A comparison of polyglycolic acid (Dexon), nylon and silk sutures in skin surgery.

Synthetic polyglycolic acid (Dexon) sutures were compared with silk or nylon in 50 patients requiring skin surgery. After some practice, polyglycolic acid is an easily handled as nylon or silk. Skin reactions occurred in 50% of the patients sutured with nylon, in 10% of those sutured with polyglycolic acid, and in none sutured with silk. Wound healing was satisfactory in all cases.

Cosmetic result of lower midline laparotomy wounds: polybutester and nylon skin suture in a randomized clinical trial.

OBJECTIVE: To compare the cosmetic outcome of lower midline laparotomy scars using either nylon or polybutester suture for skin closure. METHODS: A randomized clinical trial compared polybutester skin suture with nylon for lower midline laparotomy wounds in 50 women undergoing gynecologic surgery. Scar hypertrophy, scar width, scar color, the presence of cross-hatching marks, and a total score were assessed in all patients at 18 months following surgery and compared by nonparametric statistical tests. RESULTS: The wounds closed with polybutester suture were significantly less hypertrophic than those closed with nylon. Regardless of the suture material used, the lower part of the laparotomy scar showed an inferior cosmetic result compared with the upper part underneath the umbilicus for scar hypertrophy, scar width, and the total scar score. CONCLUSIONS: Polybutester skin suture diminishes the risk of hypertrophic scarring because of its special properties allowing it to adapt to changing tensions in the wound. Increased closure tension of the skin in the midline region above the pubic bone may be caused by a relative immobility of the skin. Scar cosmetics in this area may be improved by extra subcuticular suture support.

[Experiments on suture material prolene 10-0 and vicryl 11-0 compared with nylon 10-0 in the rabbit cornea (author's transl)].

Synthetic absorbable suture material Vicryl 11-0 and not absorbable Prolene 10-0 are tolerated well by the corneal tissue as compared with nylon 10-0. Vicryl 11-0 sutures remained intact for only 13.2 days on an average, this is too short for normal corneal surgery. As nylon shows signs of absorption with time Prolene may be preferable to nylon where permanent sutures are necessary.

Protective sensibility--its monofilament nylon threshold equivalents in leprosy patients.

An attempt has been made to definite the levels of "protective sensibility" in terms of perception thresholds to monofilament nylon induced touch/pressure stimuli. Certain problems were observed while interpreting the observations. There appears to be a range of threshold values instead of a clear cut-off point. We suggest that a monofilament nylon stimulus two times the normal threshold value for that patient be taken as cut-off point. This will make the observations of Birke and Sims (1986) and Hammond and Klenerman (1987) reasonable without having any need to exclude the cases who defy the boundaries laid by them. Since the genesis of plantar ulcer is multifactorial, it appears logical to include all patients who have a certain degree of hypoaesthesia, for special ulcer care program. The likely problems while using monofilament nylons in the field and their possible solutions have also been outlined.

Degranulation and abnormal bactericidal function of granulocytes procured by reversible adhesion to nylon wool.

Granylocyte bactericidal capacity, chemotaxis, hexose monophosphate shung activity (before and after phagocytic stimulus), and quantitative nitroblue tetrazolium reduction and enzyme content were examined in cells obtained by filtration leukaphresis (FL) and continuous-flow centrifugation (CFC). A decrease in the bactericidal efficiency of FL-produced cells compared to that of both normal and CFC-procured granulocytes was found; the decrease was 17% with a cell-to-bacteria ratio of 5:1, and 55% with a 1:1 ratio. Moreover, FL-acquired cells were often vacuolated and consistently contained less acid phosphatase and beta-glucuronidase than did normal granulocytes. When normal cells were incubated for 1-2 hr with nylon wool, 30% of the total acid phosphatase and beta-glucuronidase was released, with no evidence of cell death, thus suggesting degranulation. Similar results were obtained with glass, cotton, or polysulfone plastic fibers. Electron microscopic and peroxidase cytochemical studies of the adherence of normal granulocytes to nylon fibers were also carried out. After 30 min of incubation, cell-to-fiber attachment and cellular aggregation had occurred, although the cells per se appeared normal. After 60 and 120 min, other changes became apparent: (1) a decrease in the amount of cytoplasmic granules; (2) large, intracytoplasmic vaculoles; and (3) extracellular peroxidase on fiber surfaces. We conclude that granulocytes obtained by adherence to nylon fibers show both morphological and biochemical evidence of degranulation and diminished bactericidal capacity, and that these abnormalities may be causally related to decreased granulocyte survival in transfusion recipients.

The nylon wool adherence marker of the B cell lineage appears at the resting pre-B cell stage.

The proliferative state of functionally defined populations of size-separated pre-B cells was investigated. The results presented here support the hypothesis that pre-B cells are in the G2 phase when stable membrane IgM is first expressed. Small resting precursor cells significantly adhere to nylon wool columns, unlike larger cycling cells which can be enriched in the nonadherent fraction. These results suggest that the nylon wool "receptor' is a useful early B cell marker. Since the in vitro recruitment of both cycling and resting precursors was shown to be essentially dependent on agar mitogen(s), cumulative data suggest that the receptors for agar mitogen(s) appear shortly before surface IgM on proliferating pre-B cells. Nylon wool adherence potential develops next on their resting progeny, followed by full lipopolysaccharide reactivity as cells further mature.

Isolation of human T lymphocytes: comparison between nylon wool filtration and rosetting with neuraminidase (VCN) and 2-aminoethylisothiouronium bromide (AET)-treated sheep red blood cells (SRBC).

Isolation of human T lymphocytes by rosetting with SRBC, centrifugation on Ficoll-Hypaque gradients and lysis of SRBC, which reportedly affects the functional activities of lymphocytes, was compared to isolation of T lymphocytes by filtration through a nylon wool column. The two methods were similar in yields and purity of T lymphocytes. No selective loss of T cell subpopulations was found in T lymphocytes isolated by filtration through a nylon wool column as judged by the percentage of T gamma and T micron and the suppressor and helper activity in functional assays. Therefore, filtration through a nylon wool column is a useful method to purify T lymphocytes and has the advantage of being comparatively simple and quick to perform and of avoiding manipulations which may affect the functional activity of T lymphocytes.

Cyclic polyamide oligomers extracted from nylon 66 membrane filter disks as a source of contamination in liquid chromatography/mass spectrometry.

Background interferences are perhaps an unavoidable part of analytical detection schemes, particularly when analyzing trace level samples or when using detectors with high levels of sensitivity. In liquid chromatography, mobile phase solvents are routinely filtered using membrane filter disks to trap particulates in hopes of minimizing contamination, providing improvements in data output and instrumental operation. In this study, we report that one such filter disk leads to a significant level of contamination in LC and LC/MS experiments. Extractable compounds from nylon membrane filters generate significant background signals in UV absorption chromatograms at 214 nm, and are also detected by electrospray ionization mass spectrometry, with nominal m/z values of 453 and 679. It is shown that rinsing the nylon membranes before their use can reduce, but will not eliminate, the extractable contaminants from the mobile phase. Through MS and tandem MS analysis, we have identified these contaminants as cyclic oligomers of polyamide 66. Based on these results, it is recommended that nylon membrane filters be avoided when conducting trace level analysis, particularly when conducting LC/MS experiments.

Fungal keratitis from nylon line lawn trimmers.

Ocular trauma from nylon line lawn trimmers is becoming more prevalent. Previous case reports have described penetrating trauma caused by these tools. We managed three cases of fungal keratitis caused by injuries from nylon line trimmers. Fungal keratitis should be strongly considered as the cause of any corneal ulcer related to trauma from a nylon line lawn trimmer.

Ocular dangers in the garden. A new menace--nylon line lawn trimmers.

Nylon line lawn trimmers represent a source of potentially devastating ocular trauma. Five cases of ocular trauma associated with the use of line trimmers are reported. These case reports document new mechanisms and circumstances of injury, including trauma from debris other than nylon line fragments and injury to bystanders. Furthermore, injuries are often serious, with two of these patients requiring an evisceration or enucleation after endophthalmitis developed. In one patient, six different organisms were cultured and another patient exhibited an Aspergillus organism and Propionibacterium acnes. As nylon trimmers gain in popularity, they will become an increasingly frequent source of ocular injury. A retrospective 40-month review of 85 cases (87 eyes) of penetrating and perforating ocular trauma shows line trimmers to be the fifth leading cause of such injury.

Use of the mobile nylon bag technique to determine the digestible energy content of traditional and non-traditional feeds for swine.

The following experiment was conducted using the mobile nylon bag technique (MNBT) to determine dry matter and energy digestibility in traditional feeds as well as non-traditional feeds in order to calculate digestible energy (DE) values for use in ration formulation programmes. A total of 22 ingredients were tested in this experiment including the traditional cereal grains barley, corn, oats and wheat, as well as secondary cereal grains such as normal and low viscosity rye, low lignin and high fat oat groats, wheat heavies and # 1 wheat screenings and a new bread wheat designated as CDC Teal. Three legumes (lupines, peas and dehydrated alfalfa), three varieties of canary seed (CDC Maria, dehulled CDC Maria and Keet), and raw and micronized canola seed were tested as protein sources. Finally, three oilseed meals obtained from the Chinese feed industry (Chinese rapeseed meal, Chinese cottonseed meal and extruded full-fat soybean meal) were also included. After simulating gastric digestion the nylon bags were inserted into the duodenum of five barrows through simple duodenal T-cannulae. Eight bags were administered to each pig daily. Ten nylon bags were prepared for each feed with two bags being inserted into each of the five barrows. A total of 220 bags were inserted over a 7-day period. The overall results of this experiment indicate that the MNBT has great potential for use in determining the digestible energy content of swine feeds. For the most part, values obtained in the present experiment compared favourably with previously published values. Four feedstuffs produced digestible energy values that differed by greater than 5% from previously reported values. However, variation in chemical content provided a reasonable explanation for these discrepancies. The MNBT has several advantages compared with conventional digestibility methods in that many feeds can be tested in a relatively short duration of time with significantly fewer animals being used, only small amounts of feed are required and the test allows for energy measurements in feedstuffs that would not normally be fed to pigs as a single ingredient. Additional research on a wider variety of feedstuffs should be conducted to confirm the potential of the MNBT as a tool to accurately determine DE values for swine.

Feasibility of a nylon-mesh holder for vitrification of bovine germinal vesicle oocytes in subsequent production of viable blastocysts.

To improve the feasibility of nylon-mesh holder for vitrification of bovine cumulus-oocytes complexes (GV-COCs) having germinal vesicle, this study was conducted to demonstrate effects of sugars and protocol of exposure in vitrification on subsequent in vitro maturation, ultrastructural changes, and in vitro development in bovine immature oocytes after cryopreservation using nylon mesh. Before vitrification, GV-COCs were exposed to the cryoprotectant, which was composed of 40% (v/v) ethylene glycol, 18% (w/v) Ficoll-70, and 0.3 M sucrose (EFS40) or 0.3 M trehalose (EFT40), either by single step or in a stepwise way. The maturation rates in the stepwise exposure with EFS40 or EFT40 were significantly higher (P < 0.05) compared with the corresponding rates in the single step. In the stepwise exposure, few abnormalities were observed compared with the single-step exposure, where most oocytes showed a highly vacuolated cytoplasm with many ruptured mitochondria. Cleavage rates in fertilized oocytes previously exposed stepwise to EFS40 or EFT40 were significantly higher than those exposed by the single-step procedure. The cleaved embryos derived from the stepwise exposure to EFS40 developed to blastocysts. After transfer of blastocysts derived from vitrified GV oocytes, a female calf was born. These results indicate that vitrification of large numbers of bovine GV-COCs using a nylon-mesh holder accompanied with stepwise exposure minimizes structural damage in organelles, resulting in yield of viable blastocysts following in vitro embryo production.

Improved B cell typing for HLA-DR using nylon wool column enriched B lymphocyte preparations.

A rapid, simple procedure for preparing B lymphocytes using nylon wool has been devised. When mixtures of T lymphocytes, B lymphocytes, and monocytes are applied to nylon wool columns, B lymphocytes can then be removed, virtually free of monocytes. Such preparations (Ad) are 85 +/- 6% SIg+ and have only 3 +/- 1% monocyte contamination. The yield from peripheral blood averages 5% of all mononuclear cells. In contrast, the E rosette depletion method (E-) yields cells which are only 64 +/- 5% SIg+ and have 25 +/- 7% monocyte contamination. The superiority of the nylon method for HLA-DR typing was demonstrated in a comparative study of Ad and E- with eight normal individuals. Cytotoxic scores were higher and a large number of reactions, representing DRw groups and additional cross-reactions, was detected.

Hematopoiesis on suspended nylon screen-stromal cell microenvironments.

A three-dimensional culture system for the growth of primate and rodent bone marrow was developed in our laboratory. This method involves the seeding of stromal cells onto a nylon screen and the inoculation of fresh or cryopreserved bone marrow hematopoietic cells after stromal cell processes had extended across 3 to 4 out of every 5 mesh openings. Stromal cells attach, grow, and secrete matrix proteins which contribute to an intricate microenvironment for the support of multilineage hematopoiesis, which was observed for greater than 270 days in the rat model and for greater than 12 weeks in the human system, as evidenced by flow cytometry analysis and in vitro clonogenic assays. The adherent zones of these suspended nylon screen cultures consisted primarily of immature cells. These cultures could also be used as substrates for cytotoxicity measurements; treatment of rat bone marrow cultures of various ages with cytosine beta-D arabinofuranoside, cyclophosphamide, 5-fluorouracil, or methotrexate resulted in a dose-dependent decrease in CFU-C numbers and altered the phenotypic distribution of hematologic cells in the adherent zone. The use of a modification of this method to generate large numbers of active cytolytic cells after greater than 75 days culture of rat bone marrow-derived natural killer cells is described also. Suspended nylon screen bone marrow culture also has potential uses in genetic insertion and graft vs. host disease studies, blood component therapy, the evaluation of ex vivo purging programs, and in marrow expansion for transplantation.

Influence of forage species and diet particle size on the passage of digesta and nylon particles from the reticulorumen of steers.

To assess factors that influence the passage of digesta from the reticulorumen, ruminally fistulated steers (387 kg) were fed to appetite in a 4 x 4 Latin square design either orchardgrass (OG) (Dactylis glomerata L.) or switchgrass (SG) (Panicum virgatum L.) hays. The two hays were fed in either the long form or ground to pass a 2-cm screen. Steers fed OG diets had greater DMI (P < .05) than steers fed SG diets (11.7 vs 10.4 kg/d respectively). Particle size of ruminal digesta and feces determined by wet sieving was greater (P < .05) for the OG and long hay diets than for the SG and ground diets. Particle size of digesta collected from the anterior dorsal sac and anterior ventral sac of the rumen and from the reticulum did not vary (P > .25) with site. Conversely, ruminal concentrations of 1-mm nylon particles tended (P = .11) to be greater in the lower strata of the reticulorumen at 12 and 24 h after dosing. Neither a site effect nor a site x time-after-dosing interaction (P > .75) was observed for the 3- or 5-mm nylon particles, suggesting that a sorting process independent of specific gravity occurs in the rumen for smaller particles (before the reticulomasal orifice). Passage rate of the 1-mm nylon particles from the reticulorumen was greater (P < .01) for animals fed the SG than for those fed the OG diets, whereas for the 5-mm particles, passage rate was greater for the OG diets.(ABSTRACT TRUNCATED AT 250 WORDS)

Intestinal digestibility in dairy cows of protein from grass and clover measured with mobile nylon bag and other methods.

Ruminal, intestinal, and total tract digestibilities of CP in grass, grass silage, and clover were measured using the mobile nylon bag method. Total tract indigestibilities measured with this method were compared with indigestible CP predicted by in vitro, 14-d ruminal incubation, and 12-h followed by 14-d ruminal incubation methods. Indigestible CP was related to chemical composition and harvest date. Ruminal disappearance of CP from nylon bags was 58, 71, and 58% for grass, grass silage, and clover, respectively. Intestinal CP disappearance was 84, 81, and 91% of escape CP, respectively. Lower ruminal disappearance was compensated by higher intestinal disappearance, resulting in a small variation in total tract indigestible CP within forage. Total tract indigestible CP was 7, 6, and 4%, respectively. Average total tract indigestible CP measured with the mobile nylon bag method (6%) was lower than that estimated by the in vitro (13%), 14-d ruminal incubation (14%), or 12-h ruminal incubation followed by 14-d ruminal incubation (11%) methods. All methods ranked forages similarly, resulting in high correlation coefficients among methods within forage. Total tract indigestible CP of grass, grass silage, and clover was related to CP content and harvest date, DM and ADIN content, and DM and CP contents, respectively.

Wound infection following high saphenous ligation. A trial comparing two skin closure techniques: subcuticular polyglycolic acid and interrupted monofilament nylon mattress sutures.

A prospective, randomized trial was performed to compare the incidence of wound infection following skin closure with subcuticular polyglycolic acid and interrupted monofilament nylon mattress sutures in patients undergoing high saphenous ligation (126 patients; 75 polyglycolic acid wounds, 86 interrupted monofilament nylon mattress wounds). By 6 weeks after operation, pus had discharged from 15 (20%) polyglycolic acid wounds compared with seven (8%) interrupted monofilament nylon mattress wounds (P = 0.05), giving an overall infection rate of 13.7%. Infection occurred in 23% of the 56 procedures performed by the two surgeons with the highest infection rates compared with 8% of the 105 procedures performed by the remaining nine surgeons (P = 0.02). The excess of infections in wounds closed with subcuticular polyglycolic acid was caused by an excess of infections in operations performed by the two surgeons with the highest infection rates. The higher infection rate found with subcuticular polyglycolic acid appeared to be operator dependent.

The use of sterile adhesive tape in the closure of arthroscopic puncture wounds: a comparison with a single layer nylon closure.

A prospective trial was undertaken comparing the wound healing and infection rates in arthroscopic arthrotomy wounds closed by sterile adhesive tapes on interrupted Nylon skin sutures. Two hundred and thirty consecutive arthroscopic procedures had arthrotomy wounds closed by a single layer skin closure of either interrupted Nylon (n = 62) or sterile adhesive tapes (n = 168). All wounds healed by primary intention. Only one wound developed a superficial infection around a Nylon suture and no patient developed a synovial herniation. It is concluded that closure of arthroscopic puncture wounds with sterile adhesive tape is effective and convenient for wound management.
