Affective components and intensity of pain correlate with structural differences in gray matter in chronic back pain patients.

Although chronic back pain is one of the most frequent reasons for permanent impairment in people under 65, the neurobiological mechanisms of chronification remain vague. Evidence suggests that cortical reorganisation, so-called functional plasticity, may play a role in chronic back pain patients. In the search for the structural counterpart of such functional changes in the CNS, we examined 18 patients suffering from chronic back pain with voxel-based morphometry and compared them to 18 sex and age matched healthy controls. We found a significant decrease of gray matter in the brainstem and the somatosensory cortex. Correlation analysis of pain unpleasantness and the intensity of pain on the day of scanning revealed a strong negative correlation (i.e. a decrease in gray matter with increasing unpleasantness/increasing intensity of pain) in these areas. Additionally, we found a significant increase in gray matter bilaterally in the basal ganglia and the left thalamus. These data support the hypothesis that ongoing nociception is associated with cortical and subcortical reorganisation on a structural level, which may play an important role in the process of the chronification of pain.

Primary central white matter hypoplasia of the neocortex.

Leukodystrophies, or diseases of the white matter, represent acute or ongoing damage to the oligodendrocytes of the central nervous system. Early childhood white matter disease is most commonly observed after hypoxic ischemic insults, with acute magnetic resonance imaging changes followed by atrophy or periventricular leukomalacia. Dysmyelination occurring in the setting of inborn errors of metabolism is characterized by progressive changes with high signal intensity in white matter on magnetic resonance imaging. This report presents a patient whose magnetic resonance imaging scans demonstrated hypoplasia of myelin in the telencephalon, without clinical or magnetic resonance imaging evidence of inflammatory dysmyelination. Clinical features included intractable seizures, severe hypotonia, and dysmorphic facial features coupled with a static failure to gain developmental milestones. Together, the clinical and magnetic resonance imaging findings are evidence of a primary failure of myelination in the neocortex.

Regional white matter change in pre-symptomatic Huntington's disease: a diffusion tensor imaging study.

The pathology of Huntington's disease (HD) is characterized by diffuse brain atrophy, with the most substantial neuronal loss occurring in the caudate and putamen. Recent evidence suggests that there may be more widespread neuronal degeneration with significant involvement of extrastriate structures, including white matter. In this study of pre-symptomatic carriers of the HD genetic mutation, we have used diffusion tensor imaging to examine the integrity and organization of white matter in a group of individuals who previously demonstrated abnormalities in response to a functional magnetic resonance imaging paradigm. Our results indicate that, before the onset of manifest HD, there are regional decreases in fractional anisotropy, indicating early white matter disorganization.

Gray matter density reduction in the insula in fire survivors with posttraumatic stress disorder: a voxel-based morphometric study.

Voxel-based morphometry (VBM) is an objective whole-brain technique for characterizing regional cerebral volume and tissue concentration differences in structural magnetic resonance images. In the current study, we used VBM to examine possible cerebral gray matter abnormalities in patients with posttraumatic stress disorder (PTSD) due to fire. The subjects included 12 victims of a fire disaster with PTSD and 12 matched victims of the same fire without PTSD. Magnetic resonance images were obtained on a 1.5-Tesla General Electric scanner at Central South University, and an entire brain volume of 248 contiguous slices was obtained for each subject. Then, gray matter density in patients with PTSD and control groups was compared by using a VBM approach in SPM2. Group analysis was thresholded at P<0.001, uncorrected, at the voxel level. The following three regions of reduced gray matter volume were found in patients with PTSD compared with controls: left hippocampus, left anterior cingulate cortex (ACC), and bilateral insular cortex. It was concluded that there are structural abnormalities of the hippocampus, the ACC and the insular cortex in patients with PTSD due to fire.

White matter integrity in kleptomania: a pilot study.

This study's goal was to examine microstructural organization of frontal white matter in kleptomania. Ten females with DSM-IV kleptomania and 10 female controls underwent diffusion tensor imaging. Inferior frontal white matter was the a priori region of interest. Trace and fractional anisotropy (FA) were also calculated for frontal and posterior cortical regions in both subject groups. Kleptomania subjects had significantly higher mean frontal Trace, and significantly lower mean frontal FA than control subjects. Group differences remained significant when right and left frontal Trace and FA were analyzed. Groups did not differ significantly in posterior Trace or FA. Kleptomania may be associated with decreased white matter microstructural integrity in inferior frontal brain regions.

A fully automated method for quantifying and localizing white matter hyperintensities on MR images.

White matter hyperintensities (WMH), commonly found on T2-weighted FLAIR brain MR images in the elderly, are associated with a number of neuropsychiatric disorders, including vascular dementia, Alzheimer's disease, and late-life depression. Previous MRI studies of WMHs have primarily relied on the subjective and global (i.e., full-brain) ratings of WMH grade. In the current study we implement and validate an automated method for quantifying and localizing WMHs. We adapt a fuzzy-connected algorithm to automate the segmentation of WMHs and use a demons-based image registration to automate the anatomic localization of the WMHs using the Johns Hopkins University White Matter Atlas. The method is validated using the brain MR images acquired from eleven elderly subjects with late-onset late-life depression (LLD) and eight elderly controls. This dataset was chosen because LLD subjects are known to have significant WMH burden. The volumes of WMH identified in our automated method are compared with the accepted gold standard (manual ratings). A significant correlation of the automated method and the manual ratings is found (P<0.0001), thus demonstrating similar WMH quantifications of both methods. As has been shown in other studies (e.g. [Taylor, W.D., MacFall, J.R., Steffens, D.C., Payne, M.E., Provenzale, J.M., Krishnan, K.R., 2003. Localization of age-associated white matter hyperintensities in late-life depression. Progress in Neuro-Psychopharmacology and Biological Psychiatry. 27 (3), 539-544.]), we found there was a significantly greater WMH burden in the LLD subjects versus the controls for both the manual and automated method. The effect size was greater for the automated method, suggesting that it is a more specific measure. Additionally, we describe the anatomic localization of the WMHs in LLD subjects as well as in the control subjects, and detect the regions of interest (ROIs) specific for the WMH burden of LLD patients. Given the emergence of large NeuroImage databases, techniques, such as that described here, will allow for a better understanding of the relationship between WMHs and neuropsychiatric disorders.

Neurotoxicity of intrathecally administered bupivacaine involves the posterior roots/posterior white matter and is milder than lidocaine in rats.

BACKGROUND AND OBJECTIVES: Clinical and laboratory studies suggest that lidocaine is more neurotoxic than bupivacaine. However, histological evidence of their comparative neurotoxicity is sparse. We thus pathologically and functionally compared the intrathecal neurotoxicity of these agents. METHODS: Rats received 0.12 microL/g body weight lidocaine (0%, 2%, 10%, or 20%) or bupivacaine (0%, 0.5%, 2.5%, or 5%) in distilled water via an intrathecal catheter. The influence of high osmolarity was also examined using 5% bupivacaine in 20% glucose solution (5% BG) and a control 25% glucose solution. The L3 spinal cord, the posterior and anterior roots, and the cauda equina were examined by light and electron microscopy. Walking behavior and sensory threshold were investigated as neurofunctional tests. RESULTS: The posterior root and posterior white matter showed axonal degeneration in rats treated with 10% and 20% lidocaine and 5% bupivacaine in distilled water (5% BDW) and in 5% BG, but not in rats treated with 2% lidocaine, 0.5% and 2.5% bupivacaine, distilled water, or 25% glucose solution. The histological damages were more severe in 20% lidocaine-treated rats than in 5% bupivacaine-treated rats. The damage of posterior white matter was observed only when the posterior root was severely injured. No significant difference of histological findings was observed between 5% BDW and 5% BG. Functional abnormalities were found only in rats treated with 20% lidocaine. CONCLUSIONS: The neurotoxic lesions caused by bupivacaine and lidocaine were indistinguishable in the primary site and the extending pattern, such as axonal degeneration originating from the posterior roots and extending to the posterior white matter. The intrathecal neurotoxicity is greater in lidocaine than in bupivacaine.

Gray matter abnormalities associated with duration of untreated psychosis.

PURPOSE: A long duration of untreated psychosis (DUP) is associated with relatively poor clinical and social outcomes. In order to identify whether an anatomically mediated mechanism may give rise to poorer outcomes, it is important to identify whether a long DUP is associated with greater brain structural abnormalities. METHOD: 81 patients with first-episode psychosis (schizophrenia, affective, and other psychoses) were scanned using high resolution Magnetic Resonance Imaging. DUP was defined as the number of days between first onset of psychotic symptoms and first contact with mental health services. High-resolution MRI images and voxel-based methods of image analysis were used to investigate brain structure in these patients. RESULTS: Longer DUP was associated with gray matter reductions in left middle and inferior temporal, left occipital and left fusiform cortices, and with gray matter excess of the left basal ganglia. All findings remained significant when co-varying for exposure to antipsychotic treatment. CONCLUSIONS: Temporal gray matter reductions are more marked in patients with a long DUP. This could reflect a progressive pathological process that is active prior to treatment. Alternatively, these abnormalities could be associated with a more insidious onset of illness and a later presentation to services.

Influence of organic matter on the uptake of cadmium, zinc, copper and iron by sorghum plants.

This article describes an experiment, carried out under controlled environment conditions, to investigate the effects of a fulvic acid fraction of soil organic matter on growth, cadmium (Cd) uptake and redistribution by sorghum. In addition the uptake of copper (Cu), zinc (Zn) and iron (Fe) was also determined. Sorghum was grown in nutrient solutions with 0, 0.1, 1 and 10 mg Cd dm(-3), in the absence and presence of organic matter (32 mg C dm(-3)), for various periods up to 20 days. A decrease in sorghum biomass due to Cd toxicity was observed at 10 mg Cd dm(-3), but for concentrations of 0.1 and 1 mg Cd dm(-3) the biomass was increased compared with control, without visual toxicity symptoms. The presence of organic matter (OM) further increased biomass production. Cadmium was mainly retained in sorghum roots, as usually found in tolerant plants, but Cd accumulation in sorghum was greater than in other Gramineae, or even more tolerant plants such as lettuce. The presence of OM decreased the bioavailability of Cd that was partially retained in solution by the OM ligands. However, OM promoted the translocation of Cd to shoots, an effect that may pose a risk to public health because plant-animal transfer of Cd could be enhanced. The presence of OM decreased the uptake of Cu, Zn and Fe. The presence (vs. absence) of 0.1 mg Cd dm(-3) enhanced the uptake of Fe, both in the absence and presence of OM.

Characterization of dissolved organic matter from source to sea using fluorescence and absorbance spectroscopy.

Dissolved organic matter fluorescence, absorbance and dissolved organic carbon were measured from source to sea in the River Tyne catchment, of approximately 2935 km(2) and encompassing areas of contrasting land use. The catchment has three major tributaries: the North Tyne which has good water quality, high dissolved organic carbon concentrations and visible water colour from the high proportion of peat in its upper catchment; the South Tyne which has good water quality with typical riverine dissolved organic carbon concentrations and drains from limestone uplands; and the Derwent, a more urbanized catchment which is increasingly impacted by treated sewage effluent discharges towards its mouth. Thirty sample sites, 23 along the three main tributaries and seven within the estuary, were sampled on six occasions over the period 2002-2003. High absorbance at 340 nm and dissolved organic carbon concentration identify N Tyne waters due to the peaty headwaters, but no downstream trends in these parameters are observed in any of the tributaries, in contrast to the estuary where a rapid decrease is observed in both. Fluorescence in contrast demonstrated downstream trends in both intensity and wavelength, especially in the Derwent as it is increasingly impacted by anthropogenic dissolved organic matter. Elevated protein-like fluorescence intensity also fingerprints sewage effluent within the estuary. The absorbance coefficient at 340 nm was found to have the strongest correlation to dissolved organic carbon concentration, greater than all fluorescence intensity parameters measured. However, fluorescence analysis permits the source of the dissolved organic matter to be determined, and therefore has implications for understanding its fate in estuaries and the ocean.

Microbial reduction of 99Tc in organic matter-rich soils.

For safety assessment purposes, it is necessary to study the mobility of long-lived radionuclides in the geosphere and the biosphere. Within this framework, we studied the behaviour of (99)Tc in biologically active organic matter-rich soils. To simulate the redox conditions in soils, we stimulated the growth of aerobic and facultative denitrifying and anaerobic sulphate-reducing bacteria (SRB). In the presence of either a pure culture of denitrifiers (Pseudomonas aeruginosa) or a consortium of soil denitrifiers, the solubility of TcO(4)(-) was not affected. The nonsorption of TcO(4)(-) onto bacteria was confirmed in biosorption experiments with washed cells of P. aeruginosa regardless of the pH. At the end of denitrification with indigenous denitrifiers in soil/water batch experiments, the redox potential (E(H)) dropped and this was accompanied by an increase of Fe concentration in solution as a result of reduction of less soluble Fe(III) to Fe(II) from the soil particles. It is suggested that this is due to the growth of a consortium of anaerobic bacteria (e.g., Fe-reducing bacteria). The drop in E(H) was accompanied by a strong decrease in Tc concentration as a result of Tc(VII) reduction to Tc(IV). Thermodynamic calculations suggested the precipitation of TcO(2). The stimulation of the growth of indigenous sulphate-reducing bacteria in soil/water systems led to even lower E(H) with final Tc concentration of 10(-8) M. Experiments with glass columns filled with soil reproduced the results obtained with batch cultures. Sequential chemical extraction of precipitated Tc in soils showed that this radionuclide is strongly immobilised within soil particles under anaerobic conditions. More than 90% of Tc is released together with organic matter (60-66%) and Fe-oxyhydroxides (23-31%). The present work shows that ubiquitous indigenous anaerobic bacteria in soils play a major role in Tc immobilisation. In addition, organic matter plays a key role in the stability of the reduced Tc.

Seasonal variations and mass closure analysis of particulate matter in Hong Kong.

The chemical characteristics of ambient particulate matters in urban and rural areas of Hong Kong were determined in this study. A monitoring program starting from November 2000 to February 2001 (winter) and June 2001 to August 2001 (summer) for PM10 and PM2.5 was performed at three monitoring stations in Hong Kong. Twenty-four-hour PM10 and PM2.5 samples were collected once every 6 days at two urban sites, PolyU and KT, and every 12 days at a background site, HT, with Hi-Vol samplers. High concentrations of OC, EC (except in PolyU), water-soluble ions and elements were observed in winter among the three sampling sites for PM10 and PM2.5 fractions. Seasonal variations were significant in background HT. Dilution effect due to the increase in mixing depth and precipitation in summer reduced the concentrations of particulate matters. Long-range transport could contribute to the higher concentrations of particulate matter in the winter. Chemical mass closure calculations were performed for PM10 and PM2.5 observed. Mass closure improved when separate factors (1.4 and 1.9 respectively) were used to convert water-soluble organic carbon (WSOC) and water-insoluble organic carbon (WINSOC) into corresponding organic masses. The urban sites showed high percentages of water-soluble ions in winter and high percentages of carbonaceous species in summer. Better results were obtained for the chemical mass closure analysis in winter than in summer. High temperature and solar radiation in summer increased the rate of the complex photochemical reaction in the atmosphere. Therefore the chemical mass closure analysis would underestimate the volatized species and secondary aerosols during summer.

Leaching of heavy metals (Cu, Ni and Zn) and organic matter after sewage sludge application to Mediterranean forest soils.

In Mediterranean dry and semiarid areas, soil organic matter is often depleted due to ancient and intensive human activity. Under these conditions the use of sewage sludge as a land reclamation technique may be a means to revert desertification processes and to enhance soil function and nutrient cycling. However, applications of heavy metal-contaminated sewage sludges can significantly increase potentially toxic metal concentrations in soils and metal transfer to freshwater and plants. The aims of this study are 1) to investigate the leaching of Cu, Zn and Ni from three contrasted Mediterranean forest soils (a basic loam, a basic clay, and an acid loam) treated with sewage sludges and 2) to explore the relationships between metal mobility and soil properties and with the leaching of organic matter. The selected soils were incubated in columns (5 replicatesx3 soilsx3 treatments). Treatments were (a) soil application of low metal content sewage sludge (LMS), (b) soil application of metal-enriched sewage sludge (MES), and (c) control. The sewage sludge application represented a dose of 6 kg dry weight m-2. Soil columns were incubated at room temperature for 110 days and were irrigated weekly with deionised water to make a total of a 1130 mm. Leachates were collected and analysed for pH, EC, organic carbon Cu, Ni and Zn concentrations. The concentration of metals and organic matter in the leachates depended on the soil characteristics and on the type of sewage sludge added to the soil. Basic soils with a high amount of clay showed the highest metal retention capacity, while acid soils with low clay content showed the lowest. Of the three metals studied, Ni exhibited the greatest mobility. Zn mobility was also rather high, particularly in the acid soil. Despite the fact that basic soils showed greater OM content than the acid soil, organic carbon in leachates after sludge addition was of the same order of magnitude in all studied soils. OM mobility may enhance the leaching of metals while the OM bound to soil particles may enhance the retention of metals. The rate of leaching per unit of metal in the soil, for Cu in all soils and for Zn in the basic soils, did not increase even in soils amended with metal-enriched sewage sludge. However, the total amount of Cu, Zn, and Ni leached through the 30 cm columns of the metal-enriched sewage sludge increased in comparison to the control soils. In spite of this fact, metal concentrations were in most cases well below limit for drinking water quality criteria of Spanish legislation. Thus, after one single addition of metal-enriched sewage sludge, for the studied Mediterranean soils, the risk of heavy metal pollution of groundwater appears to be low.

Bioreactivity of particulate matter in Beijing air: results from plasmid DNA assay.

An in vitro plasmid assay was employed to study the bioreactivity of PM (particulate matter) in Beijing air. It was found that the TD20 (toxic dose of PM causing 20% of plasmid DNA damage) of Beijing PM can be as low as 28 microg ml(-1) and as high as >1000 microg ml(-1). Comparison of the physical properties, such as morphology and size distribution, and oxidative potential indicates that the PM(2.5) (particulate matter with an aerodynamic diameter of 2.5 microm or less) has a stronger oxidative capacity than PM(10) (particulate matter with an aerodynamic diameter of 10 microm or less), and that the higher number percentages of soot aggregates and lower number percentages of mineral and fly ashes are associated with the higher oxidative capacity. Although the mass of PM(10) during dust storms is commonly 5 times higher than that during non-dust storm episodes, the oxidative capacity of PM(10)s of dust storms is much lower than that of the non-dust storm PM(10)s. The water-soluble fractions and intact whole particle solutions of Beijing airborne particles produce similar plasmid assay results, demonstrating that the bioreactivity of Beijing airborne particles is mainly sourced from the water-soluble fraction. In the samples with stronger bioreactivity, the total analyzed water soluble Al, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As and Pb (ppm) concentrations are higher. The water soluble zinc shows a good negative correlation with TD20s, suggesting that the water-soluble zinc is probably the major element responsible for the plasmid DNA damage.

Effects of land-use change on deposition and composition of organic matter in Frickenhauser See, northern Bavaria, Germany.

Characterization of bulk organic matter (OM) from lacustrine sediments of Frickenhauser See (northern Bavaria, Germany) reveals considerable variation during the last two millennia. Atomic C/N ratios and total organic carbon (TOC) content are positively correlated with arboreal pollen percentages which are used as an indicator of land-use intensity. Despite possible alterations of OM during early diagenesis, differences between three major lithological units are large enough to be interpreted as human impact on the sedimentation of OM in the lake. Sediments deposited prior to deforestation in the catchment area (unit C) are characterized by mean C/N ratios of 14.5, indicating a mixed composition of organic matter derived from lacustrine and terrestrial sources (forest litter). During a period of intensive soil erosion (ca. 1000 AD until 1870 AD; unit B), low C/N ratios of around 7.7 suggest that the relative proportion of forest litter decreased in favour of the lacustrine component and soil OM. Terrestrial plant detritus is only transported into the lake through numerous turbidite events. Deforestation and agriculture also coincide with a decreasing TOC content from an average value of 10.7% to 1.5%. This decrease is explained through a dilution effect due to the high input of minerogenic matter. Stable carbon isotopes indicate eutrophication of the lake due to the mobilization of nutrients through soil erosion. Starting around 1870 AD, organic-rich sapropelic sediments are deposited (unit A). A decline of turbidite events and increased wind shelter from trees due to reduced land-use intensity led to meromictic conditions. Consequently, negative excursions in the delta13C isotope record at the onset of unit A probably reflect methanogenesis under permanently anoxic conditions.

Effect of ghrelin in dry matter intake and energy metabolism in prepartum sheep: a preliminary study.

The objective of this study was to determine the effect of ghrelin on dry matter intake and energy metabolites in ewes during the last 10d of gestation. Ewes were randomly assigned to either a treatment (n=6) or a control group (n=6). One hour after feeding (08:00 and 16:00), ewes in the treated group were given ovine ghrelin (3 microg/kg, i.m.) and the control group received an equivalent volume of saline. Treatments were continued until lambing. Daily blood samples were taken starting at assignment. The principal findings were that: (1) ewes treated with ghrelin had a higher dry matter intake only on the first day of treatment; (2) treated ewes had lower serum concentrations of non-esterified fatty acids and insulin but higher serum concentrations of growth hormone; (3) serum concentrations of beta-hydroxy butyrate and cortisol were not different between treated and non-treated ewes. In conclusion, treatment with ghrelin affected dry matter intake and energy metabolites of pre-partum ewes during the last 10 d of pregnancy.

An IR study to investigate the structural relationship of lignin-like matter and lignosulphonates obtained from animal-vegetable wastes.

Lignin-like matter (I) from rice hull residues and humin-like matter and (II) from rice hull-fecal matter compost yield, upon sulphonation by SO(3), the lignosulphonates III and IV, respectively. The solid state IR spectra of I and II are analysed and interpreted based on the functional groups content values obtained for III and IV by solution NMR spectroscopy. IR absorbance values at five selected wavelengths for I and II correlate well with functional groups content values for III and IV according to Beer's law. It is concluded that the sulphonated derivatives, aside from the -SO(3)H content, reflect well the chemical structure of the parent materials. No side reaction of I or II with SO(3), other than C sulphonation, is apparent. The above correlation may be useful for waste management purposes.

Modification of soil humic matter after 4 years of compost application.

Two soil plots, 1 ha each, were amended yearly for 4 years, respectively, with 35.8 and 71.6 Mg ha(-1) yr(-1) of mature compost (CM) obtained from food and vegetable residues. The compost, amended soils, and a control soil plot after 4 years (S4), were analyzed for humin (HUC), humic acid (HAC), fulvic acid (FAC), and non-humic carbon (NHC) content. Compared to S4, the amended soil contained more humified C (HAC, FAC and HUC) and less NHC. Further evidence of the effect of compost on soil organic matter was obtained by the analysis of the humic acid (HA) fractions isolated from both the compost and the soils. These were characterized by elemental analyses and Diffuse Reflectance Infrared Fourier Transformed spectroscopy. The HAs isolated from CM and from S4 were significantly different. The HAs isolated from the amended plots were more similar to HA isolated from CM than to HA isolated from S4. The experimental data of this work indicate that the compost application may affect significantly the soil organic matter composition, and that the approach used in this work allows one to trace the fate of compost organic matter in soil.
