Effect of beta-agonists on production of cytokines by activated T cells obtained from asthmatic patients and normal subjects.

Intracellular levels of cAMP were found to regulate T cell activity. We examined whether beta2-agonists altered cytokine production and cyclic adenosine monophosphate (cAMP) accumulation in concanavalin A (ConA)-activated peripheral T cells from asthmatic patients. Procaterol and isoproterenol weakly decreased the ConA-elicited interleukin (IL)-4 and IL-5 secretion; however, the inhibitory effect of procaterol on the ConA-induced IL-2 secretion was inferior to that of isoproterenol in normal controls and was little in asthmatics. The intracellular accumulation of cAMP by procaterol was not altered compared with that by isoproterenol. Results suggest that there is a qualitative difference between procaterol- and isoproterenol-induced cAMP accumulation in T cells.

Differential coupling to positive inotropic responses of cyclic AMP produced by stimulation of beta 1- and beta 2-adrenergic receptors.

In canine right ventricular muscle, isoproterenol, a nonselective beta-agonist, and T-1583, a selective beta 1-agonist, produced positive inotropic effects (PIE) as full agonists. Their PIEs were antagonized by atenolol, a selective beta 1-antagonist, (pA2 = 7.71 for isoproterenol and pA2 = 7.82 for T-1583). The PIE of T-1583 was also antagonized by practolol, a selective beta 1-antagonist (pA2 = 6.55). Procaterol known as a selective beta 2-agonist gave a biphasic concentration-response curve with a maximum that was approximately half those of isoproterenol and T-1583. The PIE of procaterol at low concentrations (pD2 = 7.93), which maximum was approximately 10% of the maximum of isoproterenol, was antagonized by ICI 118,551, a selective beta 2-antagonist (ICI; pA2 = 8.84) and that at high concentrations (pD2 = 5.12) by atenolol (pA2 = 7.56). Increases in cyclic AMP content produced by isoproterenol were reduced greatly by atenolol and slightly by ICI, whereas those produced by T-1583, which were smaller than those by isoproterenol, were abolished by atenolol but not changed by ICI. Procaterol (10(-7) M) produced an increase in cyclic AMP, which was abolished by ICI but not changed by atenolol, to almost the same extent as procaterol (10(-5) M) did, which was abolished by atenolol and slightly reduced by ICI. These results indicate that increased cyclic AMP through beta 1- but not beta 2-receptors couples well to PIE.

Further evidence for a homogeneous population of beta-1-adrenoceptors in bovine coronary artery.

Isolated rings from the proximal and distal ends of the bovine epicardial anterior descending coronary artery and guinea pig tracheal rings were mounted in tissue baths for the measurement of isometric contraction and relaxation, and spontaneously beating rabbit atria were prepared for measurement of chronotropic response. All tissues were exposed to phenoxybenzamine to block tissue uptake of catecholamines and alpha adrenoceptors. The arterial and tracheal rings were contracted with 25 mM K+ in order to observe agonist-induced relaxation. Beta adrenergic agonist dose-response curves were obtained in all tissues in the presence and absence of the beta-2 selective antagonist, ICI 118,551. The orders of agonist potencies in the proximal and distal coronary arteries and the rabbit atria were the same: namely, isoproterenol (ISO) greater than norepinephrine (NE) much greater than procaterol. In contrast, that in the guinea pig trachea was procaterol greater than ISO greater than NE. Procaterol was a weak, partial agonist in the bovine coronary artery and rabbit atria and a potent, full agonist in the guinea pig trachea. The ICI 118,551 pA2 values in the proximal segment of the bovine coronary artery were the same against both NE and fenoterol, 7.38. In the distal segment, the values were 7.33 and 7.14, respectively. ICI 118,551 -log KB values in the rabbit atria, 6.81 and 6.83 for NE and ISO, respectively, were slightly below those in the coronary artery segments, whereas -log KB values in the guinea pig trachea were substantially higher, 8.19 and 8.81 for fenoterol and procaterol, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Beta-2 adrenoceptor-mediated relaxation of the isolated human saphenous vein.

On isolated strips of human saphenous vein, pretreated with 5 microM phenoxybenzamine and contracted with 10 mM KCl, the beta adrenoceptor mediating the relaxant effects of isoproterenol, procaterol and norepinephrine was characterized using the selective beta-1 adrenoceptor antagonist, bisoprolol, and the selective beta-2 adrenoceptor antagonist, ICI 118,551. All three agonists produced concentration-dependent relaxations of the isolated saphenous vein with an order of potency: procaterol (pD2 value, 7.69) greater than isoproterenol (pD2 value, 7.41) much greater than norepinephrine (pD2 value, 5.30). ICI 118,551 (3 X 10(-10) to 3 X 10(-9) M) was nearly 100 times more potent than bisoprolol (10(-7) to 10(-6) M) in antagonizing the relaxant effects of isoproterenol and procaterol. The slopes of the Schild plots for the antagonistic effects of ICI 118,551 and bisoprolol against isoproterenol- and procaterol-induced relaxations were not significantly different from unity indicating interaction with a homogeneous population of beta adrenoceptors. The pA2 value for ICI 118,551 amounted to 9.11 to 9.20 and for bisoprolol to 6.50 to 6.63. In addition, the concentration-response curve for the relaxant effect of norepinephrine was significantly shifted to the right by 10(-9) M ICI 118,551, but not affected by 10(-7) M bisoprolol. These results indicate that on the isolated strips of the human saphenous vein the beta adrenoceptor mediating relaxation is of the beta-2 subtype.

Comparison of the attenuating effects of four beta-adrenoceptor agonists on rat isolated uterus and aorta.

1. The ability of four beta-adrenoceptor agonists to attenuate oxytocin (0.2, 2 and 20 nmol/L) or KCl (20, 40 and 80 mmol/L)-induced contractions of the uterus (n = 5-8 for each agonist) and the KCl (18 mmol/L)-induced contractions of the aorta (n = 9 for each agonist) from rats, pretreated with oestradiol has been compared. 2. Isoprenaline, salbutamol, terbutaline and procaterol (0.1-10 mumol/L) attenuated the contractions of the uterus and the aorta. All four agonists had similar attenuating potencies on the uterus. 3. Procaterol caused the same maximal attenuation (33%) on the aorta as the other beta-adrenoceptor agonists and is thus acting as a full beta 2-adrenoceptor agonist under these experimental conditions. Isoprenaline and procaterol were much more potent than salbutamol and terbutaline in attenuating the aorta responses. 4. This study showed that isoprenaline and procaterol were potent attenuants on both the uterus and aorta whereas salbutamol and terbutaline were potent uterine but only modest aorta attenuants. This preliminary study indicates that the responsiveness of uterine and vascular tissue to certain beta 2-adrenoceptors differs.

[Modulation of cytokine production from human mononuclear cells by several agents].

We investigated the effects of drugs, especially anti-pulmonary disease agents, on the production of cytokines from human peripheral blood mononuclear cells (PBMC). Roxithromycin (RXM), a macrolide antibiotic with the structure of 14-member macrocycline ring increased adherent cells (monocyte/macrophages), whereas it suppressed the proliferation of PBMC stimulated with phytohemagglutinin (PHA). RXM suppressed the production of IL-1 beta and TNF-alpha from lipopolysaccharide (LPS)-stimulated PBMC in a dose-dependent manner. Levofloxacin, a fluorinated quinolone, increased IL-2 production by PBMC stimulated with PHA. The production of GM-CSF and soluble IL-2 receptor was suppressed at high concentrations of LVFX. LVFX suppressed IL-1 beta production, but did not the production of TNF-alpha and IL-8 production. A beta-adrenoceptor agonists (beta-agonist), procaterol, clenbuterol, fenoterol and terbutaline suppressed the production of TNF- and IL-1 beta. TNF-alpha production was almost completely suppressed by dibutyryl cyclic AMP (dbcAMP), whereas IL-1 beta production appeared to be partially refractory even at the highest concentration examined. Both procaterol and theophylline elevated cAMP levels in LPS-stimulated PBMC, but the effect of procaterol was limited. The inhibition of the production of TNF-alpha and IL-1 beta by procaterol was additively potentiated with theophylline. Of examined phosphodiesterase (PDE) isozyme inhibitors type IV PDE inhibitors were more effective in inhibiting the production of TNF-alpha and IL-1 beta by LPS-stimulated PBMC than a nonselective, type III or type III/IV inhibitor. The addition of the beta-agonist increased the inhibitory effect of tested PDE inhibitors on the production of TNF-alpha and IL-1 beta Type IV, type III and nonselective PDE inhibitors were effective in inhibiting the production of IFN-gamma and IL-2 in a dose-dependent manner. In contrast, the production of IL-4 and IL-5 was inhibited by only the highest concentration of type IV inhibitor, and other agents had no effect on the production. Similarly, dbcAMP inhibited the production of IFN-gamma and IL-2 more potently than that of IL-4 and IL-5. The addition of the beta-agonist increased the inhibitory effect of tested PDE inhibitors on the production of IFN-gamma and IL-2 production. These findings indicate that these agents have an immunodulatory action on the production of cytokines by PBMC and also indicate that they could be potent pharmacological agents for the treatment of diseases in which several cytokines are important etiological factors.

[A second drug in nonatopic asthma insufficiently controlled with beta 2-adrenergic stimulants: budesonide versus theophylline].

We assessed whether theophylline or budesonide (an inhaled corticosteroid) is the most adequate drug to be added to the treatment of patients insufficiently controlled with beta agonists (procaterol). Two comparable groups of nonatopic asthmatic adults (n = 24), aged between 16 and 66 years (42 +/- 17), were investigated in a crossed, randomized study. After 4 weeks with procaterol + 800 micrograms of budesonide, the forced expiratory flow improved (p less than 0.01), the degree of bronchial hyperreactivity to histamine was reduced (p less than 0.001), the daily recorded symptomatology almost disappeared in all and the number of procaterol inhalations was reduced (p less than 0.05). The combination of theophylline + procaterol during 4 weeks did not improve these parameters from their baseline values. It was concluded that inhaled steroids are the drugs of choice in patients with asthma which is not controlled only with beta agonists.

Effects of ovarian hormones on beta-adrenergic receptor-mediated relaxation in the female rabbit bladder.

The effects of ovarian hormones on beta-adrenergic receptor-mediated responses in female rabbit detrusor smooth muscles were investigated. Ovariectomized mature female New Zealand white rabbits were untreated or treated with estrogen and/or progesterone for 2 weeks. The contractile responses to carbachol and KCl in the detrusor strips were not significantly different in all groups. As compared with dobutamine and GS-332, isoproterenol and procaterol significantly relaxed the detrusor strips derived from all groups on KCl-induced tonic contractions. Combined with estrogen treatment, isoproterenol, procaterol and GS-332 caused a significant increase in this muscle relaxation. Furthermore, estrogen treatment caused a significant increase in relaxation as a result of forskolin and the cyclic AMP (cAMP) production that was induced by isoproterenol, procaterol and GS-332. However, estrogen treatment did not affect the relaxant response to dibutyryl cyclic AMP. Progesterone treatment did not affect beta-adrenergic receptor-mediated responses. These results suggest that estrogen treatment causes the increased relaxant responses mediated by beta2- and beta3-adrenergic receptor subtypes, which may be related to the increased cAMP content in female rabbit detrusor smooth muscles.

Beta 2-adrenoceptor-mediated increase in the slow inward calcium current in atrial cells.

The effects of procaterol (beta 2-adrenoceptor agonist) on the slow inward calcium current (Isi) were examined in single cells of guinea-pig heart. Procaterol increased Isi in atrial cells, in the presence of atenolol (beta 1-adrenoceptor antagonist). The effect was abolished by ICI-118551 (beta 2-adrenoceptor antagonist). However, procaterol did not modify the membrane currents in ventricular cells. These results suggest that beta 2-adrenoceptor agonists bind to beta 2-adrenoceptors in atrial cells and augment Isi, but beta 2-adrenoceptors are not present in ventricular cells.

Effects of beta 2-agonists on the contractility of fatigued canine diaphragm in vivo.

We examined the effects of four beta 2-agonists, clenbuterol, metaproterenol, procaterol, and albuterol, on the contractility of fatigued canine diaphragm. Diaphragmatic contractility was assessed from changes in transdiaphragmatic pressure (Pdi) generated by supramaximal electrical stimulation of the phrenic nerve. Diaphragmatic fatigue was developed by applying an inspiratory resistive load to spontaneously breathing dogs for 30-50 min. Four doses of each beta 2-agonist were intravenously administered to dogs when the decreased Pdi became stable. Clenbuterol increased Pdi at doses of 10 and 20 micrograms/kg (P < 0.05). Metaproterenol improved Pdi of the fatigued diaphragm at low stimulation frequencies in a dose-dependent manner (P < 0.05). Procaterol increased Pdi at a low dose but decreased Pdi at higher doses, possibly due to a marked decrease in systemic blood pressure. Albuterol did not change Pdi at any dose. These results suggest that both clenbuterol and metaproterenol have a positive inotropic effect on fatigued canine diaphragm, while procaterol has a negative effect at high doses. Albuterol showed no inotropic effect.

Zatebradine inhibits tachycardia induced by bronchodilators without affecting respiratory resistance in dogs.

Bronchodilators used for bronchial asthma reduce respiratory resistance but also increase heart rate to some extent. It is often difficult to use such bronchodilators with elderly patients and patients with heart disease. The object of our study was to investigate whether a specific bradycardic agent, zatebradine, inhibited the heart rate increased by bronchodilators without affecting respiratory resistance. We evaluated the effects of zatebradine on the increases in heart rate and inhibition of the respiratory resistance in response to the bronchodilators, isoproterenol, procaterol (a beta 2-adrenoceptor agonist), 6-(3-dimethylaminopropionyl)-forskolin, NKH 477 (an adenylyl cyclase activator) and aminophylline in the anesthetized and artificially ventilated dog. When zatebradine in doses of 0.05-1.5 mg/kg i.v. decreased heart rate without affecting arterial blood pressure, it dose dependently attenuated the increase in heart rate in response to isoproterenol, procaterol, NKH 477 and aminophylline but did not affect the inhibition by these substances of the increase in respiratory resistance induced by histamine. Propranolol (0.01-0.3 mg/kg i.v.) dose dependently inhibited not only the increase in heart rate but also the inhibition of the respiratory resistance induced by isoproterenol and procaterol. The present results indicate that zatebradine selectively inhibits the increase in heart rate in response to cyclic AMP-dependent bronchodilators without affecting their bronchodilator effects in anesthetized dogs and suggest that zatebradine may be a useful drug for prevention of the tachycardia induced by bronchodilators used for patients with bronchial asthma.

Differential effects of opioid and adrenergic agonists on proliferation in a cultured cell line.

A novel clonal cell line transfected with the delta-opioid receptor (delta-OR) encoding gene was used to study agonist-activated regulation of cell proliferation. In this cell line, endogenous beta2-adrenergic receptors (beta2-ARs) are coexpressed with the exogenous delta-ORs. Upon individual acute treatments with morphine and procaterol (a selective beta2-AR agonist), both the delta-OR and beta2-AR are coupled to differential modulation of cyclic AMP (cAMP) levels in accord with the classical second messenger response patterns to these agonists in the normal cellular settings of the receptors. But chronic morphine activation of the delta-OR inhibits cellular proliferation, while chronic procaterol activation of the beta2-AR stimulates it. Chronic treatment with the individual agonists is accompanied by differential activation of the mitogen-activated protein kinase (MAPK) isozymes, extracellular-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK). The findings suggest that chronic beta2-AR activation stimulates proliferation by interacting with the ERK signalling cascade independent of a cAMP-mediated pathway. In contrast to treatment with individual agonists, chronic dual agonist treatment suppresses procaterol-induced stimulation of ERK activity and stimulation of proliferation indicating that a cross-regulatory interaction occurs between the delta-OR and beta2-AR signalling systems in the cells under these conditions.

Subcellular fractions of cyclic AMP and cyclic AMP-dependent protein kinase and the positive inotropic effects of selective beta 1- and beta 2-adrenoceptor agonists in guinea pig hearts.

We studied the effects of a full beta 1-adrenoceptor agonist (T-0509), a beta 2-adrenoceptor agonist (procaterol) and a nonselective beta-adrenoceptor agonist (isoproterenol, ISO) on subcellular cyclic AMP levels and cyclic AMP-dependent protein kinase (cyclic AMP-PK) activity in guinea pig hearts and compared them with the effect of each drug on cardiac contractility. T-0509 (10(-8)M) and ISO (3 x 10(-8)M) caused an increase of approximately 170% in dF/dtmax, whereas 10(-7) procaterol produced only a 25% increase. All these agonists significantly increased the cyclic AMP level in ventricular homogenate. Subcellular fractions were obtained by centrifugation at 100,000 g for 10 min and by Li2SO4 precipitation of the 100,000-g supernatant. In the control heart, probably salcolemmal protein, phospholamban, and 60-kDa protein in the particulate fraction and probably troponin I and troponin C in the supernatant fraction were mainly phosphorylated by the catalytic subunit of cyclic AMP-PK. In the precipitate obtained from the supernatant fraction with Li2SO4, probably all proteins described were contained. However, none of the proteins were detected in the supernatant obtained with Li2SO4. T-0509 and ISO caused significant changes in cyclic AMP levels and cyclic-PK activities in all fractions. However, procaterol increased the cyclic AMP concentrations and cyclic AMP-PK activities only in the supernatant fraction and the supernatant obtained with Li2SO4. T-0509 and ISO increased cyclic AMP level (9-16 pmol/mg protein) and cyclic AMP-PK activity ratio (0.27-0.33) significantly to the same degree in the precipitate obtained with Li2SO4, whereas the effects of T-0509 in other fractions were about twofold less than those of ISO. These results suggest that beta 1- and beta 2-adrenoceptor agonists cause differential compartmentalization of cyclic AMP and cyclic AMP-PK in the cardiac muscle.

Facilitation by beta-adrenoreceptors of stimulation-induced vasoconstriction in pithed spontaneously hypertensive rats.

In pithed bilateral adrenal demedullated spontaneously hypertensive rats, slow i.v. infusion of adrenaline (500 ng/min) significantly increased pressor responses to the electrical stimulation of the entire sympathetic outflow, but had little effect on pressor responses induced by bolus injections of noradrenaline. After pretreatment with timolol (1 mg/kg, i.v., 30 min) adrenaline infusion enhanced noradrenaline-induced pressor effects, but not stimulation-induced responses. Salbutamol (50-500 ng/min, i.v.) and procaterol (2.5 ng/min, i.v.) infusions significantly potentiated stimulation-induced pressor effects without affecting noradrenaline-induced responses. Higher rates of salbutamol (5 micrograms/min, i.v.) and procaterol (25 ng/min, i.v.) infusion significantly depressed noradrenaline-induced pressor effects and attenuated those induced by sympathetic nerve stimulation. It is concluded that the potentiation of stimulation-induced pressor responses by adrenaline salbutamol and procaterol, involves a beta-adrenoreceptor mediated facilitation of sympathetic neurotransmission.

Is there any evidence of beta 1-adrenoceptors mediating relaxation of guinea-pig lung parenchyma?

We tried to find functional evidence for the existence of beta 1- and beta 2-adrenoceptors in the isolated guinea-pig lung parenchymal strip preparation, using potent and selective beta 1- and beta 2-adrenoceptor stimulation. To obtain potent beta 1-adrenoceptor stimulation the nonselective beta-adrenoceptor agonist isoprenaline was combined with a highly selective beta 2-adrenoceptor antagonist--ICI 118,551. Potent beta 2-adrenoceptor stimulation was obtained by procaterol. Practolol (beta 1-adrenoceptor antagonist) and ICI 118,551 were used as antagonists. ICI 118,551, 10(-7) mol/l, shifted the concentration response (C/R) curve of isoprenaline to a higher concentration range. The C/R curve of procaterol was shifted in the same way and to the same degree by this concentration of ICI 118,551. The C/R curve of isoprenaline was not further shifted after blockade with a combination of ICI 118,551, 10(-7) mol/l, and practolol, 10(-6). However, in the trachea preparation, a tissue containing both beta 1- and beta 2-adrenoceptors, there was a further shift of the C/R curve of isoprenaline to a higher concentration range after blockade with a combination of ICI 118,551 and practolol in the concentrations given above. In this preparation the shift of the C/R curve of procaterol was ten times greater than that of isoprenaline after blockade with ICI 118,551, 10(-7) mol/l. We conclude that it is possible to characterize small fractions of beta 1-adrenoceptors coexisting with beta 2-adrenoceptors with the technique used. Furthermore there is still no functional evidence of the existence of beta 1-adrenoceptors in the lung parenchyma.

Relaxant and beta 2-adrenoceptor blocking activities of (+/- )-, (+)- and (-)-pindolol on the rat isolated aorta.

The KCl contracted rat aorta is relaxed by procaterol, (+/- )-, (+)- and (-)-pindolol. The relaxations to procaterol, but not to (+/- )-pindolol, were prevented by ICI 118,551 at 10(-6) M. The relaxations to (+/- )-pindolol are, therefore, not due to beta-adrenoceptor agonism. At 10(-7) M ICI 118,551, (+/- )-, (+)- and (-)-pindolol were beta 2-adrenoceptor antagonists as they inhibited the relaxant responses of the aorta to procaterol.

Binding pockets of the beta(1)- and beta(2)-adrenergic receptors for subtype-selective agonists.

We examined the subtype-selective binding site of the beta-adrenergic receptors (betaARs). The beta(1)/beta(2)-chimeric receptors showed the importance of the second and seventh transmembrane domains (TM2 and TM7) of the beta(2)AR for the binding of the beta(2)-selective agonists such as formoterol and procaterol. Alanine-substituted mutants of TM7 of the beta(2)AR showed that Tyr(308,) located at the top of TM7, mainly contributed to beta(2) selectivity. However, Tyr(308) interacted with formoterol and procaterol in two different ways. The results of Ala- and Phe-substituted mutants indicated that the phenyl group of Tyr(308) interacted with the phenyl group in the N-substituent of formoterol (hydrophobic interaction), and the hydroxyl group of Tyr(308) interacted with the protonated amine of procaterol (hydrophilic interaction). In contrast to beta(2)AR, TM2 is a major determinant that beta(1)-selective agonists such as denopamine and T-0509 bound the beta(1)AR with high affinity. Three amino acids (Leu(110), Thr(117), and Val(120)) in TM2 of the beta(1)AR were identified as major determinants for beta(1)-selective binding of these agonists. Three-dimensional models built on the basis of the predicted structure of rhodopsin showed that Tyr(308) of the beta(2)AR covered the binding pocket formed by TM2 and TM7 from the upper side, and Thr(117) of the beta(1)AR located in the middle of the binding pocket to provide a hydrogen bonding for the beta(1)-selective agonists. These data indicate that TM2 and TM7 of the betaAR formed the binding pocket that binds the betaAR subtype-selective agonists with high affinity.

Coexistence of beta 1-, beta 2-, and beta 3-adrenoceptors in dog fat cells and their differential activation by catecholamines.

The existence of a beta 3-adrenoceptor (in addition to classical beta 1- and beta 2-), its involvement in the control of lipolysis and its recruitment by catecholamines were investigated in dog adipose tissue. Isoproterenol, norepinephrine, and the beta 2-selective agonist procaterol fully activated lipolysis in adipocytes (order of potency: isoproterenol > norepinephrine = procaterol). beta 3-Adrenergic agonists stimulated lipolysis with the order of potency: BRL 37344 > CGP 12177 > SR 58611A. Propranolol and bupranolol (nonselective beta-antagonists) antagonized, with a low potency, the effect of BRL 37344, whereas the beta 1-antagonist CGP 20712A and the beta 2-antagonist ICI 118551 were without action. CGP 20712A inhibited the effect of lower concentrations of agonists (0.05 microM isoproterenol, 0.1 microM norepinephrine and 0.1 microM procaterol) with an inhibitory constant (mean Ki) of 0.0075, 0.032 and > 10 microM, respectively. Mean Ki values for the beta 2-antagonist ICI 118551 were 1.744, 1.243, and 0.019 microM. This result indicates that low concentrations of isoproterenol and norepinephrine stimulate lipolysis mainly via beta 1-adrenoceptors in dog fat cells. Inversely, the lipolytic effect of higher concentrations of agonists i.e., 1 microM isoproterenol and catecholamines, was weakly antagonized by CGP 20712A or ICI 118551 while the nonselective beta-antagonists bupranolol and propranolol suppressed the effects with the order of potency expected for a beta 3-adrenoceptor: bupranolol > propranolol. These data indicate 1) the presence of a functional beta 3-adrenoceptor that coexists with beta 1- and beta 2-adrenoceptors in dog fat cells; 2) a separation of the differential potencies of physiological amines in the activation of lipolysis through beta 1-, beta 2-, and beta 3-adrenoceptors; the lipolytic response initiated at low concentrations (submicromolar range) of norepinephrine is primarily mediated by the beta 1-adrenoceptor subtype; and 3) an activation of the beta 3-adrenoceptor that occurs at higher concentrations of catecholamines.
