Acute monoblastic leukaemia. Clinical, biological data and survival in 45 cases.

Between 1978 and 1980, 45 cases of acute monoblastic leukaemia have been diagnosed, treated and followed in our institute. Morphological diagnosis was performed according to the French-American-British classification. Tumoral syndrome (particularly extra-medullary) and hyperleucocytosis were the most striking findings at the time of diagnosis. Cytogenetic analysis performed in 31 cases before treatment has showed that abnormality of the long arm of chromosome 11 seemed to be more frequently associated with the poorly differentiated cytological subtype M5 (a). Intensive chemotherapy with zorubicin and cytosine arabinoside led to complete remission in 75% of the cases. Central nervous system prophylaxis appeared definitively useful in preventing meningeal relapse. Despite a prolongation of the median duration of complete remission which now reaches 12 months, the prognostic is still poor.

Computer-assisted long-term measurements of urinary output and other biological data.

A computerized system is described, combining automatic collection of urine in short intervals (minutes) over long periods (days) and recordings of body temperature, MABP, and heart rate in chronically instrumented conscious dogs. During the studies the dogs are housed in metabolic cages. Indwelling catheters and electrical wires are connected to a specially designed swivel and directed out of the cage to the next room. Infusions, blood sampling, and monitoring can be performed from this room without disturbance to the dogs. Three examples of recordings are given. In one of these examples the sodium excretion patterns on 5 consecutive days under continuous saline infusion in one dog is evaluated. Urine was collected every 20 min. Sodium excretion showed cyclic variations. Fourier analysis exhibited 18-h periods and 4- to 8-h periods. The described system renders, e.g., coherent time series analysis possible for a variety of simultaneously recorded physiological variables and may thus acquire considerable importance for integrative physiology.

Inhibitory effect of aryl thienyl-ketones and -thioketones on arachidonic acid-induced malondialdehyde formation in human platelets: biological data and molecular modelling.

A series of anti-thrombotic aryl thienyl-ketones and -thioketones was assayed in vitro for their inhibitory effect on malondialdehyde (MDA) production induced by arachidonic acid in human platelets. For several compounds MDA formation was strongly inhibited indicating that the anti-platelet target was situated on the cyclooxygenase pathway. A comparison between the inhibition constant Ki and the IC50 values revealed competitive inhibition kinetics. The molecular structure of one active compound was analysed by X-ray diffraction and theoretical calculations to provide information on its electronic and lipophilic properties.

[Histomorphometric and biological data on osteoporosis due to immobilization].

The effect on the bones of immobilization was studied by means of quantitative histological methods on 34 biopsies of the iliac crest after different periods of immobilization in 28 patients, 22 of whom were immobilized by medullary lesions. At the same time a biochemical study was carried out on 68 immobilized patients over a 52-week period. This histomorphometric study included measurements of the absolute volume of trabecular bone, the volume of the osteoid matrix, the osteoclastic resorption surfaces, the size of the osteocyte lacunae, the thickness of the iliac cortical zones and the medullary adipose volume. 19 subjects received in addition double labeling with tetracycline with the object of carrying out a histodynamic study of the osteoblastic activity. The decrease in the absolute volume of the trabecular bone reached 33 percent of its original value and continued up to the 25th week, and was followed by stabilization at a value slightly higher than the vertebral fracture limit. Immobilization also led to an increase in the osteoclastic resorption surfaces and, secondarily, to an increase in the periosteocyte lacunae, a thinning of the iliac cortical zones, and a decrease in the volume of the osteoid matrix. Calciura increased as did total hydroxyprolinuria in parallel with an increase in the resorption surfaces. The histological and biochemical changes suggest a histodynamic hypothesis, according to which the longevity of the basic multicellular bone unit increases, leading to a new equilibrium characterized by very slow renewal of the bone.
