few parameters. In both sexes the intensity of f1 and f2 stimuli at the frequency of 1.5 kHz was found to be significantly different at p = 0.48. The results of DPOAE measurements can be regarded as a statistical probability of standard. In view of the lack of approved calibration standards, the data obtained from the study will be considered a local set of standard values and will be used for clinical interpretation of pathological cases.

[Comparative study of the pathogenicity and antigenicity of 6 Paracoccidioides brasiliensis strains].

A comparative study of antigenicity and pathogenicity for rats of six Paracoccidioides brasiliensis strains was carried out. The antigenic capacity "in vitro" of cytoplasmic extract from each strain was determined by immunodiffusion test against 6 serum samples obtained from rats experimentally infected with Paracoccidioides brasiliensis, that had presented positive reactions with a metabolic control antigen. The cytoplasmic extracts were used at final concentration of 100 mg/ml. All of them showed 2 or 3 precipitation bands in this assay. One hundred twenty Wistar rats both sexes weighing approximately 200 g, were inoculated intracardiacally with suspensions of the yeast phase of different P. brasiliensis strains. Two concentrations containing 3 x 10(7) and 5 x 10(7) cells/ml of each isolate were prepared. The inoculated animals were divided in two groups, one was left to its spontaneous outcome and the percentages of deaths were registered and the other rats were sacrificed at 14, 28, 56 and 70 days post-infection. The following parameters were taken into account for evaluation: A) presence of macroscopic granulomas in lung, liver, spleen and kidney; B) presence of P. brasiliensis in microscopic exams of the same organs, in wet preparations and in histologic sections stained by H&E; C) culture of lung and D) immunodiffusion test using pre-mortem serum samples and the homologous antigen. The correlation between the most important parameters studied in each strain are summarized as follow: As no significant differences between the two inocula employed for each strain was observed, the before mention results are the average of those obtained with each inoculation doses.(ABSTRACT TRUNCATED AT 250 WORDS)

[A comparative evaluation of an immunoenzyme analytical method using a fluorogenic or chromogenic substrate in determining the Marburg virus antigen].

A comparative evaluation of the developed variants of indirect solid-phase enzyme immunoassay using fluorogenic and chromogenic substrates for the determination of Marburg virus antigen was carried out. The resolving capacity of this method was 3.8 x 10(-9) g of protein for the former and 3.1 x 10(-8) g of protein for the latter substrate. Cross titrations demonstrated the lack of common antigens with Ebola virus.

The correlates of fertilizing capacity in normal fertile men.

An investigation has been carried out in normal fertile men on the correlates of fertilizing capacity as defined by the zona-free hamster egg penetration test. The results demonstrate that the apparent fertility of these men is compatible with a wide range of intrinsic sperm quality as reflected by penetration rates ranging from 14% to 90% and differences in the minimum concentration of motile spermatozoa required to initiate penetration. These differences in sperm function could not be correlated with any of the conventional parameters of semen analysis or any of a large number of sperm movement characteristics analyzed by time-exposure photomicrography, with two exceptions: (1) the percentage of progressively (greater than 25 micron/sec) motile spermatozoa exhibiting an amplitude of lateral head displacement (Ah) of less than 10 micron, which was positively correlated with penetrating capacity (P less than 0.01), and (2) the percentage of progressively (less than 25 micron/sec) motile spermatozoa exhibiting an Ah of greater than 10 micron, which was negatively correlated with penetration rate (P less than 0.05). The information obtained in this study should provide a useful basis against which to compare the properties of spermatozoa in cases of suspected infertility.

Influence of increased anatomical dead space no functional residual capacity of lungs and the lung clearance index.

A study of the influence of added anatomical dead space on the functional residual capacity of the lung was carried out on a total number of 40 healthy subjects of ages ranging between 21 and 50 years. The functional residual capacity was found to decrease as additional dead space was introduced in the breathing circuit. This decrease was associated with an increase in tidal volume, lung functions, as given by the lung clearance index, were found to deteriorate with each addition of dead space. There was also an increase in pulse rate in all age groups.

A genome scan for Loci associated with aerobic running capacity in rats.

Aerobic capacity is a complex trait that defines the efficiency to use atmospheric oxygen as an electron acceptor in energy transfer. Copenhagen (COP) and DA inbred rat strains show a wide difference in a test for aerobic treadmill running and serve as contrasting genetic models for aerobic capacity. A genome scan was carried out on an F(2)(COP x DA) segregating population (n=224) to detect quantitative trait loci (QTLs) associated with aerobic running capacity. Linkage analysis revealed a significant QTL on chromosome 16 (lod score, 4.0). A suggestive linkage was found near the p-terminus of chromosome 3 (lod score, 2.2) with evidence of an interaction with another QTL on chromosome 16 (lod score, 2.9). All three QTLs showed a dominant mode of inheritance in which the presence of at least one DA allele was associated with a greater distance run. These results represent the first aerobic capacity QTLs identified in genetic models.

Concurrent performance of motor tasks and processing capacity in patients with schizophrenia.

Any task is carried out more successfully if we allocate undivided attention to it, but as demands on attentional capacity increase-for example, in concurrent or dual task conditions-performance on attended tasks becomes more impaired. Patients with schizophrenia show impaired performance on tasks requiring high levels of attentional capacity. This study examines performance of 11 patients with schizophrenia and 13 normal controls on two motor tasks (placing pegs in a pegboard and repetitive index finger tapping) under unimanual, bimanual, and dual task conditions. The patients with schizophrenia placed fewer pegs and had reduced tapping speed in unimanual and bimanual conditions. However, the decrement in bimanual performance as a percentage of unimanual performance was not significantly different for the patients and controls on either the pegboard or tapping tasks. By contrast, under dual task conditions, the performance of the patients with schizophrenia in peg placement actually improved relative to the unimanual pegboard task, whereas tapping performance deteriorated compared with the unimanual tapping, a decrement that was significantly greater for the patients. Thus the improvement in the visually guided pegboard task was at the expense of the repetitive tapping task. These results are discussed in terms of an impairment of self initiated movement with general sparing of externally triggered movements in schizophrenia and the role of frontostriatal loops in this process.
