Active site topologies of bacterial cytochromes P450101 (P450cam), P450108 (P450terp), and P450102 (P450BM-3). In situ rearrangement of their phenyl-iron complexes.

The reactions of cytochromes P450101 (P450cam), P450108 (P450terp), and P450102 (P450BM-3) with phenyldiazene result in the formation of phenyl-iron complexes with absorption maxima at 474-478 nm. Treatment of the cytochrome P450 complexes with K3Fe(CN)6 decreases the 474-478 nm absorbance and shifts the phenyl group from the iron to the porphyrin nitrogens. Acidification and extraction of the prosthetic group from each of the ferricyanide-treated enzymes yields a different mixture of the four possible N-phenylprotoporphyrin IX regioisomers. The ratios of the regioisomers with the phenyl ring on pyrrole rings B, A, C, and D (in order of elution from the high performance liquid chromatography column) are, respectively: cytochrome P450cam, 0:0:1:4; P450terp, 0:0:0:1; and P450BM-3, 2:10:2:1. The isomer ratio for recombinant cytochrome P450BM-3 without the cytochrome P450 reductase domain (2:9:2:1) shows that the reductase domain does not detectably perturb the active site topology of cytochrome P450BM-3. Potassium ions modulate the intensity of the spectrum of the phenyl-iron complex of cytochrome P450cam, but do not alter the N-phenyl isomer ratio. Computer graphics analysis of the crystal structure of the cytochrome P450cam phenyl-iron complex indicates that the active site of cytochrome P450cam is open above pyrrole ring D and, to a small extent, pyrrole ring C, in complete agreement with the observed N-phenylprotoporphyrin IX regioisomer pattern. The regioisomer ratios indicate that the active site of cytochrome P450terp is only open above pyrrole ring D, whereas that of cytochrome P450BM-3 is open to some extent above all the pyrrole rings but particularly above pyrrole ring A. The bacterial enzymes thus have topologies distinct from each other and from those of the mammalian enzymes so far investigated, which have active sites that are open to a comparable extent above pyrrole rings A and D.

[Clinical observation of CP and iodoform for intracanal medication].

To evaluate and compare the clinical effect of camphophenic (CP) and iodoform used as intracanal medication in root canal therapy for periapical periodontitis, 305 teeth suffering from periapical periodontitis in 245 subjects were divided into two groups. The experimental group of 163 teeth was treated with CP and iodoform, while the control group of 142 teeth was treated with CP alone. The clinical effect after 10 days was observed. The result showed that the response rate in the teeth treated with CP and iodoform was 92.64%, significantly higher than that in the teeth treated with CP alone (79.58%), P<0.01.

[Optical biosensor study and molecular modeling of interactions between cytochrome P450cam and cytochrome b5].

The formation of complexes of cytochrome P450cam (P450cam) with full-length cytochrome b5 (d-b5) and its tryptic water-soluble fragment (t-b5) was analyzed using a two-channel IAsys+ optical biosensor. It was found that t-b5 can form complexes with P450cam, while d-b5 does not interact with P450cam. The involvement of amine groups of P450cam in the complex formation was demonstrated. The temperature dependence of t-b5(im)/P450cam complex formation was measured. The association rate constant (k(on)) increased with temperature, while the dissociation rate constant (k(off)) practically remained unchanged. It was concluded that hydrophobic interactions play a key role in the complex formation, while electrostatic interactions are significant for complex stabilization. Based on temperature dependence the activation energy, enthalpy and entropy of complex formation were calculated. It was shown that the entropy component plays a key role in t-b5(im)/P450cam interaction. Computer modeling of P450cam/t-b5 and P450cam/d-b5 interactions was carried out. Using the method of molecular docking some hypotheses of protein-protein complexes were advanced and the best ones were selected based on geometric complementarity, calculated binding energy and probability of electron tunneling between proteins. The computer modeling has shown that only P450cam and t-b5 can form the stable complex. These results are in good agreement with the experimental data obtained with the optical biosensor.

Antimicrobial efficiency of some antiseptic products on endodontic microflora isolated from gangrenous pulp tissue.

AIM: The aims of the study are to investigate the bactericidal effect of three antiseptics (chlorhexidine solution, povidone-iodine solution, and Walkhoff solution) and to determine the minimum inhibitory concentration and their effect on different microbial species. METHODS AND MATERIALS: The study was performed on microflora derived from root canals with simple and complicated pulp gangrene and on pure strains of Enterococcus and Candida albicans. RESULTS: Chlorhexidine and povidone-iodine proved to have antibacterial and antifungal effects if used in the treatment of pulp gangrene and in cases not responding to conventional therapy. CONCLUSION: According to the obtained results, the spectrum of antibacterial agents used in infected canal irrigation can be enlarged to include the agents tested.

[Studies on the chemical constituents of essential oil of Hunan Artemisia annua].

The chemical constituents of the essential oil of Hunan wild Artemisia annua L. in Xuefeng mountain region have been studied by means of the GC-MS. The essential oil was extracted from Artemisia annua L. by steam distillation. The chemical constituents were separated and identified by GC-MS, and the relative contents of them in the oil were determined with area normalization method. Forty-five components were separated and identified which accounted for 90% of total essential oil. These components identified from the essential oil indicate that Hunan wild Artemisia annua L. in Xuefeng mountain region is a good medical plant, and worth to exploiting.

A simple test for mothball component differentiation using water and a saturated solution of table salt: its utilization for poison information service.

All 651 cases of mothball ingestion received by a poison information center were studied. At each time of inquiry, the staff member asked the caller to test whether the mothball floated or sank in water and a saturated solution of table salt. With the results of the test the staff member identified the mothball ingredient. Though most of the callers were housewives with little knowledge of medicine, the differentiations were each completed within 5 min. Later, gas chromatographic analyses revealed that no errors in differentiations had been made. Our method proved a quick preliminary test for mothball component differentiation in poison information service.

Mycoplasma gallisepticum as a model to assess efficacy of inhalant therapy in budgerigars (Melopsittacus undulatus).

One hundred budgerigars (Melopsittacus undulatus) were infected in the nares (0.02 ml/naris), eye (0.02 ml/eye), and throat (0.2 ml) with Mycoplasma gallisepticum (MG) R strain (3.175 x 10(7) colony-forming units/ml). Fifty birds were treated with sterile broth and served as the controls; 25 of those were exposed to an inhalant, and the others were not treated. Infected birds were divided into four groups; 1) no treatment, 2) penicillin in drinking water, 3) inhalant, 4) both penicillin and inhalant. At days 3, 7, 10, 14, and 21 postinfection (PI), birds were assessed for clinical signs of disease. Five birds in each group were euthanatized at each interval PI; quantitative cultures were performed on lavages from the nares and trachea and on throat swabs; tracheas and air sacs were examined histopathologically. No clinical signs, lesions, or cultural isolations occurred in any control birds. All infected birds developed clinical signs and lesions of the trachea and air sac, but none died. The most severe clinical signs were seen in birds that were infected with MG and received no other intervention or birds that received penicillin in conjunction with infection. Increased respiratory tract lesions were associated with penicillin treatment; aerosol therapy resulted in fewer lesions.

Tissue necrosis after using an arsenical endodontic preparation: a case report.

A case is reported of severe loss of alveolar bone in the maxilla, as a result of leakage into the tissues of an arsenical paste from the pulp chamber of an endodontically 'treated' tooth. The history of the use of arsenic in dentistry is discussed, as are the hazards inherent in the use of this outmoded and dangerous medicament.

[Evaluation of endodontic treatment with modified Walkhoff technique in teeth with necrotic pulp and/or periapical lesions].

A treatment has been carried out in a total of 112 dental cases with necrotic pulp and/or periapical lesions using yodoformic paste. The technique employed was that of a conventional endodontic therapy of the root and ductal filling, using yodoformic paste and gutta-percha. The clinical and radiological follow up checks, three, six and twelve months after treatment, show improvement in the periapical lesions, with bone regrowth and complete disappearance of the radio-lucity within a variable time span, depending on the gravity of the damage, but never greater than twelve months. The immediate post-operative well being and absence of complications in the majority of cases also be highlighted.
