Isolation of Coxiella burnetii by a centrifugation shell-vial assay from ticks collected in Cyprus: detection by nested polymerase chain reaction (PCR) and by PCR-restriction fragment length polymorphism analyses.

Ticks are the principal vectors and reservoirs of Coxiella burnetii. The identification of isolates is necessary for understanding the clinical diversity of Q fever in different geographic areas. This is the first report of isolation of C. burnetii from ticks by the shell-vial assay and by nested polymerase chain reaction (PCR) assay for the detection of this pathogen in ticks. Of 141 ticks collected in Cyprus (Rhipicephalus sanguineus and Hyalloma spp.), 10% were found to be infected with C. burnetii. Three ticks were positive by hemolymph test, and 11 triturated ticks were positive by nested PCR. Three isolates were obtained by the centrifugation shell-vial technique. Analysis by PCR, then restriction fragment length polymorphism showed that the 3 Cyprus isolates had identical restriction profiles to reference strains Nine Mile and Q212. The methods described are useful in studying the epidemiology and ecology of C. burnetii.

Sporicidal kinetics of Bacillus subtilis spores by heated scallop shell powder.

Scallop shell powder heated at 1,000 degrees C for 1 h exhibited sporicidal action against Bacillus subtilis spores. The sporicidal kinetics of this action were analyzed with the use of a nonlogarithmic model. Apparent death rate constants (k) were obtained under various conditions. The value of k increased with powder concentration but became constant beyond the concentration representing the solubility of Ca(OH)2. A linear inverse relationship between k and temperature was found, and from this relationship the activation energy required for the death of B. subtilis spores in the heated shell powder slurry could be determined.

Shell disease in river cooters (Pseudemys concinna) and yellow-bellied turtles (Trachemys scripta) in a Georgia (USA) lake.

A disfiguring shell disease was detected in river cooters (Pseudemys concinna) and yellow-bellied turtles (Trachemys scripta) from Lake Blackshear, Georgia (USA). The turtles used were part of a mark-recapture study conducted from September 1991 to June 1993. Histologic changes on four turtles included acute segmental necrosis of the epidermis, followed by ulceration, necrosis of the underlying dermis and dermal bone, and exaggerated remodeling of bone. Additional findings included visceral inflammatory lesions and bacterial infection, sepsis and marked trematode ova granulomatosis. The cause of the shell lesions was not determined.

A simple estimation method for the embryonic toxicity of plastic wrapping sheets using an egg shell-free culture system of chick embryos.

To estimate the embryonic toxicity of food wrapping films, the shell-free chick embryos were cultured using plastic wrapping sheets like a suspending hammock. Polyethylene (PE) wrapping sheets with no additives showed much higher survivability than polyvinylchloride (PVC) wrapping sheets including plasticizers. Furthermore when a PVC wrapping sheet was autoclaved at 110 degrees C for 30 min, the embryonic survivability significantly decreased compared with the untreated PVC wrapping sheet. These observations suggest that the egg shell-free culture method using a plastic wrapping sheet is a useful estimation method for the embryonic toxicity of food wrapping film.

Cellular sites for dynorphin activation of kappa-opioid receptors in the rat nucleus accumbens shell.

The nucleus accumbens (Acb) is prominently involved in the aversive behavioral aspects of kappa-opioid receptor (KOR) agonists, including its endogenous ligand dynorphin (Dyn). We examined the ultrastructural immunoperoxidase localization of KOR and immunogold labeling of Dyn to determine the major cellular sites for KOR activation in this region. Of 851 KOR-labeled structures sampled from a total area of 10,457 microm2, 63% were small axons and morphologically heterogenous axon terminals, 31% of which apposed Dyn-labeled terminals or also contained Dyn. Sixty-eight percent of the KOR-containing axon terminals formed punctate-symmetric or appositional contacts with unlabeled dendrites and spines, many of which received convergent input from terminals that formed asymmetric synapses. Excitatory-type terminals that formed asymmetric synapses with dendritic spines comprised 21% of the KOR-immunoreactive profiles. Dendritic spines within the neuropil were the major nonaxonal structures that contained KOR immunoreactivity. These spines also received excitatory-type synapses from unlabeled terminals and were apposed by Dyn-containing terminals. These results provide ultrastructural evidence that in the Acb shell (AcbSh), KOR agonists play a primary role in regulating the presynaptic release of Dyn and other neuromodulators that influence the output of spiny neurons via changes in the presynaptic release of or the postsynaptic responses to excitatory amino acids. The cellular distribution of KOR complements those described previously for the reward-associated mu- and delta-opioid receptors in the Acb shell.

Outbreaks of Salmonella serotype enteritidis infection associated with eating raw or undercooked shell eggs--United States, 1996-1998.

During the 1980s and 1990s, Salmonella serotype Enteritidis (SE) emerged as an important cause of human illness in the United States. The rate of SE isolates reported to CDC increased from 0.6 per 100,000 population in 1976 to 3.6 per 100,000 in 1996 (Figure 1). Case-control studies of sporadic infections and outbreak investigations found that this increase was associated with eating raw or undercooked shell eggs (1). From 1996 to 1998, the rate of culture-confirmed SE cases reported to CDC declined to 2.2 per 100,000; however, outbreaks of illness caused by SE continue to occur. This report describes four SE outbreaks during 1996-1998 associated with eating raw or undercooked shell eggs and discusses measures that may be contributing to the decline in culture-confirmed SE cases.

Distribution among sample test results when testing shelled corn lots for fumonisin.

The statistical distribution known as the compound gamma function was studied for suitability in describing the distribution of sample test results associated with testing lots of shelled corn for fumonisin. Thirty-two 1.1 kg test samples were taken from each of 16 contaminated lots of shelled corn. An observed distribution consisted of 32 sample fumonisin test results for each lot. The mean fumonisin concentration, c, and the variance, s2, among the 32 sample fumonisin test results along with the parameters for the compound gamma function were determined for each of the 16 observed distributions. The 16 observed distributions of sample fumonisin test results were compared with the compound gamma function using the Power Divergence test. The null hypothesis that the observed distribution could have resulted from sampling a family of compound gamma distributions was not rejected at the 5% significance level for 15 of the 16 lots studied. Parameters of the compound gamma distribution were calculated from the 32-fumonisin sample test results using the method of moments. Using regression analysis, equations were developed that related the parameters of the compound gamma distribution to fumonisin concentration and the variance associated with a fumonisin test procedure. An operating characteristic curve was developed for a fumonisin sampling plan to demonstrate the use of the compound gamma function.

A role for dopamine D1 receptors of the nucleus accumbens shell in conditioned taste aversion learning.

The involvement of dopamine (DA) in conditioned taste aversion (CTA) learning was studied with saccharin or sucrose as the conditioned stimulus (CS) and intraperitoneal lithium as the unconditioned stimulus (US). The dopamine D(1) antagonist R(+)-7-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrochloride (SCH 23390) (12.5-50 microg/kg, s.c.), given 5 min after the CS, impaired the acquisition of CTA in a paradigm consisting of three or a single CS-lithium association. SCH 23390 failed to impair CTA acquisition given 45 min after, 30 min before, or right before the CS. (-)-trans-6,7,7a,8,9,13b-hexahydro-3-chloro-2-hydroxy-N-methyl-5a-benzo-(d)-naphtho-(2,1b) azepine (SCH 39166) (12.5-50.0 microg/kg, s.c), a SCH 23390 analog that does not bind to 5HT(2) receptors, also impaired CTA. No significant impairment of CTA was obtained after administration of the specific D(2)/D(3) antagonist raclopride (100 and 300 microg/kg, s.c.). The ability of SCH 23390 to impair CTA learning was confirmed by its ability to reduce the conditional aversive reactions to a gustatory CS (sweet chocolate) as estimated in a taste reactivity paradigm. SCH 39166 impaired CTA also when infused in the nucleus accumbens (NAc) shell 5 min after the CS. No impairment was obtained from the NAc core or from the bed nucleus stria terminalis. The results indicate that D(1) receptor blockade impairs CTA learning by disrupting the formation of a short-term memory trace of the gustatory CS and that endogenous dopamine acting on D(1) receptors in the NAc shell plays a role in short-term memory processes related to associative gustatory learning.

[First experience with using a series of generalized golden proportions for studying the shell of brachiopod and mollusks].

Symmetry of shells of clams (Veneridae: Katelysia, Venus, Periglypta; Fimbriidae: Corbis fimbriata) and brachioipods (Cancrinella undata, Echinoconhus punctatus, Reticulatia inflatiformis and Neophricodothyris waageni) was studied geometrically with spiral symmetry and connected mathematical methods (complex proportions and Fibonacci numbers). Location regularity of the elements of concentrical sculpture connected with shell growth is interpreted as unrolling of logarithmic spiral in the process of growth. Correlations of radius of sculpture circles were studied with the raw of generalized golden sections of V.I. Korobko and G.N. Primak. The discovered regularities of circles location being similar to some peculiarities of phyllotaxis are governed by the rules of the raw of generalized golden sections. Two spiral folds could keep correspondence between the increase in mass of growing animal and the increase in intensity of water drawing by ciliate apparatus.

Outbreaks of Salmonella serotype enteritidis infection associated with eating shell eggs--United States, 1999-2001.

A Salmonella serotype Enteritidis (SE) epidemic emerged in the 1980s, when increasing numbers of infections were detected in the Northeastern and Mid-Atlantic regions of the United States. In the early 1990s, while SE rates in the Northeast began to decline, the SE epidemic expanded to the Pacific region. Nationwide, the number of SE isolates reported to CDC peaked at 3.8 per 100,000 population in 1995. Although rates of culture-confirmed SE infection reported to CDC declined to 1.9 by 1999 (Figure 1), rates did not decline further through 2001, and outbreaks continue to occur. Investigations of outbreaks and sporadic cases have indicated repeatedly that, when a food vehicle is identified, the most common sources of SE infection are undercooked and raw shell eggs. This report describes two SE outbreaks associated with eating shell eggs and underscores the need to strengthen SE-control measures.

Wear-through of metal-backed acetabular shell in total hip arthroplasty.

Full-thickness wear-through of the metal-backed acetabular shell after total hip arthroplasty is unlikely in patients who receive adequate follow-up after operation. We report such a case that occurred after neglect of a dislodged polyethylene liner from the cup of a total hip prosthesis. The patient was a 71-year-old retired male laborer with a 40-year history of alcohol dependence that may have contributed to the decline in his condition. During a period of almost 7 years, clinical findings such as clicking sound, leg length shortening, eccentric position of the femoral head relative to the acetabular shell, and metallic mark in the polyethylene liner had been identified on radiographic examination. However, delayed recognition of the clinical importance of these findings resulted in full thickness wear-through and subsequent bony deficiency.

Evaluation of the shell vial pre-CPE method using monoclonal antibodies for the diagnosis of human cytomegalovirus infection in the field of pediatrics.

A method of pre-cytopathological effects (CPE) detection in shell vial cultures using monoclonal antibodies (mAb) was evaluated for detection of cytomegalovirus (CMV) in pediatric specimens. The diagnostic value of antibody detection by enzyme immunoassay (EIA) was also evaluated. CMV was isolated from 47 of 1179 specimens tested by the routine tissue culture method. Positive results were obtained in 51 of these 1179 specimens using the pre-CPE method. The pre-CPE shell vial assay was demonstrated to be a rapid and sensitive method of detecting CMV. Detection of the virus by the pre-CPE method was considered much more reliable than by EIA IgM assays for pediatric patients.

Medical expert systems developed in j.MD, a Java based expert system shell: application in clinical laboratories.

Growing complexity of diagnostic tests, combined with increased workload, stringent laboratory accreditation demands, continuous shortening of turn-around-time and budget restrictions have forced laboratories to automate most of their iterative tasks. Introduction of artificial intelligence by means of expert systems has gained an important place in this automation process. Different parts of clinical laboratory activity can benefit from their implementation and the present project deals with one aspect, namely the clinical interpretation of diagnostic tests. This paper describes how j.MD, a new Java based expert system shell, was used to reprogram the expert system for interpretation of amylase isoenzyme patterns that has been in use for many years in our laboratory, and that was originally programmed in Pro.MD, a Prolog based expert system shell. One of the most important advantages of the j.MD system is its bidirectional link with the laboratory information system. This project shows how expert systems for the interpretation of complex diagnostic tests that demand specific expertise can become an integrated part of the automated clinical chemistry lab.

The distorted shell method for clustering for syndrome classification.

Syndrome classification may be described as the arrangement of individuals into groups on the basis of their phenotypic resemblance. This paper describes how phenotypic resemblance may be quantified and demonstrates a numerical method called distorted shell clustering, which isolates groups of phenotypically similar individuals representing syndromes. This new method takes into consideration apparent biological properties of syndromes. It allows for overlapping phenotypes between syndromes, and differing character association and variability within syndromes. This method is compared to four other clustering methods by using suspects for a syndrome of known etiology (Down syndrome). The numerical results based on the phenotype then can be compared with the actual diagnosis. Only the distorted shell method classifies patients, without error, into two major clusters: the Down and the non-Down, while maintaining a high level of efficiency.

Microwave synthesis of core-shell gold/palladium bimetallic nanoparticles.

The microwave-assisted polyol reduction method was applied to the synthesis of core-shell gold/palladium bimetallic nanoparticles by the simultaneous reduction of the AuIII and PdII ions. The thickness of the palladium shell was calculated to be approximately 3 nm, and the gold core diameter is 9 nm. The structure and composition of the bimetallic particles were characterized by high-resolution transmission electron microscopy equipped with a nanoarea energy-dispersive X-ray spectroscopy attachment, transmission electron microscopy, X-ray diffraction, and X-ray photoelectron spectroscopy.
