Hormone-responsive cells derived from human dental papilla: characterization in vitro and in vivo in diffusion chambers.

Cells of the dental papilla are capable of odontoblastic, fibroblastic, and endothelial differentiation and formation of dentin and the dental pulp. In the present study dental papilla cells, obtained from human tooth buds (HDP cells), were cultured in vitro through 3 to 7 passages. After exposure to prostaglandin E2 there was a marked decrease in intracellular cyclic AMP (cAMP) levels as compared to hormone-free controls. Parathyroid hormone and calcitonin had stimulatory effects with 1 and 2 log increases in cAMP, respectively. The HDP cells showed moderate activity of alkaline phosphatase, 1 log higher than that of hamster kidney fibroblasts (BHK 13) and 1 log lower than that of osteoblastic osteosarcoma cells (ROS 17/2). When cultured for 4 or 8 wk in diffusion chambers (DC) implanted in athymic mice, many of the HDP cells underwent odontoblastic morphodifferentiation with very long, single processes extending into the matrix. This matrix contained banded and unbanded collagen fibers. Neither light nor electron microscopy of the DC content revealed mineral deposits. These results suggest that HDP cells have an intrinsic potential for partial odontoblastic differentiation; inductive signals like those originating from odontogenic epithelium are probably essential for the completion of hard tissue formation.

Effect of impression materials and techniques on the marginal fit of metal castings.

The effect of two impression materials used in three different techniques was tested for the accuracy of the marginal fit of metal castings. Sixty identically prepared Ivorine teeth were divided into groups of 20. The following methods were selected to make impressions: (1) putty in a metal stock tray with simultaneous elastomeric impression wash, (2) copper band relined with autopolymerizing acrylic resin and subsequent light-body elastomeric impression material, and (3) copper bands with modeling compound. Faciolingual sections of the Ivorine teeth with cemented metal castings were examined by scanning electron microscopy (SEM) at x 100 magnification. Measurements of the thickness of cement layers were calculated both manually and by use of a recently developed computerized method. The differences in the gingival marginal gaps between various impression materials and techniques at both the facial and lingual surfaces were not statistically significant.

Effect of retraction procedures on the periodontium in humans.

On the basis of wound healing and gingival recession caused by the three procedures, the copper-band retraction method was the most satisfactory. This tentative conclusion is based on the following reasons: 1. Retraction methods must be evaluated relative to the impression procedure and fit of the restoration. The long-range effects of the marginal fit are probably the most important factor for enhancing periodontal health. 2. This study involved only healthy periodontia of adolescent patients. Different healing might be observed in tissues characterized by gingivitis or periodontitis. 3. A broader study involving a greater range of procedures and conditions is recommended to evaluate each retraction technique. 4. This study involved teeth which had an adequate zone of attached gingiva. More complicated healing and perhaps altered sequences might be observed if the procedures were performed on gingival margins of alveolar mucosa, thin gingival walls, or areas of root prominence and thin cortical bone.

Rapid genotyping of Escherichia coli O157 isolates by random amplification of polymorphic DNA.

Although several typing methods have been described for Shiga-like toxin-producing Escherichia coli O157, the methods are somewhat cumbersome. Using 22 isolates of Escherichia coli O157 and five other Escherichia coli isolates, two primers (M13 core sequence and 970-11) were found to give excellent differentiation between isolates using random amplified polymorphic DNA (RAPD). Using only the presence or absence of variable bands, a matrix of 20 variable characters was identified. From these characters, similarity coefficients were calculated and a phenogram constructed. All of the Escherichia coli O157 isolates were easily distinguished from the non-O157 Escherichia coli isolates. Using a 95% similarity cutoff, we found 13 RAPD types among the 22 Escherichia coli O157 isolates. Isolates thought to be identical by toxin and phage typing as well as by epidemiological association were distinguished, and others thought to be distinct by lack of epidemiological association were identical. RAPD using M13 and 970-11 primers is a potentially useful typing tool for Escherichia coli isolates of serotype O157 and possibly other Escherichia coli isolates.

The influence of restorative treatment approaches and the use of local analgesia, on the children's discomfort.

AIM: The aim is to investigate possible differences in discomfort during treatment with the atraumatic restorative treatment (ART) or the Conventional restorative method with and without local analgesia (LA). METHODS: The study group consisted of 6 and 7 year old children with no dental experience (mean age 6.98, SD +/- 0.52) randomly divided into four treatment groups: Conventional method with and without LA and ART with and without LA. One or two proximal lesions in primary molars were treated. The heart rate and the behaviour (Venham) were measured. STATISTICS: Statistical analysis was performed in SPSS version 10.0. RESULTS: In a first session 300 children were treated and 109 children for a second time in the same way as at the first visit. During the first session ART without LA gave the least discomfort while the Conventional method without LA gave the most discomfort. During the second treatment the least discomfort was observed with ART without LA and the most discomfort in the Conventional way with LA. CONCLUSION: There is a constant preference for hand instruments; the bur is increasingly accepted. The experience with LA is the reverse.

Tissue displacement methods in fixed prosthodontics.

The various methods to accomplish gingival displacement have been described. The techniques have been noted and their relative advantages and disadvantages summarized. The practicing dentist can use this review to make better informed decisions regarding the method he or she chooses for the treatment of a particular patient.

A rapid dowel core construction technique.

The use of composite resins for dowel cementation and core construction is well established. A short, easy, time-saving method is proposed for simultaneous dowel cementation and core fabrication using a composite core construction material.
