Influence of turbidity on photometric assays: a blank sample must always be used.

The authors studied the influence of sample turbidity on photometric assays at different wavelengths. Turbidity was created by using a mixture of hydrazine sulfate and hexamethylene tetramine, and a dye solution (p-nitrophenol, cobalt nitrate). This work demonstrates the necessity of using a bland sample, for example in the glucose assay (glucose oxidase/peroxidase). The influence of turbidity on an assay depends upon the instrument. Knowledge of this influence is essential for the evaluation of the viability of an assay in hyperlipemic serum.

[Contribution of neutrophils to Aspergillus infection].

Neutrophil disfunction, caused by a decreased production of effective radical oxygen species by myeloperoxidase (MPO) and NADPH oxidase within the neutrophil, may result in susceptibility to opportunistic fungal infections. In vitro, MPO produces OCl, which kills bacteria and viruses. In the case of MPO deficiency, susceptibility to Candida albicans infection was observed. Furthermore, it was demonstrated that MPO knockout mice were primarily susceptible to C. albicans infection. With regards to NADPH oxidase deficiency, such a patient was found to have severe chronic granulomatous disease (CGD) due to Aspergillus infection. This deficiency may have resulted from a gene mutation and/or abnormality of the NADH oxidase components, particularly cytochrome b558 (gp91phox and p22phox) in membrane and cytosol factors p47phox, p67phox, p40phox and others in neutrophil. Thus, irregular regulation of transcription factors for gene expression of phox molecules and MPO permits susceptibility to Aspergillus and Candida infections.

[Synthesis of conjugates of L-lysine alpha-oxidase with antibodies].

The conjugation of the drugs with vector molecules enables to obtain therapeutic preparation, which may be transported to the selected target organ. In the present work the methods of conjugation of antineoplastic enzyme L-lysine alpha-oxidase with antibodies were elaborated. Conjugates were worked out through the attachment of amino groups on the antibody surface either with the aldehyde groups which were created in L-lysine alpha-oxidase molecule (0.2% of initial enzymatic activity) or with the aldehyde groups of cross-linking molecules. Maximal (78%) L-lysine alpha-oxidase activity in conjugates was observed when oxidized peroxidase which contained the aldehyde groups was used as crosslinking agent. The glutaraldehyde method yielded 70% of initial enzyme activity.

Evaluation of an automated glucose-oxidase procedure.

We evaluated a slight modification of the automated oxidase/peroxidase method for glucose determination in blood with use of 4-aminophenazone [J. Clin. Pathol. 22, 246 (1969)]. Nonsugar reducing substances did not interfere, except for ascorbic acid in very high concentrations. Lipemic sera gave slightly lower values. Values for reproducibility, specificity, and sensitivity were closer to such values for the hexokinase method than was the case for some other commonly used methods.

[A mechanized enzymic method for the determination of uric acid (author's transl)].

A continuous flow method is described for the determination of uric acid in serum using uricase and peroxidase, with omicron-dianisidine as the chromogen. The method can be used with single channel systems and with the multichannel system SMA 12/60 (Technicon). The new method was compared with six other manual and mechanized methods for the dosage of uric acid, and the results were analyzed statistically. The results from all the enzymic methods showed a good correlation, whereas the "chemical oxidation" methods showed systematic deviations.
