Analysis of eye lens-specific genes in congenital hereditary cataracts and microphthalmia of the miniature schnauzer dog.

The congenital hereditary cataracts and microphthalmia in the miniature schnauzer dog are inherited by an autosomal recessive mode. To understand the genetic basis of these diseases, the authors purified and analyzed leukocyte deoxyribonucleic acid (DNA) from affected and normal animals using a candidate gene approach. Because the genes that encode the lens-specific proteins, specifically, alpha, beta, and gamma crystallins and the membrane protein (MP26), are known to maintain the structure and function of the lens, the authors used complimentary DNA (cDNA) fragments that corresponded to the above genes to search for the mutations at their loci in the affected animals. They found no evidence of the gene deletion and rearrangement in any of the five loci. In addition, the hybridizable sequences of the dog DNA to the specific probes for the human chromosome 4 and 18 loci, which are reported to be involved in the abnormality of the human eye, seem to be unaffected. These data support the notion that the hereditary cataracts and microphthalmia in the dog may be associated with genes other than those reported for several animal systems.

A miniature voltmeter for monitoring small amplitude electrical activity in the hypothermic arrested myocardium.

A measurement system which is capable of detecting microfibrillation in the hypothermic, arrested heart has been developed. It is shown that this system can reliably measure microfibrillation signals as low as 10 microV in amplitude. Using this system, it has been shown that even after the heart has been rendered visibly arrested using cold potassium, it may still be producing a low level of electrical activity. It has also been shown that the presence of this microfibrillation during the period of the cold potassium administration and visible arrest, is very highly correlated with the decrease in high energy phosphates (ATP), mitochondrial injury, and a decreased cardiac function post-operatively. Using the information obtained from this first measurement system, a miniature, battery powered voltmeter has been designed and fabricated. This voltmeter has been tested using the initial measurement system as reference and produces the same information concerning the small signal electrical activity (i.e.: same waveform and RMS value). Because this voltmeter is battery powered and is, therefore, completely isolated from all electrical equipment in the operating theatre, it can be used to monitor the presence, or absence, of microfibrillation in the human heart. Of greatest importance, however, is the fact that the voltmeter can serve as a monitoring device which will indicate when the potassium should be administered in order to render the heart both mechanically and electrically arrested.

[The effect of "nonquantal" acetylcholine on miniature end-plate currents in the mouse].

The development of postsynaptic potentiation (PSP) and desensitization (DS) under the nonquantal secretion (NS) of acetylcholine (ACh) was studied using characteristics of miniature end-plate currents (MEPCs) in mice diaphragm muscle. The H-effect (the measure of the NS level) fell to zero during 3 hours after acetylcholinesterase (AChE) inhibition by armine at 20 degrees C. The MEPCs decay time constant (tau) decreased in parallel with the H-effect, though there was no reduction of the MEPCs amplitude in this case; tau did not change when NS was absent (early denervation). The maximal prolongation of tau after AChE inhibition was the same both in innervated (NS was normal) and denervated (NS was absent) muscle. The reduction of tau accelerated with the temperature rise and occurred after AChE inhibition by neostigmine. There was no changes in tau when AChE was active. It is suggested that nonquantal ACh decreases tau due to the DS development. NS induces no significant PSP after the AChE inhibition, but can do it later according to DS development, so that signs of DS may partially be masked by PSP.

[The characteristics of the action of calcium ions on miniature end-plate currents after the disruption of mediator hydrolysis].

Effect of calcium on the miniature end-plate currents (MEPC) at the frog neuromuscular junction was studied by the voltage-clamp technique. Rise of the calcium concentration in the Ringer solution up to 9 mmol/l caused a decrease of the MEPC amplitude which was related to the reduction of the end-plate channel conductance. Calcium had no effect on the time course of MEPCs at the active acetylcholinesterase (AChE) but accelerated MEPC decay by 26% after AChE inhibition by neostigmin or armin. It is supposed that the shortening effect of calcium on the MEPC decay phase is based on the ability of calcium to modulate the block of ionic channels by acetylcholine or to accelerate the process of desensitization of the postsynaptic membrane.

Postnatal ossification centers of the atlas and axis in miniature schnauzers.

Postnatal ossification of the atlas and axis was studied in Miniature Schnauzers by examining alizarin-stained bone clearings, cleaned dry bones, radiographs, histologic sections, and arterially perfused bone clearings. Sixty-two pups (1 day to 16 weeks old) and 4 adults were examined. In 1-day-old pups, the atlas consisted of 3 separate ossification centers: a left and right neural arch center and midventrally, the intercentrum 1, which formed the body of the atlas. The axis contained 4 separate ossification centers: a left and right neural arch center; centrum 2 in the main part of the axis body; and centrum 1, which formed the caudal part of the dens and the cranial part of the axis body. By 6 weeks of age, the epiphysis on the caudal end of the axis body had begun to ossify. At this time, the intercentrum 2, which developed as a cuboidal ossification center intercalated between centrum 1 and centrum 2 in the middle of the cranial half of the axis body, also began to ossify. The centrum of the proatlas, which formed the apex of the dens, was first seen ossified in a 9-week-old pup. These 10 ossification centers were seen as constant and separate elements. In all dogs, the dens developed from 2 separate ossification centers: the centrum of the proatlas formed the cranial one-quarter, and centrum 1 formed the caudal three-quarters. Dens dysplasia is unlikely to be a result of failure of development of one of the ossification centers for the dens.(ABSTRACT TRUNCATED AT 250 WORDS)

[Atypical miniature end plate currents in neuromuscular synapses of the rat under normal conditions, after acetylcholinesterase inhibition and cooling].

In the end-plates of rat diaphragm among atypical miniature end-plate currents (MEPCs) 2.9% were giant and 5.1% were slowly rising. The frequency of the giant MEPCs was decreased when temperature was lowered and increased when acetylcholinesterase (AChE) was inhibited; the latter effect was reversed if d-tubocurarine was added. Frequency of the slowly rising MEPCs changed insignificantly by all conditions. It is suggested that a highly temperature-dependent presynaptic mechanism of giant MEPC generation does exist which is activated by acetylcholine (ACh). Data about changes in the time course of the slowly rising MEPCs by AChE inhibition and lowering of temperature make it possible to suggest that the slowly rising MEPCs may be accounted for either slow release of ACh quanta or release of quanta on large distances from synaptic cleft and postsynaptic cholinoreceptors. The latter is possible if ACh quanta are released from synaptic Schwann cell to periaxonial space.

[The value of miniature compression screws in maxillofacial surgery].

According to the well-known principle of compression, a new kind of screw has been developed. It is a development of the miniaturized Michelet-Champy osteosynthesis device which we have used for the last 10 years in maxillofacial surgery. These traction-screws were initially elaborated for rigid stabilization after sagittal ramus osteotomies such as Obwegeser-Dal Pont. Usually, three screws are inserted at each side, via a transjugal approach. It is then possible to remove the intermaxillary fixation device at the end of the surgical procedure. However, these screws are also useful in other cases: for other kinds of mandibular osteotomies (visor osteotomy for instance), in traumatology (oblique mandibular or condylar fractures) or for fixation of onlay bone grafts on the facial structures. When iliac bone grafts, which are essentially cancellous bone are used, it is possible to use a small metallic ring to avoid penetration of the head of the screw into the graft. The screws can be inserted by a cutaneous punctiform incision when it is impossible to pass the screwdriver through the buccal incision. In all cases, this material is particularly easy to insert, allows good fixation, permits a solid interosseous compression, and is particularly well tolerated. The main drawback is the difficulty of choosing the right length of the screws because only two lengths are available at the present time. Despite this problem, these mini traction-screws seem to be useful for maxillofacial surgeons.

[An attempt to estimate various characteristics of neuro-muscular transmission from the rising phase of miniature end-plate currents].

A simple expression describing the relation between the duration of rising phase and amplitude of miniature end-plate currents (MEPC) is suggested. Parameters of MEPC of the rat diaphragm with different degrees of acetylcholinesterase inhibition by galanthamine were used to estimate the lower levels of acetylcholine diffussion coefficient (0.86.10-6 cm2/s) and channel opening rate constant (21000 s-1).

[Effect of antibodies to glial glycolipid antigens on miniature end-plate potentials].

Less than 4% of miniature end-plate potentials (MEPPs) in frog cutaneous-pectoral muscles, normal or exposed to nonimmune rabbit serum were atypical. These MEPPs were of normal amplitude, but their time course was nearly twice as slow as that of normal MEPPs. The exposure of muscles to rabbit anti-galactocerebroside serum induced more than three-fold increase in the fraction of atypically slow MEPPs. Inhibition of acetylcholinesterase caused an appearance of giant MEPPs, the fraction of slow MEPPs being unchanged. Possible mechanisms of increase in fraction of atypical slow MEPPs based on modification of the synaptic Schwann cell functioning are discussed.

[Hematologic indicators in patients with morbid obesity after surgical creation of the miniature stomach].

Surgical establishment of miniature stomach was conducted in 28 patients with severe alimentary-constitutional obesity (mean body weight 155.7 kg). After a 18-36-month follow-up the patients underwent hematological examination. It was found that a 134% excess of body weight registered preoperatively dropped to 54%. Reduced levels of blood red cells, hemoglobin, serum iron were not registered. Neither was anemia. This shows that physiological function of the whole gastrointestinal tract was not damaged, the blood loss appeared insignificant. Substitution therapy with hemostimulants during the following two years may be avoided.

[Postsynaptic potentiation of miniature currents of muscle fiber endplates in the rat diaphragm. The effect of an acetylcholinesterase inhibitor, temperature and curare].

Miniature end-plate currents (MEPC) were recorded in voltage-clamped end-plates of the rat diaphragm. A positive correlation between the amplitude and half-decay time of individual MEPC was found in control, after acetylcholinesterase (AChE) inhibition the correlation increased: correlation coefficients--0.29 and 0.49, respectively (28 degrees C). Addition of curare after the AChE inhibition caused a decrease in the amplitude and duration of MEPC, but had no effect on the correlation between these parameters. Cooling down to 18 degrees C led to the essential reduction of this correlation in control as well as after the AChE inhibition. Effect of the prolongation of the half-decay time due to the AChE inhibition was more significant at 28 degrees C than at 18 degrees C. The Q10 value for duration of the rising MEPC phase (about 2) was less than that for the half-decay time (about 3). The role of factors determining the MEPC duration is discussed. The results are explained by the postsynaptic potentiation in terms of the cooperative action of agonists on cholinoreceptors and of peculiarities of the acetylcholine diffusion from the synaptic cleft.

[Characteristics of the time course of miniature endplate currents in different parts of the neuromuscular junction in the frog].

Extracellular glass microelectrodes were used for recording miniature endplate currents (MEPC) from proximal and distal parts of frog sartorius synapse. The amplitude and half-time of decay (t1/2) of MEPC in proximal parts exceeded those in distal parts. These differences were eliminated by tubocurarine and increased under the influence of armin. In 80% of experiments the positive correlation between the amplitude and t1/2 of MEPC was revealed, which reflects repetitive binding of acetylcholine (ACh) molecules with cholinoreceptors (ChR). Positive correlation was observed in all investigated proximal parts of terminal, and only in half of distal ones. The electron microscopic study showed that postsynaptic folds in proximal parts occur more frequently, are poorly branching and rather shallow. A conclusion is made that the more prolonged time course of MEPC in proximal synapse parts is due to the more pronounced repetitive binding of ACh molecules with ChR of the postsynaptic membrane, which may be explained by high density of ChR and special, features of the synaptic cleft geometry in proximal parts.

[Effect of hyaluronidase on the end-plate miniature currents and potentials in the frog].

The effect of 0.1% hyaluronidase on miniature end-plate potentials and currents (MEPP and MEPC) was studied in the frog cutaneous-pectoris muscle. The action of hyaluronidase on armin-pretreated muscles caused a decrease in the amplitude, duration of half-decay time and rising phase of MEPPs and MEPCs. The positive correlation between the amplitude and half-decay time of MEPPs and MEPCs was diminished. Hyaluronidase treatment of preparations with active acetylcholinesterase caused the half-life time of MEPCs to increase without any changes in the amplitude and rising phase of MEPCs. It is suggested that enzymatic destruction of a part of the glycocalix of cells forming the neuromuscular junction and of a part of the extracellular matrix results in a weakening of the nonspecific acetylcholine binding, thus facilitating the acetylcholine diffusion into the synaptic cleft.

[Miniature currents of the endplates of the muscle fibers of the diaphragm of the rat after inhibition of acetylcholinesterase with galanthamine].

Miniature end-plate currents (MEPC) in rat diaphragm were studied with voltage-clamp technique when synaptic acetylcholinesterase (AChE) was inhibited with different concentrations of galanthamine. The MEPC amplitude and time course were increased progressively with galanthamine concentrations in the range of 3.16 X 10(-8) - 10(-6) g/ml. The decay of MEPC was always exponential. The input resistance of muscle fibres increased. Galanthamine (10(-5) g/ml) produced a curare-like action: the amplitude and duration of MEPC were less as compared with those at galanthamine concentration 10(-6) g/ml, the decay of MEPC became biphasic. During washing out of the drug, the duration of MEPC began to increase and then to diminish, returning to the initial value 3 hours later. The decay of MEPC became exponential. A positive correlation was found between half-decay time and amplitude of MEPC both in the presence and in the absence of anticholinesterase. It is supposed that the functional role of synaptic AChE in limiting the postsynaptic effect of acetylcholine is not so significant as it is usually considered, therefore it is possible to use the parameters of MEPC for the estimation of functional AChE activity.

[Tetanic potentiation of miniature end-plate potential frequency at frog neuromuscular junction in manganese solutions].

Using a sciatic nerve-sartorius muscle preparation of the frog, we have studied the effect of Mn on the increase in miniature end-plate potential (MEPP) frequency that occurs during tetanic stimulation (50 Hz, 2-3 min) of the nerve in nominally Ca2+-free, Mn2+ and Mg2+ solutions. During stimulation the frequency increased over the first minutes to reach an asymptote. The time course for the increase was analyzed following the model proposed by Barton, Cohen and Van der Kloot (1983). The ratio of the Ca2+ bound to the receptor at intervals during the tetanus, b, to the Ca2+ bound before stimulation was begun, b0, was calculated from MEPP frequencies. b/b0 indicates changes in intraterminal Ca2+ concentration produced by the tetanus. In solutions made with no added Ca2+ but containing 1 mM MgEGTA, the increase in b/b0 during stimulation showed a linear or convex time course. Similar time courses were obtained in solutions containing Mn2+ or Mg2+ as the sole divalent cation. On the other hand, when solutions contained Ca2+, the time course for the increase followed a sigmoidal curve. The present results suggest that Mn2+ enters the nerve terminal during stimulation and raises the intracellular Ca2+ concentration, which in turn promotes transmitter release.

[Analysis of a model of the miniature endplate current].

The numerical model of the miniature end-plate current MEPC described earlier was systematically analyzed to obtain an optimal set of parameters. This set permits simulating a number of experimental effects: time course of normal MEPC, cholinesterase inhibition, alpha-bungarotoxin action, potential dependence of MEPC, etc. The time course of the simulated "giant" MEPC fitted the experimental data only partially. A good correspondence between the model and experimental data underlies the conclusion that this model reflects well the relative contribution of several processes to MEPC generation.

Enzyme immunoassays with the miniature centrifugal fast analyzer.

We studied the EMIT (Enzyme Multiplied Immunoassay Technique, Syva) procedures for the assay of phenytoin and phenobarbital in serum, adapting them to the miniature Centrifugal Fast Analyzer. For different concentrations of drug, each rate of reaction decreased continuously with time, tending to converge on a single common value. The rate was most affected by the concentration of drug almost immediately after the reagents were mixed, less so thereafter. The antibody evidently is present in sufficient excess to bind all the enzyme-labeled drug ordinarily present, but the antibody-bound enzyme was only 75% inhibited; this helps explain the appreciable residual activity when no drug is present. The reaction course was the same whether the serum and enzyme-labeled drug were added to the antibody sequentially or simultaneously, which suggests that antibody is bound to drug appreciably faster than to enzyme-labeled drug. The reaction rates 15 to 30 s after mixing were used as the measure of the drug concentrations. These results were confirmed by noting the rates at successive 15-s intervals. The analyzer yielded a run-to-run CV of 10% for phenobarbital at 30 mg/liter, and 9% for phenytoin at 15 mg/liter, as compared to the 15% quoted by Syva.
