Effect of intracochlear aminooxyacetic acid on cochlear potentials and endolymph composition.

Summary--Aminooxyacetic acid (AOAA; 1 MM) when perfused through the scala tympani of guinea pigs produced a biphasic reduction in the endocochlear potential, together with a shift in the input-output functions of the cochlear microphonics and the compound action potential of the cochlear nerve. Since the ionic (Na+, K+) content of the scala media was not alerted, it appears that such changes did not underlie the reduction recorded in the endocochlear potential. The negative direct current potential recorded in the organ of Corti did not appear to be affected by the drug. Attempts to antagonize the effects of AOAA with pyridoxine failed.

Transport of 5-hydroxydopamine and horseradish peroxidase through the perilymph-endolymph barrier.

The distribution of 5-hydroxydopamine (5-OHDA) and horseradish peroxidase (HRP) in the cochlea was examined by light and electron microscopy after perilymphatic perfusion. Reaction product of 5-OHDA was observed along the endolymphatic surface of the cochlear duct in both 10-minute and 30-minute perfusions, while that of HRP was observed only in the 30-minute perfusion. Both tracers were found in pinocytotic vesicles of Reissner's membrane, Claudius' cells, and Hensen's cells. No labeling was seen in tight junctions of the epithelial cells lining the scala media. The epithelial cells of the spiral prominence showed various degrees of degeneration in the 5-OHDA experiment. These morphologic findings suggest that 5-OHDA and HRP can enter the endolymphatic space from the perilymphatic space by pinocytosis and that the degree of transport depends on the size of the particles.

Effects of exposure to noise on permeability to potassium of the endolymph-perilymph barrier in guinea pigs.

Healthy guinea pigs were exposed to broadband noise at 115 dBA for 7 days. When perfusion of the scala vestibuli was carried with artificial perilymph containing 43K, the 43K concentrations in the perilymph of the scala tympani were very low in both control and noise-exposed guinea pigs. The transport rate constant for K+ was computed by utilizing compartmental analysis. The results indicate that exposure to noise at 115 dBA for 7 days does not alter the permeability to K+ of Reissner's membrane or the lateral wall of the cochlear duct but does decrease K+ conductance of the organ of Corti.

[Effect of potassium canrenoate on the EP and Na+, K+ activities in endolymph].

The perilymphatic space was perfused with artificial perilymph containing 5 x 10(-3)M potassium canrenoate. The EP, K+ and Na+ activities in the scala media were measured with double barrelled K+ or Na+ selective microelectrode. Following the onset of the perfusion, the EP gradually declined and was stable after about 20 minutes. K+ activity also declined gradually but Na+ activity was unchanged. When the EP became stable, the artificial ventilation was stopped. The EP dropped to a large negative potential and K+ activity decreased gradually, but Na+ activity increased by degrees. The same results were observed in the untreated animals when the ventilation was stopped. There are no pathological changes both in the TEM view of the stria vascularis and in the SEM view of the hair cell. These results suggest that this drug may affect K+ conductance of the stria vascularis specifically.
