Translational control mechanisms modulate gene expression in a variety of cellular and viral systems. Using hypertonic conditions to block protein synthesis in vivo, we observed that the synthesis of several major equine herpesvirus type 1 proteins was selectively inhibited. Although sensitivity to hypertonic conditions was graded across a continuum, messages coding for proteins of 203, 130.5, and 31.5 kDa were significantly more resistant to higher salt concentrations in vivo than those coding for polypeptides of 148, 116, and 74 kDa. Similar results were observed in vitro when potassium acetate was used to block initiation. In addition, Northern blot analyses demonstrated that steady-state levels of cellular mRNAs declined beginning at about 6 hr after infection. Taken together, these results indicate that the expression of several major equine herpesvirus type 1 genes was controlled in part at the post-transcriptional level.

BACKGROUND: To study the epidemiological pattern of Hodgkin's disease (HD) in children of Southern Iran, a total of 120 cases of biopsy-proven Hodgkin's disease, less than 18 years old, submitted for histological diagnosis during 1987-1997 were classified, using the Rye's Classification. MATERIAL/METHODS: Data on age, sex, number in households, socio-economic status, clinical presentation, signs and symptoms, para-clinical data at the time of diagnosis, treatment plan, recurrence and the presence of metastases were derived from the patients' history, and analyzed. RESULTS: The largest subgroups were those of Mixed cellularity (66.7%), followed by Nodular sclerosis (23.3%), Lymphocytic predominance (8.3%) and Lymphocytic depletion (1.7%). The male-to-female ratio was 2.87. Patients with Nodular sclerosis were generally older than those in other groups. The mean age in all histopathological classes was higher in females. CONCLUSIONS: The epidemiological pattern of the HD in southern Iran is similar to that of other developing countries and the northern part of Iran, in which mixed cellularity is the most common subtype. However, this pattern differs from the pattern seen in the United State and Europe, since Nodular sclerosis is the most common subtype in these regions.

We have previously demonstrated that glucocorticoids induce differentiation of human bone marrow stromal cells (BMSC) into cells expressing mature osteoblast phenotype. As glucocorticoids have marked effects on extracellular matrix protein synthesis in bone, and because proteoglycans are important components of bone matrix and may condition the differentiation and biological activities of osteoblasts, we studied the effects of dexamethasone (Dex) on the synthesis of small proteoglycans [decorin (DCN) and biglycan (BGN)] in adult human BMSC and human osteoblasts (HOB). First passaged HOB and BMSC were treated with either ethanol or 10(-7) M Dex for 7 days. After treatment, the cells were metabolically labeled with either [35S]SO4 alone or [35S]SO4 and [3H]leucine together for 24 h. Conditioned media were collected, and cell layers were extracted with 4 M guanidine HCl. The extracts and conditioned media were subjected to gel filtration and ion exchange chromatography. Fractions containing radiolabeled proteoglycans were analyzed either directly or after immunoprecipitation by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Dex treatment resulted in a dramatic increase in DCN and an associated decrease in BGN in both the conditioned medium and the cell layer of HOB cultures. In Dex-treated BMSC cultures, BGN was decreased in both the conditioned medium and cell layer, whereas DCN was stimulated in the majority of cultures. Northern blot analysis indicated that steady state messenger RNA (mRNA) concentration of DCN was increased by Dex in all of the HOB cultures and in seven of eight BMSC cultures analyzed. The steady state mRNA level of BGN was decreased by Dex in both HOB and BMSC cultures. The regulation of DCN and BGN mRNA by Dex in both HOB and BMSC (when responsive) was dose dependent. Time-course analysis indicated that as little as 1 day of treatment with Dex was sufficient to decrease BGN mRNA and increase DCN mRNA (when observed) levels in BMSC; the regulation spanned a 4-week interval, during which the extracellular matrix of BMSC was mineralized. The effect of Dex on the steady state mRNA levels of DCN and BGN in HOB was also apparent after 1 day of treatment. These accumulated results suggest that Dex modulates the synthesis of small proteoglycans in both human bone marrow stromal osteoprogenitor cells and mature osteoblasts.
Giant nuclear transcripts, and in particular the RNAs of the globin gene domains which are much larger than their canonical pre-mRNAs, have been an enigma for many years. We show here that in avian erythroblastosis virus (AEV)-transformed chicken erythroleukaemic cells, where globin gene expression is abortive, the whole domain of alpha-globin genes is transcribed for about 33 kb in the globin direction and that this RNA is part of the nuclear matrix. Northern blot hybridisation with strand-specific riboprobes, recognising genes and intergenic sequences, and RT-PCR with downstream primers, show that the continuous full domain transcript (FDT) starts in the vicinity of a putative LCR and includes all the genes as well as known regulatory sites, the replication origin, and the DNA loop anchorage region in the upstream area. Absent in chicken fibroblasts, the globin FDT overlaps the major part of the ggPRX housekeeping gene that is transcribed in the opposite direction. RT-PCR and in situ hybridisation with genic and extra-genic globin probes demonstrated that the globin FDT is a component of the nuclear matrix. We suggest that the globin FDTs keep the domain in an active state, and the globin RNAs on the processing pathway are a component of the nuclear matrix. They may take part in the dynamic nuclear architecture when productively processed, or turn over slowly when globins are not synthesised.

The expression of ten proto-oncogenes was studied in cell lines derived from transplantable mouse teratomas. The cell lines represent different forms of early embryonic cell specialization. The analysis included two embryonal carcinoma (EC) lines (PCC3 and F9), and four differentiated cell lines derived from teratocarcinoma, namely trophoblastoma (3-TDM), parietal endoderm (PYS-2), visceral endoderm (PSA5-E) and skeletal myoblasts (Cl10). The expression of c-oncogenes was studied by analysing poly(A)+RNA for complementary sequences by dot blot and Northern blot hybridization. The results were related to the rate of cell multiplication and the state of differentiation by examining [3H]thymidine incorporation, growth curves and tissue-specific differentiation markers. Expression of c-myc and c-Ki-ras was found in all cell lines. In dot blot assays, poly(A)+RNA from all cell lines also hybridized with v-abl and v-sis probes. A marked decrease in c-myc expression was found in teratoma-derived myoblasts differentiating into myotubes. A similar reduction was found when 'nullipotent' F9 cells were induced by retinoic acid (RA) to form primitive endoderm. However, reduction of the growth rates of the parietal and visceral endodermal cell lines were not accompanied by decreased expression of c-myc or c-Ki-ras. Hybridization signals obtained with a v-sis probe was low in all teratoma-derived cell lines tested, except for the myogenic cell line Cl10. Both in exponentially growing and differentiated cultures of this line, two size classes of transcripts hybridized strongly to the v-sis probe. However, these transcripts, 7 and 3 kb, most likely represent endogenous retroviral transcripts and not c-sis transcripts. Expression of c-myb, c-mos, c-fes, c-src and c-erb A and c-erb B could not be detected in any of the cell lines studied.
Recent studies have found that many young women just beginning their sexual lives use alcohol prior to intercourse. A large number appear to drink heavily enough prior to sex to compromise their ability to use contraceptives. The question emerges whether there is a relationship between drinking before intercourse, the nonuse of birth control methods, and unplanned pregnancies. The present research describes 43 instances of intercourse which resulted in unplanned pregnancies in 14- to 21-year-olds. Variables examined included alcohol use prior to sex, amount of alcohol consumed, the use of other drugs, the planning of intercourse, respondents' stated reasons for nonuse of contraceptives, and other general demographic data.
Six biopsies (4 of human fibrosarcomas, 1 of a B-cell lymphoma and 1 of a normal lymph node from a melanoma patient) and 6 cell lines (derived from 5 different human osteosarcomas and from 1 rhabdomyosarcoma), together with control cells, were examined for the expression of c-sis, c-fos and c-myc. The expression of c-sis/PDGF-B-related proteins was also examined in cultured cells (not in biopsies). In situ hybridization studies further showed that the occurrence and level of expression of c-sis mRNA and c-sis/PDGF-B-related proteins were significant in the tumor cells. Expression of c-fos and c-myc mRNA did not correlate with c-sis expression. Southern blot analysis of c-sis, c-fos and c-myc of 20 DNAs of cell lines derived from human sarcoma or biopsies showed an identical pattern for BamH1 and EcoR1 restriction fragments of c-sis (except for 1 fibrosarcoma biopsy), implicating no gene rearrangement as a cause of enhanced proto-oncogene expression. The nucleotide sequence of c-sis is highly homologous to that of the viral v-sis oncogene which is capable of transforming infected cells. We conclude that enhanced expression of c-sis in the sarcomas we have examined is involved in the initiation and/or maintenance of the cell transformed state.

Treatment of HepG2 cells with forskolin led to 60-100% stimulation of system A activity, measured as the Na+-dependent uptake of alpha-(methylamino)isobutyric acid. The stimulation was reproducible with cholera toxin and dibutyryl cAMP, and inhibitable by H7, a non-specific protein kinase inhibitor. The stimulatory effect was eliminated by cycloheximide and actinomycin D. The forskolin effect was associated with an increase in the maximal velocity of the transport system, with no change in substrate affinity. These cells express three different subtypes of system A (ATA1, ATA2, and ATA3). Treatment with forskolin increased the steady-state levels of ATA1 and ATA2 mRNAs, but decreased that of ATA3 mRNA.
Since the majority of suicide studies are ex post facto studies of demographic data collected by pathologists anc coroner's investigators, the role of the forensic scientist in determining the accuracy of statistical analyses of death is extremely important. This paper discusses how two salient features of a decedent, surname and residence location, can be misleading in determining the ethnic/racial classification of the decreased. Because many Southern California Indians have Spanish Surnames and most do not reside on an Indian reservation it is shown that the suicide statistics may represent an over-estimation of actual Mexican-American suicidal deaths while simultaneously representing an under-estimation of the suicides among American Indians of the region.
