Telomerase activity in soft-tissue and bone sarcomas.

The telomerase enzyme is a reverse transcriptase capable of replacing the telomeric DNA sequences that are lost at each cell division. Telomerase activation permits extended cell proliferation beyond normal senescence checkpoints, and accordingly, telomerase activity has been detected in a wide range of malignant cells and tissues but is absent in terminally differentiated somatic cells. To date, the majority of cancer-related telomerase analyses have been performed on carcinomas that originate from epithelial cells, and few reports have included tumors originating from nonepithelial cells. In this study, we used the PCR-based telomeric repeat amplification protocol (TRAP) to assay telomerase activity in nuclear protein extracts obtained from a range of malignant and benign connective tissue lesions. In total, 62 histologically diagnosed specimens were analyzed including 37 sarcomas, 7 benign mesenchymal tumors, 12 normal tissue samples, and 6 carcinoma metastases obtained from bone. Thirty (81%) of the 37 primary sarcoma samples contained telomerase activity, and four of the six carcinoma metastases were also positive. Conversely, telomerase activity was detectable in only one of seven benign lesions and in none of the 12 normal connective tissue controls. Tumors of connective tissue origin can sometimes be difficult to categorize and to evaluate microscopically with regard to clinical management. As is the case in carcinomas, the presence of telomerase activity appears to be indicative of malignancy in mesenchymal tumor biopsy material and therefore may be useful as an adjunct to the pathologist in the assessment of borderline cases.

[Antitumor action of Ser. marcescens nuclease covalently bound to soluble dextrans].

The antitumor effect of Ser. marcescens nuclease, modified by covalent binding using the method of diazocoupling with m-aminobenzyloxymethyl dextran of molecular weight 20 000, 40 000, 60 000, exceeds 3--4 times the antitumor effect of the native enzyme in relation to Ehrlich ascites carcinoma in contact tumor cells exposure. In intramuscular injection of the enzyme the nuclease preparation modified by dextran of molecular weight 40 000 showed the greatest antitumor activity against Ehrlich carcinoma and sarcoma 37.

Synthetic sulfur-containing amino acids. Inhibition of transport systems in S37 ascites tumor cells.

The preparation of a series of synthetic sulfur-containing amino acids is described. These compounds included heterocyclic analogues of L-cysteine, DL-norcysteine, and DL-homocysteine. The amino acids were assessed for their ability to inhibit the neutral amino acid transport systems of the Sarcoma 37 ascites tumor cell and their inhibitions were compared with those of L-methionine and L-ethionine. Transport studies indicated that the amino acids synthesized were capable of inhibiting the uptake of [3H]-L-histidine in the S37 cell.

[Changes in tumor cell sensitivity to the action of triterpene glycosides in liposomes].

The effect of the increased levels of cholesterol in ascites tumor cells of mouse sarcoma 37 on the cytochemical activity of 4 triterpene glycosides was studied. A definite correlation between the increase in the cholesterol level in the cells and the glycoside activity was noted. The effect character of some triterpene glycosides depended on the pH value of the medium.

N-acetyltransferase activity as a possible criterion in prognosing the anti-tumor effect of cyclophosphamide.

The results obtained in the present study indicated that a reduction in the activity of N-acetyltransferase (EC 2.3.1.5) occurs in cyclophosphamide-treated mice with sarcoma 37. A difference in variations of the level of N-acetyltransferase activity in outbred mice with and without tumors was observed in cyclophosphamide-treated animals. It was established that the effect of cyclophosphamide on the activity of N-acetyltransferase depends on the initial level of the enzyme activity (phenotype of acetylation) as well as on the genotype which determines the N-acetyltransferase activity.

[Potentiating effect of disodium salt of 2,6-dimethyl-1,4-dihydropyridine-3,5-bis-carbonyl hydroxyacetic acid on the activity of various antitumor agents].

The disodium salt of 2,6-dimethyl-1,4-dihydropyridine-3,5-bis-carbonyl hydroxyacetic acid (I) used in the mg/kg dose decreases the cyclophosphane toxicity in mice and potentiates the cytostatic activity of cyclophosphane, 5-fluorouracil and arabinosyl cytosine against leukemia P388, murine sarcoma 37 and Walker's carcinosarcoma. Administered alone I exhibits no antitumour activity. The potentiation of the antitumour effect of drugs appears independent of the administration schedule. Biochemical evidence indicates that I does not block DNA synthesis in leukemic cells in vitro, but significantly enhances the DNA-blocking effect of cyclophosphane in the same cells in vivo.

[Oxygen status and angioarchitectonics of different types of tumors and their changes after radiotherapy].

The study of the oxygen tension and blood supply to the implanted tumors of different histological structures demonstrated a correlation between pO2 and blood supply of sarcoma 37 after a single and fractionated irradiation. As to Lewis carcinoma and B-16 melanoma, the correlation was only found in tumors of a small size: as the tumor grew up to 14-15 mm such a correlation was absent. It was also established that the radiation-induced changes in the vessels and the oxygenation depended upon the histological structure of the tumor.

[Temperature phase transitions in cancer cells].

Light scattering (90 degrees) of Ehrlich ascites tumor and sarcoma 37 cell suspension in the temperature range 25-60 degrees C was studied (heating velocity was 3 grad/min). It is found that the scattering curve has the peak at 46 degrees C and two plots which are typical of phase transitions in the membranes; the first plot in the range 40-46 degrees C is reversible and the second one at 46-51 degrees C is irreversible. It is proposed that 46 degrees is a critical temperature for the membranes structure stability and viability of the studied cells.

[Cytostatic effect of cyclophosphane on esophageal tumors and epithelium of mice].

Investigation of cytostatic activity of cyclophosphamide in sarcoma 37 and esophagus epithelium in albino mice with respect to the diurnal rhythm of mitotic activity and the number of labeled nuclei was performed. Apparently the tumour cells in the G1-phase and at the beginning of the S-phase of the mitotic cycle were the most sensitive to the inhibitory effect of this drug. During the completion of the DNA-synthesis period the cell resistance to the action of the cytostatic increased. Cells at the G1-phase of the mitotic cycle were sensitive to the inhibitory action of cyclophosphane in the esophageal epithelium.

[Experimental combined administration of cyclophosphane with serotonin, insulin and glucose].

Experiments on mice with sarcoma 37 evidenced that the use of cyclophosphan in combination with serotonin, insulin, insulin + glucose and serotonin + glucose enable it to obtain a more significant antineoplasic effect than this is possible by the treatment with cyclophan alone. A combined application of these agents produced a more profound inhibition of the glycogen-forming function and restitution of the glycogen cintent in the liver of the test animals occurred quicker than when cyclophosphan alone was used. In oncological practice it is recommended to use combinations of cyclophosphan with serotonin, insulin, insulin + glucose and serotonin + glucose.

Inhibiting effect of vitamins C and B12 on the mitotic activity of ascites tumors.

The mitotic activity of the transplantable mouse tumors, Sarcoma 37, Krebs-2, and Ehrlich carcinomas, in the ascites form, were inhibited after treatment with a mixture of vitamins C and B12 with no apparent toxic side effects. These vitamins when administered alone, at the same dosage, did not seem to have any apparent effect on mitosis or the morphology of the cells studied. Microscopic examination of the stained ascites fluid taken from the mice treated with the vitamin mixture showed few tumor cells, and these in various stages of disintegration. Also, an increase in lymphocytes, monocytes and neutrophils were noticed; however, later in the experiment, no tumor cells could be found and monocytes and macrophages were abundant.

Fine needle aspiration cytology vs. needle core biopsy of soft tissue tumors. A comparison.

Fine needle aspiration (FNA) cytology is an effective modality in the diagnosis of soft tissue lesions. Previous studies compared FNA with surgical biopsies and reported high sensitivity and specificity rates. No study has evaluated the diagnostic yield of FNA to that of needle core biopsies (NCBs). One hundred seventeen FNAs were performed by our clinical staff for soft tissue lesions (1980-1992). The FNAs were divided into three categories: diagnostic 53 (37 sarcoma, 16 benign); unsatisfactory, 44; normal/inflammatory cells present, 20. Of these, 59 had concomitant NCBs: 37 on patients with sarcoma on FNA, 22 on unsatisfactory FNA and none on those with normal/inflammatory cells (followed clinically). There was 100% correlation between FNA and NCB when sarcoma was diagnosed. In 7 of these cases, the NCB further specified the type of sarcoma. No NCBs were employed to confirm the diagnosis of benign tumor on FNA; excisional biopsies showed a total correlation. The 22 NCBs for unsatisfactory FNA yielded 15 sarcomas, 2 fibromatosis and 5 benign lesions. Seventeen patients with unsatisfactory FNA had surgical biopsies (3 sarcoma, 14 benign), and 5 were lost to follow-up. In our experience, diagnostic FNA gave a yield identical to that of NCB, and the latter did not contribute to patient management. Core biopsies may have the advantage of subtyping selected sarcomas diagnosed by FNA. Unsatisfactory FNA should be evaluated further by a repeat aspirate or NCB. Performance of FNA by cytopathologists can reduce the number of unsatisfactory specimens and allow repeat aspiration. In our study most of the unsatisfactory FNAs were from retroperitoneal and pelvic lesions, were performed under radiographic guidance and could have been minimized by immediate cytologic assessment. Many of these lesions, however, may be as readily accessible via NCB as they are via FNA. Utilization of each of these modalities and of their full benefits ultimately depends on the proper clinical setting and the experience of the clinician and pathologist.

Inhibition of S37 ascites cell amino acid transport systems by alpha-chloromethylketone analogs.

Alanine chloromethylketone and leucine chloromethylketone were synthesized and their effects on amino acid transport in sarcoma 37 mirone ascites tumor (S37) cells were studied. Alanine chloromethylketone preincubation weakly inhibited system A. Leucine chloromethylketone preincubation strongly inhibited both amino acid transport systems L and A. Leucine chloromethylketone was also a competitive inhibitor of leucine transport. Labeled leucine chloromethylketone was concentrated by S37 cells. Leucine chloromethylketone preincubation inhibition was concentration dependent and partial protection of transport was afforded by leucine. Steady-state retention of amino acids was decreased more than the initial velocity of transport by leucine chloromethylketone preincubation. Glutathione was also depleted. Labeled leucine chloromethylketone was incorporated in a plasma membrane protein fraction comigrating on a DEAE-cellulose column (DE52) with gamma-glutamyltranspeptidase activity. There was a modest increase in vital staining after treatment of S37 cells with leucine chloromethylketone, and glucose uptake was also inhibited. Whilst several effects occur during treatment of S37 cells with leucine chloromethylketone, it is suggested than one prominent effect is alkylation of amino acid transport system components.

GLI gene expression in bone and soft tissue sarcomas of adult patients correlates with tumor grade.

The GLI gene encodes a transcription factor harboring five zinc finger motifs that bind to DNA in a sequence-specific manner. The gene was originally identified because of its amplification in a human glioblastoma, and previous studies have shown it to be amplified in a significant proportion of mesenchymal tumors, such as childhood sarcomas. Here we evaluate GLI gene expression in bone and soft tissue sarcomas of adult patients. Samples from 40 patients (37 sarcomas and 3 benign mesenchymal tumors) and samples of 15 normal mesenchymal tissues were examined for GLI gene amplification and expression by Southern hybridization, reverse transcription-PCR of tissue RNA, and immunohistochemistry, using a new polyclonal GLI antibody developed against an epitope outside of the zinc finger region. In contrast to childhood sarcomas, amplification of the GLI gene was not observed in sarcomas of adult patients. Although GLI gene expression in sarcomas was significantly higher than that in normal mesenchymal tissues (P < 0.0001), the levels were very variable. Attempts to correlate the expression data with different pathophysiological parameters only showed a significant relationship to tumor grade. Based on these data, increased levels of GLI gene expression may be indicative of the aggressiveness of the tumor.

Growth factor receptor and related oncogene determination in mesenchymal tumors.

BACKGROUND: Sarcoma are rare malignant neoplasms that originate from a mesenchymal cell line. The epidermal growth factor receptor (EGF-R) has been identified in these malignant neoplasms by immunohistochemical techniques. METHODS: This investigation has evaluated the gene amplification and expression of EGF-R and the homologous oncogene c-erbB-2 in soft tissue and osseous sarcomas by Southern and northern blot analysis. RESULTS: Amplification of EGF-R and c-erbB-2 was identified in 2 of 117 (1.7%) and 6 of 105 (5.7%) of the sarcomas, respectively. Increased expression of EGF-R and c-erbB-2 was identified in 21 of 43 (49%) and 35 of 94 (37%) sarcomas, respectively. CONCLUSIONS: The expression of these two genes in sarcomas appears to occur independently and not be associated with tumor histologic characteristics, grade, size, DNA content, or proliferative activity.

Genetic imbalances in 67 synovial sarcomas evaluated by comparative genomic hybridization.

We used comparative genomic hybridization (CGH) to evaluate DNA sequence copy number changes in 67 synovial sarcomas of both monophasic and biphasic histological subtypes. Changes (mean among aberrant cases: 4.7 aberrations/tumor; range: 1-17), affecting most often entire chromosomes or chromosome arms, were detected in 37 sarcomas (55%). Gains and losses were distributed equally, but different chromosomes were affected with variable frequencies. The most frequent aberrations, each detected in 9-11 of 67 tumors, were gain of 8q and gain at 12q (12q14-15 and 12q23-qter), loss of 13q21-31, and loss of 3p. Other frequent changes (in 7 or 8 cases) included gains at 2p, 1q24-31, and 17q22-qter, and losses at 3cen-q23 and 10q21. High-level amplifications were seen in 7 cases. A total of 16 regions were detected. Two of them, 8p12-qter and 21q21-qter, seen in 4 and 2 tumors, respectively, were recurrent. No aberrations specific to histological subtype were identified. However, genetic changes in the monophasic tumors were more complex and numerous (mean among aberrant cases: 5.3 aberrations/tumor; range: 1-17) than in the biphasic tumors (mean: 2.5 aberrations/tumor; range: 1-5), and high-level amplifications occurred more frequently. All but 1 of the sarcomas showing high-level amplification were of the monophasic subtype. These findings may reflect differences in the pathogenesis and biological behavior of both histological subtypes of synovial sarcoma.
