Age-related changes in plasma oestrogen concentration, behavioural responsiveness to oestrogen, and reproductive success in female gray-tailed voles, Microtus canicaudus.

Age-related increases in the incidence of vaginal cornification were associated with increases in the reproductive success of female gray-tailed voles previously isolated from males. The pregnancy rate of females first paired with males at 30-50 days of age was significantly lower than that of females first paired at 90-120 or 150-200 days of age. The improvement was due to increases in propensities to display receptive behaviours and decreases in the incidence of sterile matings. Although plasma oestrogen concentrations increased with age and were higher in receptive than unreceptive females, plasma oestrogen values alone did not account completely for differences in receptive behaviours amongst females of different ages. Females ovariectomized at 30-50 days of age rarely displayed receptive behaviours when treated with large doses of oestradiol benzoate, whereas nearly all females similarly treated were receptive if ovariectomies were performed between 150 and 200 days of age. The receptivity rate of females ovariectomized between 90 and 150 days was intermediate between the rates of the other two groups.

Microhabitats occupied by Loxosceles intermedia and Loxosceles laeta (Araneae: Sicariidae) in Curitiba, Paraná, Brazil.

A survey was done of the environments and substrata occupied by Loxosceles intermedia Mello-Leitão, 1934 and Loxosceles laeta (Nicolet, 1849) (Araneae: Sicariidae) in urban woods and anthropic constructions in Curitiba. In total, 2,099 trees, 364 decaying logs as well as stones and roots, built-up areas, and wasteland in six urban parks and 60 residences were inspected. In total, 1,775 m2 of vegetation was inspected, but spiders and their vestiges were collected only in and around buildings in urban parks and residences. L. intermedia was more common than L. laeta and occurred both indoors and outdoors, whereas L. laeta was more common indoors in wooden houses. The two species did not occur in the same microhabitats, although both preferred paper, wood, and construction materials. Spiders collected in urban parks were heavier than those collected in residences, although only males collected in urban parks were larger than those from residences. The lack of vestiges indicative of a previous occupation in the urban parks suggested that both species occupied primarily the anthropic environment where they found numerous substrata that offered thermal isolation and suitable conditions for web fixation, ecdysis, and reproduction.

[The antagonistic activity of bifidobacteria in vitro and in vivo studied by using gnotobiological technology].

The antagonistic activity of 4 strains of bifidobacteria (B. adolescentis 2 F1, B. longum Z4, B. breve R2 and B. bifidum G1), isolated from the vagina of healthy females of the reproductive age, with respect to Escherichia coli, Klebsiella ozaenae, Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa and Gardnerella vaginalis were studied in vitro and in vivo. The in vitro experiments revealed that all above-mentioned bifidobacteria were capable of inhibiting the growth of all indicator bacterial strains. Still of all the bifidobacteria under study had different levels of activity. B. adolescentis strain 2 F1 exhibited the highest inhibiting activity in vitro. In contrast to in vitro experiments, in vivo experiments with B. breve R2 demonstrated its high antagonistic activity with respect to E. coli. The data thus obtained indicate that in the study of antagonistic activity the use of the in vivo model as also expedient, for it is mainly in vivo that probiotic preparations show their activity.

[Trichoderma produces an inhibitor of the human immunodeficiency virus].

The ability of protein isolated from (Trichoderma Rifai) and azydothymidine to inhibit the reproduction of HIV-virus was compared. The obtained experimental data have verified that Trichoderma Rifai protein is a promising human immunodeficiency virus (HIV) inhibitor.

K-ras activation in neoplasms of the human female reproductive tract.

The role of cellular oncogenes in the development of epithelial tumors of the human female reproductive tract has not previously been extensively studied. DNAs isolated from ten human uterine, 13 ovarian, and four cervical neoplasms and from three cell lines derived from endometrial adenocarcinoma were investigated by dot blot hybridization after polymerase chain reaction amplification of ras gene sequences and in some cases by NIH 3T3 transfection. Transforming activity was found in two of nine endometrial adenocarcinomas, but none of seven ovarian carcinomas and none of four cervical carcinomas showed transforming activity. K-ras sequences with a GGT----GAT mutation in codon 12 were demonstrated in both transformants derived from endometrial carcinoma. K-ras codon 12 mutations were similarly detected in six of 13 endometrial carcinomas (one GAT and GCT, one GTT and GCT, two GAT, two GTT) and two of 13 ovarian tumors (GAT and GCT, GAT), both mucinous adenocarcinomas. Point mutation of K-ras in codon 12 is thus comparably frequent in uterine endometrial carcinomas and in colorectal carcinomas and may have similar significance as an event that contributes to progression of these tumors. Cervical carcinomas and ovarian tumors in general, with the possible exception of mucinous adenocarcinoma of the ovary, do not appear to have this characteristic.

[Genome expression of the DNA-containing human papovavirus in transformed human cells].

In the course of study of the transformed cells of line 63 the phenomenon of cyclically repeated transitory infection of the level of separate cells accompanied by the periodically isolated DNA-containing virus has been shown. Virus reproduction was judged by the determination of the infectious and hemagglutinating activities and radioactivity. The application of activation methods (co-culturing, somatic hybridization and cells treatment with mytomycin C) led to the increase of virus synthesis in the transformed cells of line 63 during spontaneous production of it. We failed to express viral genome in refractor phase.

Comparative study on the inhibitory effect of rimantadine on the reproduction of epidemic strains of influenza A virus H3N2 and H1N1.

The sensitivity to rimantadine of the reproduction of influenza virus A/Texas/1/77 (H3N2) and A (H1N1) isolates Nos 897, 778 and 782, isolated from patients during an epidemic influenza outbreak in Moscow in the winter of 1977/1978 was compared. A (H3N2) virus proved to be more sensitive than the A (H1N1) viruses tested, as manifested by decreases of the infectious titre in ovo and of the haemagglutinin titre in MDCK cells.

[Effect of mutations on the dynamics of numbers of individuals with different genotypes in an isolated heterogeneous population].

The overall (throughout the range) dynamics of the number of diploid individuals possessing n possible gene alleles in a given autosomal locus is studied in an isolated heterogeneous population. The significant influence of new mutations on overall dynamics is shown. It is demonstrated that, under certain conditions, the selection mechanism might have a greater effect on the dynamics of number of individuals with different genotypes in the isolated heterogeneous population than on mutation and "death-reproduction" mechanisms.

[Experience in introducing the method of placental therapy in vitiligo in Ukraine].

The preparation "melagenin" stimulating reproduction of melanocytes and synthesis of melanotic pigment was isolated from human placenta in Cuba. As many as 366 pediatric patients with vitiligo who ranged from 4 to 15 years old were followed up for 1 year. In 62 cases the area of depigmentation amounted to 71 to 100%. Treatment with melagenin proved to be effective in 83% of vitiligo patients. Indications and contraindications have been determined.

A comparison of individual joint contributions to multijoint position reproduction acuity in overhead-throwing athletes.

BACKGROUND: Measures of joint position sense often test single-axis motions isolating a single joint. Such methods hamper our ability to apply findings to functional multijoint activities. Using a functional, active, multijoint test, we measured upper-extremity position reproduction in overhead-throwers. We compared acuity among four joints and examined individual joint contributions or patterns of contribution among joints to overall task acuity. METHODS: We used an electromagnetic tracking device to measure reproduction of two functional upper-extremity positions. We calculated absolute, constant and variable error at four upper-extremity joints around multiple axes of rotation: three axes at the scapulothoracic and glenohumeral joints and two axes at the elbow and wrist. To represent individual joint acuity, we calculated 3-dimensional variable error. Additional 3-dimensional variable error scores using deviation in hand position (with reference to the thorax) represented overall task acuity. We used principle component analyses to identify contributions of individual joints or patterns of contribution among joints to overall task acuity for absolute, constant and variable error. FINDINGS: Scapulothoracic and glenohumeral joints displayed better acuity compared with elbow and wrist joints. When examined separately, absolute, constant and variable error scores did not indicate any individual joint contributed more to task acuity. When we examined the principle components of all error score measures together, a distinct proximal-to-distal pattern of joint contribution to overall task acuity emerged. INTERPRETATION: Proximal joints display better reposition acuity compared with distal joints, however proximal joints contribute more to acuity of the overall task. Our results indicate that upper-extremity joints do not function independently in repositioning tasks and measures of absolute, constant and variable errors combined over multiple joints may better represent upper-extremity function.

Two pine endo-beta-1,4-glucanases are associated with rapidly growing reproductive structures.

Two cDNA clones encoding endo-beta-1,4-glucanases (EGases) were isolated from a radiata pine (Pinus radiata) cDNA library prepared from immature female strobili. The cDNAs PrCel1 (inus adiata cellulase ) and PrCel2 encode proteins 509 and 515 amino acids in length, respectively, including putative signal peptides. Both proteins contain domains conserved in plant and bacterial EGases. The proteins PRCEL1 and PRCEL2 showed strong similarity to each other (76% amino acid identity), and higher similarity to TPP18 (73 and 67%, respectively), an EGase cloned from tomato (Lycopersicon esculentum) pistils, than to any other reported EGases. Northern-blot analyses indicated that both genes displayed a similar pattern of expression. The only significant difference was in the level of expression. In situ hybridizations were used to demonstrate that, within differentiating pine reproductive structures, PrCel1 expression was greatest in microsporangia in pollen strobili and near the developing ovule in the seed strobili. Expression was also found in vegetative tissues, especially in regions experiencing cell elongation, such as the elongating region of root tips. Both proteins have an ability to degrade carboxymethylcellulose in vitro. Genomic-blot analysis indicated the presence of a family of EGase genes in the radiata pine genome, and that PrCel1 and PrCel2 are transcribed from distinct one-copy genes.

Comparative study on isolation calls emitted from hamster pups.

Waveforms of isolation calls emitted from hamster pups, which were Syrian hamsters, Djungarian hamsters, and Chinese hamsters, were compared in a basic study on improving reproduction by decrease of cannibalism, because it was reported that maternal behavior was induced by isolation calls in rodents. Isolation calls of hamster pups, isolated from their mother and receiving cold stress, were collected by Real-Time Spectrogram (RTS), and calculated to spectrograms and power spectra by SIGNAL. Isolation calls consisted of ultrasonic vocalizations (USVs) and audible vocalizations (ADVs) in each species. Waveforms of isolation calls emitted by the hamster pups, were shown to have several characteristic features. In this study, the species specificity of isolation calls was shown in hamster pups. It would seem that the species specificity originates in the differences of sensitivity to cold stress via the autonomic nerve in hamsters.

Microsatellite analysis of paternity and reproduction in Arctic grizzly bears.

We report data from analyses of microsatellite loci of 30 grizzly bear family groups which demonstrate that each cub in a litter can be sired independently, and we derive estimates of maximum reproductive success for males, from an Arctic population in northwestern Alaska that is minimally affected by human activities. These analyses were made possible by the use of single-locus primers that amplified both of an individual's alleles at eight microsatellite loci and by detailed knowledge of maternal/offspring relationships that allowed the identification of paternal alleles. No single male was responsible for more than approximately 11% of known offspring, and no more than 49% of breeding-age males successfully bred. These data contribute to an understanding of the genetic and demographic basis of male reproductive success, which is of vital importance in the maintenance of small, isolated grizzly bear populations.

Expression of an antimicrobial peptide identified in the male reproductive system of rats.

Bin1b is a beta-defensins-like molecule originally isolated from the rat epididymis. Owing to its bactericidal activity, Bin1b may have therapeutic properties suitable for the treatment of sexually transmitted diseases. The amino terminus of the mature Bin1b peptide contains a conserved myristoylated Gly residue. We studied the requirement of the terminal myristoylated Gly residue in the bactericidal activity of Bin1b and found that the terminal myristoylated Gly residue is not essential for the bactericidal activity. In addition, we expressed the tandem repeats of Bin1b in Escherichia coli and found that two tandem repeats of Bin1b protein were successfully expressed. The bacterially expressed tandem Bin1b repeats may be used in a diverse array of biochemical and cell biological studies.

Reproduction of mucosal disease with cytopathogenic bovine viral diarrhoea virus selected in vitro.

Isolates of non-cytopathogenic bovine viral diarrhoea (BVD) virus from 18 persistently infected calves from one herd were compared by using monoclonal antibodies directed against the major viral glycoprotein gp53. All the isolates displayed an almost identical reaction pattern. Based on this antigenic analysis three cytopathogenic BVD and three non-cytopathogenic BVD viruses closely related to the non-cytopathogenic BVD herd isolate were selected. Six of the persistently infected calves were inoculated with a pool of the three closely related cytopathogenic BVD viruses and two with a pool of the three non-cytopathogenic BVD viruses. In addition three animals were infected with one closely related cytopathogenic BVD strain (Indiana) and two animals with the antigenetically different cytopathogenic BVD viral strain A1138/69. Regardless of the inoculation route all the animals superinfected with closely related cytopathogenic BVD viruses developed the characteristic lesions of mucosal disease within 14 days of infection. Animals which were inoculated with non-cytopathogenic BVD viruses which closely resembled the herd isolate, or with cytopathogenic BVD viruses which did not resemble the herd isolate did not develop any signs of disease. However, the latter group produced antibodies to the superinfecting virus.

Study of the role of Chlamydia, Mycoplasma, Ureaplasma and other microaerophilic and aerobic bacteria in uterine infections of mares with reproductive disorders.

In six healthy mares and 24 mares showing reproductive disorders swab samples were taken from the fossa clitoridis to isolate Taylorella equigenitalis, and from the uterus to isolate mycoplasmas, ureaplasmas and other aerobic bacteria. Swab samples were also taken from the uterus for Chlamydia antigen ELISA and Chlamydia PCR studies. The uterus of 27 mares was examined cytologically, and biopsy samples were taken from the endometrium for histological examinations and for immunohistochemical examinations aimed at the detection of chlamydiae. T. equigenitalis, mycoplasmas, ureaplasmas and chlamydiae could not be detected from any of the mares examined. Aerobic facultative pathogenic bacteria were isolated from mares with endometritis in four cases. In 18 out of 22 mares with endometritis (82%) no infective agents could be demonstrated. Further studies are needed to elucidate the relative importance of non-infectious causes of endometritis and of anaerobic bacteria often detectable in the uterus in the aetiology of the reproductive disorders observed.

Isolation and culture of bovine oviductal epithelial cells for use in the anatomy and physiology laboratory and undergraduate research.

This article presents methods for the isolation and culture of epithelial cells from the bovine oviduct for use in both research and the teaching laboratory and provides examples of ways that an oviductal cell culture can be incorporated into an undergraduate research program. Cow reproductive tracts are readily available from area butchers, and the procedure for isolation of the epithelium is simple and inexpensive. The cells can be observed immediately after isolation or can be cultured for up to 72 h under simple conditions for observation over several days. For experimental use, epithelial cells are cultured in standard cell culture medium, where they continue to divide and actively secrete substances into the medium. The ease with which the tissue can be collected and cells isolated makes the oviductal epithelium ideal for use in both the teaching laboratory and research projects in which undergraduates serve as investigators.

Pharmacological properties of the extract of thymus gland (Thymomodulin-TFX) and its effect on reproduction.

The effect of Thymomodulin-TFX on pentetrazole convulsions, tremorine-induced tremor, pain response to intraperitoneal acetic acid injection, hexobarbital sleeping time, isolated guinea pig ileum, isolated rat uterus, rabbit skeletal muscle response, diuresis and corneal response was tested. In addition the effect of TFX on reproduction of albino rats was investigated. In doses up to 20 mg/kg, 8 times higher than clinical doses, TFX did not reveal any unwanted effects. The results of tests widen the security margin for TFX's usage.
