Radioactive isotope labeled diffusible components of a bone morphogenetic substratum.

Bone matrix gelatin labeled with radioactive glucose, cysteine, proline, tyrosine or tryptophan was enclosed in one chamber while an adjoined second chamber was left empty to become filled with interstitial fluid. The paired diffusion chambers were implanted into the anterior abdominal wall of allogeneic rats for 14 days. Release of the matrix gelatin (BMG) from the loaded chamber was followed by a bone formation on the outer membrane of the unloaded chamber. 3H-cysteine labeled components of BMG were disseminated into the interstitial fluids and throughout the host tissues 5 times more rapidly than 3H-proline labeled BMG. The relative rates of loss of radioactivity from the residue was cysteine greater than tryptophan greater than tyrosine greater than proline. Electron microscopic autoradiography revealed random dissemination of collagenous and non-collagenous derivatives of BMG. 3H-glucose distribution was coordinated with ruthenium red staining coarse granules and may have originated from BMG. A component of interstitial fluid staining positively with Coomassie brilliant blue and PAS methods was identified by SDS polyacrylamide gel electrophoresis of interstitial fluid aspirated from empty chambers attached to chambers loaded with bone matrix gelatin. The character of the bone morphogenetic property in bone gelatin has not yet been defined in physico-chemical terms but present evidence points to the view that it is associated with a protein molecule.

[Formation and migration of radioactive microemboli to the lung during radiovenography].

Two patients with lower extremity thrombophlebitis were studied by radionuclide venography with macroaggregated albumin labelled with 99mTc. Data analysis was performed with a new method. It generated sequential series of imagen (1-2 sec/frame) of the transit of the tracer through leg, thigh, and abdomen, allowing a more precise definition of temporal changes on regional radioactivity. This method rendered the first real evidence of the binding of the tracer to pre-existent venous thrombi, of the dislodgement of these and their gradual fragmentation during their transit to the lungs. The patients had no complaints during or after the study. Lung scanning, performed 24 hours later, showed slight alteration on regional lung perfusion in one case, and none in the other. It is concluded that radioactive microemboli to the lungs is a minor complication of radionuclide venography, which does not justify prevention. This could only nullify the sensitivity of the procedure.

Cardiac output measurements with thermodilution and radioactive microspheres. A comparative study in cats.

Thermodilution and radioactive microsphere measurements are both commonly used methods for determination of cardiac output in experimental animals. However, no comparison has been made of these methods for cardiac output determination, which is why this study was made. In cats 65 simultaneous measurements of cardiac output with the two methods were performed. No difference in mean values was found and the coefficient of correlation was 0.955 (P less than 0.001). Only eight out of the 65 measurements differed more than 10%. Thermodilution measurements are suitable for repeated determinations and when immediate data are wanted. On the other hand radioactive microsphere measurements are preferable when data on tissue blood flow and blood flow distribution are desired as well. The high correlation found seems to allow cardiac output measurements with these two methods to be grouped together.

Effect of ovarian steroids on uptake of radioactive amino acids by brain tissues.

Effects of two major ovarian hormones, estrogen (E) and progesterone (P), on the uptake of amino acids by the brain nuclei of ovariectomized mice were examined. They were divided into four groups: (1) oil controls, (2) P-treated, (3) estradiol benzoate (E2B)-treated, and (4) E2B-primed P-treated mice. Two hr after the last hormonal treatment they were given a single s.c. injection of a mixture of 3H-leucine and 3H-methionine, and then sacrificed 2 hr later. Intensity of the uptake of radioactivity was measured on autoradiograms of the stained tissue sections. Group 1 showed a relatively high uptake of radioactivity by the SO, PV and SPH, compared with that by the remaining brain cells. Group 2 had a slight enhancement of the uptake by the VM, SPH and CC, compared with that in group 1. Group 3 showed a significant enhancement of the uptake by most of the preoptico-hypothalamic nuclei except for the VM, DM and PM, compared with that in group 1. Group 4 had slightly enhanced uptake by the POM, POL, SCH, SO, PV, VM, ARC, SPH and CC, compared with that in group 1. E-priming affected the uptake; it enhanced the uptake by the VM and SPH, and inhibited it by the PVA, AH, LH and CC. Uptake by the PM and EC remained unchanged in all groups. The present results suggest that two major steroids, E and P, influence protein synthesis in most of the brain regions including the preoptic and hypothalamic areas.(ABSTRACT TRUNCATED AT 250 WORDS)

[Search for phosphopeptides in the feces of axenic rats fed radioactive ovine casein].

Radioactive ovine casein was obtained by injecting 100 microCi of 14C-Ser into the jugular vein of an ewe. The milk collected 17 and 24 h after this injection contained 12 p. 100 of the radioactivity injected in protein form. The seryl residues were specificially labelled. This casein was used as the only protein source fed to axenic rats; 0.30 p. 100 of the tracer ingested was found in the feces of those rats. Since phosphoserine represented 25 p. 100 of the total casein seryl residues, the phosphopeptides may not be selectively unabsorbable.

[Retention of radioactive aerosols (products of radon-222 decay) by various types of anti-dust masks].

The paper presents results of measurements of the efficiency of retention of radon decay products, occurring as radioactive aerosols, by anti-dust masks. The efficiency of three types of masks: PM-1, PM-3, PP-2 "Delta" was studied under various conditions of equilibrium between radon and products of its decay in the air. It was concluded that the efficiency of retention of radon decay products, occurring as radioactive aerosols, by the mentioned types of masks, comes to 75--87%. Anti-dust PM-1 (F-14 filter) and PM-3 (F-13 filter) masks are recommended as most efficient.

Radioactive phosphorus and external radiation as an adjuvant to surgery for ovarian carcinoma.

At the Johns Hopkins Hospital between the years 1967 and 1973, 40 patients with primary ovarian carcinoma were treated with radioactive phosphorus. Of these, 19 were treated with external radiation in addition. The morbidity from the radioactive phosphorus alone was negligible. Dosages of less than 5000 rads to the pelvis were usually well tolerated even when given in combination with the isotope. Pathologic changes in the bowel are discussed.

[In vitro incorporation of radioactive sulfur into proteoglycans of cartilage. Effects of various nonsteroidal anti-inflammatory agents].

Radioactive sulphur (35SO4) is incorporated in the proteoglycans of chicken embryonic cartilage. In fact, the molecular weight of the macromolecules which bind the sulphur corresponds to monomeric proteoglycans and proteoglycan-hyaluronic acid complexes. This incorporation does not take place when the cartilage has been previously killed. The capture of 35S is stimulated by human serum, in a dose-dependent way. At doses less then, equal to and greater than the maximal plasma concentrations observed following a single oral dose (600 mg), acetyl salicylic acid and sodium salicylate inhibit the incorporation of radioactive sulphur in the cartilage. Under the same conditions, tiaprofenic acid and oxametacine have no effect on this metabolic parameter.

Combined autoradiography and immunohistochemistry for simultaneous localization of radioactively labeled steroid hormones and antibodies in the brain.

A combined technique of autoradiography and immunohistochemistry is described for localization of radioactively labeled ligands and antibodies to neuropeptides, protein hormones and neurotransmitter synthesizing enzymes in the brain. This method permits the simultaneous visualization of radioactively labeled cells and neuropeptide-producing cells in the same tissue preparation. Autoradiograms are fixed with weak paraformaldehyde solution prior to photographic processing for subsequent immunoperoxidase staining. With this procedure 3H-estradiol is localized in neurophysin I and arginine vasopressin-producing cells of the mouse supraoptic and paraventricular nuclei, neurophysin-producing cells of the guinea pig hypothalamus, and dopamine-beta-hydroxylase-containing neurons of the rat lower brainstem; 3H-dihydrotestosterone in pituitary gonadotropes and thyrotropes; and 3H-1,25-(OH)2 vitamin D3 in pituitary thyrotropes and pancreatic B-cells.

[Radioactivity of an acid-soluble liver fractions after administration of 3H-thymidine for the purpose of explaining the reasons for delayed DNA labeling].

When 3H-thymidine is injected 24 hours before a partial hepatectomy, the radio-activity of acid-soluble fraction in the mouse liver begins to rise after the operation. When 3H-thymidine is injected to animals immediately after the operation or to unoperated animals, no such a rise is recorded. The maximal radioactivity of acid-soluble fraction of the liver is observed 28 hours after hepatectomy, at the moment preceding the commencement of DNA synthesis in a great number of hepatocytes. This increase is in conformity with the subsequent rise of DNA radioactivity. It is suggested that cells of the regenerating liver reutilize the degradation products of labeled DNA which become a source of DNA late labeling.

[Detection of platelet autoantibodies. Comparison of an antiglobulin consumption assay with a radioactive direct Coombs test (author's transl)].

131 patients (124 with idiopathic thrombocytopenic purpura) have been investigated for platelet autoantibodies with an antiglobulin consumption assay (ACA) and a direct Coombs test (DCT) using 125I-antiglobulin. The two assays have been simultaneously performed on 165 platelet-suspensions. The results of ACA (expressed as normal or abnormal amounts of platelet bound IgG or PBlgG) and those of DCT (expressed as positive or negative) were concordant in 128 out of 165 cases (77.6 per cent). In 37 of 165 cases (22.4 per cent) the DCT was negative while PBgG was found to be slightly elevated with ACA. There was a significant correlation (r = 0.44) between PBlgG levels and radioactivity index calculated in DCT. The relationship between these data was graphically expressed as a curve constructed on semilog paper meaning that PBlgG levels corresponding to high radioactivity index were more uncertain than those corresponding to lower index. The quantitative expression of DCT would be improved by refering the results to a standard curve and not only to normal platelets. In the case, results of DCT would be probably equivalent to those of ACA.

Essential thrombocythemia: response during first year of therapy with melphalan and radioactive phosphorus: a polycythemia Vera Study Group report.

Thirty-one patients with essential thrombocythemia were randomized to receive either melphalan or radioactive phosphorus as myelosuppressive therapy. Twenty-seven patients were evaluable for response. Of 13 patients treated with melphalan, 11 had a complete response (platelet count less than 450,000/mm3) at 3 and 6 months. This response rate was significantly better than the response to radioactive phosphorus. The response rates were similar at 12 months. No significant toxicity was observed with either regimen.

Bone blood flow in the femora of anesthetized and conscious dogs in a chronic preparation, using the radioactive tracer microsphere method.

The radioactive tracer microsphere method is applicable to the study of the hemodynamics of bone. As observed previously by others, areas of predominantly cancellous bone produce higher blood flow measurements than do areas of predominantly cortical bone. Bone containing fatty marrow has lower blood flow rates than does bone containing hematopoietic marrow. Blood flow to the femoral neck was the greatest measured (26.74 +/- 1.95 ml/min/100 gm), while the diaphyseal region was the area with the lowest flow (4.67 +/- 0.46 ml/min/100 gm). There was no significant difference between consecutive measurements using up to five differently labeled radioactive microspheres. This enabled observations of the effects of an experimental procedure on bone blood flow in the same animal. Additionally, there was no statistically significant differences between regional blood flows measured 40 days apart in the same animal. Bone blood flow measurements were slightly lower in anesthetized than in conscious dogs. The RTM method is the one most suitable for the estimation of bone blood flow under either acute or chronic conditions.

[Treatment of diffuse toxic goiter with radioactive iodine in combination with prolonged antithyroid therapy].

The authors analyzed clinically the results of treatment of 237 patients with diffuse toxic goiter with radioactive iodine in combination with prolonged antithyroidal therapy (12--18 months). The advantages of combined therapy consisted in the use of relatively low 131I doses, a great frequensy of clinical remissions of thyrotoxicosis (83%), in the absence of thyrotoxicosis remissions after the administration of a low radioactive iodine dose and a low percentage of stable hypothyroidism development (0.8%) as a complication of radioiodine therapy.

Synthesis and biosynthetic utilization of radioactive photoreactive fatty acids.

A synthesis of a homologous series of highly radioactive photoreactive fatty acids has been developed. Oxidative cleavage of the cis double bonds of unsaturated fatty acids by NaIO4/OsO4 produced the omega-oxo fatty acids; subsequent reduction by [3H]NaBH4 afforded a series of highly radioactive [omega-3H]hydroxy fatty acids to which a variety of photoreactive probes can be readily attached. In this study, the m-diazirinophenoxy group was coupled to the omega-hydroxy groups of the fatty acids, yielding the omega-(m-diazirinophenoxy) derivatives of nonanoic, undecanoic, tridecanoic, and pentadecanoic acid. These photoreactive derivatives were incorporated by L-cells into their major phospholipids; in addition, a number of proteins were shown to be acylated by these fatty acids. The photoreactive group was shown to remain intact as indicated by the cross-linking of the resulting photoreactive phospholipids to other phospholipids as well as to a number of L-cell proteins upon photolysis. This procedure thus identifies integral membrane proteins and directly produces photoaffinity derivatives of those proteins normally acylated by fatty acids.

[Recovery and purification of leakage radioactive krypton].

A recovery-purification system for radioactive Kr was constructed by enclosing an 85Kr testing apparatus with an airtight vessel. Experiments were carried out to examine the recovery rate of leakage Kr and the purity of Kr recovered with the system. The concentration of the leakage Kr in the airtight vessel was decreased to a detection limit of 0.3 ppm by the system. The recovery rate corresponded to 99.976%. The concentration of the Kr recovered was over 99.9908%. It shows that almost 100% of the radioactive Kr leaked out into the airtight vessel could be recovered and the He content in the Kr recovered could be reduced to less than 100 ppm.

Autoradiographic study of the distribution of bound radioactivity in the respiratory tract of Syrian hamsters given N-[3H]nitrosodiethylamine.

Adult Syrian golden hamsters received a single intragastric dose of N-[3H]nitrosodiethylamine. Their tracheas, extrapulmonary stem bronchi, and lungs were processed for high-resolution light-microscopic autoradiography to monitor the distribution of bound radioactivity. In the trachea and extrapulmonary stem bronchi, mucous cells contained the most bound radioactivity, while in the lobar and segmental bronchi and bronchioles, Clara cells were the major site of binding. In conjunction with earlier conducted studies on the pathogenesis of N-[3H]nitrosodiethylamine-induced respiratory tract tumors, these findings indicate that metabolic competence and a preexisting capacity for proliferation are important factors in determining the target cell types of this compound.
