64Cu-labeled folate-conjugated shell cross-linked nanoparticles for tumor imaging and radiotherapy: synthesis, radiolabeling, and biologic evaluation.

UNLABELLED: Long-circulating nanoparticles functionalized with ligands for receptors overexpressed by tumor cells have promising applications for active and passive tumor targeting. The purpose of this study was to evaluate 64Cu-radiolabeled folate-conjugated shell cross-linked nanoparticles (SCKs) as candidate agents to shuttle radionuclides and drugs into tumors overexpressing the folate receptor (FR). METHODS: SCKs were obtained by cross-linking the shell of micelles obtained from amphiphilic diblock copolymers. SCKs were then functionalized with folate, fluorescein thiosemicarbazide (FTSC), and 1,4,8,11-tetraazacyclotetradecane-N,N',N'',N'''-tetraacetic acid (TETA). The specific interaction of SCK-folate with the FR was investigated on KB cells. The biodistributions of 64Cu-TETA-SCK and 64Cu-TETA-SCK-folate were evaluated in athymic mice bearing small-size KB cell xenografts (10-100 mg), whereas the intratumor distributions were investigated by autoradiography in 0.3- to 0.6-g KB cell xenografts. RESULTS: A global solution-state functionalization strategy has been introduced for attaching optimum numbers of targeting and imaging agents onto the SCKs for increasing the efficiency of interaction with cell-surface receptors. Epifluorescence microscopy confirmed the specific interaction of FTSC-SCK-folate with the FR in vitro. 64Cu labeling of TETA-SCKs led to the radiolabeled compounds with 15%-20% yield and >95% radiochemical purity. The biodistribution results demonstrated high accumulation of 64Cu-labeled SCKs in organs of the reticuloendothelial system (RES) (56.0 +/- 7.1 %ID/g and 45.7 +/- 3.5 %ID/g [percentage injected dose per gram] in liver at 10 min after injection for folated and nonfolated SCKs, respectively) and a prolonged blood circulation. No increase of SCK tumor uptake deriving from folate conjugation was observed (5.9 +/- 2.8 %ID/g and 6.0 +/- 1.9 %ID/g at 4 h after injection for folated and nonfolated SCKs, respectively). However, tumor accumulation was higher in small-size tumors, where competitive block of SCK-folate uptake with excess folate was observed. Autoradiography results confirmed the extravasation of radiolabeled SCKs in vascularized areas of the tumor, whereas no diffusion was observed in necrotic regions. CONCLUSION: Despite high RES uptake, the evaluated 64Cu-labeled SCKs exhibited long circulation in blood and were able to passively accumulate in tumors. Furthermore, SCK-folate uptake was competitively blocked by excess folate in small-size solid tumors, suggesting interaction with the FR. For these reasons, functionalized SCKs are promising drug-delivery agents for imaging and therapy of early-stage solid tumors.

Thyroidal modulation following hypo- and hyper-thermia in the soft-shelled turtle Lissemys punctata punctata Bonnoterre.

The aim of the current study was to investigate the effects of ambient temperature on thyroid activity of the soft-shelled turtle (Lissemys punctata pucntata). Turtles exposed to low ambient temperature (10 degrees C for 15 days) showed a significant decrease in relative thyroid weight, follicular cell size (cell became squamous from cuboidal type) and epithelial height in both the peripheral and central follicles of the gland, with the appearance of homogeneous colloid materials in the follicular lumen. Thyroid peroxidase activity declined significantly. In contrast, high ambient temperatures (32/34 degrees C for 15 days) caused reverse changes to those observed after exposure with low ambient temperature. No significant difference was marked in thyroid activity between 32 and 34 degrees C temperature treatments. The findings provide evidence that low ambient temperature inhibits thyroid activity and high ambient temperature stimulates the gland activity in soft-shelled turtles. Ambient temperature acts presumably via the hypothalamo-hypophysial (TRF-TSH) axis which in turn alters thyroid function in turtles.

Synthesis and characterization of Fe/Nd2O3 core-shell nanoparticles by hydrogen plasma-metal reaction.

Fe/Nd2O3 core-shell nanoparticles (CSNs) with a mean diameter of 35 nm were produced successfully by using hydrogen plasma-metal reaction (HPMR) method. This core-shell structure was confirmed by high resolution transmission electron microscopy (HRTEM), energy dispersion X-ray spectroscopy (EDS), X-ray photoelectron spectral (XPS), and induction-coupled plasma (ICP) spectroscopy. The magnetic properties were measured by vibrating sample magnetometer (VSM). It was found that the mole ratio of Nd to Fe on the nanoparticle surface is 1.2:1, about 7 times of that of the whole nanoparticle. The saturation magnetization Ms and remanence Mr of Fe/Nd2O3 nanoparticles decrease prominently from Fe nanoparticles, whereas the coercivity Hc drops only less than 5% of Fe nanoparticle. These CSNs have potential applications in magnetic and catalytic fields.

The effect of sustained-release varnish of chlorhexidine in dental plastic shells on salivary Streptococcus mutans.

The suppression of S. mutans is a predominate factor in preventing tooth decay. Sustained-release-delivery varnish of chlorhexidine was applied to dental plastic shells and administered to eight volunteers to be worn while sleeping. Salivary bacterial samples were taken on days 0, 7, 14, 21, 28, 35. A statistically significant reduction in S. mutans counts was recorded during the course of the study while a non-significant reduction in the total bacterial counts was found. Our results indicate that the application of chlorhexidine in the form of slow-release varnish in plastic shells is an effective intraoral drug delivery system resulting in reduction of S. mutans.

[Non-bullous erythroderma (congenital) ichthyosiforme with perinuclear shells].

A congenital dermatosis with an unusual course is described in a 5-year-old girl. During the first months of life she was found to have erythrokeratodermatic plaques, which developed into a non-bullous (congenital) ichthyosiform erythroderma (CIE) within 1 year. Since then, the clinical picture has remained stable. Histologically, the upper epidermal layers showed ballooning and vacuolization of the keratinocytes, glycogen storage, and focal parakeratotic keratinization. Conspicuous ultrastructural alterations consisted in the formation of shells of fine-filamentous, cross-linked material of unknown nature, binuclear keratinocytes, perinuclear edema formation, and the occurrence of lipid droplets in the horny layer. The n-alkane value was within the normal range. Our histological and ultrastructural findings are largely in accordance with those in a case of "congenital reticular ichthyosiform erythroderma". The disorder seems to represent a special genotype in the heterogeneous group of the non-bullous CIEs. We suggest naming it "non-bullous (congenital) ichthyosiform erythroderma with perinuclear shells".

The structure of the shell and polar plugs of the egg of the whipworm, Trichuris trichiura (Nematoda: Trichuridae) from the Samango monkey (Cercopithecus albogularis).

The structure of the shell of the egg of Trichuris trichiura was examined using light microscopy as well as scanning and transmission electron microscopy. The results confirmed its three-layered structure and provided evidence that the cores of the polar plugs, which could be seen to be extensions of the shell's middle layer, could be lost en bloc, either mechanically or chemically, and in this way probably provided an exit for the first stage larva at hatching.

Peanut shell colitis.

A 40-year-old physician experienced abdominal pain, loose stools, hematochezia, and anal discomfort with defecation associated with the daily consumption of 15 to 30 whole peanuts, including the shells. Thorough evaluation revealed only nonspecific colitis of the distal portion of the sigmoid colon and inflamed hemorrhoids. Discontinuation of whole peanut ingestion was associated with symptomatic, endoscopic, and histological resolution. In this patient, undigested peanut shells seem to have caused a nonspecific colitis, perhaps as the result of mechanical abrasion of the colonic mucosa.

Immobilization and reproducibility in radiotherapy for cancer of the larynx and pharynx using simplified shell and bite-block.

In this paper we describe the comparative study concerning the immobilization during radiotherapy for cancer of the larynx and pharynx with or without shell and fixing device. Immobilization was continuously monitored by semi-conductor detector. Reproducibility was investigated by means of verification film. The result significantly indicated that better immobilization was obtained in the patients treated with use of the shell (p much less than 0.001). Reproducibility of the field arrangement was also sufficient throughout the course of radiotherapy.

[Morphological characteristics of the shell of the ova of Capillariidae (Nematoda, Capillariidae)].

Peculiarities of the surface architectonics of the egg shell in 18 species of capillariids (Nematoda, Capillariidae) were studied. Capillariids were found in mammals of 5 orders: Insectivora, Chiroptera, Muridae, Carnivora and Arctiodactyla. A correlation between the structure of the external surface of egg shell of these nematodes on one hand, and localization in the organism of the definitive host and its biology on the other, was established. On the basis of these characters capillariids of the genera Capillaria, Eucoleus, Calodium, Liniscus, Thominx and Scrjabinocapillaria were arranged into 6 groups: 1 - the genus Capillaria (luman of the stomach and gut), 2 - the genus Eucoleus (mucous membrane of oesophagus and stomach), 3 - the genus Calodium (parenchima of the liver), 4 - the genus Liniscus (urinary bladder), 5 - the genus Thominx (lungs), 6 - the genera Thominx and Scrjabinocapillaria (lumen of the stomach). The classification suggested makes more precise and supplements the known systems of nematodes of the fam. Capillariidae.

Application of a fluid-filled spherical sandwich shell as a biodynamic head injury model for primates.

In this paper, two areas are investigated: first, the effect of pulse duration on skull fracture and brain injury and, second, under what conditions a scaling can be considered between subhuman primates and human. The skull is modeled as a spherical sandwich shell representing the general structure of the skull bones as being the compact inner and outer tables separated by a porous diploë layer. The brain and surrounding cerebrospinal fluid are modeled as an acoustic fluid with properties similar to those of water. The fluid-shell system is subjected to a local axisymmetric and arbitrary time-dependent external pulse and the transient response of the system is determined by numerical methods. The effect of pulse duration is investigated by choosing the pulse as the Heaviside function. Pulses with short duration are found to be less damaging as far as skull fracture and brain injury are concerned. Human head impact tolerance curves are obtained. For the second part of the investigation, a more realistic pulse containing both exponential and sine components is chosen to study the responses of the models of the human head and four species of primates. The numerical results are obtained for the same magnitude of the pulse as well as for equal magnitudes of initial acceleration after impact.

Ultrastructural observations on the chorionic epithelium, parathyroid glands and bones from chick embryos developed in shell-less culture.

Three-day-old chick embryos were cultured in Petri dishes until they reached developmental stages 37 or 39 (Hamburger & Hamilton, 1951). The electron microscopical study of the chorio-allantoic membranes showed that, even in the absence of the shell and the corresponding calcium supply, 'capillary-covering' and 'villus-cavity' cells differentiated well. The parathyroid glands from the cultured embryos showed ultrastructural signs indicative of active synthesis and secretion of the parathyroid hormone. This correlates well with the significant hypocalcemia (5.4 mg/100 ml) observed in these embryos. In all cultured embryos mineralization of bones was greatly reduced as shown by alizarin bulk staining and confirmed by histological and electron microscopical analysis. The ultrastructural characteristics of osteoblasts and osteocytes, as well as of the bone matrix, appeared normal. The defect in mineralization appeared thus to be due to a deficiency in the availability of calcium and not to a delay in bone differentiation. This implies that the yolk sac appears to lack calcium regulatory capacity since it cannot compensate for the absence of shell by increasing its own contribution in order to assure an adequate mineralization of the embryonic bones.

Evaluation of sampling plans used in the United States, United Kingdom, and The Netherlands to test raw shelled peanuts for aflatoxin.

The United States is a large producer and exporter of peanuts. The United Kingdom and The Netherlands are major importers of U.S. peanuts. Each country has a different guideline or legal limit for peanut products containing aflatoxin. Peanuts are tested for aflatoxin in each country by using specifically designed aflatoxin sampling plans to determine if the aflatoxin concentration in a lot of raw shelled peanuts is less than the guideline or legal limit. For raw shelled peanuts, the U.S. plan has the highest sample acceptance limit of 15 ng total aflatoxin/g, the UK plan has a sample acceptance limit of 10 ng total aflatoxin/g, and the Dutch Code of Practice (called the Dutch plan) has the lowest sample acceptance limit at 3 ng aflatoxin B1/g. The U.S. plan uses a maximum of 3 sampling units, each weighing 21.8 kg; the UK plan uses a single sampling unit of 10 kg; and the Dutch plan uses 4 sampling units, each weighing 7.5 kg. The sampling variance is lowest for the U.S. plan and highest for the Dutch plan. The sample preparation variance is lowest for both the Dutch and UK plans and highest for the U.S. plan, primarily because of the mill type used to comminute the kernels in the sample. For a given distribution among lot concentrations, the U.S. plan accepts the greatest number of lots and the Dutch plan rejects the greatest number of lots. The average aflatoxin concentration among accepted lots is highest for the U.S. plan and lowest for the Dutch plan.(ABSTRACT TRUNCATED AT 250 WORDS)

Production of chitinase by Pseudomonas aeruginosa K-187 using shrimp and crab shell powder as a carbon source.

Shrimp and crab shell powder prepared by treating shrimp and crab processing waste with boiling and crashing was used as a substrate for isolating alkali-tolerant chitinolytic microorganisms. Strain K-187 appeared to be the chitinase-producing strain with the most potential. The organism was identified as a strain of Pseudomonas aeruginosa. Maximum chitinase activity was obtained when the strain was grown aerobically in a medium consisting of 3.0% shrimp and crab shell powder, 0.1% CMC, 0.1% (NH4)2SO4, 0.1% K2HPO4, 0.1% MgSO4.D7H2O and 0.1% ZnSO4 (pH 9), at 45 degrees C after 3 days. The optimum pH and temperature of the enzyme reaction were 7 and 40 degrees C, respectively. The chitinase was stable at pH from 5 to 10 and was stable under 60 degrees C.

[Influence of heating degrees on internal quality of calcined shell drugs].

The internal quality of 4 kinds of shell drugs calcined under different conditions was investigated and compared in terms of the yielding ratios and total calcium content of the calcined product, Ca2+ content of the decoction and total water-soluble extractive rate. It was found that at calcining temperatures higher than 800 degrees C the internal quality of the shell drugs would change markedly.

Drugs from the peptide venoms of marine cone shells.

Australian cone shell venoms are being investigated as an exciting new source of bioactive peptides as part of a new collaborative project between the 3D Centre and AMRAD. Initial studies have already revealed a number of new and novel acting peptides amongst the hundred or so small, heavily constrained peptides present in the venom of each cone shell. The aim of the project is to develop peptidomimetic drugs based on a selection of these native peptides.

Glaucoma filtration surgery and the shell tamponade technique.

1. Excessive outflow of aqueous through the newly formed sclerostomy is the key factor in precipitating hypotony and other serious complications. 2. Glaucoma shell tamponade is used to temporarily impede the outflow of aqueous at the filtration site in order to allow the eye to stabilize. 3. The "torpedo" dressing is critically important to the success of the shell tamponade.

The distribution of calcium salt precipitates in the core, periphery and shell of cholesterol, black pigment and brown pigment gallstones.

Calcium bilirubinate, palmitate, carbonate and phosphate have been identified in the cores of cholesterol and pigment gallstones, suggesting a role for precipitated calcium salts in the early events of gallstone formation. Previous studies that compared the calcium salt contents of cholesterol and pigment stones required destruction of gallstone structure. We have used scanning electron microscopy with windowless energy-dispersive x-ray microanalysis to determine the prevalence of calcium salts in a series of cholesterol (n = 105), black pigment (n = 35) and brown pigment (n = 6) gallstones obtained from 146 consecutive patients undergoing cholecystectomy. These techniques provide specific identification of cholesterol and individual calcium salts as they occur within the core, periphery and shell of gallstones without destroying stone structure. Calcium precipitates more than 0.5 micron in diameter can be detected in a cholesterol background at a detection limit of 0.01% by weight. Calcium salts were detected in the centers of 88% of cholesterol and 100% of black (p < 0.05 vs. cholesterol) and brown pigment stones. Calcium bilirubinate was identified in the cores of 54% of cholesterol and in all pigment stones (p < 0.001 black pigment vs. cholesterol). Calcium palmitate was detected in all brown pigment stones, in 39% of cholesterol stones (p < 0.001 vs. brown pigment) and in 31% of black stones (p < 0.01 vs. brown pigment). Peripheral calcium salts were detected less in cholesterol (19%) than in black or brown stones (100%, p < 0.05). Fourteen percent of cholesterol and black pigment stones were surrounded by shells containing mostly calcium carbonate.(ABSTRACT TRUNCATED AT 250 WORDS)
