Capsid protein VP1 deletions in JC virus from two AIDS patients with progressive multifocal leukoencephalopathy.

PCR on 52 cerebrospinal fluid (CSF) specimens and 33 brain biopsies obtained from HIV-1 positive patients utilized pairs of primers from both the early region (JTP) and late region (JLP). In these patients, in whom progressive multifocal leukoencephalopathy (PML) was suspected on the basis of clinical symptoms and magnetic resonance imaging (MRI) studies, eight CSFs (15%) and 14 brain biopsy specimens (42%) contained JCV DNA sequences. In two patients' samples, the CSFs were positive for JCV DNA in the VP1 region using the primer pair for the VP1 region (JLP), but the fragment amplified migrated more rapidly than the 129-bp product obtained from prototype JCV(Mad-1) or the fragment amplified from the antigenic variant of JCV known as Mad-11. These patients died 3-4 months after onset of progressive neurological symptoms. Cycle sequencing of the fragments revealed overlapping deletions of 24 and 27 nucleotides. These strains were of different genotypes, designated strain 107 and strain 206. Computer analysis of the VP1 amino acid sequence predicts that the eight or nine amino acid residue deletions represent a surface loop with a high antigenic index. These naturally occurring deletion mutants are the first examples of a phenomenon observed experimentally in the mouse polyoma virus capsid protein VP2.

Interaction of Heliothis armigera nuclear polyhedrosis viral capsid protein with its host actin.

In order to find the cellular interaction factors of the Heliothis armigera nuclear polyhedrosis virus capsid protein VP39, a Heliothis armigera cell cDNA library was constructed. Then VP39 was used as bait. The host actin gene was isolated from the cDNA library with the yeast two-hybrid system. This demonstrated that VP39 could interact with its host actin in yeast. In order to corroborate this interaction in vivo, the vp39 gene was fused with the green fluorescent protein gene in plasmid pEGFP39. The fusion protein was expressed in the Hz-AM1 cells under the control of the Autographa californica multiple nucleopolyhedrovirus immediate early gene promoter. The host actin was labeled specifically by the red fluorescence substance, tetramethy rhodamine isothicyanete-phalloidin. Observation under a fluorescence microscopy showed that VP39, which was indicated by green fluorescence, began to appear in the cells 6 h after being transfected with pEGFP39. Red actin cables were also formed in the cytoplasm at the same time. Actin was aggregated in the nucleus 9 h after the transfection. The green and red fluorescence always appeared in the same location of the cells, which demonstrated that VP39 could combine with the host actin. Such a combination would result in the actin skeleton rearrangement.

[Expression of baculovirus recombinant HPV16L1 and L2 capsid proteins in insect cells].

BACKGROUND: To express capsid proteins of HPV16 in insect cells system. METHODS: Recombinant baculovirus stocks were generated by cotransfection with baculovirus DNA and baculovirus recombinant transfer plasmids. The expression of aimed proteins in insect cells were confirmed by using SDS-PAGE and Western blot. RESULTS: Two stable strains of recombinant baculovirus expressing HPV16L1 protein alone and co-expressing HPV16L1 plus L2 proteins were obtained. The L1 and L2 ORFs of HPV16 produced proteins of 57000 and 97000, respectively. The yield of L1 protein was about 25%-30% of the total proteins of the insect cell. CONCLUSIONS: L1 and L2 proteins of HPV16 could be expressed with high efficiency in insect cells via recombinant baculoviruses.

Significance of plasma IgA and IgG antibodies to Epstein-Barr virus early and viral capsid antigens in Thai nasopharyngeal carcinoma.

Epstein-Barr virus (EBV) is an important causal factor of human nasopharyngeal carcinoma (NPC). High levels of serum IgA and IgG antibodies to EBV early and viral capsid antigens (IgA/EA, IgA/VCA, IgG/EA and IgG/VCA) have been reported in NPC patients. Since specific serum IgA/EA, IgA/VCA and IgG/EA are claimed to be useful serological markers for NPC. In order to evaluate whether plasma IgA/EA, IgA/VCA, IgG/EA and IgG/VCA antibody levels are useful markers for diagnosis and prognosis of Thai NPC, we examined the prevalence of these antibodies in 79 NPC patients, and 127 age-matched controls (47 healthy subjects (HS), 32 cases of other disease (OD) and 48 cases of other cancer (OC)) by using an indirect immunofluorescence assay. The prevalence of plasma IgA/EA, IgA/VCA, and IgG/EA in NPC patients (55.7, 68.4 and 68.4%) was significantly higher than in the HS (0.0, 0.0 and 20.5%,), OD (0.0, 0.0 and 3.1%) and OC (0.0, 0.0 and 20.8%) groups (p<0.05). The prevalence of plasma IgG/VCA in NPC patients (93.7%) was significantly different from those for the OD and OC groups (71.9 and 43.8%) but not for the HS group (89.4%). In NPC patients, the geometric mean titers (GMT) of plasma IgA/EA, IgA/VCA and IgG/EA were increased with an advanced clinical stage of disease but not IgG/VCA. In contrast, GMT of IgG/VCA was increased with aggressive type of disease (histological type) but not IgA/EA, IgA/VCA, and IgG/VCA. The results of our study suggest that plasma IgA/EA, IgA/VCA and IgG/EA antibodies may be useful markers for diagnosis and assessing prognosis of Thai NPC.

[IgA antibodies against Epstein-Barr virus capsid antigen].

The authors describe infection with the Epstein-Barr virus (EBV) which is associated with the formation of various types of antibodies--heterophil (HP) antibodies as well as antibodies against individual EBV antigens. They evaluate the contribution of a recently elaborated and introduced method used for detection of antibodies against the capsid antigen of the EBV class IgA (anti-VCA IgA) by indirect immunofluorescence to the diagnosis of acute and chronic EBV infections. Anti-VCA IgA are useful in the diagnosis of EBV infections if the patient has no longer HP and anti-VCA IgM, but only anti-VCA IgG. If, however, in addition to anti-VCA IgA also anti-VCA IgA are present, their presence can be the only detectable sign of a recent EBV infection and also a sign of reactivation of chronic EBV infection.

[A comparison of the detection of IgM antibodies against the viral capsid antigen (VCA) of EBV (Epstein-Barr virus) in various groups of patients, using indirect immunofluorescence, indirect ELISA and reverse ELISA. Study of the diagnostic efficacy of the anti-VCA EBV IgM ELISA-VIDITEST kit].

OBJECTIVES: To test diagnostic efficiency of a novel method for detection of IgM antibodies to VCA EBV. To compare sensitivity and specificity of detection of IgM antibodies to VCA EBV from patients at various stages of EBV infection by various serological methods. MATERIAL AND METHODS: IgM antibodies to VCA EBV were detected using IgM ELISA Viditest anti-VCA EBV IgM assay (Vidia, Ltd., Czech Republic) and comparative serological methods (indirect immunofluorescence assay, indirect ELISA (Human, SRN) and reverse ELISA (DiaSorin, Italy) in four independent diagnostic laboratories on panels of sera from 1) infectious mononucleosis patients, 2) patients with serological markers of EBV reactivation, 3) seropositive individuals lacking serological markers of active EBV infection, 4) seronegative individuals, 5) patients with IgM antibodies against another herpesvirus and 6) patients positive for rheumatoid factor. The sera yielding discrepant results were retested in the reference laboratory using the reference methods (indirect immunofluorescence and reverse ELISA). Overall, 854 IgM anti-VCA-positive or -negative sera were evaluated. RESULTS: The sensitivity and specificity of the IgM Viditest anti-VCA EBV assay were 94.7 and 96.1 %, respectively. All of the three tests compared were similarly reliable in detecting IgM anti -VCA EBV antibodies in the samples from infectious mononucleosis patients. On the other hand, indirect fluorescence assay proved clearly superior to ELISAs for detection of IgM anti-VCA antibodies in the samples with serological pattern of EBV reactivation, typically showing significantly lower IgM anti-VCA titers compared with those from primary infection. CONCLUSIONS: The multilaboratory comparative study proved that the novel IgM ELISA-Viditest anti-VCA EBV assay (Vidia, Ltd., Czech Republic) which shows comparable diagnostic efficiency for detection of IgM EBV antibodies to viral capsid antigen (VCA) as compared to the other assays tested, i.e. ELISA (Human, Germany) and reverse ELISA (DiaSorin, Italy), is suitable for use in serological diagnosis of EBV.

High prevalence of immunoglobulin A antibody against Epstein-Barr virus capsid antigen in adult patients with lupus with disease flare: case control studies.

OBJECTIVE: Systemic lupus erythematosus (SLE) is a severe autoimmune disease with rare remission and recurrent flare. Epstein-Barr virus (EBV) infection has been reported to be strongly associated with SLE in the United States, but with an inconclusive role in Asia. We investigated the role of EBV infection in patients with SLE in Taiwan, with one of the highest population densities in Asia. METHODS: We conducted case-control studies to test whether EBV infection was associated with adult SLE in Taiwan. In the first study, 36 adults with SLE and 36 sex and age matched controls were enrolled for examination of serum IgG, IgM, and IgA antibody against EBV-virus capsid antigen (EBV-VCA). In the second study, another 36 adult lupus cases and 36 matched controls were enrolled to confirm the high prevalence of IgA antibody against EBV-VCA found in the first study. Further, both groups of SLE patients were combined to analyze the association between the existence of IgA antibody against EBV-VCA and disease activity (determined by SLEDAI score) and disease flare in patients with SLE. RESULTS: In the first study, IgA antibody against EBV-VCA was the only marker with significantly higher prevalence in adults with SLE compared to healthy adults (36.1% vs 5.6%; p < 0.005). In the second study, we confirmed that the prevalence of IgA antibody against EBV-VCA was indeed higher in adults with SLE (38.9% vs 2.8%; p < 0.001). With further analysis (pooling analysis), adult SLE patients with IgA antibody against EBV-VCA had higher disease activity compared to SLE patients without IgA antibody against EBV-VCA (SLEDAI 7.8 +/- 6.6 vs 3.3 +/- 2.1; p < 0.001). SLE patients with flare showed much higher prevalence of IgA antibody against EBV-VCA compared to those without flare (81.3% vs 25.0%; p < 0.001). CONCLUSION: This is the first evidence that IgA antibody against EBV-VCA is strongly associated with disease flare in SLE patients. It suggests that EBV reactivation may contribute toward the disease flare of SLE.

[Evaluation of IgG and IgA type antibodies to capsid and early antigens of the Epstein-Barr virus in the diagnosis of nasopharyngeal carcinoma].

BACKGROUND: The Epstein-Barr virus (EBV) is related with nasopharyngeal carcinoma (NPC) in that, in this neoplasm, high levels of antibodies are found vs different EBV antigens. METHODS: The determinations of IgG and IgA type antibodies were evaluated vs capsid antigens (VCA) and early antigen (EA) of the EBV by indirect immunofluorescence in 14 patients diagnosed with NPC, in 12 patients with other tumors of the head and neck and in 61 blood donors. RESULTS: The detection of IgA type antibodies with a sensitivity and specificity of 100% and 92% for the VCA antigen and of 86% and 100% for the EA antigen are the most useful tests in the early diagnosis of nasopharyngeal carcinoma. The determination of IgG type antibodies were of a more limited usefulness with sensitivities and specificities of 86% and 67% for IgG anti-VCA and 100% and 83% of IgG anti-EA. CONCLUSIONS: The detection of antibodies vs VCA and EA antigens is useful in the differential diagnosis of NPC with other tumors of the head and neck.

Nucleotide sequences of the VP1 capsid proteins of wild poliovirus types 1 and 3 from epidemic areas of Brazil.

The nucleotide sequences encoding the capsid protein VP1 were determined for the wild polioviruses of serotypes 1 and 3 endemic to the northeastern region of Brazil. Compared with the corresponding Sabin vaccine strain sequences, the wild isolates differed at 20% (type 1) and 22% (type 3) of their nucleotide positions, and in 7% (type 1) and 11% (type 3) of their amino acid residues. The highest degree of amino acid heterogeneity occurred within the amino-terminal residues of the VP1 proteins. Intratypic amino acid differences also occurred in VP1 surface residues that form parts of antigenic sites for neutralizing antibodies.

[Expression of the Epstein-Barr virus P150 viral capsid antigen in Escherichia coli for the use as antigen in diagnostic tests].

We have attempted to produce the P150 viral capsid antigen of Epstein-Barr Virus (EBV) in Escherichia coli. This protein was found, by immunoprecipitation, to be a clinically relevant antigen, especially for the determination of the IgA-titer in patients with Naso Pharyngeal Carcinoma (NPC). Since the expression of the entire P150 coding region was unsuccessful, we synthesized only the antigenic parts of this protein. After purification of P150 expressed by PUR290CXH580 with column chromatography (sepharose 2B-CL), the resulting product reacts particularly well with IgA antibodies of NPC patients indicating its diagnostic value for NPC.

[Simultaneous detection of IgM antibodies against the hepatitis A virus and the viral capsid antigen of Epstein-Barr virus in acute hepatitis].

The simultaneous detection of IgM antibodies to hepatitis A virus (anti-HAV IgM) and IgM antibodies to viral capsid antigen (anti-VCA IgM) of Epstein-Barr virus (EBV) in patients with acute viral hepatitis has led us to systematically study serological markers of EBV in patients with anti-HAV IgM positive acute hepatitis and to test for anti-HAV IgM in sera of patients with acute hepatitis associated with serological evidence of current primary EBV infection. All patients studied were HBsAg negative and were not drug-addicts, nor homosexuals. In 15 consecutive patients with anti-HAV IgM positive acute hepatitis, anti-HAV IgM and anti-VCA EBV IgM antibodies were simultaneously detected in 9 cases. Of these 9 patients, antibodies to nuclear antigen were positive in 8 cases, antibodies to early antigen were positive in 7 cases and rheumatoid factor was positive in 4 cases. In 5 consecutive patients with acute hepatitis associated with serological evidence of current primary EBV infection, anti-HAV IgM was not detected. Simultaneous presence of anti-VCA EBV IgM, early antigen IgG antibodies and nuclear antigen antibodies in 7 patients with acute hepatitis associated with anti-HAV IgM suggests reactivation of EBV or reactivation of clones secreting antibodies anti-EBV in HAV infections. Furthermore, these results show that anti-VCA IgM only cannot be considered to be a specific marker of early EBV infection in patients with acute hepatitis.

[Characterization of antigenic types of circulating rotaviruses in Mendoza, Argentina based on typing of the external VP7 capsid protein].

Rotavirus is one of the most common etiologic agents of acute diarrhea in childhood. Understanding the immunologic mechanisms involved in rotavirus diseases, including knowledge on seasonal and geographic antigenic variations may be crucial for vaccine development. A monoclonal antibody based ELISA specific for antigenic domains on the outer capsid protein VP7 has been developed and used widely in the past years. We studied the rotavirus VP7-serotype epidemiology causing diarrhea in children who consulted at two main hospitals of Mendoza, Argentina over a 20 month period. A total of 227 cases of diarrhea were identified, 45 of which (20%) were rotavirus positive. We're able to serotype 43 viruses (96%), 42 VP7-type 1 and one VP7-type 3. The VP7-type 3 was detected towards the end of the second year, possibly representing a new incoming VP7-type. Three electropherotype patterns were identified, two corresponding to VP7-type epidemiology in Mendoza, Argentina seems to be characterized by a relatively homogeneous pattern of circulation with a strong predominance of VP7-type 1 viruses, at least during the 20 month period studied, in contrast to what has been reported in larger, more cosmopolitan cities like Buenos Aires.

Prevalence of IgA specific antibodies to Epstein-Barr virus capsid and early antigens in nasopharyngeal carcinoma.

Ninety-one patients with nasopharyngeal carcinoma (NPC), and 164 age-matched healthy controls were tested for presence of IgA antibodies to Epstein-Barr virus capsid antigen (VCA) and early antigen (EA) in their sera by indirect ELISA using "EBViral DETECT" commercial test kit. IgA anti-VCA was found in 76 (83.5%) of NPC patients and 16 (9.8%) of the controls. Meanwhile, IgA anti-EA was found in 72 (79.1%) of NPC patients and 21 (12.8%) of the controls. In a parallel study by indirect immunofluorescence test (IIF), IgA anti-VCA was found in 77 of 91 (84.6%) NPC patients and 22 of 142 (15.5%) controls. The prevalence rates of anti-VCA as screened by ELISA and IIF were very similar suggesting that neither one of the two tests can be used alternatively depending on the purpose and facilities in each individual laboratory. IgA antibodies to VCA and EA were more prevalence in NPC patients than those in the controls, the finding which again supported the association between EBV and NPC as was suggested in many other reports.

[Expression of a gene encoding the porcine rotavirus VP7 capsid using a nondefective recombinant adenoviral genome].

A nondefective recombinant human adenovirus 5 (Ad5) carrying the gene encoding for the porcine rotavirus outer capsid protein VP7 in the E3 region of the Ad5 genome has been obtained. mRNA for VP7 in recombinant Ad5KV14/VP7 infected cells was demonstrated by means of RT-PCR. Radioimmunoprecipitation of cell extracts infected with Ad5KV14/VP7 on early (12 h p.i.) and late (24 h p.i.) stages of adenovirus infection with rabbit polyclonal antiserum raised to purified rotavirus virions revealed a recombinant protein possessing the same electrophoretic mobility, 37 kDa, as the native rotavirus K VP7. One-dimensional peptide mapping also demonstrated the identity of the recombinant and native VP7.

Construction of four double gene substitution human x bovine rotavirus reassortant vaccine candidates: each bears two outer capsid human rotavirus genes, one encoding P serotype 1A and the other encoding G serotype 1, 2, 3, or 4 specificity.

Previously, four human x bovine rotavirus reassortant candidate vaccines, each of which derived ten genes from bovine rotavirus UK strain and only the outer capsid protein VP7-gene from human rotavirus strain D (G serotype 1), DS-1 (G serotype 2), P (G serotype 3), or ST3 (G serotype 4), were developed [Midthun et al., (1985): Journal of Virology 53:949-954; (1986): Journal of Clinical Microbiology 24:822-826]. Such human x bovine reassortant vaccines should theoretically provide antigenic coverage for the four epidemiologically most important VP7(G) serotypes 1, 2, 3, and 4. In an attempt to increase the antigenicity of VP7-based human x animal reassortant rotavirus vaccines which derive a single VP7-encoding gene from the human strain and the remaining ten genes from the animal strain, we generated double gene substitution reassortants. This was done by incorporating another protective antigen (VP4) of an epidemiologically important human rotavirus by crossing human rotavirus Wa strain (P serotype 1A), with each of the human x bovine single VP7-gene substitution rotavirus reassortants. In this way four separate double gene substitution rotavirus reassortants were generated. Each of these reassortants bears the VP4-encoding gene from human rotavirus Wa strain, the VP7-encoding gene from human rotavirus strain D, DS-1, P, or ST3, and the remaining nine genes from bovine rotavirus strain UK. The safety, antigenicity, and protective efficacy of individual components as well as combinations of strains are currently under evaluation.

Demonstration of papillomavirus capsid antigen in human conjunctival neoplasia.

To investigate the association of human papillomavirus with conjunctival neoplasia, we identified 50 resected papillomas from 47 patients. Papillomas were composed of papillary or, less commonly, flat proliferations of predominantly nonkeratinizing squamous epithelium with admixed goblet cells. Koilocytosis was focally present in 30 tumors (60%). Atypia that ranged from mild to severe was present in ten lesions (20%). In addition, we examined specimens of conjunctival dysplasia or carcinoma from 61 patients. The lesions were predominantly flat proliferations of atypical epithelial cells. Twenty biopsies performed for suspected sarcoidosis were used as controls. Papillomavirus capsid antigen was demonstrated using an immunoperoxidase technique in nuclei of mature superficial epithelial cells of 23 papillomas (46%) and five dysplasias or carcinomas (8.2%) but not in the control biopsy specimens. These results suggest that papillomavirus may play a role in the etiology of conjunctival papilloma, dysplasia, and carcinoma.

Serological diagnosis of Epstein-Barr virus infection by novel ELISAs based on recombinant capsid antigens p23 and p18.

A new pair of Epstein-Barr virus ELISAs (Biotest Anti-EBV VCA IgG and VCA IgM ELISA) was evaluated for usefulness for routine diagnosis of acute EBV infections. The ELISAs are based on two viral capsid antigens (VCA), p23 (BLRF2, full-length) and p18 (BFRF3, carboxy-half), that are combined by autologous gene fusion. In total, 179 sera were tested in direct comparison with classical VCA immunofluorescence assays (IFA). With the help of clinical data and additional reference serology, i.e., heterophile antibodies, anti-EA IgG (IFA) and anti-EBNA-1 IgG (ELISA), the patients were divided into the following categories: seronegatives (46), acute primary infections (67), previous infections (39), suspected reactivations (20) and constellations with intermediate serological patterns (7). The VCA IgG and VCA IgM ELISAs showed overall agreement to IFA of 95.0% and 94.4%, respectively. The calculated analytical performance (sensitivity; specificity) of VCA IgG and VCA IgM was 94.0%; 97.8% and 97.1%; 96.5%, respectively. A certain delay in seroconversion of anti-p23-p18 IgG may account for a significant difference in sensitivity of the VCA IgG ELISA between primary (88.4%) and previous infections (100%). In summary, the new recombinant VCA ELISAs yielded good correlation to VCA IFA and in combination with EBNA-1 IgG allow rapid, sensitive, and specific diagnosis of infectious mononucleosis or EBV immune status in general.

Immunoglobulin A against viral capsid antigen of Epstein-Barr virus and indirect mirror examination of the nasopharynx in the detection of asymptomatic nasopharyngeal carcinoma.

To evaluate the efficacy of population screening for early stage nasopharyngeal carcinoma (NPC) in southern China, the authors recruited 42,048 and 10,402 apparently healthy subjects residing in a high incidence and a low incidence area, respectively; all subjects were between the ages of 30 and 59 years. The subjects' serum specimens were tested for immunoglobulin (Ig) A antibody against viral capsid antigen (IgA/VCA) of Epstein-Barr virus (EBV). Of the subjects from the high incidence area, 2823 were found to be seropositive. In follow-up, they had yearly examinations of the nasopharynx by indirect mirror with or without biopsy; 41 were found to have histologically confirmed asymptomatic NPC during the first 2 years of follow-up. The tumors in most of these cases were localized and were at earlier stages than tumors of symptomatic cases of NPC seen in the same region before the screening. The yearly indirect mirror examination of the nasopharynx seems to have effectively identified most of the tumors at the stage of asymptomatic disease. The risk of harboring NPC was found to be different among the different sex and age subgroups of seropositive individuals. By limiting such screening to those who are at exceedingly high risk, the cost of the screening can be kept within the spending of the public health authority, and the effectiveness of the screening also is improved.
