Intra- and extracellular element concentrations of rat renal papilla in antidiuresis.

The element concentrations in various intra- and extracellular compartments of the tip of the rat renal papilla were determined during antidiuresis using electron microprobe analysis. Urinary concentrations (means +/- SEM) were: urea, 1509 +/- 116; potassium, 268 +/- 32; sodium, 62 +/- 19 mmoles X 1(-1); and osmolality, 2548 +/- 141 mOsm X kg-1. Electrolyte concentrations in the interstitial space were: sodium, 437 +/- 19; chloride, 438 +/- 20; and potassium, 35 +/- 2 mmoles X kg-1 wet wt. The vasa recta plasma exhibited almost identical element concentrations. The values in the papillary collecting duct cells were: sodium, 28 +/- 1; chloride, 76 +/- 3; potassium, 135 +/- 3; and phosphorus, 316 +/- 7 mmoles X kg-1 wet wt. Similar concentrations were observed in the papillary epithelial cells. In interstitial cells potassium and phosphorus concentrations were virtually identical to those of the collecting duct cells, whereas sodium and chloride concentrations were higher by about 30 mmoles X kg-1 wet wt. The element composition of the various papillary cells is, thus, not substantially different from that of proximal tubular cells. This finding demonstrates that cellular accumulation of electrolytes is not the regulatory mechanism by which papillary cells adapt osmotically to their high environmental osmolality and sodium chloride concentration.

Surface analysis of ProFile instruments by scanning electron microscopy and X-ray energy-dispersive spectroscopy: a preliminary study.

AIM: The purpose of this preliminary study was to analyze surface irregularities on ProFile nickel-titanium (NiTi) rotary instruments before use, after sterilization by two different methods and after instrumentation of molar root canals. METHODOLOGY: Five new ProFile NiTi rotary instruments with 0.04 taper, size 20, five instruments with 0.04 taper, size 25 and five with 0.06 taper, size 20 were analyzed by scanning electron microscopy (SEM) and X-ray energy-dispersive spectroscopy (EDS). The instruments were then divided into two groups: three of each taper and size were sterilized in a dry heat sterilizer, whilst two of each taper and size were sterilized in an autoclave. After inspection by the same techniques, the files were used to instrument six molar root canals, and again analyzed by SEM/EDS. RESULTS: Together with the usual machining defects, a variable amount of material containing mainly carbon and sulphur was found on the surface of new instruments. This material was not removed by standard cleaning and sterilization procedures. The use of the files for instrumentation of root canals eliminated a considerable amount of the adhered substance, but deposits rich in calcium and phosphorus accumulated on the remaining impurities. CONCLUSIONS: The adherence of dentine on surface deposits found on new ProFile instruments requires special cleaning, prior to sterilization.

The friction and wear patterns of orthodontic brackets and archwires in the dry state.

Frictional resistance at the bracket-archwire interface has been demonstrated to impede tooth movement when sliding mechanics are used. Thus, the coefficients of friction of titanium and stainless steel brackets used in conjunction with stainless and ion-implanted beta-titanium archwires were investigated using a single contact interface between the brackets and archwires. The wear patterns between the brackets and the.016- in flat archwire surfaces were also examined using scanning electron microscopy and energy dispersive x-ray analysis. Stainless steel brackets tested with. 016-in flat stainless steel wire surfaces recorded the lowest coefficient of static friction mean (0.289), whereas titanium brackets paired with.016-in flat ion-implanted beta-titanium wire surfaces produced the highest mean (0.767). Stainless steel brackets had significantly (P <.05) lower coefficients of friction than titanium brackets for all wires except.020-in round stainless steel wires. Ion-implanted beta-titanium wires generally had significantly larger coefficients of friction than stainless steel wires. The increased friction of the titanium and ion-implanted beta-titanium alloys is also reflected in the severity of their wear patterns. An inverse relationship between friction and archwire surface dimension was generally found for ion-implanted beta-titanium wires. Round stainless steel wires demonstrated lower coefficients of kinetic friction than the flat stainless steel wire surfaces.

Effect of fluoride application on tensile bond strength of self-etching adhesive systems to demineralized dentin.

STATEMENT OF PROBLEM: It has been reported that the bond strength of composite to demineralized dentin is lower than that to sound dentin. This can be a problem in the success of so-called sealed restorations. PURPOSE: The aim of this study was to evaluate the effect of fluoride application on the tensile bond strength of self-etching adhesive systems to demineralized dentin. MATERIAL AND METHODS: One hundred twenty extracted bovine incisors were ground flat with 600-grit silicone carbide paper. Ninety teeth were then immersed in acetate buffer (pH 4.0) to form demineralized dentin. These teeth were randomly divided into 3 groups of 30 each: surfaces treated with fluoride solution (Group NF), surfaces treated with fluoride solution followed by a water rinse (Group RF), and control surfaces with no pretreatment (Group C). The remaining 30 teeth comprised a group with normal dentin surfaces (Group S). Each group was further divided into 3 subgroups of 10 each to test the bond strength test of Clearfil SE Bond, Unifil Bond, and Mac-Bond II. Tensile bond strengths (in MPa) were measured with a universal testing machine at a crosshead speed of 0.5 mm/min. Mean bond strengths were analyzed by 2-way analysis of variance and Fisher's PLSD (P =.05). SEM observations of the surfaces before and after priming and at resin-dentin interfaces in each group were performed. Elemental analysis of the dentin surfaces before priming was also carried out. RESULTS: The bond strengths of the adhesives to demineralized dentin in Groups NF, RF, and C were significantly lower than that of the normal dentin in Group S (P <.05). The mean bond strengths of the 3 adhesives in Group NF were higher than those in Groups RF and C, but a significant difference was observed only when Clearfil SE Bond was used (P <.05). In SEM images the open dentinal tubules on the surface and the resin tag formation at the resin-dentin interface were apparent in Group NF but were not observed in Groups RF and C. On elemental analysis, considerable amounts of fluoride and calcium were detected in the surfaces of Group NF. CONCLUSION: Within the limitations of this study, the surface treatment with fluoride solution supported the resin tag formation at the resin-dentin interface and slightly improved the bond strength of the self-etching adhesive systems tested to demineralized dentin. This result indicated that the fluoride treatment to demineralized dentin might contribute to the success of the sealed restoration.

Arc Welders' pneumoconiosis: application of advanced scanning electron microscopy.

Study of lung tissue from necropsy of a 58-year-old arc welder with arc welders' pneumoconiosis, confirmed by history, chest radiography, and pathology, demonstrates the versatility and usefulness of new techniques in scanning electron microscopy (SEM). Secondary electron imaging, the most familiar SEM mode, showed heavy cellular infiltrates in alveoli, the interstitium, and within the interstices of loose whorled fibrotic nodules. Backscattered electron imaging, in which contrast is proportional to elemental atomic number, revealed intracellular metal particles not otherwise visible. Microprobe analysis, energy-dispersive x-ray spectrometry, mapped elemeental iron over the particle image and identified traces of silicon in the whorled nodules. Arc welders' pneumoconiosis appears to be more than a benign siderosis resulting from particulate iron deposition. Simultaneous exposure to other components of welding fumes may alter the pathologic picture, inducing a more complicated fibrotic reaction. The more recently developed advanced techniques of SEM are well suited to the study of pneumoconioses and other problems of heterogenous tissue and mixed chemical systems.

Physical and chemical characterization of mn phosphate/sulfate mixture used in an inhalation toxicology study.

The use of methylcyclopentadienyl manganese tricarbonyl (MMT) in unleaded gasoline has given rise to numerous debates on the potential public health risk associated with manganese emissions. In fact, combustion products are mainly Mn phosphate, Mn sulfate, and Mn phosphate/sulfate mixture. Our research group did several inhalation studies in order to assess the toxicity of each Mn species. The objective of this study is to determine the physical and the chemical characteristics of a mixture of Mn phosphate/sulfate used in one of these inhalation toxicology studies. First, the mixture was analyzed by X-ray diffraction in order to obtain the specific peak of Mn phosphate and Mn sulfate. These peaks were used as reference. Second, samples of the mixture were collected on filters in the inhalation chamber at a concentration level of 3000 microg/m(3). They were analyzed by scanning electron microscopy (SEM), analytical transmission electron microscopy (ATEM), and x-ray energy-dispersive spectrometry (EDS) to show their size, morphology, and chemical composition. Results indicate that 33% of the particles were found to be agglomerated, while free particles accounted for 44% for Mn phosphate and 23% for Mn sulfate.

Morphological changes of human teeth with Er:YAG laser irradiation.

OBJECTIVE: The purpose of this study was to investigate morphological changes in human enamel and dentin structures irradiated with the use of an Er:YAG laser. SUMMARY BACKGROUND DATA: Despite the high number of research projects concerned with digital applications of lasers, few studies have targeted the effects of Er:YAG laser irradiation on human enamel or dentin structures. METHODS: Enamel and dentin surfaces of 40 extracted human teeth were irradiated for 5 seconds with a 2.94 microns wavelength, at an output of 300 mJ. Histological and scanning electron microscope studies were then performed to determine the morphological changes in the enamel and dentin structures of those teeth. RESULTS: The Er:YAG laser beam produced defects without signs of serious thermal damage to enamel or dentin. Undesirable thermal effects such as surface cracking or carbonization were not observed. Histological examinations revealed the presence of a thin basophilic line at the bottom and along the walls of defects in the dentin. These morphological changes were evidence of minimal thermal damage to surrounding tissue. CONCLUSIONS: These findings suggest that it is possible to remove the enamel and dentin using Er:YAG laser irradiation with minimal thermal changes. However, the mechanism involved in the ablation of enamel and dentin by Er:YAG laser irradiation has not been clarified in this study. Therefore, more research is necessary before the Er:YAG laser can be used clinically in the future.

Pulmonary silicatosis. A case diagnosed by needle-aspiration biopsy and energy-dispersive x-ray analysis.

The diagnosis of silicatosis was made by observing birefringent crystals in needle-aspiration biopsy specimens of bilateral basilar pulmonary masses in a clay worker, initially suspected of having metastatic neoplasm. Subsequently the crystals were examined by scanning electron microscopy and energy-dispersive x-ray analysis, which gave supportive evidence for their silicate composition. This experience indicates that needle-aspiration biopsy in conjunction with scanning electron microscopy and energy-dispersive x-ray analysis may be used in the diagnosis of benign pulmonary diseases, especially those of the inhalational fibrogenic variety.

Platelet studies in patients with hypertrophic cardiomyopathy.

Because of a shared abnormality of platelet and muscle in some neuromuscular diseases, platelets were investigated in five cases of hypertrophic cardiomyopathy, a disease likely to be primarily myogenic. Morphometric and chemical differences from normal were found consisting of increased cell size, decreased phosphorus concentrations and increased permeability to cations. The findings suggest membrane and energy metabolism aberrations.

Bone bonding ability of a new biodegradable composite for internal fixation of bone fractures.

Hydroxyapatite particles and poly(L-lactide) composites for internal fixation of bone fractures have been developed based on the hypothesis that incorporation of hydroxyapatite particles in a poly(L-lactide) matrix might enhance bone bonding. This study evaluated the bone bonding ability of these biodegradable composites. Two types of hydroxyapatite and poly(L-lactide) composite were used in this study: calcined hydroxyapatite/poly(L-lactide) and uncalcined hydroxyapatite/poly(L-lactide). Rectangular plates (2 x 10 x 15 mm) of each composite or poly(L-lactide) were implanted into the metaphysis of the tibiae of 33 male rabbits, and the failure load was measured by conducting a detaching test 8, 16, and 25 weeks after implantation. The failure loads of calcined hydroxyapatite/poly(L-lactide), uncalcined hydroxyapatite/poly(L-lactide), and poly(L-lactide), respectively, were 13.60, 13.95, and 0.46 N at 8 weeks; 29.84, 24.09, and 2.86 N at 16 weeks; and 25.50, 29.67, and 2.43 N at 25 weeks. Histologic observation revealed that the composites formed direct contact with the bone. The results in this study indicate that the composites improved the strength of the interface between bone and plate. This improved interfacial strength lead to a substantial decrease in the frequency of implant loosening in the treatment of fractured bones by internal fixation.

Acute calcification in alkali-injured rabbit cornea treated with synthetic inhibitor of metalloproteinases (SIMP).

PURPOSE: The aim of this study was to evaluate the ultrastructure of acute calcification observed in alkali-injured rabbit corneas treated with synthetic inhibitor of metalloproteinases (SIMP). METHODS: A 12-mm alkali burn was made on the right corneas of 16 rabbits. Eight eyes were treated with topical phosphate-buffered SIMP, and eight control eyes were given the buffer only. Six corneas (five in the SIMP group and one in the control group) developed chalky white opacification in the central area of the cornea within 6-30 days. The corneas were excised for the following examinations: (a) light microscopy, (b) energy-dispersive x-ray analysis, (c) electron microscopy, and (d) high-angle x-ray diffraction. RESULTS: The results confirmed that there was stromal calcification in these corneas, as shown by positive van Kossa staining, needle-shaped calcium crystal aggregates, and increased calcium in the opaque areas of the corneas. Proteoglycans in the corneal stroma of both the SIMP group and the control group significantly decreased after alkali burns. Very few proteoglycan filaments could be identified in the calcified areas of the stroma. X-ray diffraction reflections with a Bragg spacing of 3.44 +/- 0.04 A were observed from all the calcified corneas treated with SIMP. The intermolecular spacings of collagen fibrils in the calcified stroma (1.24 +/- 0.34 nm) were significantly lower than those in the noncalcified stroma (1.64 +/- 0.04 nm) and the control corneas (1.64 +/- 0.02 nm). The results indicate that the collagen molecules within the fibrils of the calcified stroma are packed more closely than normal. CONCLUSION: The comparison of the x-ray diffraction patterns from the calcified corneas and chicken bone suggests that the calcium deposits in these corneas are in a form similar to those found in bone. The higher rate and more extensive calcification in the SIMP-treated corneas may result from the inhibitory action of SIMP on debris processing, but the mechanism is unknown.

Isolation and characterization of hot particles from Chernobyl fallout in southwestern Finland.

Three types of activity composition have been found in airborne hot particles that were transported long distances from the Chernobyl accident. Their characterization is based on the analysis of single particles isolated from Pinus Sylvestris needles. The average activity of the particles was 130 Bq at the time of the accident. The most common type of particle contains the radioactive species 141Ce, 144Ce, 95Zr and 95Nb; the second type includes 103Ru and 106Ru along with the previous isotopes; and the third contains 103Ru and 106Ru only. Cesium-134 and -137 were present only in very small amounts. The activity composition of the Chernobyl reactor core fuel was similar to the composition of the first and second type particle; apparently the core fuel was only partially volatilized. The main bulk composition of the particles is shown to be U. The average aerodynamic size of the identified hot particles is 10 microns. The particles are rectangular or pentagonal in shape.

Detection of titanium in human tissues after craniofacial surgery.

Generally, titanium fixation plates are not removed after osteosynthesis, because they have high biocompatability and high corrosion resistance characteristics. Experiments with laboratory animals, and limited studies of analyses of human tissues, have reported evidence of titanium release into local and distant tissues. This study summarizes our results of the analysis of soft tissues for titanium in four patients with titanium microfixation plates. Energy dispersive x-ray analysis, scanning electron microscopy, and electrothermal atomic absorption spectrophotometry were used to detect trace amounts of titanium in surrounding soft tissues. A single metal inclusion was detected by scanning electron microscopy and energy dispersive x-ray analysis in one patient, whereas, electrothermal atomic absorption spectrophotometry analyses revealed titanium present in three of four specimens in levels ranging from 7.92 to 31.8 micrograms/gm of dry tissue. Results from this study revealed trace amounts of titanium in tissues surrounding craniofacial plates. At the atomic level, electrothermal atomic absorption spectrophotometry appears to be a sensitive tool to quantitatively detect ultra-trace amounts of metal in human tissue.

Fracture and fatigue behavior of shot-blasted titanium dental implants.

This investigation studies the effect of the shot-blasting treatment on the cyclic deformation behavior of a commercially pure titanium, with two microstructures: equiaxed and acicular. The fatigue tests were carried out in artificial saliva medium at 37 degrees C. Cyclic deformation tests have been carried out up to fracture, and the fatigue crack nucleation and propagation have been analyzed. The results show that the shot-blasting treatment improves the fatigue life in the microstructures studied, and that the equiaxed was better in mechanical properties than the acicular. The cause of this improvement in the mechanical properties is due to the compressive stress on the material surface for the shot-blasted specimens. Hardness tests were carried out to determine the value of these internal stresses.

Electron probe analysis of human skin: determination of the water concentration profile.

The water concentration profile across rapidly frozen human epidermis has been measured using electron probe analysis and analytical electron microscopy. Determinations were made within the cytoplasm of individual cells. From the basal layer, the water content remained relatively constant or decreased slightly across the viable tissue and decreased approximately linearly across the stratum corneum. A considerable discontinuity in water content occurred between these two regions over the stratum corneum-stratum granulosum junction and the last granular cell layer. The dominance of the water profile by a discontinuity suggests water loss is governed by a partitioning process, presumably a partitioning into the lipid domain. A water discontinuity offers important functional advantages in the conservation of substances within the body and in protection from intruding molecules.

Heterogeneous abnormalities of platelet dense granule ultrastructure in 20 patients with congenital storage pool deficiency.

Studies on platelet dense granule structure were carried out in 20 patients with various types of congenital storage pool deficiency (SPD), including 15 with specific deficiencies of dense granules and dense granule substances (delta-SPD), and five with combined deficiencies of dense and alpha-granules (alpha delta-SPD). Dense granules were identified by their high affinity for uranyl ions (uranaffin reaction), by their ability to accumulate the fluorescent dye mepacrine, and by their inherent electron opacity on unfixed, unstained whole mount preparations. By all these methods, dense granules were markedly decreased in seven albino patients with the Hermansky-Pudlak syndrome (HPS) variant of delta-SPD. These findings suggest that the basic defect in these patients is a specific abnormality in organelle development which prevents the formation of an intact granule structure, a quantitative abnormality which may differ from that in animals with related pigment disorders. In contrast, eight non-albino patients with delta-SPD had, on average, only a slightly reduced number of uranaffin-positive and mepacrine-positive granules, but a shift in uranaffin-granule distribution towards those lacking a dense core ('empty granules'), suggesting a more qualitative type of dense granule defect. These results are consistent with previous evidence suggesting a decreased uptake of ATP across the granule membrane in delta-SPD. In addition, on whole mounts, these patients' platelets contained substantial numbers of electron dense chains and clusters which contained P and Ca, but with a P/Ca ratio less than that of typical dense granules, and which were retained, along with a larger amount of ATP, after thrombin treatment of the platelets. The various findings in these patients raise the possibility that these structures may represent microvesicles, derived from the Golgi apparatus, which provide a transport mechanism for concentrating adenine nucleotides and calcium in dense granules and which is impaired in some patients with SPD. Additional defects may account for the more extensive granule abnormalities observed in alpha delta-SPD.
