Fe2+-catalyzed heterolytic RO-OH bond cleavage and substrate oxidation: a functional synthetic non-heme iron monooxygenase system.

The reaction of [Fe22+(H2Hbamb)2(N-MeIm)2], [1], a binuclear, non-heme iron complex, with 2-methyl-1-phenylprop-2-yl hydroperoxide (MPPH) shows that [1] induces heterolytic cleavage of the peroxy O-O bond. Catalytic atom transfer reactions (1:MPPH:PhSMe 1:596:6011) resulted in the highly efficient (99 +/- 1%), catalytic oxidation of phenyl methyl sulfide to phenyl methyl sulfoxide/sulfone (T.N. = 500/11 respectively) and cyclohexane to cyclohexanol/cyclohexanone (T.N. = 230/5 respectively) showing the highly efficient, catalytic capacity of [1] to carry out oxygen insertion chemistry.

Comparative study of the cytotoxicity induced by antioxidant epicatechin conjugates obtained from grape.

We studied the cytotoxicity of epicatechin conjugates obtained by depolymerization of grape polymeric flavanols in the presence of cysteamine or cysteine and the resulting conjugates purified by ion exchange and/or reversed-phase high-resolution chromatography and compared it to their antioxidant capacity. The studies were carried out on fibroblast and keratinocyte cell lines. The cytotoxic effects of these products were observed at concentrations 3-7-fold higher than the antioxidant concentration after exposure for 24, 48, and 72 h. The compounds with a gallate group were more toxic than the corresponding products without one. It is interesting to note that the esther ethyl derivative exhibited low cytotoxicity but had the most potent antioxidant activity. The results indicated that effective antioxidant activity can be obtained from these products in a concentration range that is safe for the normal cell. This finding suggests new pharmaceutical applications and may also help us to identify the potential therapeutic dose.

In vivo saturation of the transport of vinblastine and colchicine by P-glycoprotein at the rat blood-brain barrier.

PURPOSE: To determine concentration-dependent P-gp-mediated efflux across the luminal membrane of endothelial cells at the blood-brain barrier (BBB) in rats. METHODS: The transport of radiolabeled colchicine and vinblastine across the rat BBB was measured with or without PSC833, a well known P-gp inhibitor, and within a wide range of colchicine and vinblastine concentration by an in situ brain perfusion. Thus, the difference of brain transport achieved with or without PSC833 gives the P-gp-mediated efflux component of the compound transported through the rat BBB. Cerebral vascular volume was determined by coperfusion with labeled sucrose in all experiments. RESULTS: Sucrose perfusion indicated that the vascular space was close to normal in all the studies, indicating that the BBB remained intact. P-gp limited the uptake of both colchicine and vinblastine, but the compounds differ in that vinblastine inhibited its own transport. Vinblastine transport was well fitted by a Hill equation giving IC50 at approximately 71 microM, a Hill coefficient (n) approximately 2, and a maximal efflux velocity Jmax of approximately 9 pmol s(-1) g(-1) of brain. CONCLUSIONS: P-gp at the rat BBB may carry out both capacity-limited and capacity-unlimited transport, depending on the substrate, with pharmacotoxicologic significance for drug brain disposition and risk of drug-drug interactions.

Dietary protein intake and skeletal-muscle protein metabolism in rats. Studies with salt-washed ribosomes and transfer factors.

1. Aspects of skeletal muscle protein synthesis in vitro were studied in young rats given a low-protein diet for up to 10 days and during re-feeding with an adequate diet. 2. Partially purified muscle transfer factors (transferases I and II), crude and purified (NH(4)Cl-washed) ribosomes and a pH5 enzyme fraction were prepared for this purpose. 3. A marked decrease in the capacity of crude ribosomes to carry out cell-free polypeptide synthesis occurred within 4 days of feeding the low-protein diet. 4. The capacity of salt-washed ribosomes to promote amino acid polymerization, in the presence of added transfer factors and aminoacyl-tRNA, was only slightly decreased by the dietary treatment. 5. However, the capacity of salt-washed ribosomes to bind (14)C-labelled aminoacyl-tRNA was decreased by feeding the low-protein diet. 6. The capacity of the pH5 enzyme fraction to promote amino acid incorporation in a complete cell-free system was decreased within 2 days of feeding the low-protein diet. There is no evidence that the change is associated with aminoacyl-tRNA synthetase or binding enzyme activities of the pH5 fractions. 7. These changes are discussed in relation to the diminished rate of protein synthesis in the intact muscle cell when rats are given a low-protein diet.

Comparative study with EMG, pressure algometry and manual palpation in tension-type headache and migraine.

According to International Headache Society classification criteria, the presence of pericranial muscle disorder in tension-type headache should be evaluated using one of the following methods: EMG, pressure algometry or manual palpation. The purpose of this study was to compare the results of these three methods in 15 patients with episodic tension-type headache, 29 with chronic tension-type headache and 22 presenting migraine without aura compared to those obtained in healthy individuals. Algometric and EMG recordings at the frontalis muscle during mental arithmetic were more impaired in episodic and chronic tension headache patients than in controls and migraine patients. Chronic tension headache patients were significantly impaired at the trapezius muscle in all three tests compared to controls. Our data indicate that when two or three tests were carried out the diagnostic capacity was significantly improved in comparison to only one test. Moreover, since a different pattern could be seen with pain and without pain, the existence of headache at the time of testing should be taken into consideration.

A Kleinian approach to group psychotherapy.

This article presents clinical material from a group that is now in its seventh year. The clinical material illustrates an aspect of the working through of depressive anxieties. The theoretical framework for understanding the material and my interventions is Bion's application of Klein's ideas. The clinical vignettes show how a group struggles with the split between good and bad characteristic of the paranoid-schizoid position (Ps) and the more integrated view of objects as both good and bad, characteristic of the depressive position (D). This more integrated view carries with it the capacity for both love and hate. One member's early memory of cruelty mobilizes other members' guilt toward their objects as well as their reparative wishes.

Biogenesis of mitochondrial membranes in Neurospora crassa during cellular differentiation: changes in oxidative phosphorylation and synthesis of mitochondrial phospholipids.

Changes in the capacity of mitochondria to carry out oxidative phosphorylation and in the rate of synthesis and incorporation of phospholipids into mitochondria were measured during the germination of conidiospores of Neurospora crassa. The competence of isolated mitochondria to carry out coupled respiration was very low during the first 3 h growth, but it increased rapidly, reaching maximal levels at 5 to 6 h growth. Changes in mitochondrial function were the same in cells grown in 2% sucrose- or 15% glucose-supplemented medium. The rate of synthesis of mitochondrial phospholipids was very low during the first 2 h growth and increased to maximal levels between 3 and 5 h. The rate of synthesis of mitochondrial phospholipids was approximately three times higher in cells grown in 15% glucose than in those grown in 2% sucrose. The maximal rate of synthesis of mitochondrial phospholipids occurred during spore germination and preceded attainment of full competence for oxidative phosphorylation. The lipid-rich condition of the mitochondrial resulting from the high rate of synthesis of phospholipids in glucose-grown cells is postulated to be related to the whorled inclusions observed in thin sections of Neurospora cells.

Increased stimulatory capacity of spleen cells from adult thymectomized mice in mixed lymphocyte reactions.

Conflicting results have been reported concerning the effect of adult thymectomy (A-Tx) on the mixed lymphocyte reaction (MLR) response. Our purpose was to test the reverse question, namely the effect of A-Tx on the stimulatory capacity of mouse lymphocytes carrying allelic disparities at the Mls and the H-2 loci. Spleen cells from A-Tx CBA donors were found to be significantly more stimulating than cells from normal donors. That A-Tx could eliminate suppressor T cells involved in MLR regulation or in preventing back stimulation is not likely according to our data. An increment in expression or in accessibility of Mls or H-2 antigenic determinants induced by A-Tx is no more likely, since a smilar increase in lymphocyte proliferation was observed in syngeneic cultures. The most probable explanation remains that A-Tx discloses a strongly stimulatory subpopulation, perhaps acting in part nonspecifically. The hypothesis of an enrichment in B cells versus a loss of T cells is not compatible with our findings, since B-cell-enriched fractions from various origins never attained the stimulating ability disclosed by A-Tx cells. Moreover, the cell involved is theta-positive, although slightly nylon-wool-adherent, and thus shares several properties with immature T cells involved in auto-rosette formation.

Impact of hormone replacement therapy on microvascular function in healthy and Type 2 diabetic postmenopausal women.

AIMS: Hormone replacement therapy (HRT) has been previously reported to modulate vascular function and cardiovascular risk. Its impact on the macrocirculation has previously been explored, however, little data is available on its impact on the microcirculation. This study aimed to determine the impact of HRT on microvascular function in healthy and Type 2 diabetic postmenopausal women (n=20 and 17, respectively). METHODS: Microvascular function was assessed by skin maximum hyperaemia, skin hyperaemic response to iontophoretically applied acetylcholine (endothelial-dependent vasodilator) and sodium nitroprusside (endothelial-independent vasodilator), capillary pressure and the microvascular filtration capacity. Microvascular assessments were carried out at baseline and repeated following 6 months' oral hormone replacement therapy (1 mg oestradiol/0.5 mg norethisterone or 1 mg unopposed oestradiol for hysterectomized women). RESULTS: Following 6 months' therapy there were no significant changes in microvascular assessments in the healthy women. In the diabetic women there was a reduction in the skin hyperaemic response to acetylcholine [median pretreatment peak response: 1.95 (25th, 75th centiles: 1.54, 2.30) V vs. post-treatment peak response: 1.53 (1.30, 1.91) V (P=0.011, Wilcoxon's signed rank test)] and sodium nitroprusside [median peak response 1.59 (1.37, 1.99) vs. 1.35 (0.92, 1.63) V (P=0.011)] with HRT, but no other changes. CONCLUSION: These data suggests that HRT does not affect microvascular function in healthy women, but adversely affects it in diabetic women. These findings may help to explain why HRT fails to provide the predicted cardiovascular protection, and raises the possibility that HRT influences microangiopathy progression in diabetic women.

Role of the Yersinia outer membrane protein YadA in adhesion to rabbit intestinal tissue and rabbit intestinal brush border membrane vesicles.

The Yersinia virulence plasmid confers on strains of Yersinia pseudotuberculosis and Y. enterocolitica an adhesive potential superior to the one encoded by the chromosome alone. We have evaluated the role of the plasmid-encoded outer membrane protein YadA (formerly called Yopl) in adhesion. Insertional inactivation of the yadA gene (formerly called yopA), which encodes YadA, led to a reduction in the capacity of plasmid-carrying strains of Y. pseudotuberculosis 0:III and Y. enterocolitica 0:9 to adhere to intestinal tissue, brush border membranes and polystyrene surfaces. The adhesive characteristics of the mutants were comparable to those of their plasmid-cured counterparts. When the yadA gene from Y. pseudotuberculosis serotype 0:III or Y. enterocolitica serotype 0:3 or 0:8 was cloned into an Escherichia coli strain, increased ability to adhere to intestinal tissue, brush border membrane vesicles and polystyrene was transferred concomitantly. The introduction of the yadA gene from Y. pestis, which is unable to express YadA due to a one base pair deletion, did not change the adhesive characteristics of E. coli. Expression of YadA in the outer membrane may, therefore, make an important contribution to intestinal adherence of the two enteropathogenic members of the Yersinia species, Y. pseudotuberculosis and Y. enterocolitica.

Urban growth and decline: San Jose and St. Louis in the 1960's.

The population changes in San Jose and St. Louis between 1960 and 1970 exemplify the two broad trends-urban formation followed by metropolitan dispersal-that have shaped 20th-century urbanization in this country. The fact that these developmental trends were expressed through demographic processes found to be common to both cities, despite their contrasting recent experiences, suggests that generalizations can be made about the complex forces underlying urbanization. The formation of metropolitan San Jose's population parallels the traditional process whereby a region's growth comes to be focused, through migration, on a few urban centers. The modern variant is not characterized by a rural-to-urban shift, however, but by migration flows among urban areas, and particularly to a few most-favored areas, such as San Jose. Migratory growth has left a powerful demographic legacy in San Jose. This legacy is also instructive for studying the migratory formation of any new city's population. Its demographic character determines its demographic destiny, whose likely variations we can now perceive with some clarity. San Jose's population is both youthful and chronically migratory. The presence of many prospective parents and relatively few elderly persons lays a broad foundation for the population's continued growth through natural increase, despite the national downturn in fertility (14). Even without further net in-migration, the population of new cities like San Jose would continue to grow at an above-average rate. The hypermobility of San Jose's population (that is, its propensity for further migration) also has an important bearing on the future. With about 21 migrants entering and 17 departing each year per hundred residents, San Jose's rapid migratory growth rests (as it would in other new cities) on a precarious arithmetic balance. A significant dip in local employment growth could easily reduce net migration to a small fraction of its present high level. Even a slight decline would result in the inflow's no longer exceeding the high volume of outflow. Demographic analysis alone cannot foresee such an employment downturn, but if it happened, the migratory downturn probably would be swift. Hypermobility also works the other way; and given San Jose's focal position in California's expanding metropolitan structure (with its virtually endless supply of migratory growth), net migration could resume with equal swiftness. The outward dispersal of population from central cities that has occurred in St. Louis has been accelerating in other cities as well, and will remain a prominent feature of U.S. urban growth. It may seem paradoxical that in a period noted for something called "urban growth" there are so many declining central cities, but that is merely one indication that the "central city" no longer is the real city, except in name. Real city or not, the central city can expect to come into political conflict with other jurisdictions created in the process of dispersion. In cities like St. Louis, where population is dispersing but old political boundaries are fixed, the problems of the central city are separated from the resources in the suburbs. Transitional problems associated with persistent and severe outmigration also arise: accumulation of disadvantaged citizens, declining demand for city housing, and a diminished replacement capacity in the population. Carried far enough, the last of these problems results in natural decrease, and thereafter the population's decline acquires its own dynamic. As noted earlier, the white population in St. Louis has reached this point: The number of persons dying now exceeds the number being born. For two reasons, this natural decrease can do little other than intensify. First, a substantial proportion of whites are either entering or already within the high-mortality age brackets. The white population's crude death rate therefore will continue to rise. Second, prospective parents are becoming scarce among St. Louis's whites, and the national evidence that parents in general will choose to have smaller families continues to mount. The white population's crude birth rate is therefore likely to fall, barring a dramatic increase in fertility or a strong and sustained inflow of childbearing families. Nor is St. Louis's black population likely to grow substantially. It is expanding steadily through natural increase, but black migration out of the city is more than enough to cancel that increase.

Patient compliance with inhaled medication: does combining beta-agonists with corticosteroids improve compliance?

Patient compliance with an inhaled corticosteroid may be greater if it is combined with a beta-agonist. This study compared compliance with an inhaled corticosteroid (budesonide), and a short-acting inhaled beta-agonist (terbutaline sulphate), and a Turbuhaler inhaler containing a combination of the two drugs. In an open, multicentre, parallel group study 102 asthmatic patients were randomly divided into two groups, either receiving the two drugs in separate Turbuhalers or combined into one Turbuhaler. A twice daily regimen was prescribed and a preweighed metered-dose inhaler (MDI) of salbutamol was provided for rescue use. Compliance was measured using the Turbuhaler Inhalation Computer (TIC), which recorded the time and date of each inhalation over a 12 week period. Forced expiratory volume in one second (FEV1) and forced vital capacity (FVC) measurements were carried out at week 0, 6 and 12. Results from 72 patients were analysed. The average compliance was 60-70%. Treatment was taken as prescribed on 30-40% of the study days, and over-usage occurred on less than 10% of days. Only 15% of patients took the drugs as prescribed for more than 80% of the days. Compliance was no greater in patients using the combined inhalers. Other ways of improving patient self-management need further investigation.

Effects of organic and inorganic mercurials on thyroidal functions.

Acute effects of methylmercuric chloride and mercuric chloride on thyroidal functions were examined. The organic mercurial concentration of 4 x 10(-5) M inhibited by 50% of Na+K+ATPase in the membraneous preparation from the hog thyroid, and 6 x 10(-7) M of the inorganic mercurial showed the same extent of the inhibition. The Mg2+ ATPase activity in the preparation was neither affected by CH3HgCl up to a concentration of 2 x 10(-3) M, nor by HgCl2 up to 1 x 10(-4) M. After an intraperitoneal injection to mice of 5 micrograms of mercurial per gram body weight daily for 2 consecutive days, the 4-hour and the 24-hour uptakes of 131I by the thyroids were partially reduced by both organic and inorganic mercurials. A significant reduction in percentages of labeled iodothyronines was demonstrated to suggest that mercurial may cause a coupling defect in the synthesis of iodothyronines. Incubation of hog thyroglobulin with 8 x 10(-3) M of methylmercuric chloride caused no observable aberration in slab disc electrophoreogram, but the protein was apparently denatured by the same concentration of mercuric chloride suggesting that thyroglobulin may carry a large binding capacity against either mercurial, but the inorganic mercurial can be more potent denaturant of the protein. The in vitro lysosomal hydrolysis of the mercurial-pretreated rat thyroglobulin which was labeled with 125I in vivo and fortified with the carrier hog thyroglobulin was not affected, but the direct addition of either mercurial in the medium resulted in a significant inhibition of the proteolytic action. Iodotyrosine deiodinase in the thyroid was inhibited by both mercurials in in vitro and in vivo systems. A partial reduction in the serum bound 131I-iodide in both mercurial treated groups was observed at 4 hours and 24 hours after the radioiodide administration. The blood thyroxine levels estimated by radioimmunoassay were quite reduced in the inorganic mercurial treated group and also moderately reduced in the methylmercurial treated group, indicating that the hormone secretion was affected by mercurials.

Demographic analysis of the variation in the rates of multiple maternities in Sweden since 1751.

To elucidate the causes and mechanisms of twinning and higher multifetal maternities, we have taken advantage of the statistical sources of Sweden, where continuous statistics for the whole population are the oldest available. We found strong secular and regional fluctuations. The rates of multiple maternities were the highest during the last three decades of the 18th century, when the twinning rate was more than 17 per 1,000, the triplet rate was more than 3 per 10,000, and the quadruplet rate was almost 7 per 1 million maternities. During 1849-1873 the twinning rate in Sweden was 14.2 per 1,000, but this rate showed great regional differences, being 18.0 per 1,000 on the island of Gotland and 12.6 per 1,000 in the county of Alvsborg. During this period the twinning rate in the countryside in the county of Stockholm was 20.4, but in the city of Stockholm it was only 14.1 per 1,000. In Sweden after the 1930s there was a marked decrease in the twinning rate, which by the 1960s had fallen to only about half of what it had been two centuries earlier. The corresponding reductions for triplet and quadruplet rates were about 75%. The aim of this paper was to study the temporal and regional variations in multiple maternities in Sweden from 1751 to 1960 based on demographic and some socioeconomic data for the counties. We confirmed our earlier studies that maternal age and parity cannot satisfactorily explain the secular and regional differences in the twinning rates. In contrast to studies in France (1901-1968), we found no unequivocal association between the twinning rates and the crude birth rates. The correlation coefficients between the twinning rate and the crude birth rate showed statistically significant regional and temporal variations. After eliminating the temporal trends, regional differences in the correlation coefficients remained. The twinning rates for the counties seem to converge toward a common low level, 10-12 per 1,000. The observed convergence toward relatively similar levels may be caused by the increased matrimonial migration distances and decreased endogamy of the citizens as a consequence of better communications. The increased urbanization and industrialization that started in the last decades of the 19th century broke up the old static agrarian isolates and caused Sweden, within 2-3 generations, to develop from a poor nation to one of the most prosperous in the world. A more urban and affluent lifestyle, a better diet, and increased stress and sedentary occupations may have reduced the physical capacity of mothers to carry gestations with multiple embryos or fetuses to completion.

Overexpression of c-kit in a leukemic cell population carrying a trisomy 4 and its relationship with the proliferative capacity.

The expression of c-kit and its ligand, the stem cell factor (SCF), was studied in five cases of acute myeloid leukemia. One of these had a trisomy of chromosome 4, where the c-kit oncogene is located. In this case, the c-kit oncogene was overexpressed, but matched by a low expression of its ligand, SCF. The molecular evaluation of the growth rate by c-myc and the histone H3 expression indicated that the growth fraction of this cell population was very low. In one of the other leukemic cell populations studied, characterized by a low expression of c-kit and an elevated expression of the SCF, the growth fraction was also very low. Our results suggest that at least for some receptor oncogenes, the simple overexpression cannot be taken as an indication that the oncogene is involved in the deregulation of cell proliferation.

[Acarofauna of house dust in Barcelona and surrounding cities].

The study of the house-dust acarofauna has aroused great interest since Voorhorst and colls. (41), in 1964, showed the continual presence of Dermatophagoides pteronyssinus in this habitat; and considered by these authors to be mainly responsible for the allergenic capacity of housedust. Studies carried out by other authors around the world confirm the universal character of this acarus and its prevailing presence in house-dust biocenosis. Dermatophagoides pteronyssinus is usually accompanied with other species of the same Pyroglyphidae family (see table II and III), and by species belonging to other acarian families and groups (table I). In this work 182 house-dust samples have been analyzed; these samples were collected in Barcelona and its surrounding. All have been found to be positive to acari. Samples of 5 g of house-dust were used for the study. The number of acari present in each sample showed a great variation both in relation to the total of acari present and to the number of isolated Pyroglyphidae (table VII and VIII). Dermatophagoides pteronyssinus appeared to be the prevailing species, found in 175 of the 182 samples examined and representing 39.70% of the total population of the acari collected. Other Pyroglyphidae isolated were D. farinae (95 samples), Euroglyphus maynei (75 samples), D. microceras (19 samples), D. passericola (1 sample) and Malayoglyphus carmelitus (1 sample) (table VI). The frequency and density in which other species of acari appeared in house-dust samples studied are shown in table VII. If we compare our results with those obtained by others (1, 7, 11, 21, 23, 26, 28, 29, 38) we see a great difference according to the frequency of acarian infestation in house-dust. We think that the causes of this variation are the diverse methods and techniques employed in the isolation and enumeration of acari. The low infestation percentages given by some authors could be due to the following: a) a study of insufficient amounts of house-dust samples; b) the lack of a method used for processing the samples; c) the interaction of both factors. The discovery of three single D. passericola specimens found in one sample lets us assume that the pressence of this acarus in house-dust has been sporadic. We think it could originate from birds' feathers or nests. Despite the very high number of Tarsonemus sp. collected (37.17% of the acari total) the representative of this species is actually very low, since almost all of them were found in two different samples taken from the same house.

[Role of bacterial protease degrading secretory immunoglobulin A in Klebsiella persistence].

During the study of 186 K. pneumoniae clinical strains the presence of SIgA-protease activity was detected in 109 isolates (58.6%). The PCR test system permitting the detection of iga gene DNA in K. pneumoniae strains isolated in purulent inflammatory processes of different localization was developed. For the first time a conjugative plasmid with a mol. wt. of 100 MD was detected in K. pneumoniae clinical isolates. This plasmid controlled the capacity of inactivating SIgA, carried the markers of antibiotic resistance and was capable of expression in a wide circle of hosts belonging to the family Enterobacteriaceae. The results of in vitro experiments revealed that the strains containing plasmid SIgA were cytotoxic with respect to cells HEp-2 and more resistant to the digestive action of enzymes produced by the phagolysosomes of guinea pig peritoneal cells. As shown on the in vivo biological model with the use of the genetically linked pair of Salmonella typhimurium 274 vaccine strains, differing in SIgA-protease activity, plasmid pSigA ensured more prolonged survival of bacteria in mice in comparison with the nonplasmid variant. The study of SIgA-protease activity of K. pneumoniae strains circulating among patients with a view to the detection of clones causing the development of persistent infection was shown to have good prospects.

[Immunomodulating effect of protein fractions isolated from bifidobacteria].

The screening of the immunomodulating activity (IMA) of different protein fractions isolated from bifidobacteria was carried out and the capacity of these fractions for changing the proliferative activity of immunocompetent cells was evaluated. Soluble proteins were extracted from lyophilized and sonicated bacterial mass of B. bifidum strain 1 in Na2HPO4 (pH 8) in a water bath at 65 degrees C for 30 minutes. After the formation and removal of nucleic acid sediment the resulting supernatant fluid was dialyzed, its adsorption spectra were analyzed and the fluid was fractionated in a specially proposed device for preparative electrophoresis. Protein fractions were tested for IMA on spleen cells of CBA mice in the reaction of lymphocyte blast-transformation by the level of the inclusion 3H thymidine. The analysis of IMA of protein fractions revealed that their high-molecular components produced a pronounced dose-dependent effect on the proliferative activity of spleen cells. The fractions containing low-molecular components were either inactive (fraction 4) or active only in the maximum dose (fraction 5).
