Pollen tube Growth and Self-Incompatibility in Heuchera micrantha var. diversifolia (Saxifragaceae).

A crossing study and an analysis of pollen tube growth were conducted in diploid and autotetraploid Heuchera micrantha var. diversifolia to distinguish between the possible mechanisms that could promote the high outcrossing rates observed and that could also result in the absence of fruit set following self-fertilization. The crossing study indicated that no fruit set occurred after self-pollination, whereas fruit set occurred in all of the hand-pollinated outcrosses. After 4 d, the self-pollinated flowers shriveled and abscised. Pollen tube growth following hand pollination was assessed in selfed and outcrossed flowers using fluorescence microscopy. The self-pollinated flowers exhibited far fewer pollen tubes than did the outcrossed flowers. Furthermore, in self-pollinated flowers, some of the pollen tubes extended into the style; fewer than one-half of the pollen tubes reached the base of the style and still fewer reached the ovules. The variable length of pollen tube growth, the uniform timing of floral abortion after self-pollination, and the absence of variability among individuals in the level of fruit set following self-pollination are all consistent with a system that lies somewhere between classic gametophytic self-incompatibility and late-acting self-incompatibility as the mechanism that is most likely operating in H. micrantha var. diversifolia. A similar "nonstandard" system may be present in other Saxifragaceae, such as Tolmiea and Lithophragma, as well as in Ribes, the sister group of Saxifragaceae. Our data also indicate that ploidal level (diploid vs. autotetraploid) has no influence on the extent or mechanism of self-incompatibiltiy in autopolyploid H. micrantha var. diversifolia.

Incidence of pain conditions in dental practice in a Danish county.

From September 1981 to February 1982, an epidemiologic study was carried out on the incidence of pain conditions among patients between 18 and 70 yr of age in 30 dental practices in Ribe County, Denmark. A total of 35 464 patients consulted the 50 participating dentists during the study period. 823 patients consulted the dentist due to pain. The most frequent cause of pain was caries sequelae (pulpitis (35%) and acute apical periodontitis (31%)). There was a significant difference in the distribution of the diagnoses between patients with regular and non-regular dental care. Although the data in the present study may not be representative for the entire Danish population, it seems reasonable to assume that in Denmark there are at least 70 000 annual consultations to a dentist due to pain. Furthermore, a minimum of 4000 patients can be expected to be absent from work annually, due to illness caused by dental pain.

Embelin, an inhibitor of X chromosome-linked inhibitor-of-apoptosis protein, blocks nuclear factor-kappaB (NF-kappaB) signaling pathway leading to suppression of NF-kappaB-regulated antiapoptotic and metastatic gene products.

Identifying the active chemical ingredients of ancient medicines and the molecular targets of those ingredients is an attractive therapeutic objective. Embelin, identified primarily from the Embelia ribes plant, is one such compound shown to exhibit chemopreventive, anti-inflammatory, and apoptotic activities through an unknown mechanism. Because nuclear factor-kappaB (NF-kappaB) regulates several genes associated with inflammation, proliferation, carcinogenesis, and apoptosis, we postulated that embelin might mediate its activity through modulation of NF-kappaB activation. We found that embelin inhibited tumor necrosis factor (TNF) alpha-induced NF-kappaB activation. Both inducible and constitutive NF-kappaB activation were abrogated by embelin. In addition, NF-kappaB activated by diverse stimuli such as interleukin-1beta, lipopolysaccharide, phorbol myristate acetate, okadaic acid, hydrogen peroxide, and cigarette smoke condensate also was suppressed. We found that embelin inhibited sequentially the TNFalpha-induced activation of the inhibitory subunit of NF-kappaBalpha (IkappaBalpha) kinase, IkappaBalpha phosphorylation, IkappaBalpha degradation, and p65 phosphorylation and nuclear translocation. Embelin also suppressed NF-kappaB-dependent reporter gene transcription induced by TNFalpha, TNF receptor-1 (TNFR1), TNFR1-associated death domain protein, TNFR-associated factor-2, NF-kappaB-inducing kinase, and IkappaBalpha kinase but not by p65. Furthermore, we found that embelin down-regulated gene products involved in cell survival, proliferation, invasion, and metastasis of the tumor. This down-regulation was associated with enhanced apoptosis by cytokine and chemotherapeutic agents. Together, our results indicate that embelin is a novel NF-kappaB blocker and potential suppressor of tumorigenesis.

LC/PDA/ESI-MS Profiling and Radical Scavenging Activity of Anthocyanins in Various Berries.

Anthocyanin extracts of two blueberries, Vaccinium myrtillus (bilberry) and Vaccinium ashei (rabbiteye blueberry), and of three other berries, Ribes nigrum (black currant), Aronia melanocarpa (chokeberry), and Sambucus nigra (elderberry), were analyzed by high-performance liquid chromatography coupled with photodiode array detection and electrospray ionization - mass spectrometry (LC/PDA/ESI-MS). Both bilberry and rabbiteye blueberry contained 15 identical anthocyanins with different distribution patterns. Black currant, chokeberry, and elderberry contained 6, 4, and 4 kinds of anthocyanins, respectively. The radical scavenging activities of these berry extracts were analyzed by using 2,2-diphenyl-1-picrylhydrazyl (DPPH). All these extracts showed potent antiradical activities.

[3H]-thymidine uptake and lipid peroxidation by tumor cells on embelin treatment: an in vitro study.

Using a rapid technique to assess drug-induced cell toxicity, a fibrosarcoma cell line was exposed in vitro to increasing concentrations of embelin, a benzoquinone derivative of Embelia ribes Burm., and simultaneously inoculated with [3H]-thymidine. After regular time intervals, the cells were examined for incorporation of the labelled thymidine in DNA, lipid peroxide and glutathione levels. A dose-dependent decrease in labelled thymidine uptake, lipid peroxide and glutathione levels was observed on embelin administration.

Management of occupational risk to reproduction in a Danish county.

In Denmark pregnant women are entitled to receive maintenance allowances if the working conditions are suspected to be harmful to the fetus. The aim of the study was to evaluate these preventive measures taken to reduce occupational hazards to reproduction in Denmark. In 1985 1,247 pregnancies were identified in Ribe county during a 6-month period. Three hundred and forty-five women employed at the beginning of the pregnancy in manufacturing industries, construction work, health services, or agriculture were interviewed by telephone concerning their working conditions and the measures taken to reduce possible occupational risks. The response rate was 85%. On the basis of the interviews the risks and the preventive measures were assessed. Out of 200 women who had no changes in the working conditions during pregnancy, 46 (23%) had a working situation which was considered potentially harmful. For 57 the working situation was changed during the first months of pregnancy because of an assumed risk. The remaining 37 were out of work or received maintenance allowances because of occupational hazards. In this study a greater awareness than expected about possible occupational risks was found, but in some instances occupational risks were overlooked or neglected.

Screening of Mongolian plants for influence on amylase activity in mouse plasma and gastrointestinal tube.

Mongolian plants were screened for their influence on alpha-amylase activity in mouse plasma. Methanolic extracts of Geranium pratense, Rhodiola rosea, Ribes pullchelum and Vaccinium uliginosum inhibited the enzyme activity in isolated mouse plasma by greater than 40% and the effect was concentration dependent. Vaccinium uliginosum also showed a depressive effect on elevation of postprandial blood glucose to some extent.

Isolation of high molecular weight DNA suitable for BAC library construction from woody perennial soft-fruit species.

We have developed a novel nuclei extraction method that allows for the extraction of high molecular weight DNA from leaves of woody perennial soft-fruit species that contain high levels of carbohydrates and polyphenolics. The method utilizes a modified buffer system including 4% (w/v) polyvinylpyrrolidone (PVP)-10 and a combination of nylon filters and Percoll gradients to purify nuclei extracts prior to embedding in agarose plugs. The effectiveness of the method was demonstrated on leaves of red raspberry (Rubus idaeus) and blackcurrant (Ribes nigrum), two soft-fruit species that have shown to be recalcitrant to standard genomic DNA extraction methods. Extracted DNA was readily digested by restriction enzymes and, as shown for raspberry, suitable for bacterial artificial chromosome (BAC) library construction.

A study of some important vitamins and antioxidants in a blackcurrant jam with low sugar content and without additives.

Blackcurrant (Ribes nigrum) jam was manufactured with the aim of producing a jam with a low sugar content, and without any additives. Four temperatures were investigated, namely 60 degrees C, 76 degrees C, 92 degrees C and 97 degrees C. Processing time varied between 1-20 min. After processing, the highest content of ascorbic acid was found in the jam processed at 97 degrees C for 1 min, which contained 63.3 +/- 2.6 mg ascorbic acid/100 g jam. At all combinations investigated more than 60% of the original amount of ascorbic acid was retained after manufacturing and packaging. The jam made at 92 degrees C was stored in a shelf-life study for 13 months. The jam was then stored at 8 degrees C, ambient temperature and at 37 degrees C. At ambient temperature the jam was stored both in dark and in daylight, at 8 degrees C and at 37 degrees C the jam was stored in dark. After 13 months of storage, at 8 degrees C, 60% of the amount of ascorbic acid and 29% of the amount of anthocyanins were retained. In the jam stored at higher temperatures less of both was retained. The beta-carotene in the jam was found to be stable throughout the whole shelf-life study. Exposure to light did not have any effect on any of the components studied. The degradation of anthocyanins was best described by a second-order reaction and the activation energy was determined to be 90 kJ/mol. A jam of blackcurrant may be considered as a good source of vitamins and antioxidants after one year, if certain precautions concerning manufacture and storage conditions are taken.

[Baker's cysts in children. A retrospective study at the hospitals in the county of Rebe from 1992 to 1998].

Popliteal cysts or Baker's cysts in childhood is a rare disease usually found by the parents. The purpose of this study is to study the recurrence rate after primary surgical resection and conservative treatment of the cysts. We retrospectively found 20 children with Baker's cysts treated at Ribe Amts Hospitals during the period 1992-1998. Thirteen patients were treated conservatively and seven had a surgical resection of the cysts performed. Of the 13 conservatively treated patients the cysts was still present in six patients (46%). In seven patients the cysts had disappeared within nine months. In the surgically treated group the cysts recurred in three patients (43%). Surgical intervention for Baker's cysts in children should only be performed after thorough consideration, due to a high recurrence rate.

Molluscicidal activity of Lawsonia inermis and its binary and tertiary combinations with other plant derived molluscicides.

Molluscicidal activity of leaf, bark and seed of Lawsonia inermis against Lymnaea acuminata and Indoplanorbis exustus was studied. Highest toxicity was observed in the seed of Lawsonia inermis. Toxicity of binary (1:1) and tertiary (1:1:1) combinations of the essential oil of cedar (Cedrus deodara Roxh) and neem (Azadirachta indica A. Juss), powder from bulb of garlic (Allium sativum Linn), and oleoresin extracted from rhizome of ginger (Zingiber officinale Rosc) with Lawsonia inermis and Embelia ribes fruit powder were studied against L. acuminata and I. exustus. L. inermis seed powder in combination with Cedrus deodara oil and Azadirachta indica oil was more toxic than their individual components and other combinations.

Relevance of radiation-induced bystander effects for environmental risk assessment.

A novel mechanism involving a medium borne signalling factor has been identified following irradiation of populations of cells to doses ranging from 5 mGy-5 Gy gamma-rays or to as little as 1 alpha particle traversal in a culture containing hundreds of cells. The factor can be released into culture medium and can induce responses in unexposed cultures. It has been called a "radiation-induced bystander factor". The effect is obviously relevant to risk assessment as it happens at very low doses. It could also offer new avenues for development of drugs aimed not at cell destruction but at restoring the tissues own control and coordination of response following DNA damage. The effect is clearly induced by radiation and probably by other substances. While these effects are now accepted to happen both in vitro and in vivo, their relevance and function is unknown. The investigation and modelling of the mechanism and the variation in level and type of effect in relation to genetic background and clinical history are key questions which need to be addressed in the field. The key driving hypothesis of the work being done by our laboratory is that radiation-induced bystander effects (RIBE) reflect emergent control in complex tissues and communicating cell systems, which can be harnessed for therapeutic gain.

Antioxidant status, lipoprotein profile and liver lipids in rats fed on high-cholesterol diet containing currant oil rich in n-3 and n-6 polyunsaturated fatty acids.

Plant-based n-3 polyunsaturated fatty acids (PUFA) possess a prospective antiatherogenic potential. Currant oil from Ribes nigrum L. is one of the few plant oils containing PUFAn-3 (15.3 mol%) in addition to PUFAn-6 (60.5 mol%). This study was aimed at comparing the effects of currant oil with those of lard fat, rich in saturated (43.8 mol%) and monounsaturated (47.0 mol%) fatty acids, on antioxidant parameters, the lipoprotein profile and liver lipids in rats fed on 1 % (w/w) cholesterol diets containing either 10 % of currant oil (COD) or lard fat (LFD). After 3 weeks of feeding, the COD induced a significant decrease in blood glutathione (GSH) and an increase in Cu(2+) induced oxidizability of serum lipids, but did not affect liver GSH and t-butyl hydroperoxide-induced lipoperoxidation of liver microsomes. Although the COD did not cause accumulation of liver triacylglycerols as LFD, the lipoprotein profile (VLDL, LDL, HDL) was not significantly improved after COD. The consumption of PUFAn-3 was reflected in LDL as an increase in eicosapentaenoic and docosahexaenoic acid. These results suggest that currant oil affects positively the lipid metabolism in the liver, above all it does not cause the development of a fatty liver. However, adverse effects of currant oil on the antioxidant status in the blood still remain of concern.

Coregulation of lux genes and riboflavin genes in bioluminescent bacteria of Photobacterium phosphoreum.

Investigation of the expression of the riboflavin (rib) genes, which are found immediately downstream of luxG in the lux operon in Photobacterium phosphoreum, provides more information relevant to the evolution of bioluminescence, as well as to the regulation of supply of flavin substrate for bacterial bioluminescence reactions. In order to answer the question of whether or not the transcriptions of lux and rib genes are integrated, a transcriptional termination assay was performed with P. phosphoreum DNA, containing the possible stem-loop structures, located in the intergenic region of luxF and luxE (OmegaA), of luxG and ribE (OmegaB), and downstream of ribA (OmegaC). The expression of the CAT (Chloramphenicol Acetyl Transferase) reporter gene was remarkably decreased upon the insertion of the stem-loop structure (OmegaC) into the strong lux promoter and the reporter gene. However, the insertion of the structure (OmegaB) into the intergenic region of the lux and the rib genes caused no significant change in expression from the CAT gene. In addition, the single stranded DNA in the same region was protected by the P. phosphoreum mRNA from the S1 nuclease protection assay. These results suggest that lux genes and rib genes are part of the same operon in P. phosphoreum.

Evaluation of a Danish teaching program in breast self-examination.

This study investigated the effect of a BSE training program on women's knowledge, attitudes, and behavior regarding BSE. Postal questionnaires were sent to 629 women who had participated in 1998-2000 in the BSE training program run by Ribe County, Denmark, and to a local matched control group of the same size selected through personal registration numbers. Response rates were 77% and 56%, respectively. A significantly greater number of women who had attended BSE training reported that they knew how and when to do BSE, and what they should do if they discovered breast changes (97% compared to 66% in control group). Similar proportions in each group felt confident of finding any breast changes (57%) and believed that routine BSE can influence the chances of recovery from breast cancer (90%). There were also no significant differences between the intervention and control groups in the reporting of anxiety as a result of performing BSE (24% and 17%, respectively). The intervention group was significantly more likely to perform BSE regularly (66% compared to 52% in control group) and to use a more correct technique (44% compared to 20% in control group). It was concluded that a formal training program increases the likelihood of regular BSE performance with a correct technique.

[The significance of organized screening for uterine cervix cancer in Denmark during 1968-1987].

The policy concerning organization of screening for cervical cancer has varied in the Danish counties. Organized programmes started in the municipality of Frederiksberg in 1962, in the municipality of Copenhagen and the county of Maribo in 1967, and in the country of Copenhagen in 1968. On the other hand organized screening programmes have never existed in the counties of Vestsjaelland, Viborg, Ringkøbing and Ribe. We have used this "natural experiment" to study the influence of organized screening on the incidence and mortality of cervical cancer in Denmark. Women aged 30-59 years during the period 1968-87 are included in the study. The effect of organized screening has been estimated employing multiplicative Poisson models, where age, period, region and organized screening have been considered. Women living in counties where organized screening started three or more years ago have a statistically significantly reduced risk of cervical cancer. The relative risk for the incidence in this group is 0.77 (95% confidence interval 0.69-0.86), and the relative risk for the mortality is 0.75 (95% confidence interval 0.61-0.92). Only a part of the decrease in the incidence and mortality of cervical cancer which can be attributed to the organized screening activity. The additional decrease can be attributed to the considerable unorganized screening activity during the past 20 years, and to changes in exposure to risk factors.
