Silica stone--development due to long time oral trisilicate intake.

We report a case of a 30-year-old female who had a long-term history of trisilicate-containing antacid intake for gastric discomfort. She had experienced repeated attacks of renal colic. Neither intravenous pyelography nor ureteroscopy could define the presence of calculi. The metabolic evaluations were normal. However, X-ray diffraction revealed a silicate stone. We suggest that the attack of renal colic in those patients with long-term history of trisilicate intake should arouse the possibility of silica urolithiasis.

Recovery of viruses from water by a modified flocculation procedure for second-step concentration.

A reduction in virus recovery efficiencies stemming from a change in the commercial processing of powdered beef extract was reversed by the addition of Celite analytical filter aid. Supplementing beef extract with this silicate is recommended as a modification to the organic flocculation procedure for second-step concentration in monitoring for waterborne viruses. Considerable differences in virus recovery were found among lots of beef extract and Celite preparations; this indicates that the performance of each lot of these substances should be checked before use.

Sorption-desorption of cadmium in aqueous palygorskite, sepiolite, and calcite suspensions: isotherm hysteresis.

Sorption isotherms have been widely used to assess the heavy metal retention characteristics of soil particles. Desorption behavior of the retained metals, however, usually differ from that of sorption, leading to a lack of coincidence in the experimentally obtained sorption and desorption isotherms. In this study, we examine the nonsingularity of cadmium (Cd) sorption-desorption isotherms, to check the possible hysteresis and reversibility phenomena, in aqueous palygorskite, sepiolite and calcite systems. Sorption of Cd was carried out using a 24-h batch equilibration experiment with eight different Cd solution concentrations, equivalent to 20-100% of maximum sorption capacity of each mineral. Immediately after sorption, desorption took place using successive dilution method with five consecutive desorption steps. Both Cd sorption and desorption data were adequately described by Freundlich equation (0.81<r(2)<0.99). The sorption and desorption reactions, however, did not provide the same isotherms, indicating that hysteresis occurred in Cd sorption-desorption processes. The extent of hysteresis was quantified based on the differences obtained from sorption and desorption isotherms regarding the amount of Cd sorbed, the Freundlich exponent, and the Cd distribution coefficient. The results revealed that, sepiolite possessed the most hysteretic behavior among the minerals studied. Calcite showed much smaller hysteresis compared to the other two silicate clays at low Cd surface load, but its hysteresis indices significantly increased, and exceeded that of palygorskite, as the amount of Cd in the systems increased. The average amount of Cd released after five desorption steps, was 13.8%, 2.2% and 3.6% for the palygorskite, sepiolite and calcite, respectively, indicating that a large portion of Cd was irreversibly retained by the minerals.

Sepiolite asbestos from Franklin, New Jersey: a case study in medical geology.

Asbestiform sepiolite has been found in a zinc deposit at Franklin, New Jersey. The host rock is Precambrian Franklin marble. Pseudomorphous foliated texture and cross-cutting relationships indicate replacement of talc by sepiolite. Sepiolite is manganoan, while talc contains little manganese, suggesting differences in manganese substitution in these minerals and providing evidence against solid-state replacement. Sepiolite from Franklin is of moderate crystallinity and consists of soft, flexible mass-fiber. Comparison with other sepiolite samples suggests that increased crystallinity among sepiolites parallels increased fiber length, while disorder appears to be associated with flexibility. Length and aspect ratio (length/width) distributions support toxicological equation of asbestiform sepiolite with the regulated asbestos minerals. The data presented here suggest that talc can be unstable in a low-temperature hydrothermal environment, altering under certain conditions to form sepiolite. In a wider context, sepiolite may be expected to precipitate at the end-stages of hydrothermal mineralization in some talc deposits and in carbonate and calc-silicate rocks. Many occurrences of sepiolite in these rocks may have been overlooked due to similarities with other minerals. The fact that sepiolite has been previously unrecognized and misidentified at Franklin is a case in point. Since there is evidence suggesting that sepiolite is fibrogenic and carcinogenic, its potential occurrence in consumer talcs and crushed-stone products has environmental health implications. Similarities in composition and other analytical parameters may cause sepiolite to be mistaken for fibrous talc or chrysotile in environmental samples.

Joint analyses by laser-induced breakdown spectroscopy (LIBS) and Raman spectroscopy at stand-off distances.

Raman spectroscopy and laser-induced breakdown spectroscopy (LIBS) of solid samples have both been shown to be feasible with sample-to-instrument distances of many meters. The two techniques are very useful together, as the combination of elemental compositions from LIBS and molecular vibrational information from Raman spectroscopy strongly complement each other. Remote LIBS and Raman spectroscopy spectra were taken together on a number of mineral samples including sulfates, carbonates and silicates at a distance of 8.3 m. The complementary nature of these spectra is highlighted and discussed. A factor of approximately 20 difference in intensity was observed between the brightest Raman line of calcite, at optimal laser power, and the brighter Ca I LIBS emission line measured with 55 mJ/pulse laser power. LIBS and Raman spectroscopy have several obstacles to devising a single instrument capable of both techniques. These include the differing spectral ranges and required detection sensitivity. The current state of technology in these areas is discussed.

Atomic scale modelling of the cores of dislocations in complex materials part 2: applications.

In an accompanying article, we have described a methodology for the simulation of dislocations in structurally complex materials. We illustrate the applicability of this method through studies of screw dislocations in a structurally simple ionic ceramic (MgO), a molecular ionic mineral (forsterite, Mg2SiO4), a semi-ionic zeolite (siliceous zeolite A) and a covalent molecular crystalline material (the pharmaceutical, orthorhombic paracetamol-II). We focus on the extent of relaxation and the structure of the dislocation cores and comment on similarities and points of disparity between these materials. It is found that the magnitude of the relaxation varies from material to material and does not simply correlate with the magnitude of the principal elastic constants in an easily predictable fashion, or with the size of the cohesive lattice energy or length of the Burgers vector, which emphasises the need to model the non-linear forces and atomic structure of the core.

Interaction between erythrocyte plasma membrane and silicate dusts.

Lysis of erythrocytes in vitro leading to release of hemoglobin has often been used as a convenient test system for predictive toxicological evaluation of noxious particulates encountered as pollutants in occupational and environmental atmospheres. However, in spite of several studies with silica and silicate dusts, the exact biochemical mechanism for membrane damage is not clear. Therefore, detailed biochemical studies were conducted. Screening of a large number of fugitive dust samples indicated that the dissolution of silica under assay conditions has a qualitative relation to hemolytic potency. The strong capacity of slate mine dust to cause hemolysis was studied in detail. The kinetics of lysis indicated that on prolonged contact the adsorption of hemoglobin altered the lytic index. Coating of dust with serum, albumin or polyvinylpyrrolidone (PVP) and some lipids reduced lytic potency markedly, while EDTA was ineffective. Altering the surface chemistry of dust by heating, alkali treatment, washing with water, and acid washing reduced hemolysis in increasing order. Thus, chemical interactions between dusts and membranes are involved in hemolysis, and it may be concluded that the interaction of dust constituents with biomembranes is one of the likely mechanisms involved in the toxicity of particulate air pollutants.

A study of Spanish sepiolite workers.

BACKGROUND: Sepiolite is an absorbent clay that is used as pet litter. It forms thin crystals, which are a transition between chain and layered silicates. Inhalation studies in animals have shown no evidence of pulmonary damage. This paper reports a cross sectional study of the total work force of the largest sepiolite production plant in the world. METHODS: Two hundred and eighteen workers (210 men and eight women) were studied. Height, age, and smoking history were recorded. Chest radiographs were read according to the International Labour Office (ILO) classification by two readers. Readings were used to construct a numerical score, which was then used in statistical analyses. Forced expiratory volume in one second (FEV1) and forced vital capacity (FVC) were divided by the square of the height. Casella size selective personal samplers were used in randomly selected operatives to collect dust eight years before the rest of the study was carried out. These samples were evaluated gravimetrically. Total dust samples were examined by optical and electron microscopes. Results were analysed by bivariate linear regression, chi 2 tests, and analysis of variance. RESULTS: When allowance was made for smoking habit workers exposed to dry dust showed a significantly greater decline in FEV1 with age than workers with little exposure to dry dust. A similar pattern applied to FVC. Radiographic score showed deterioration with age but no clear differences from other variables. High concentrations of dust were found in the bagging department and also in the classifier shed. CONCLUSIONS: The major finding was that lung function deteriorated more rapidly in those who had had more exposure to dust, but there was no evidence of any accompanying radiographic change.

Exit wave reconstructions using through focus series of HREM images.

The through focus exit wave reconstruction technique uses a series of high resolution electron microscopy images to reconstruct the complex electron wavefunction at the exit plane of the specimen. The feasibility of the through focus exit wave reconstruction method and its most important limitations are discussed. It is shown that-provided all aberrations of the microscope are well corrected for-a large improvement in the interpretability of the information can be obtained.

Kinetics of in vivo degradation of sepiolite-collagen complexes: effect of glutaraldehyde treatment.

A kinetic study of the in vivo degradation of sepiolite-collagen complexes after subcutaneous implantation in rats was performed. A foreign-body reaction was the characteristic host tissue response against the implants. The resorption of the implanted materials was analyzed by measuring both the weight and the collagen persistences. This last was measured by using (14C)acetylated collagen, which was revealed to be not modified upon radioactive labeling, in terms of its ability to form a complex with sepiolite. The persistence of the implants is controllable by treatment of the collagen component with glutaraldehyde. Thus, for 1% glutaraldehyde-treated collagen complexes, 100% of persistence was observed after several months of implantation, this value decreasing to a few days for nontreated collagen samples. The collagen-sepiolite complex showed a low immunological response, almost null for 1% glutaraldehyde-treated collagen complexes, which was analyzed by measuring anti-collagen antibodies levels. Based on the performed studies, sepiolite-collagen complexes can be considered a resorbable material.

Synthesis and degradation of nucleic Acid components by formamide and cosmic dust analogues.

We show the unprecedented one-pot synthesis of a large suite of pyrimidines (including cytosine and uracil) and purines from formamide in the presence of cosmic-dust analogues (CDAs) of olivines. Since the major problem in the origin of informational macromolecules is the instability of their precursors, we also investigate the stabilizing effect of CDAs on the intrinsic instability of oligonucleotides in formamide.

Cell morphology, proliferation and collagen synthesis of human fibroblasts cultured on sepiolite-collagen complexes.

The growth and morphology as well as collagen biosynthesis of human fibroblasts obtained and cultured on sepiolite-collagen complexes have been studied. No differences on cell morphology and growth properties nor collagen synthesis were observed when compared with standard culture substrates. The type I/type III ratio of biosynthesized collagen by fibroblasts cultured on sepiolite-collagen complexes was about 5-6 with no difference when compared to control conditions. This normal behavior was also observed for the type I/type III procollagens. According to these studies the sepiolite-collagen complexes do not modify the studied features of the fibroblasts.

Effect of crystallization heat treatment on the microstructure and biaxial strength of fluorrichterite glass-ceramics.

The purpose of this study was to evaluate the effect of crystallization heat treatment on the flexural strength of potassium fluorrichterite glass-ceramics for biomedical applications. After melting and casting, discs were sectioned from the glass ingots, randomly divided into 10 groups, and heat treated at various temperatures ranging between 890 degrees C and 925 degrees C for various durations. One group was air-abraded after heat treatment. XRD revealed the presence of fluorrichterite and fluormica for all heat-treated groups. SEM confirmed the presence of both fluorrichterite and fluormica crystals. Crystal size increased with temperature and duration of the heat treatment. The mean flexural strength after air abrasion was not significantly different than that of the other groups except the group heat treated at 925 degrees C. The mean flexural strength was maximum after heat treatment at 900 degrees C for 1 h (497.6 +/- 25.5 MPa) and decreased linearly with the longer durations of heat treatment at 900 degrees C.

Correlation of absorption of sulfamethazine boluses with dissolution using a new dissolution apparatus for veterinary tablets.

A rotating-basket apparatus for dissolution testing of veterinary bolus tablets was designed and constructed. Sulfamethazine boluses containing different disintegrating agents were evaluated in vitro and by blood level data following administration to cattle. The dissolution t50 and various pharmacokinetic parameters showed directly compressible starch and carboxymethylstarch to be the most effective disintegrants in the concentrations employed while magnesium aluminum silicate and microcrystalline cellulose were about equal but less effective than the previous disintegrants. A bolus formulation containing no disintegrant gave even less satisfactory results. A correlation was established between the dissolution t50 and the time to peak plasma level and also between the t50 and the area under the plasma-time curve for the first 36 hr.

In vitro uptake of oral contraceptive steroids by magnesium trisilicate.

Some steroids used in oral contraceptives were adsorbed significantly by magnesium trisilicate. The adsorption affinity followed the sequence: ethindrone greater than mestranol greater than norethindrone greater than ethinyl estradiol. Adsorption data obtained at relatively low initial concentrations fitted a Langmuir plot; the values for monolayer adsorption ranged between 0.24 and 0.32 mg/g. At higher concentrations of the steroids, multilayer adsorption occurred. The results of desorption experiments made at 37 degrees in water and 0.05 N HCl suggested that desorption was incomplete and depended on the amount of steroid adsorbed. During the dissolution testing of a brand of contraceptive tablets containing norethindrone acetate, the presence of 0.5% (w/v) magnesium trisilicate in the medium resulted in almost complete reduction in the amount of the steroid remaining in solution after 1 hr.

Effect of concomitant administration of magnesium trisilicate on GI absorption of dexamethasone in humans.

The oral absorption of dexamethasone in humans was compared to its absorption when coadministered with magnesium trisilicate. The bioavailability of dexamethasone was estimated by measuring the suppressive effect of the drug on the daily excretion of endogenous steroids. Administration of 1 mg of dexamethasone to six healthy male volunteers significantly decreased the urinary excretion of 11-hydroxycorticosteroids. However, the coadministration of magnesium trisilicate with dexamethasone decreased its absorption, as indicated by the increased urinary excretion of 11-hydroxycorticosteroids. The decrease in absorption was attributed to drug adsorption on the antacid surface. These results confirm previous in vitro findings.

Influence of viscosity on absorption from nitrofurantoin suspensions.

Nitrofurantoin, 200 mg, was administered orally to 11 subjects in an aqueous reference dispersion and in five suspensions having the same rheogram. Algin, carbomer, guar gum, methylcellulose, and colloidal magnesium aluminum silicate were the five suspending agents employed. Complexation was demonstrated by dialysis between nitrofurantoin and methylcellulose and between nitrofurantoin and carbomer; however, physiological availability was not altered by the interaction. The viscosity increase slowed absorption and urinary excretion, thus delaying the time of the maximum excretion rate without a decrease in bioavailability. A clinically acceptable urinary nitrofurantoin concentration was maintained for at least 2 hr longer by a viscosity increase.

Sepiolite, a potential excipient for drugs subject to oxidative degradation.

Sepiolite, a member of the fibrous mineral group of clays, is relatively free of surface ferric iron and does not accelerate significantly the oxidative degradation of hydrocortisone. The compatibility of sepiolite with drugs that can undergo oxidative degradation is in sharp contrast to the catalytic effect of attapulgite, another fibrous mineral, which contains a significant amount of surface ferric iron and which therefore accelerates oxidative degradation. Sepiolite adsorbs hydrocortisone by a weak adsorption mechanism, which was shown by IR spectroscopy to be chiefly due to hydrogen bonding. However, accelerated oxidative degradation of the adsorbed hydrocortisone does not occur. Maximum adsorption occurs at pH 7-8.5. Desorption occurs readily by washing with water. Sepiolite has similar rheological properties to attapulgite. The results of this study suggest that sepiolite may be useful as a pharmaceutical excipient for drugs that undergo oxidative degradation.
