Prognostic relevance of Ki-67 antigen expression in 329 cases of oral squamous cell carcinoma.

Prognostic relevance of Ki-67 staining in oral squamous cell carcinomas was investigated by immunohistochemical expression in 329 cases of squamous cell carcinoma of the oral cavity due to a formamide pretreatment. The growth fraction (Ki-67 labelling index, LI) was correlated to histopathological grading, lymphocytic reaction, stroma/tumour proportion, pattern of invasion, mitotic rate, degree of keratinisation, tumour size, thickness of tumor, depth of invasion, lymph node involvement and five-year survival rate. Highly significant inverse correlation was found between the Ki-67 LI, the stroma/tumour proportion and the degree of keratinisation whereas no correlation could be established between the Ki-67 LI and all the other histological and clinical paramters. Ki-67 staining alone has no prognostic relevance in oral cancer.

Effect of various xenobiotics and steroids on aryl hydrocarbon hydroxylase activity of intestinal and hepatic microsomes from male rats.

The effects of four solvents and 37 xenobiotics and endogenous steroids on intestinal and hepatic microsomal aryl hydrocarbon hydroxylase (AHH) activity were determined. In general, hepatic AHH was more sensitive than intestinal AHH to inhibition by a wide variety of xenobiotics. Acetone, ethanol, dimethyl formamide, triamcinolone acetonide, 17 alpha-propyltestosterone, metronidazole, cimetidine and butylated hydroxyanisole (BHA) activated or stimulated intestinal AHH activity while inhibiting hepatic AHH activity at the same concentrations. Ethoxyquin, progesterone, pregnenolone acetate, deoxycorticosterone acetate, tamoxifen and SKF 525-A exerted markedly different concentration-dependent inhibitions of intestinal and liver AHH activities. The most potent inhibitors of AHH activity were captan, BHA, chlorpromazine, thioridazine, disulfiram and menadione. The ability of steroids to inhibit AHH activity correlated well with lipophilicity.

Evaluation of chemotherapy in a murine model for bladder cancer.

A transplantable transitional cell murine bladder tumor induced by N-[4-(5-nitro-2-furyl)-thiazolyl] formamide (FANFT) was characterized by tumor growth, survival time and response to chemotherapy drugs, cis-dichloro-trans-dihydroxy-bis-iso-propylamine platinum IV (CHIP), cis- diaminedichloro platinum II (DDP), cyclophosphamide (CTX) and methotrexate (MTX). Nontreated tumor-bearing mice were observed to survive 43 +/- 7 days (mean +/- SEM) with an average tumor burden of 8.45 +/- 0.65 g (mean +/- SEM) of solid tumor tissue. Lung metastasis was observed in 3 animals after 42-49 days post implantation. Microscopically, the primary tumor and the lung metastasis were structurally similar. In response to chemotherapy, tumor growth was significantly retarded (p less than 0.005) in the DDP-treated group, and survival was significantly increased in the CTX-treated group (p less than 0.001). Lung metastasis was observed in all treatment and control groups. This model has specific reproducible characteristics which make it a useful murine tumor model to study locally invasive bladder cancer.

Water-soluble and insoluble crystallins of the developing human fetal lens, analyzed by agarose/polyacrylamide thin-layer isoelectric focusing.

Eight human fetal lenses, selected on basis of normality, of a gestational age of 119 to 231 days were analyzed by thin-layer isoelectric focusing (IEF) in agarose/polyacrylamide gels. This method was adapted for the separation of lens crystallins into HM-, alpha-, beta- and gamma-crystallins. It is especially suitable for analysis under non-denaturing conditions of high-molecular-weight crystallins and of insoluble crystallins (WI) solubilized in formamide. The latter could be separated into HM-, alpha- and gamma-crystallins. During fetal development, a considerable increase of gamma-crystallin proportion was observed due to new synthesis. This increase was balanced by a decrease of alpha-crystallin proportion.

Enhanced expression of Harvey-ras oncogene in FANFT-induced transitional cell carcinoma.

The level of mRNA expression and DNA structure of c-H-ras were examined in a transitional cell carcinoma (TCC) induced in C3H/He mice and Fisher rats by N-(4,5-nitro-2-furylthiazolyl)formamide (FANFT) and carried as serially transplanted tumors. Neither amplification nor gross rearrangements were detected by Southern analysis in the FANFT tumor. Increased expression of c-H-ras was detected by Northern and in situ hybridization in both the FANFT-induced mouse and rat TCC, as compared to normal urothelium. No expression of six other cellular oncogenes was detected in the tumor. It appears that the increase in H-ras expression in the FANFT tumor is specific to this oncogene and that increased expression of the H-ras gene may play a role in bladder carcinogenesis.

Inhibition of experimental bladder tumor induction by systemic BCG treatment.

Transitional cell carcinoma of the urinary bladder was induced in rats by ingestion of 0.188% N-[4-5-nitro-2-furyl)-2-thiazolyl] formamide. During the induction period of 8 months, one animal group received systemic BCG treatment. After 12 months, the BCG-treated rats showed a significantly lower bladder tumor weight and stage than the corresponding controls. The results are discussed with regard to the possibility of BCG prophylaxis of human bladder tumors.

Effect of solvent mixture on the properties of temperature- and pH-sensitive polysaccharide-based hydrogels.

Novel partially biodegradable, temperature- and pH-sensitive polysaccharide-based hydrogels (NDF) were synthesized from modified dextran (dextran-maleic acid, Dex-MA) and N-isopropylacrylamide precursors over a wide range of mixed solvent ratios of dimethyl formamide (DMF) to water. N-Isopropylacrylamide monomers were chosen to impart thermo-responsive capability to Dex-MA, while Dex-MA was chosen to impart pH-responsive capability to N-isopropylacrylamide. The pH-sensitive precursor (Dex-MA) was synthesized by reacting dextran with maleic anhydride in the presence of triethylamine catalyst. To fabricate multi-stimuli hybrid hydrogel networks, both Dex-MA and N-isopropylarylamide precursors were photo-cross-linked via UV at a fixed Dex-MA to N-isopropylarylamide feed ratio over a wide range of DMF to water mixed solvent ratios. The newly synthesized PNIPAAm/Dex-MA hybrid hydrogels (NDF) were characterized by Fourier transform infrared spectroscopy for chemical structure determination, differential scanning calorimetry for thermal analysis and scanning electron microscopy for morphological study. The properties of the hybrid hydrogels, such as thermo-induced deswelling, pH-sensitivity, ionic strength sensitivity and thermo-reversibility, were also examined. The swelling data obtained clearly showed that newly synthesized multi-stimuli NDF hydrogels exhibited multi-responsive capability to external stimuli like temperature and pH. The morphological data obtained showed that this new class of PNIPAAm/Dex-MA hybrid hydrogels had a wide range of unique three-dimensional porous network structures that depended on the composition ratio of the mixed DMF/water solvent during cross-linking reaction. This unique but versatile 3D porous network structures of NDF hydrogels were correlated to the data from thermo-induced swelling behavior, thermo-reversibility, pH-dependent swelling and ionic strength sensitivity.

Role of renal metabolism and excretion in 5-nitrofuran-induced uroepithelial cancer in the rat.

5-Nitrofurans have been used in the study of chemical carcinogenesis. There is substantial evidence that N-[4-(5-nitro-2-furyl)-2-thiazolyl] formamide (FANFT) is deformylated to 2-amino-4-(5-nitro-2-furyl)thiazole (ANFT) in the process of FANFT-induced bladder cancer. Paradoxically, ANFT is less potent as a uroepithelial carcinogen than FANFT when fed to rats. Feeding aspirin with FANFT to rats decreases the incidence of bladder cancer. Isolated kidneys were perfused with 5-nitrofurans to determine renal clearances and whether aspirin acts to decrease urinary excretion of the carcinogen. In FANFT-perfused kidneys, FANFT was deformylated to ANFT and excreted (1.06 +/- 0.22 nmol/min) at a rate eightfold higher than excretion of FANFT. In kidneys perfused with equimolar ANFT, excretion of ANFT was 0.25 +/- 0.05 nmol/min, which suggests a coupling of renal deformylation of FANFT to excretion of ANFT in FANFT-perfused kidneys. Neither aspirin nor probenecid altered the urinary excretion or half-life of FANFT or ANFT. In rats fed 0.2% FANFT as part of their diet, coadministration of aspirin (0.5%) increased urinary excretion of ANFT during a 12-wk feeding study, which suggests decreased tissue binding or metabolism of ANFT. Kidney perfusion with acetylated ANFT (NFTA), a much less potent uroepithelial carcinogen, resulted in no ANFT excretion or accumulation, which indicates the specificity of renal deformylase. Renal deformylase activity was found in broken cell preparations of rat and human kidney. These data describe a unique renal metabolic/excretory coupling for these compounds that appears to explain the differential carcinogenic potential of the 5-nitrofurans tested. These results are consistent with the hypothesis that aspirin decreases activation of ANFT by inhibiting prostaglandin H synthase.

Relationship of DNA adducts derived from 2-acetylaminofluorene to cell proliferation and the induction of rodent liver and bladder tumors.

Pharmacokinetic models have been developed to assist in extrapolating results from rodent bioassays. However, in numerous circumstances, it is necessary to combine such models with cellular response models to fully define interspecies and dose extrapolations. Interactions between pharmacokinetic target tissue end points (DNA adduct formation) and cellular proliferation in liver and urinary bladder carcinogenesis is illustrated with the results from the ED01 study involving 2-acetylaminofluorene administered to female mice. The interaction of genotoxic and cell proliferative effects are also illustrated in a co-carcinogenesis study with low doses of N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide and high doses of sodium saccharin. The application of such interactions to humans is illustrated for the case of cigarette smoke-induced bladder cancer.

Limitations of in situ hybridization for the detection of bluetongue virus in blood mononuclear cells.

In situ nucleic acid hybridization was tested for the ability to detect bluetongue virus (BTV) nucleic acids in blood mononuclear cells. A standard protocol was devised and applied to the demonstration of BTV genetic sequences in cultured bovine mononuclear cells that had been infected in vitro. In situ hybridization using biotinylated single-stranded RNA probes, in the presence of 50% formamide at 50 C, demonstrated an intense, positive signal in 0.001-0.01% of the BTV-infected cultured mononuclear cells. The protocol was applied to isolated mononuclear cells from an experimentally infected heifer. No infected cells were observed by this method, although the blood specimens were obtained during peak viremia.

Detection and differentiation of bovine group A rotavirus serotypes using polymerase chain reaction-generated probes to the VP7 gene.

Dot and Northern blot hybridization assays were developed to detect and differentiate group A bovine rotavirus serotypes using radiolabeled serotype 6 (Nebraska calf diarrhea virus [NCDV] and United Kingdom [UK] strains) or serotype 10 (Crocker [Cr] strain) VP7 gene probes. Partial length VP7-specific cDNA encompassing areas of major sequence diversity were generated by the polymerase chain reaction (PCR) using either cloned VP7 genes (NCDV and UK strains) or reverse transcribed mRNA (Cr strain) as templates. Radiolabeled probes prepared from the PCR-generated cDNA were tested at various stringency conditions to optimize the hybridization assays. At high stringency conditions (52 C, 50% formamide, 5 x standard saline citrate), the NCDV, UK, and Cr probes serotypically differentiated bovine rotavirus isolates in RNA samples prepared from cell culture propagated viruses or in fecal specimens from infected gnotobiotic calves. The sensitivity and specificity of NCDV and Cr VP7 probes were characterized in dot blot hybridization assays, and the probes were estimated to detect at least 1 ng of viral RNA. The serotyping results obtained using VP7 probes were similar to those obtained using serologic assays. Further development of these assays may provide a useful means for the rapid detection and differentiation of bovine rotavirus serotypes in fecal samples from calves in the field.

Use of species-specific oligonucleotide probes to detect Mycoplasma gallisepticum, M. synoviae, and M. iowae PCR amplification products.

Three digoxigenin-labeled oligonucleotide probes, complementary to the variable region of the 16S ribosomal RNA (rRNA) gene of Mycoplasma gallisepticum, M. synoviae, and M. iowae were designed. The oligonucleotides were used in a dot blot hybridization assay. The target DNA is a 780-bp fragment of the 16S rRNA gene of avian mycoplasmas amplified by a single set of primers (multispecies polymerase chain reaction [PCR]). The oligonucleotide probes were specific for their corresponding PCR products at hybridization conditions of 56 C and 50% formamide. The detection limit of the dot blot hybridization assay was approximately 70, 50, and 30 colony-forming units for M. gallisepticum, M. synoviae, and M. iowae, respectively, per 4 microliters of PCR. In general, the oligonucleotide probe dot blotting assay was a more sensitive and effective method of detecting PCR products than detection by gel electrophoresis.

Rapid embedding of tissues in Lowicryl K4M for immunoelectron microscopy.

Lowicryl K4M (K4M) was recently introduced as an embedding medium for immunocytochemistry at the electron microscope level (BL Armbruster, E Carlemalm, R Chiovetti, RM Garavito, JA Hobot, E Kellenberger, W Villiger (1982):J Microsc 126:77 and E Carlemalm, M Garavito, W Villiger (1982):J Microsc 126:123). While earlier protocols of fixation and embedding required 4-6 days, the present method has reduced the processing time by accelerating both dehydration of tissues and polymerization of K4M so that tissues can be prepared for sectioning within 4 hr. The immunocytochemical labeling density was quantitated in order to determine relative antigen preservation in tissues embedded by the accelerated protocol as compared to slower K4M embedding techniques and to tissues embedded in glutaraldehyde-cross-linked bovine serum albumin (BSA). Thin sections of Bufo marinus kidney were labeled with rabbit antibody to Na+,K+ATPase alpha chain catalytic subunit isolated from B. marinus kidney microsomes (M Girardet, K Geering, JM Frantes, D Geser, BC Rossier, JP Kraehenbuhl, C Bron (1981):Biochemistry 20:6684). B. marinus retinas were labeled with rabbit anti-opsin. After fixation in paraformaldehyde(3%)-glutaraldehyde(3%), tissues were washed in buffer, dehydrated in 50, 75, and 90% dimethyl-formamide (DMF, 10 min each); K4M:DMF, 1:2 (15 min); K4M:DMF, 1:1, (20 min); K4M (25 min); K4M (30 min) at room temperature and transferred in fresh K4M to BEEM capsules for exposure to ultraviolet light (GE 15 watt, Black-lite, 10 cm, 45 min or less) at 4 degrees C. Thin sections were labeled successively with antibody, biotinylated sheep anti-rabbit F(ab')2 and avidin-ferritin. Ferritin labeling densities were determined by point counting. High labeling densities were observed with both antibodies, equaling or exceeding levels of labeling by slower protocols or embedment in BSA.

Detection of 5-bromodeoxyuridine (BrdUrd) incorporation by monoclonal antibodies: role of the DNA denaturation step.

Immunochemical detection of cells that incorporate 5-bromodeoxyuridine (BrdUrd) requires prior denaturation of DNA in situ to make BrdUrd binding sites accessible to the antibodies. A technique is described in which the DNA denaturation step is facilitated by a) prior dissociation of histones from DNA and b) the use of low ionic strength buffer in which the cells are suspended during heating. Dissociation of histones is achieved by cell treatment with 0.08N HCl at 0 degree C, which a) increases accessibility of DNA to propidium iodide (and following the denaturation to the antibodies); b) lowers stability of DNA to thermal denaturation; c) decreases differences between various cell types due to variability in chromatin structure; and d) ensures more complete DNA denaturation. Cell heating (80-95 degrees C) at low ionic strength (1 mM Na+) eliminates the need for formamide and results in extensive and rapid DNA denaturation. The method was applied in Friend leukemia, L1210 and HL-60 cell lines, and to bone marrow, experimental animal tumor and primary human tumor cells.

Thiourea and cyanamide as inhibitors of thyroid peroxidase: the role of iodide.

Thiourea, methylmercaptoimidazole, propylthiouracil, and thiouracil are all potent inhibitors of thyroid peroxidase (TPO)-catalyzed iodination. Unlike the cyclic thioureylenes, thiourea at 5 mM has no effect on guaiacol oxidation. If iodide is added to guaiacol assays containing thiourea, enzyme activity is lost. The latter observation may be explained as follows. In the presence of iodide, the iodinating species [TPO.Ioxid], oxidizes thiourea to formamidine disulfide. This product decomposes to cyanamide at neutral pH. We have shown cyanamide to be an inhibitor of the peroxidative and iodinating functions of TPO. Studies in rats demonstrate that doses of thiourea which completely inhibit in vivo protein-bound iodine formation have no irreversible effect on TPO, as measured by guaiacol peroxidation after removal of the thyroids. The major in vivo action of cyanamide is similar to that of thiourea. The data suggest that the primary in vivo and in vitro mode of action of thiourea is the reversible Ioxid-trapping mechanism. The anomalous inhibition of guaiacol peroxidation seen in the presence of thiourea plus iodide derives from the formation of formamide disulfide, followed by its nonenzymic decomposition to cyanamide.

Absence of p53 point mutations in parathyroid adenoma and carcinoma.

Primary hyperparathyroidism is a common disorder characterized by aberrant growth and function of solitary or multiple parathyroid glands. Many, if not all, parathyroid adenomas are examples of benign clonal neoplastic growth. The molecular events associated with the development of parathyroid neoplasia have not been well characterized. We examined benign and malignant parathyroid tissues for structural abnormalities of the p53 tumor suppressor gene. To screen for mutations in the p53 gene, we analyzed polymerase chain reaction-amplified DNA by denaturing gradient gel electrophoresis. DNA was isolated from 26 benign parathyroid adenomas and 3 parathyroid carcinomas, and polymerase chain reaction was used to amplify DNA fragments corresponding to the 4 evolutionarily conserved domains within exons 5, 7, and 8 of the p53 gene in which the majority of point mutations have been identified. Amplified DNA fragments were electrophoresed through polyacrylamide gels with linearly increasing gradients of the denaturants urea and formamide. After electrophoresis, the gels were examined for the presence of abnormally migrating bands, which represent DNA with altered melting points due to nucleotide sequence changes. Amplified fragments were of the expected size in DNA from 26 parathyroid adenomas and 3 parathyroid carcinomas. Denaturing gradient gel electrophoresis studies failed to disclose evidence of mutations in exons 5, 7, and 8 of the p53 gene in these neoplasms. We conclude that p53 point mutations do not appear to be a primary event responsible for neoplastic growth in parathyroid tissue.

Detection of small amounts of human DNA in human-rodent hybrids.

A method of measuring semi-quantitatively small amounts of human DNA in irradiated human x mouse and irradiated human x Chinese-hamster somatic cell hybrids is described. One method uses molecular hybridization of cell DNA bound to nitrocellulose filters with a cRNA probe to Cot 0--1 human DNA. Alternatively hybrid cell DNA is reassociated in solution with a Cot 0--1 fraction of nick-translated human DNA. Formamide buffers give specificity to the reaction. The detection limit of the filter method is 0.2--0.5% equivalents and reassociation kinetics 0.005--0.1% equivalents of a human genome. Experiments with cell hybrids suggest that a fragment of repetitive DNA may be retained along with the selected genes after a cell fusion. In one case however, of a hybrid cell in which malignancy is suppressed, highly repetitious sequences were not found.

Synthesis and characterization of orally active nonpeptide vasopressin V2 receptor antagonists.

The present study was undertaken to evaluate whether a novel series of 2,6-diaza-5-oxobicyclo[5.4.0]undeca-1(7),8,10-triene derivatives exhibited antagonistic activity for vasopressin V1 and V2 receptors. Most of these compounds were synthesized and showed a high affinity potential for V2 receptor and low to moderate affinity potential for V1 receptor. The most potent and V2-selective compound, N-[4-[2,6-diaza-6-[2-(4-methylpiperazinyl)-2-oxoethyl] -5- oxobicyclo[5.4.0]undeca-1(7),8,10-trien-2-yl]-carbonyl]pheny l][2-(4- methylphenyl)phenyl]-formamide (11b), exhibited IC50's of 2.9 nM for the V2 receptor and 200 nM for the V1 receptor, respectively. When administered orally to rat, 11b showed an approximately 18-fold increased urine volume in comparison with control rat.
