[The antioxidant treatment of patients with acute intestinal infections due to gram-negative microorganisms].

It was established that administration of S. typhimurium lysate causes activation of the processes of free-radical oxidation. Introduction of unithiol in combination with magnesium sulfate returns the intensity of these processes to the initial level. Unithiol and alpha-tocopherol acetate protect the animals form lethal effects of S. sonnei lysate. Use of a combination of pharmacological agents consisting of unithiol, magnesium sulfate and alpha-tocopherol acetate in the treatment of patients with acute intestinal infections due to gram-negative pathogens (shigellae, salmonellae, proteus, klebsiellae, pseudomonas) resulted in decrease of the length of the intoxication and diarrhea syndrome, decrease of the frequency of repeat bacteria elimination and protracted forms of these infections, more rapid normalization of immune system functions.

[Effect of sulfhydryl compounds on the automatism of the pacemakers].

Experiments were performed on the isolated rabbit hearts and also on the hearts with complete atrioventricular block; a study was made of the effect of an excess or deficiency of the sulfhydryl groups on the automatism of the cardiac pace makers. Unithiol and cysteine in concentrations of 1-10(-6)-1-10(-4) g/ml were used as donors of sulfhydryl groups; deficiency of these groups was induced by the alloxan administration in concentrations of 1-10(-5)-5-10(-5) g/ml. Changes in the sulfhydryl group content produced no marked effect on the automatism of the synoatrial node. An excess of sulfhydryl groups promoted poststimulation depression of the automatism of the potential pace makers of the cardiac ventricles and could lead to the origination of Luciani's periods. On the contrary, in case of a deficiency of the sulfhydryl groups there was a sharp elevation of the automatism of the ventricular pace makers, the atrioventricular conduction became disturbed, and the poststimulation depression of the automatism became considerably diminished. Disturbances of the cardiac activity caused by the sulfhydryl group deficiency were completely eliminated by unithiol or cysteine.

[Effect of protectors on morphofunctional changes in rat kidneys in tourniquet shock].

The protective effect produced on the kidneys in tourniquet shock by thiol agents possessing antioxidant properties (unithiol, cystamine) was compared with that of a preparation of cell capsules of L. bulgaricus (CCLB). Tourniquet shock was modeled under ether Raush anesthesia by applying 8 turns of a rubber tourniquet to the hind limbs for 6 hours. Thirty minutes before removal of the tourniquet animals of the first group were given an injection of 1 ml of physiological solution, animals of the second group--20 mg/kg of unithol, those of the third group received an injection of 20 mg/kg of cystamine hydrochloride. Animals of the fourth group received 50 mg/kg of CCLB in 1 ml of physiological solution 7 days before the experiment. A marked protective effect of CCLB on the kidneys in tourniquet shock was established. According to the degree of the protective effect on the tissue of kidneys in tourniquet shock, the studied agents come in the following order: CCLB, cystamine, unithiol.

[Growth and metastasis of Lewis' lung carcinoma in mice under the pharmacological action on metabolism and the effects of kinins].

In experiments on C57 Bl mice the effects of growth and metastatic proliferation of Lewis lung carcinoma inoculated intramuscularly on metabolism and kinin activity have been studied during the application of pharmacologic agents (parmidin, cellulose sulphate, unithiol). Causing a weak inhibition of the primary tumour growth, parmidin if administered repeatedly in given doses, inhibits partially metastatic process, which is manifested in an essential decrease of the quantity, size and weight of lung metastases. Parmidin also inhibits an adverse effect of cellulose sulphate and unithiol on metastatic process. It is believed that the action of parmidin is of an indirect character and is mainly associated with the elimination of the kinin component of microcirculatory disorders in the lungs.

[Comparative evaluation and analysis of the bradykinin-potentiating properties of D-penicillamine and other thiol and non-thiol kininase inhibitors].

In experiments with an isolated guinea-pig ileum D-penicillamine (3,3-dimethylcysteine) potentiated the spasmogenic effect of bradykinin and exerted no influence on the analogous effects of acetylcholine, histamine and barium chloride. These properties distinguish the drug from other thiol compounds -- unithiol and cysteine. The bradykinin-potentiating action of D-penicillamine excludes the effect on the cholinergic and histaminergic systems, while the bradykinin-potentiating effect of unithiol and cysteine is reversed by atropine and tavegil. 1, 10-Phenanthroline and 8-hydroxyquinoline do not possess the bradykinin-potentiating properties, and, when administered in high doses, reduce the spasmogenic effect of the polypeptide.

Chelation therapy for methylmercury(II) poisoning. Synthesis and determination of solubility properties of MeHg(II) complexes of thiol and dithiol antidotes.

Methylmercury(II) complexes of the most widely studied antidotes for mercury poisoning have been prepared, and both the water solubility and 1-octanol/water partition coefficients determined for these complexes and the L-cysteine complex. New complexes of N-acetyl-D,L-penicillamine, 2-mercaptosuccinic acid, meso-dimercaptosuccinic acid, and Unithiol have been synthesized and characterized, and are found to have the formulations MeHgSCMe2CH(NHCOMe)CO2H, MeHgSCH(CO2H)CH2CO2H, MeHgSCH(CO2H)CH(CO2H)SHgMe, and Na[MeHgSCH2CH-(SHgMe)CH2SO3], respectively. Trends in octanol/water partition coefficients are consistent with reported studies of the effectiveness of antidotes for MeHg(II) poisoning and redistribution of MeHg(II) on administration of antidotes, particularly for British anti-Lewisite, Unithiol, and meso-dimercaptosuccinic acid.

Prognosis of mercury poisoning in mercury refinery workers.

The prognosis of chronic metallic mercury poisoning in two groups of patients from the mercury refinery of a mercury mine was evaluated by reexamination which included an interview, physical and neurological examination and determination of urinary mercury. Group I consisted of 70 male patients, who had been exposed to metallic mercury for 1.6-17.8 years, 15 of whom had been diagnosed 10 years earlier as having severe chronic metallic mercury poisoning, and the rest moderate chronic metallic mercury poisoning. At the time of reexamination, they had been removed from mercury exposure for 2 months-17 years. None of them had been treated with any chelating agent. Group 2 comprised 84 male patients from the same mercury mine, who had been exposed to metallic mercury for 2-10 years after 1962 and had been previously diagnosed as having mild chronic metallic mercury poisoning. They were reexamined after 2 months of hospital admission and chelation treatment with unithiol or sodium dimercaptosuccinate (Na-DMS). Based on clinical evaluation, the condition of the patients in both groups had all improved, even in the severe cases. The overall prognosis of chronic metallic mercury poisoning in mercury refinery workers was encouraging after termination of mercury exposure. Chelation therapy with unithiol or Na-DMS was evidently beneficial for reducing urine mercury and some symptoms, but not for neurological and stomal signs.

[Action of heavy metal salts on the development of sea urchin embryos and on protein synthesis by the cells of transplantable mouse tumors].

Zu, Cu, Cd, Pb and Hg salts blocked the cleavage divisions in the sea urchin embryos and, in lesser concentrations, affected gastrulation and induced postgastrulation defects. The early embryo- and cytotoxic effects of Zn salts appear to be based on the inhibition of protein synthesis. Unithiol exerted a protective effect. All salts under study inhibited protein synthesis in the Ehrlich's ascite carcinoma and ascite hepatoma 22a cells; unithiol and other suldhydryl compounds exerted in this case a good protective effect against Cd salts only.

[The correction of free-radical oxidation processes in patients with chronic cholecystitis subjected to ionizing radiation exposure].

An activation was found of lipid peroxidation system as well as suppression of antioxidant defence tending to be more profound in blood erythrocytes, in 47 patients with chronic cholecystitis having been in the zone of abnormal radiation. Correction of the above alterations revealed unithiol, clinical and biochemical effects to be pronounced compare with those or tocopherol. Direct antioxidant effect or unithiol, its capability of serving as an antidote of heavy of heavy metals are believed to account for the above observation.

Variations of the gravitational field as a motive power for rhythmics of biochemical processes.

Variations of the gravitational field affected by the Sun and the Moon while the Earth's moving along the orbit seem to be a powerful source of many rhythmical processes typical of biochemical processes. Studies carried out in AARI revealed the obvious relationships between the dynamics of some biochemical reactions and lambda(D)-function describing the regular variations of the gravitational field under combined influence of the Sun and the Moon. The following of them are examined as examples: the rate of the unithiol oxidation in vitro, concentration of the thiol compounds in human urine, some hematological indicators (rate of the erythrocytes sedimentation, hemoglobin content). Compatibility of run of the biochemical indicators and lambda(D)-function is indicative of essential influence of the regular variations of the gravitational field on rhythmics of the biochemical processes. As this takes place, the solar activity acts like to the instability factor. Balance of the solar activity effects and the varying gravitational field effect alter in time depending on location in the solar activity cycle.

[Use of antioxidant defense of tissues in free microsurgical transplant of complex-composite flaps].

In experiment on 25 epigastric muscular flaps of rabbits the possibilities of the flap tissues damage prophylaxis in the ischemia period were studied up as well as their defense from peroxidal and the free-radicals oxidation of the membrane phospholipids in the reperfusion period. There were noted suppression of the lipid's peroxidation processes, the raise of the flap's tissues antioxidant defense, the cellular and subcellular membranes stabilization with preservation of the vessels endothelium integrity after preliminary intraarterial infusion of the lipin, unithiol, ascorbic acid and isoptin mixture into the flap. The data obtained witness the perspective of the studied preparations application during microsurgical transplantation of composite complexes of tissues.

[Detection of pulsed cosmic radiation].

Dynamics of the oxidation rate of unithiol (sodium dimercaptopropansulfonate) by sodium nitrite was studied under perfect ecological conditions in Antarctica. Short-term spikes (signals) in optical density of the examined solution were found. Special features of these signals, such as the high penetrating ability, the obvious dependence of number of signals on the Sun' longitude, and the high speed of propagation (V > or = 300,000 km/s), indicate the astrophysical origin and possible gravitational causality of these signals.

Are 2,3-dimercapto-1-propanesulfonic acid or prussian blue beneficial in acute thallotoxicosis in rats?

Unithiol (2,3-dimercapto-I-propanesulfonic acid, DMPS) and prussian blue (potassium ferric hexacyanoferrate (II), PB), given alone or in combination, were evaluated as antidotes to treat acute thallotoxicosis in male Sprauge-Dawley rats. Animals were poisoned with 20 mg thallium (TI)/ kg bw PO on day 0 using thallous sulfate. On day 1 (24 h later), treatments began and were continued through day 4 as follows: 50 mg PB/kg bw PO, 2/ d; 5 mg DMPS/kg bw IP, 6/d (day 1), 4/d (day 2), 2/d (days 3-4); or the combination. Animals were sacrificed 24 h after the last treatment (day 5), and TI concentrations in kidney, liver, heart, brain, whole blood and feces determined by electrothermal atomic absorption spectroscopy. The relative accumulation of TI was kidney>>heart>liver approximately equal brain. PB limited incorporation of TI in all tissues. DMPS failed to significantly decrease TI in any organ, but significantly decreased TI in whole blood. PB+DMPS treatment significantly decreased the TI content in all organs, but not to a greater extent than PB alone. PB and PB+DMPS treatments significantly increased TI in feces, whereas DMPS alone produced little effect. This study confirms that PB is beneficial in the treatment of acute thallotoxicosis in rats. The failure of DMPS to significantly affect TI in target organs suggests it is not useful in treating TI poisoning.

[Curative effect of TCM-WM therapy on Wilson disease with different clinical phenotypes and polymorphisms of ATP7B gene].

OBJECTIVE: To investigate the curative effect of TCM-WM therapy on Wilson disease (WD) with different clinical phenotypes and polymorphisms of ATP7B gene. METHODS: One hundred and twenty-two WD patients with different clinical phenotypes were treated with unithiol 20 mg/kg per day and Gandou tablet ([symbol: see text]) for 1 month. Exon 18 of ATP7B gene extracted from patients and 20 healthy volunteers were amplified with PCR technique, mutations and polymorphisms were screened by SSCP analysis. RESULTS: Four kinds of abnormal migration bands in PCR-SSCP were seen in 37 patients, mutation frequencies of three different clinical phenotypes and curative effect between mutation and non-mutation patients did not show any significant difference, P > 0.05. But total effective rate of patients with Wilson type or pseudosclerosis type was significantly higher than that of patients with hepatic type, P < 0.05. CONCLUSIONS: The exon 18 of ATP7B gene of Chinese WD patients may be one of the regions with higher mutation frequency, most of them are compound heterozygotes and patients with different clinical phenotypes have different response to TCM-WM therapy. It indicate that specific mutations at least in part play a role in influencing clinical phenotypes and curative effect.

[The efficacy of treating glomerulonephritis patients with a hematuria component].

UNLABELLED: A clinical evaluation was carried out of treatment efficacy of patients suffering of glomerulonephritis with hematuria component. TREATMENT: membrane stabilizing therapy including unithiol, dimephosphon (delagil) and alpha-tocopherol acetate. Duration of this treatment was 1 month; 2 months--with alpha-tocopherol and delagil; 6 months--with delagil. TREATMENT was carried out in 80 patients. Six-month follow-up was evaluated in 62 patients. In 48 patients the treatment was effective as concerns proteinuria and in 4--only hematuria. Comparing the results of such treatment with indomethacin treatment where a positive dynamics was observed in 10 of 38 patients and only in 2 patients improvement concerned proteinuria and erythrocyturia. CONCLUSION: membrane stabilizing therapy is the method of choice in comparison with traditional indomethacin treatment.
