Explaining the phase behaviour of the pharmaceutically relevant polymers poly(ethylene glycol) and poly(vinyl pyrrolidone) in semi-fluorinated liquids.

Phase behaviour studies of low molecular weight poly(ethylene glycol) (denoted PEG 600 and PEG 1000, corresponding to molecular weights of 600 and 1000 g mol(-1), respectively) have been carried out in 2H,3H-perfluoropentane (HPFP) with and without added poly(vinyl pyrrolidone). The concentration and temperature dependencies of their phase behaviour and the effect of moisture on these systems have been established. Furthermore, the solubility of PEG 600 in binary mixtures of HPFP and perfluoropentane (PFP), as well as HPFP and perfluorodecalin (PFD) have been considered at high HPFP contents. A phase separation phenomenon in fluorinated non-aqueous media is reported for the first time: PEG 600 and PEG 1000 both show a lower critical solution temperature type phase separation boundary. The size of the PEGs was obtained from small-angle neutron scattering (radius of gyration) and pulsed-gradient spin-echo NMR (hydrodynamic radius) measurements. It is shown that polymer conformation follows a regular trend with solution concentration; the size increases from <10 A at 3 wt% in HPFP to 45 +/- 2 A at 20 wt% PEG. On changing the solvent composition by substitution of HPFP by PFP or PFD, the size decreases, consistent with a decrease in the hydrogen-bonding capacity of the solvent mixture. Computer modelling indicates an interaction between the PEG oxygen and the hydrogen of HPFP, an interaction that is absent for the fully fluorinated solvents. This indicates that hydrogen bonding is the driving force for polymer solubility in these solvents.

Predicting solvent accessibility: higher accuracy using Bayesian statistics and optimized residue substitution classes.

We introduce a novel Bayesian probabilistic method for predicting the solvent accessibilities of amino acid residues in globular proteins. Using single sequence data, this method achieves prediction accuracies higher than previously published methods. Substantially improved predictions-comparable to the highest accuracies reported in the literature to date-are obtained by representing alignments of the example proteins and their homologs as strings of residue substitution classes, depending on the side chain types observed at each alignment position. These results demonstrate the applicability of this relatively simple Bayesian approach to structure prediction and illustrate the utility of the classification methodology previously developed to extract information from aligned sets of structurally related proteins.

Solvent effect on the photochemical properties of symmetrically substituted trans-3,3',5,5'-tetramethoxystilbene.

Photochemical properties of trans-3,3',5,5'-tetramethoxystilbene (TMST) have been studied in various polar solvents. The Stokes shift of trans-TMST was found to be increased with increasing solvent polarity. The fluorescence lifetime of trans-TMST experienced a large solvent effect changing from 2.3 ns in cyclohexane to 16.6 ns in acetonitrile. These results indicate that the excited singlet state of trans-TMST has a charge-transfer (CT) character. On the basis of the obtained results, the interior polar environment of a water-soluble TMST dendrimer is discussed.

Absorption spectra of first-row transition metal complexes of bacteriochlorins: a theoretical analysis.

A theoretical study on a family of divalent transition metal bacteriochlorin complexes (M-BC, where M = Mn, Fe, Co, Ni Cu, and Zn) has been carried out to elucidate their potentialities as active molecules in photodynamic therapy (PDT). To draw a complete picture of their electronic properties, both for the ground and excited states, these complexes have been studied by the means of density functional theory (DFT). The time-dependent DFT (TDDFT) approach was used to interpret the electronic spectra, while solvent effects were taken into account by explicitly considering both two water molecules coordinated to the central metal atom and the contribution from the solvent bulk. Particular attention has been devoted to the analysis of the so-called Q bands, since these can be particularly important for medical applications. Metal substitution and environment (solvent) effects have been analyzed, and good agreement is found between computed and available UV-vis spectra. These theoretical data, especially those relative to the metallobacteriochlorins not yet completely characterized at the experimental level, could give some hints for future medical applications.

[Hyperchloremic acidosis druing plasma volume replacement].

OBJECTIVES: Review of the physiological and clinical consequences of hyperchloraemic acidosis observed during plasma volume replacement using crystalloids and colloids. DATA SOURCES: Data were searched in the Medline database after 1990 using the following key words: metabolic acidosis, crystalloids, colloids, albumin, gelatin, hydroxyethyl starch. DATA EXTRACTION: Publications before 1990 were selected for their historical value. Most of articles published after 1990 and all types including case report were accepted. DATA SYNTHESIS: Large volume infusion of isotonic solution can cause hyperchloraemic acidosis. Colloid plasma substitutes using saline solvent may be responsible for the same kind of acidosis with acidaemia. The anion gap is not modified in this case because of chloride increase. Physiological mechanism may be described using the Henderson-Hasselbach equation or the strong ion difference decrease (Stewart concept). Excessive chloride infusion is a major factor in this acid-base disorder and the term hyperchloraemic acidosis should be preferred to dilutional acidosis. When perioperative acidosis occurs, careful and complete analysis of acid-base disturbance should be made. The association of a normal anion gap, normal lactatemia, hyperchloraemia and acidaemia does not need specific treatment. Acidosis corrects spontaneously and slowly following chloride normalization. But any factor that may increase acidosis should be avoided. CONCLUSION: The use of balanced solution like lactated-Ringer solution instead of isotonic saline solution for fluid resuscitation, except for specific contra-indication as intracranial hypertension, may avoid hyperchloraemic acidosis. Potential risk of this acidosis led to the conception of a new colloid using balanced crystalloids solution as the solvent (Hextend).

Photophysical study of a family of [Ru(phen)2(Me n dpq)]2+ complexes in different solvents and DNA: a specific water effect promoted by methyl substitution.

The synthesis and spectroscopic characterisation of a family of ruthenium complexes [Ru(phen)(2)(Me(n)dpq)](2+)(n= 0, 1, 2) are reported and the photophysical effect of the progressive methyl substitution on the dpq ligand ascertained in a range of solvents and in the presence of DNA: [Ru(phen)(2)(dpq)](2+)(1), [Ru(phen)(2)(Medpq)](2+)(2) and [Ru(phen)(2)(Me(2)dpq)](2+)(3)(where dpq is dipyrido[3,2-f:2',3'-h]-quinoxaline; Medpq is 2-methyldipyrido[3,2-f:2',3'-h]-quinoxaline; Me(2)dpq is 2,3-dimethyldipyrido[3,2-f:2',3'-h]-quinoxaline and phen is 1,10-phenanthroline). The increase in electron density following substitution renders the quinoxaline nitrogen atoms more basic in the ground state to yield pK(a) values of -1.9, -2.3, and -2.7 for 3, 2, and 1, respectively. The methyl groups have a pronounced effect on the excited state photophysics of 1-3. In organic solvents, the non-radiative decay constant correlates well with the solvent polarity parameter pi*, with the effect being more pronounced with increasing methyl substitution. On the contrary, in aqueous solution there is a ca. four-fold decrease in the non-radiative decay constant upon methyl substitution. This "specific water effect" may be explained in terms of deactivation of the excited state by hydrogen bonding interactions between water and the quinoxaline nitrogen atoms, which is reduced on moving from 1 to 3. The excited state lifetimes and luminescence intensities for substantially increase when bound to DNA in aerated aqueous solutions, with a six-fold increase, compared to a more moderate three-fold increase for 2 and 3.

Enhancing oxidative resistance of Agrobacterium radiobacter N-carbamoyl D-amino acid amidohydrolase by engineering solvent-accessible methionine residues.

N-Carbamoyl D-amino acid amidohydrolase (D-NCAase) that catalyzes the stereospecific hydrolysis of N-carbamoyl D-amino acids to their corresponding D-amino acids is valuable in pharmaceutical industry. Agrobacterium radiobacter D-NCAase is sensitive to oxidative damage by hydrogen peroxide. To investigate the role of methionine residues in oxidative inactivation, each of the nine methionine residues in A. radiobacter D-NCAase was substituted with leucine, respectively, by site-directed mutagenesis. Except for two mutants (Met5Leu and Met31Leu) with similar activities, seven mutants (Met73Leu, Met167Leu/Met169Leu, Met184Leu, Met220Leu, Met239Leu, Met244Leu, and Met239Leu/Met244Leu) were found to have reduced activities. In the presence of H(2)O(2), three mutants (Met239Leu, Met244Leu, and Met239Leu/Met244Leu) with substitution of highly solvent-accessible methionines by leucines retained their activities. The other mutants were also considerably resistant to chemical oxidation than was the wild-type enzyme. Thus, substitution of solvent-accessible methionine residues with leucine to enhance oxidative stability of D-NCAase is practical but might be with compromised activity.

Tolerance of a protein to multiple polar-to-hydrophobic surface substitutions.

Hydrophobic substitutions at solvent-exposed positions in two alpha-helical regions of the bacteriophage P22 Arc repressor were introduced by combinatorial mutagenesis. In helix A, hydrophobic residues were tolerated individually at each of the five positions examined, but multiple substitutions were poorly tolerated as shown by the finding that mutants with more than two additional hydrophobic residues were biologically inactive. Several inactive helix A variants were purified and found to have reduced thermal stability relative to wild-type Arc, with a rough correlation between the number of polar-to-hydrophobic substitutions and the magnitude of the stability defect. Quite different results were obtained in helix B, where variants with as many as five polar-to-hydrophobic substitutions were found to be biologically active and one variant with three hydrophobic substitutions had a t(m) 6 degrees C higher than wild-type. By contrast, a helix A mutant with three similar polar-to-hydrophobic substitutions was 23 degrees C less stable than wild-type. Also, one set of three polar-to-hydrophobic substitutions in helix B was tolerated when introduced into the wild-type background but not when introduced into an equally active mutant having a nearly identical structure. Context effects occur both when comparing different regions of the same protein and when comparing the same region in two different homologues.

Chemical capacitance of nanoporous-nanocrystalline TiO2 in a room temperature ionic liquid.

The electrochemical behaviour of nanoporous TiO(2) in a room temperature ionic liquid (RTIL), 1-ethyl-3-methylimidazolium bis((trifluoromethyl)sulfonyl)amide (EMITFSI), was investigated by cyclic voltammetry (CV) and impedance spectroscopy. Exponentially rising currents in voltammetry were attributed to the charging/discharging of electrons in the TiO(2) film and a charge transfer mechanism. The main features of the voltammetry and impedance followed the same trends in the ionic liquid as in other organic solvents and also in aqueous electrolytes. In the presence of lithium ions, the onset potential of the charge accumulation increased due to the change of the initial position of the TiO(2) conduction band. The results show that substitution of organic solvents contained in solar cells, supercapacitors or other electrochemical devices is in general feasible, though requires some adjustment in the electrolyte composition for optimal performance.

Solvent effects and mechanism for a nucleophilic aromatic substitution from QM/MM simulations.

The nucleophilic aromatic substitution (SNAr) reaction between azide ion and 4-fluoronitrobenzene has been investigated using QM/MM and DFT/PCM calculations in protic and dipolar aprotic solvents. The effects of solvation on the transition structures, the intermediate Meisenheimer complex, and the rate of reaction are elucidated. The large rate increases in proceeding from protic to dipolar aprotic solvents are only reproduced by the QM/MM methodology.

Isotopic light scattering of lipid vesicles. Water permeation and effect of alpha-tocopherol.

The influence of the index of refraction of the solvent on light scattering properties of lecithin bilayer vesicles is described. Large vesicles (diameter 300 nm) are considered where one lamella separates the intravesicular compartment from the external medium. Stationary and transient cases are distinguished with special emphasis on the isotopic substitution of the solvent, i.e. H2O vs. D2O. Theoretical calculations based on the Mie theory of light-scattering are in accord with results from experiments. The two stationary cases considered serve to calibrate the numerical calculations and illustrate the capability of the method. Transient experiments allow the determination of permeation rates; in particular the D2O/H2O permeability coefficients can be obtained. Single component vesicular lecithin bilayers and ones containing tocopherol are compared. In the crystalline state the incorporation of tocopherol increases the fluidity of the lipid bilayer in parallel with the water permeation rate.

Lipase and esterase-catalyzed acylation of hetero-substituted nitrogen nucleophiles in water and organic solvents.

The lipase- and esterase-catalyzed acylations of hydroxylamine and hydrazine derivatives with octanoic acid and ethyl octanoate are described. The influence of solvent and nucleophile on the initial reaction rate was investigated for a number of free and immobilized enzymes. Initial rates were highest in water, but the overall productivity was optimal in dioxane. Octanoic acid (250 g/L) was converted for 93% into the hydroxamic acid in 36 h with only 1% (w/w) Candida antarctica lipase B (Novozym 435) in dioxane at 40 degrees C. This translates to a catalyst productivity of 68.5 g. g(-1). day(-1) and a space time yield of 149 g. L(-1). day(-1), unprecedented figures in the direct reaction of an acid with a nitrogen nucleophile in an organic solvent.

Ammonium chloride promoted Ugi four-component, five-center reaction of alpha-substituted alpha-isocyano acetic acid: a strong solvent effect.

[reaction: see text] Conditions have been developed for the multicomponent synthesis of di- and tetrapeptide (7) based on the unique reactivity of alpha-isocyano acetic acid (4 and its alpha-substituted derivatives) by an Ugi four-component, five-center reaction. Simply mixing 4, a carbonyl compound (aldehyde or ketone, 8), and a secondary amine (9) (ratio: 1:1:2) in toluene in the presence of 1.5 equiv of ammonium chloride afforded the desired product in good to excellent yield as a mixture of two diastereomers.

Solvent-controlled intramolecular [2 + 2] photocycloadditions of alpha-substituted enones.

The regio- and stereoselectivity of intramolecular [2 + 2] photocycloadditions of 2'-hydroxyenones are shown to be solvent-dependent. In the presence of aprotic solvents, 2'-hydroxyenones undergo photocycloadditions in a manner consistent with the presence of an intramolecular hydrogen bond between the carbonyl group and the tether's hydroxy functionality. In protic solvents, intermolecular interactions appear to disrupt the intramolecular hydrogen bond, providing products with complementary diastereoselectivity. If the facial accessibility of the alpha-tethered olefin is limited, the cycloadditions proceed to give head-to-tail or head-to-head regioisomers, depending on the nature of the solvent employed.

Migration of dehydroabietic and abietic acids from paper and paperboard food packaging into food-simulating solvents and Tenax TA.

An investigation was undertaken to establish the concentration in paper products of dehydroabietic (DHA) and abietic (AA) resin acids, present in rosin, which are major toxicants of pulp- and paper-mill effluent. Their migration was studied from paper and paperboard products into various food-simulating solvents and the substitute fatty food simulant Tenax TA (modified polyphenylene oxide). DHA and AA were detected in five of 10 virgin paper products and in all 10 recycled paperboard products for food-contact use at concentrations of 14-500 and 110-1200 microg/g, respectively. In virgin paper products, the highest migration was into 95% ethanol or heptane, with negligible or no migration into other solvents. In recycled paperboard products, migration was highest into 95% ethanol, but was also observed into 20% ethanol, water and heptane. Migration to Tenax TA was also observed and the migration level increased with time. The maximum migration levels of DHA and AA into food simulants were 0.853 and 3.14 microg/g, respectively. The results suggest that, in the worst case, the daily intake of DHA and AA from paper and paperboard products was 50 times lower than the tolerable daily intake of rosin.

Transition-Metal Derivatives of the Cyclopentadienylphosphine Ligands. 11. Reactivity of the Dinuclear Bridged Rhodium(II) Complexes toward Nitrogen-Containing Ligands.

We report observations on the reactivity of the dinuclear bridged metal-metal-bonded carbonyl [Rh(II)(CO)(&mgr;-CpPPh(2))](2)(2+) (2(2+)) and of the bis(solvate) cations [Rh(II)(solv)(&mgr;-CpPPh(2))(2)](2)(2+) (3(2+)) with nitriles, amines and pyridine and in general with nitrosyl cation and nitrite anion. By reaction of nitriles and pyridine with 2(2+) we obtained monosubstituted [Rh(2)(CO)L(&mgr;-CpPPh(2))(2)](2+) (4(2+)) and disubstituted [Rh(L)(&mgr;-CpPPh(2))](2)(2+) (5(2+)) (L = MeCN, PhCN, pyridine). These complexes (5(2+)) were also obtained directly from 3(2+). In the reactions with 2(2+) the difference in reactivity of the two rhodium(II) sites suggests a specific role of the metal-metal bond. With secondary and primary amines, reductions of 2(2+) to [Rh(I)(CO)(&mgr;-CpPPh(2))](2) (1) were also observed, and the selectivity with respect to substitution or reduction was strongly solvent dependent. Lithium diisopropylamide induces quantitatively the reduction of 2(2+) to 1 and apparently the substitution of the solvent in 3a(2+). Finally, the new compounds [Rh(II)(THF)(&mgr;-CpPh(2))](2)(2+) (3a(2+)), [Rh(II)(&mgr;-H(2)NC(5)H(4)N)(&mgr;-CpPh(2))(2)](2+) (6(2+)) and [Rh(II)(NO(2))(&mgr;-CpPh(2))](2) (7(2+)) were obtained from 2(2+) and PhIO, from 3(2+) and aminopyridine, and from 2(2+) and nitrite anion, respectively. The compounds were characterized by elemental analysis and the usual spectroscopic methods including COSY NMR experiments. Reaction of NOBF(4) with 1 led efficiently to the compound 2(2+) again through a redox process, instead of a substitution.

Solvolysis of para-substituted cumyl chlorides. Brown and Okamoto's electrophilic substituent constants revisited using continuum solvent models.

Brown and Okamoto (J. Am. Chem. Soc. 1958, 80, 4979) derived their electrophilic substitutent constants, sigma(p)+, from the relative rates of solvolysis of ring-substituted cumyl chlorides in an acetone/water solvent mixture. Application of the Hammett equation to the rates for the meta-substituted cumyl chlorides, where there could be no resonance interaction with the developing carbocation, gave a slope, rho(+) = -4.54 ( identical with 6.2 kcal/mol free energy). Rates for the para-substituted chlorides were then used to obtain sigma(p)+ values. We have calculated gas-phase C-Cl heterolytic bond dissociation enthalpy differences, Delta BDE(het) (= BDE(het)(4-YC(6)H(4)CMe(2)Cl) - BDE(het)(C(6)H(5)CMe(2)Cl)), for 16 of the 4-Y substituents employed by Brown and Okamoto. The plot of Delta BDE(het) vs sigma(p)+ gave rho(+) (SD) = 16.3 (2.3) kcal/mol, i.e., a rho(+) value roughly 2.5 times greater than experiment. Inclusion of solvation (water) energies, calculated using three continuum solvent models, reduced rho(+) and SD. The computationally least expensive model used, SM5.42R (Li et al. Theor. Chem. Acc. 1999, 103, 9) gave the best agreement with experiment. This model yielded rho(+) (SD) = 7.7 (0.9) kcal/mol, i.e., a rho(+) value that is only 24% larger than experiment.
