"Delayed" phase separation in a gelatin/dextran mixture studied by small-angle light scattering, turbidity, confocal laser scanning microscopy, and polarimetry.

Small-angle light scattering, turbidity, and confocal laser scanning microscopy were used to study microstructure formation and evolution in a gelatin/dextran mixture. There was a time-delay of up to tens of minutes between reaching the quench temperature and the onset of phase separation, because demixing only occurred once a certain amount of ordering of the gelatin molecules, measured by polarimetry, was attained. The accompanying phenomenon of gelation retarded the development of the microstructure to different extents, depending on the quench temperature. At low temperatures, the structure was rapidly trapped in a nonequilibrium state with diffuse interfaces, characteristic of the early and intermediate stages of phase separation. At higher temperatures, coarsening continued for a certain amount of time before the structure was trapped. The duration of the coarsening period increased with increasing temperature and the interface between the phases became sharp, characteristic of the late stages of phase separation. Because the ordering process continued after the target quench temperature was reached, the effective quench depth continued to increase after the initial phase separation. At high quench temperatures, the system was able to respond to the thermodynamic requirements of the increasing effective quench depth by undergoing secondary phase separation to form a droplet morphology within the preexisting bicontinuous one.

Characterization of phase separation in film forming biopolymer mixtures.

Enhanced, tailor-made films can be achieved by combining the good gas barrier of the hydrophilic high amylose maize starch (hylon) with the water resistance of the hydrophobic protein zein. Two polymers are not always miscible in solution, and the phase separation behavior of the mixture is therefore important for the final film structure and its properties. Phase separation of a mixture of these two biopolymers was induced either by cooling, which was observed as growing droplets of the hylon phase which in some cases also formed small aggregates, or by solvent evaporation and studied in real-time in a confocal laser scanning microscope. Solvent evaporation had a much stronger effect on phase separation. During the early stage of phase separation, hylon formed large aggregates and subsequently smaller droplets coalesced with other droplets or large hylon aggregates. The later part of the separation seemed to take place through spinodal decomposition.

Comparing Nafion and ceramic separators used in electrochemical purification of spent chromium plating solutions: cationic impurity removal and transport.

This study focuses on the electrolytic regeneration of spent chromium plating solutions. These solutions contain a significant amount of chromium and a lesser amount of other heavy metals, which makes them a significant environmental concern and an obvious target for recycling and reuse. The type of separator used is extremely critical to the performance of the process because they are the major resistance in the transport-related impurity (Cu(II), Ni(II), and Fe(III)) removals from contaminated chromic acid solutions. A Nafion 117 membrane and a ceramic diaphragm separator traditionally used in the industry were tested for comparison. It was found that the mobilities of Cu(II) and Ni(II) were similar and higher than that of Fe(III) using both separators. The mobility of each cation was smaller in the Nafion membrane than in the ceramic diaphragm. The measured conductivity of the ceramic diaphragm was slightly higher than that of Nafion membrane. However, the Nafion membrane was much thinner than the ceramic diaphragm resulting in the system using the Nafion membrane having higher impurity removal rates than the system using the ceramic diaphragm. The removal rates were approximately equal for Cu(II) and Ni(II) and lowest for Fe(III). Both current and initial concentration affected the removal rates of the impurities. Modeling results indicated that a system using a Nafion separator and a small catholyte/anolyte volume ratio was better than a system using a ceramic separator for removing impurities from concentrated plating solutions if the impurities transported into the catholyte are deposited or precipitated.

A new method for separating HFC-134a from gas mixtures using clathrate hydrate formation.

A new separation method using gas hydrate formation is proposed for separating HFC-134a from gas mixtures containing N2 and HFC-134a. The feasibility of this separation method was investigated from various points of view. First, to determine the mixed hydrate stability region, three-phase equilibria of hydrate (H), liquid water (Lw), and vapor (V) for HFC-134a + N2 + water mixtures with various HFC-134a vapor compositions were closely examined in the temperature and pressure ranges of 275-285 K and 0.1-2.7 MPa, respectively. Second, the compositions of the hydrate and vapor phases at a three-phase equilibrium state were analyzed for identical mixtures at 278.15 and 282.15 K to confirm the actual separation efficiency. Third, kinetic experiments were performed to monitor the composition change behavior of the vapor phase and to determine the time required for an equilibrium state to be reached. Furthermore, X-ray diffraction confirmed that the mixed HFC-134a + N2 hydrates were structure II. Through an overall investigation of the experimental results, it was verified that more than 99 mol % HFC-134a could be obtained from gas mixtures after hydrate formation and subsequent dissociation processes. Separation of HFC-134a using hydrate formation can be carried out at mild temperature and low-pressure ranges. No additive is needed to lower the hydrate formation pressure.

Separation and analysis of diastereomers and enantiomers of cypermethrin and cyfluthrin by gas chromatography.

Synthetic pyrethroid (SP) insecticides are of environmental significance because of their high aquatic toxicity. Due to their chirality, SP compounds contain multiple diastereomers and enantiomers. However, due to great structural similarities and lack of isomer standards, gas chromatographic (GC) analysis of SP diastereomers or enantiomers is poorly developed. In this study, we used a HP-5 column to separate the diastereomers and a beta-cyclodextrin-based enantioselective column (BGB-172) to separate the enantiomers of cypermethrin (CP) and cyfluthrin (CF). Resolved peaks were identified by comparing chromatograms of isomer-enriched CP products. Diastereomers of both CP and CF were separated on the HP-5 column. On the BGB-172 column, enantiomers of all cis diastereomers were separated, while those of trans diastereomers were not separated. The elution order appears to be regulated by configuration, a finding which may allow peak identification in the absence of isomer standards. When coupled with electron capture detection, the developed methods had low detection limits and may be used for analysis of SP diastereomers and enantiomers in environmental samples.

A simple method to separate red wine nonpolymeric and polymeric phenols by solid-phase extraction.

Simple polyphenols and tannins differ in the way that they contribute to the organoleptic profile of wine and their effects on human health. Very few straightforward techniques to separate red wine nonpolymeric phenols from the polymeric fraction are available in the literature. In general, they are complex, time-consuming, and generate large amounts of waste. In this procedure, the separation of these compounds was achieved using C18 cartridges, three solvents with different elution strengths, and pH adjustments of the experimental matrices. Two full factorial 2(3) experimental designs were performed to find the optimal critical variables and their values, allowing for the maximization of tannin recovery and separation efficiency (SE). Nonpolymeric phenols such as phenolic acids, monomers, and oligomers of flavonol and flavan-3-ols and anthocyanins were removed from the column by means of an aqueous solvent followed by ethyl acetate. The polymeric fraction was then eluted with a combination of methanol/acetone/water. The best results were attained with 1 mL of wine sample, a 10% methanol/water solution (first eluant), ethyl acetate (second eluant), and 66% acetone/water as the polymeric phenols-eluting solution (third eluant), obtaining a SE of ca. 90%. Trials with this method on fruit juices also showed high separation efficiency. Hence, this solid-phase extraction method has been shown to be a simple and efficient alternative for the separation of nonpolymeric phenolic fractions and the polymeric ones, and this method could have important applications to sample purification prior to biological testing due to the nonspecific binding of polymeric phenolics to nearly all enzymes and receptor sites.

Separation of mixtures at nano length scales: blow torch and levitation effect.

A new conceptual basis for the separation of multicomponent molecular mixtures is proposed. A separation method where different components of the mixtures are driven in opposite directions is realized by a judicious combination of two effects, viz., levitation and blow torch effects. Monte Carlo simulations of two Lennard-Jones binary mixtures with different-sized components are shown to be separated well if at least one of the components lies in the anomalous regime and the others lie in the linear regime. A separation factor of 10(8) is obtained on nano length scales as compared to 10(3), obtainable through conventional methods of separation on macrolength scales.

Directed assembly and separation of self-assembled monolayers via electrochemical processing.

Separated domains of 1-dodecanethiolate were fabricated via solution displacement of preformed 1-adamantanethiolate self-assembled monolayers on Au{111}. Subsequently, the 1-adamantanethiolate domains were desorbed selectively, and the substrate was exposed to a 1-octanethiol solution, creating artificially separated self-assembled monolayers of 1-dodecanethiolate and 1-octanethiolate. The molecular order of each lattice type and the apparent height differences imaged with scanning tunneling microscopy and the two distinct cathodic peaks observed with cyclic voltammetry indicated distinct separated domains of each lattice type in the separated self-assembled monolayers. By manipulating the intermolecular interaction strengths of the patterned molecules, we are able to control the structure and properties of the separated self-assembled monolayers via the exploitation of competitive adsorption and the utilization of electrochemical processing, which can be extended to other self-assembly patterning techniques such as microdisplacement printing.

Phase separation in class II organically modified silicate films as probed by phase-imaging atomic force microscopy.

Phase imaging tapping mode atomic force microscopy has been used to examine the presence and extent of phase separation in organically modified silicate films. A total of seven films were prepared using two different synthetic routes (cohydrolysis/condensation and separate hydrolysis/condensation) with three different organoalkoxysilanes and tetraethoxysilane. Films made from separately hydrolyzed sols were rougher and had larger phase separated domains compared to films prepared from cohydrolyzed sols. Likewise, films prepared from sols containing organoalkoxysilanes with nonpolar bulky substituents, such as ethyl and isobutyl, were significantly rougher and more phase separated relative to those prepared from a more miscible nitrile alkoxysilane.

Replaceable cross-linked polyacrylamide for high performance separation of proteins.

An alternative sieving matrix, replaceable cross-linked polyacrylamide (rCPA), was developed for sodium dodecyl sulfate capillary gel electrophoresis (SDS-CGE) separation of proteins. This rCPA could be conveniently pressurized into separation capillaries under a pressure of 80 psi. SDS-CGE separations using this matrix generated high resolutions for a wide range (approximately 4 kD to approximately 300 kD) of proteins. When compared to the most frequently used sieving matrixes, the rCPA permitted the highest resolutions with comparable or increased separation speed for protein separations.

High gradient magnetic separation versus expanded bed adsorption: a first principle comparison.

A robust new adsorptive separation technique specifically designed for direct product capture from crude bioprocess feedstreams is introduced and compared with the current bench mark technique, expanded bed adsorption. The method employs product adsorption onto sub-micron sized non-porous superparamagnetic supports followed by rapid separation of the 'loaded' adsorbents from the feedstock using high gradient magnetic separation technology. For the recovery of Savinase from a cell-free Bacillus clausii fermentation liquor using bacitracin-linked adsorbents, the integrated magnetic separation system exhibited substantially enhanced productivity over expanded bed adsorption when operated at processing velocities greater than 48 m h(-1). Use of the bacitracin-linked magnetic supports for a single cycle of batch adsorption and subsequent capture by high gradient magnetic separation at a processing rate of 12 m h(-1) resulted in a 2.2-fold higher productivity relative to expanded bed adsorption, while an increase in adsorbent collection rate to 72 m h(-1) raised the productivity to 10.7 times that of expanded bed adsorption. When the number of batch adsorption cycles was then increased to three, significant drops in both magnetic adsorbent consumption (3.6 fold) and filter volume required (1.3 fold) could be achieved at the expense of a reduction in productivity from 10.7 to 4.4 times that of expanded bed adsorption.

Mother-child separations among homeless and housed families receiving public assistance in New York City.

We examined the incidence, characteristics, and predictors of separations of children from mothers in 543 poor families receiving public assistance, 251 of whom had experienced homelessness during the previous 5 years. Forty-four percent of the homeless mothers and 8% of housed mothers were separated from one or more children. A total of 249 children were separated from 110 homeless families and 34 children from 23 housed families. Children were placed with relatives and in foster care but were rarely returned to their mothers. Maternal drug dependence, domestic violence, and institutionalization predicted separations, but homelessness was the most important predictor, equivalent in size to 1.9 other risk factors. We infer that policies regarding child welfare and substance abuse treatment should be changed to reduce unnecessary placements. Studies of homeless children who remain with families may be biased if separated children are excluded.

Separating X-bearing human spermatozoa through a discontinuous Percoll density gradient proved to be inefficient by double-label fluorescent in situ hybridization.

PURPOSE: Double-label fluorescence in situ hybridization (FISH) was used to evaluate the efficiency of separating X- and Y-chromosome-bearing spermatozoa through 12-step discontinuous Percoll gradients. METHODS: Liquefied normal semen samples from 10 healthy donors were overlaid onto 25% Percoll and centrifuged. Parts of the sperm pellet were saved as control, while the remaining portion was separated by 12-step Percoll gradient. After centrifugation, the spermatozoa in the 80% Percoll layer were collected. The X:Y ratio of the control and separated spermatozoa was verified by double-label FISH (CEP SOX/SGY probes) and scored blindly by one observer. Differences in the X:Y ratios between matched groups were analyzed by paired t tests. RESULTS: The overall average labeling efficiency was 99.2%. A significant enrichment (P = 0.02) of X-bearing spermatozoa was obtained in Percoll separated fractions (mean X:Y ratio = 52.2:46.4) compared with the control group (X:Y ratio = 49.5:48.4). Discontinuous Percoll gradients also decreased the proportion of aneuploid spermatozoa (from 1.0 to 0.8%), but the differences were nonsignificant. CONCLUSIONS: Discontinuous Percoll separation did increase the X:Y ratio significantly, but the enrichment of X-bearing spermatozoa is insufficient for clinical use in preconceptional sex selection.

The contribution of photosynthetic pigments to the development of biochemical separation methods: 1900-1980.

The role of photosynthetic pigments in the development of separation methods in biochemistry during the period 1900-1980 is described beginning with M. Tswett who introduced separation of chlorophylls and carotenoids on columns and coined the term chromatography in 1906. In Uppsala, T. Svedberg developed the ultracentrifuge in the 1920s. A. Tiselius improved electrophoresis in the 1930s and developed chromatography of proteins in the 1940s and 1950s. Others of 'The Uppsala school in separation science' include J. Porath, P. Flodin and S. Hjertén who further developed various gel chromatographic methods. Hjertén introduced free zone electrophoresis in narrow tubes, a forerunner of capillary electrophoresis. Two proteins, phycoerythrin and phycocyanin, were used as test substances in all these methodological studies. Aqueous two-phase partitioning as a separation method was introduced in 1956 by the author. In this work, chloroplast particles were used, and the method was applied for the separation and purification of intact chloroplasts, inside-out thylakoid vesicles and plasma membranes. My research was carried out in cooperation with G. Blomquist, G. Johansson, C. Larsson, B. Andersson and H.-E. Akerlund during a 20-year period, 1960-1980.

Behavioral and hormonal responses to separation in infant rhesus monkeys and mothers.

Effects of social stimuli on behavioral and physiological responses were examined in infant rhesus monkeys at 4 and 9 months of age. Infants and mothers were removed from the social group and housed as dyads. Following this period, infants were removed and separated under four counterbalanced conditions: (a) totally isolated--placed in a holding cage for 24 hr; (b) mother present, no contact--housed in a single cage in view of their mother, no contact; (c) mother present, contact--similar to above, with mother in proximity to the infant; and (d) peer present--separated but in proximity to a peer. In the first experiment, the infants rarely vocalized when totally isolated but showed high rates of vocalization in the presence of the mother, both with and without contact. In the mother-present conditions, they failed to show a plasma cortisol response. In contrast, totally isolated infants showed a significant elevation in plasma cortisol. At 9 months of age, these infants were separated for 3 days under two different conditions: mother present and totally isolated. Once again, the infants that were totally isolated showed little vocalization but significant elevations in plasma cortisol. In contrast, infants separated in the presence of their mothers showed high vocalization rates but no cortisol response. The concepts of protest and despair are discussed as they relate to behavioral and physiological differences observed following different separation paradigms.

One role or two? The function of psychological separation in role conflict.

Student athletes vary in how much they view their academic and athletic role identities as separate from and interfering with each other. The authors investigated the relation of these perceptions to psychological well-being in 200 intercollegiate athletes. Measures included role separation, interference, identity, and well-being. Correlations indicated that interference related negatively to well-being, whereas viewing the roles as distinct related positively to well-being. Regression analyses of demographic and role identity variables also showed a positive association between role separation and well-being, and a significant Separation x Interference interaction. Specifically, role interference was negatively related to well-being for those who viewed the two roles as distinct but unrelated for those who did not. The buffering effects of role separation are discussed.

A limit on spin-charge separation in high-Tc superconductors from the absence of a vortex-memory effect.

There is a long-standing debate about whether spin-charge separation is the root cause of the peculiar normal-state properties and high superconducting transition temperatures of the high-Tc materials. In the proposed state of matter, the elementary excitations are not electron-like, as in conventional metals, but rather the electron 'fractionalizes' to give excitations that are chargeless spin-1/2 fermions (spinons) and charge +e bosons (chargons). Although spin-charge separation has been well established in one dimension, the theoretical situation for two dimensions is controversial and experimental evidence for it in the high-Tc materials is indirect. A model with sharp experimental tests for a particular type of separation in two dimensions has recently been proposed. Here we report the results of those experimental tests, placing a conservative upper limit of 190 K on the energy of the proposed topological defects known as visons. There is still debate about the extent to which this experiment can settle the issue of spin-charge separation in the high-Tc copper oxides, because some forms of the separation are able to avoid the need for visons. But at least one class of theories that all predict a vortex-memory effect now are unlikely models for the copper oxides.

Cycles and separations: the histotopography of the urinary concentrating process.

We have analyzed the histotopography of the renal medulla of the rat in terms of cycles and separations. Cycles are pathways by which solute leaving the medulla in an ascending structure (AVR, AHL) is returned to a deeper medullary level. Separations are based on spatial incontiguity and special characteristics of the interstitium and blood supply. The two concepts are complementary: the compartmentalization resulting from separations imparts specificity to the cycles. Structural lateral heterogeneity, consisting in distinct domains organized around vascular bundles, is present in one form or another in all three medullary zones. Such compartmentalization probably leads to heterogeneity in interstitial solute concentrations, a state of affairs inconsistent with the requirements of a "central core". In all such considerations of exchanges between compartments, the lack of a unitary interstitium must be borne in mind. Instead, three general types of interstitium may be distinguished: corresponding roughly to those of the OS and VB, the interbundle region of the IS, and the IM. Among the histotopographic features of the renal medulla not usually included in models of the urinary concentrating mechanism but likely to have functional significance are the association of CD with completely distinct populations of AVR and AHL in the OM and IM; a clear-cut separation throughout the medulla between cycles involving long loops and those involving short loops; the lack of an effective countercurrent association between ascending and descending limbs of short loops in the IS; and a pronounced separation of the venous drainage of the IM from that of the OM.
