Procyanidin B-2, extracted from apples, promotes hair growth: a laboratory study.

BACKGROUND: We have previously reported that several selective protein kinase C (PKC) inhibitors, including procyanidin B-2, promote hair epithelial cell growth and stimulate anagen induction. OBJECTIVES: We discuss the hypothesis that the hair-growing activity of procyanidin B-2 is related to its downregulation or inhibition of translocation of PKC isozymes in hair epithelial cells. METHODS: We examined the effect of procyanidin B-2 on the expression of PKC isozymes in cultured murine hair epithelial cells as well as PKC isozyme localization in murine dorsal skin at different stages in the hair cycle. RESULTS: We observed that procyanidin B-2 reduces the expression of PKC-alpha, -betaI, -betaII and -eta in cultured murine hair epithelial cells and also inhibits the translocation of these isozymes to the particulate fraction of hair epithelial cells. Our immunohistochemical analyses demonstrated that PKC-alpha, -betaI, -betaII and -eta are specifically expressed in the outer root sheaths of both anagen and telogen hair follicles. The hair matrix at the anagen stage showed no positive staining for these PKC isozymes. Moderate to intense staining for PKC-betaI and -betaII in the epidermis and hair follicles was observed in a telogen-specific manner; however, expression of PKC-alpha and -eta during the telogen stage was not conspicuous. Gö 6976, an inhibitor of calcium-dependent (conventional) PKC, proved to promote hair epithelial cell growth. CONCLUSIONS: These results suggest that PKC isozymes, especially PKC-betaI and -betaII, play an important role in hair cycle progression and that the hair-growing mechanisms of procyanidin B-2 are at least partially related to its downregulation of PKC isozymes or its inhibition of translocation of PKC isozymes to the particulate fraction of hair epithelial cells.

Counting apples with oranges: a limitation of the discrimination index.

OBJECTIVES: Three consecutive diets of the Professional and Linguistic Assessment Board's multiple choice question examination in Medicine were analysed in two different ways, taking into account the responses of the candidates to the three separate specialities within medicine. DESIGN: First, discrimination indices were obtained from calculations involving all 60 questions, each with five completions. Then, the 60 questions were analysed separately in the three constituent disciplines of Surgery (15 questions), Obstetrics and Gynaecology (12 questions) and Medicine (33 questions). SETTING: Professional and Linguistic Assessment Board. RESULTS: By the first analysis, performed by considering the whole paper to be a single entity, discrimination indices for medicine were greater than for the other two disciplines: one candidate obtained a random score, i.e. one that was no better than could have arisen, with 95% confidence, by chance as predicted by the hypergeometric distribution, and reliability coefficients were high (0.87). In the second analysis, the paper was considered as three separate components and discrimination indices calculated for each discipline separately. Discrimination indices were similar in all three disciplines, there were 26 random scores, and reliability coefficients were reduced-moderately in Medicine, considerably in the other two disciplines. CONCLUSIONS: These findings emphasize the importance of defining the territory of the subject and ensuring that all the items are drawn from that territory, as well as of using a sufficiently large number of items.

An apple is more than just a fruit: cross-classification in children's concepts.

This research explored children's use of multiple forms of conceptual organization. Experiments 1 and 2 examined script (e.g., breakfast foods), taxonomic (e.g., fruits), and evaluative (e.g., junk foods) categories. The results showed that 4- and 7-year-olds categorized foods into all 3 categories, and 3-year-olds used both taxonomic and script categories. Experiment 3 found that 4- and 7-year-olds can cross-classify items, that is, classify a single food into both taxonomic and script categories. Experiments 4 and 5 showed that 7-year-olds and to some degree 4-year-olds can selectively use categories to make inductive inferences about foods. The results reveal that children do not rely solely on one form of categorization but are flexible in the types of categories they form and use.

Permeability barrier in the mantle epithelium lining the testis in the apple-snail Pomacea canaliculata (Gastropoda: Ampullariidae).

Intercellular junctions are studied in the epithelium lining the testis of the freshwater snail Pomacea canaliculata by conventional staining and lanthanum tracer techniques. The junctional complex consists of belt desmosomes and septate junctions. Septate junctions are of the pleated-sheet type and they are constantly associated with mitochondria. Gap and tight junctions appear to be absent. These septate junctions seem to be the structural correlate of an epithelial permeability barrier that separate the testis from the extrapallial space where the shell elements are deposited. These junctions may contribute to a functional barrier in the male gonad of Pomacea canaliculata. The results indicate that freshwater prosobranchs have junctional structures very close to those found in other molluscs.

Gene expression profiles of different breast cancer cells compared with their responsiveness to fermented mistletoe (Viscum album L.) extracts Iscador from oak (Quercus), pine (Pinus), white fir (Abies) and apple tree (Malus) in vitro.

Cytotoxicity assays in vitro (MTT test) showed that the different breast cancer cell lines Kpl-1, MCF-7 and Mfm-223 respond differently to the mistletoe (Viscum album L.) preparations Iscador. Quercus (Qu), Abies (A), Malus (M) and Pinus (P). In order to determine the differences in the responsiveness of the cells more exactly, the gene expression profiles were determined by cells, which were treated with Mistletoe extracts, compared with untreated control cells. Such differences can be analysed in more detail by looking at the gene expression using Human Whole Genome microarray chips (41,000 genes). The results of the transcriptome analyses suggested that Iscador preparations influenced the overregulation of genes regarding immune defense, stress response, apoptosis and cell-cell adhesion pathways. Within the Mfm-223-Zellen was the Genexpression in MCF-7 and Kpl-1. The MCF-7 cells were affected on the genes which are involved in cell-cell contacts whereas Kpl-1 responded to the mistletoe extracts by changing the mRNA levels of the immune and stress response pathways. Concerning the effects of the mistletoe extract, we conclude that Iscador Qu and M have a greater influence on the immune defense and stress response genes whereas Iscador A tends to affect the cell-cell adhesion and cytoskeleton pathways. In summary, cDNA microarray analyses give us information on whether a cancer cell is sensitive to mistletoe extracts in relation to how many genes are significantly overrepresented after mistletoe treatment, and whether a particular mistletoe extract is more effective on a specific cancer cell than the other preparation.

[Apple-Macintosh compatible software for documentation, management and evaluation of ultrasound findings in obstetrics].

The advantage of using a computer to automate routine calculations and print out charts of the obstetrical ultrasound examination is obvious. This report describes a software designed to simplify the documentation and analysis of ultrasound data in obstetrics. The system is easy to use, even for persons with little computer knowledge. The programme was written in FoxBase+/Mac (Fox Software, Inc., USA). FoxBase+/Mac takes full advantage of the easy-to-learn, easy-to-use Macintosh interface and is also very fast. Another advantage of this software is that it can be used in teaching. Non-experienced examinators can double-check the correctness of their scanning planes by observing the ultrasound pictures with the markers indicating the right measurement sites and the lists of standard values of biometrical parameters for the corresponding gestational age on the screen. In routine obstetrical ultrasound examinations it takes less than 5 min to enter the foetal biometry data and print out reports. These reports are informative and easy to interpret.
