Bromothymol blue and carbohydrate-sensitive plating media.

A new plating medium using bromothymol blue (BTB) indicator is described and compared with eosin-methylene blue (EMB), MacConkey, and Endo media. These media were tested with L-arabinose by plating fermenting and nonfermenting mutant strains of Escherichia coli. The minimum concentrations of L-arabinose that permitted differentiation of these strains were determined. Different concentrations were required for differentiating confluent patches of cells, isolated colonies, and closely spaced or adjacent colonies. L-Arabinose, L-rhamnose, D-lactose, and D-galactose were tested with modified enteric media and with BTB medium, again to determine minimum usable concentrations. BTB media and reformulated conventional media allowed detection of acidification, aerobically, at one-fifth to one-hundredth the (1%, wt/vol) concentration of carbohydrate used in standard indicator plates.

Flow injection manifold for the direct spectrophotometric determination of bismuth in pharmaceutical products using Methylthymol Blue as a chromogenic reagent.

A new simple and rapid flow injection method is reported for the direct determination of bismuth in pharmaceutical products. Methylthymol Blue (MTB) was used as a chromogenic reagent and the absorbance of the colored Bi(III)-MTB complex produced was monitored at 548 nm. The various chemical and physical variables were optimized and a study of interfering ions was also carried out. Linear calibration graphs were obtained from 0 to 100 mg l-1 Bi(m) (120 injections per hour). The precision was very good (sr = 1.3%) and the limit of detection was cL = 0.150 mg l-1. The average accuracy was also very good (er = 0.75%) and was evaluated by comparison of the results obtained with those claimed by the manufacturers. The method was found to be adequately selective, considering the ions that the samples contain.

Spectrophotometric determination of indinavir in bulk and pharmaceutical formulations using bromocresol purple and bromothymol blue.

A simple, sensitive and selective method for the determination of indinavir (IND) in bulk and in pharmaceutical formulations is described. The method is based on extraction of this drug into chloroform as ion-pair with sulphonphthalein dyes as bromocresol purple (BCP) and bromothymol blue (BTB). The optimum conditions of the reactions were studied and optimized. The absorbance of the yellow products was measured at 427 nm for IND-BCP and 420 nm for IND-BTB. The calibration graphs were linear over the range 4.0-60.2 microg x ml(-1) of drug in chloroform, using the two dyes. The composition of the ion-pairs was established by the molar ratio method. For IND the molar ratio was determined to be 1:1 by measurement of first derivative signals at 273 nm. A calibration graph was established for 3.0-70.6 microg x ml(-1) of IND for first derivative spectrophotometry. The developed method was applied successfully for the determination of IND in pharmaceutical formulations. The data obtained were compared the data given by first derivative spectrophotometry. No differences were found.

[Comparison of 2 field methods for urinary assay of chloroquine].

Two methods of urinary chloroquine assay were tested in pregnant women in the town of Banfora (Burkina Faso): the method with bromothymol blue (Bergqvist) and the method with methyl-orange (Haskins and Mount or HMM II). Urinary assay of chloroquine was performed with both methods in 45 women chosen at random whether or not under chemoprophylactic treatment (21 taking a weekly prophylaxis of 300 mg of chloroquine, and 24 controls). The HMM II method proved to be more sensitive (100%) and more specific (91.7%) than the Bergqvist method (80.9% and 83.3% respectively); it was also more reliable with regard to positive (91.3% versus 80.9%) and negative (100% versus 91.3%) predictive values. Moreover, the quantitative appreciation of the levels of chloroquine excretion proved to be superior with the HMM II method. Finally, this method is faster to perform, easier to use and cheaper, making it the method of choice for field assay of chloroquine in urine.

Extractive spectrophotometric methods for determination of lercanidipine.

Extractive spectrophotometric methods are described for the determination of lercanidipine (LER) either in pure form or in pharmaceutical formulations. The methods involve formation of coloured chloroform extractable ion-pair complexes with bromothymol blue (BTB) and bromocresol green (BCG) in acidic medium. The extracted complexes showed absorbance maxima at 417 and 416 nm for BTB and BCG, respectively. The optimization of the reaction conditions was investigated. Beer's law is obeyed in the concentration ranges 6.0-42.0 microg x ml(-1) or 7.1-43.8 microg x ml(-1) with BTB or BCG, respectively. The composition of the ion-pairs was found to be 1:1 by Job's method. The specific absorptivities, molar absorptivities, Sandell sensitivities, standard deviations and percent recoveries were evaluated. Also, LER was determined by measurement of its first derivative signals at 245 nm. Calibration graph was established for 4.2-58.0 microg x ml(-1) of LER. The methods have been applied to the determination of drug in commercial tablets. Results of analysis were validated. No interferences were observed from common pharmaceutical adjuvants.

Extractive-spectrophotometric determination of disopyramide and irbesartan in their pharmaceutical formulation.

Picric acid, bromocresol green, bromothymol blue, cobalt thiocyanate and molybdenum(V) thiocyanate have been tested as spectrophotometric reagents for the determination of disopyramide and irbesartan. Reaction conditions have been optimized to obtain coloured comoplexes of higher sensitivity and longer stability. The absorbance of ion-pair complexes formed were found to increases linearity with increases in concentrations of disopyramide and irbesartan which were corroborated by correction coefficient values. The developed methods have been successfully applied for the determination of disopyramide and irbesartan in bulk drugs and pharmaceutical formulations. The common excipients and additives did not interfere in their determination. The results obtained by the proposed methods have been statistically compared by means of student t-test and by the variance ratio F-test. The validity was assessed by applying the standard addition technique. The results were compared statistically with the official or reference methods showing a good agreement with high precision and accuracy.

Sensitive extractive spectrophotometric methods for the determination of trazodone hydrochloride in pharmaceutical formulations.

Two simple, rapid and sensitive extractive spectrophotometric methods have been developed for the assay of trazodone hydrochloride (TRH) in pure and pharmaceutical formulations. These methods are based on the formation of chloroform soluble ion-association complexes of TRH with bromothymol blue (BTB) and with bromocresol purple (BCP) in KCl-HCl buffer of pH 2.0 (for BTB) and in NaOAc-AcOH buffer of pH of 3.6 (for BCP) with absorption maximum at 423 nm and at 408 nm for BTB and BCP, respectively. Reaction conditions were optimized to obtain the maximum color intensity. The absorbance was found to increase linearly with increase in concentration of TRH, which was corroborated by the calculated correlation coefficient values (0.9996, 0.9945). The systems obeyed Beer's law in the range of 0.2-14.5 and 0.2-14.1 microg/ml for BTB and BCP, respectively. Various analytical parameters have been evaluated and the results have been validated by statistical data. No interference was observed from common excipients present in pharmaceutical formulations. The proposed methods are simple, accurate and suitable for quality control applications.

[Evaluation of the methylthymol blue and orthocresolphthalein direct methods of determination of serum calcium].

Two methods for the direct colorimetric determination of serum calcium (BMT and OCFT) have been evaluated and compared with atomic absorption spectrophotometry (AAS). Both colorimetric methods resulted simple to use, but temperature-sensitive: values for the ratio absorbance at 10 degrees C/absorbance at 45 degrees C were 1.6 (BMT) and 1.8 (OCFT). When readings were taken at 37 degrees C good precision was achieved with both methods (between-the-series coefficients of variation were 3.5 divided by 2.2% for OFCT and 1.9 divided by 3.4% for BMT). No interference from bilirubin (up to 180 mumol/l) was observed with both methods; slight positive interference from haemoglobin (1 g/l) was observed only with OCFT method. Results from the two colorimetric methods closely agreed in the range 1.0 divided by 4.5 mmol/l: statistical analysis of comparison data gave BMT = 0.0963 + 0.9521 OCFT, r = 0.9820. From the results of direct comparison with AAS (103 serum specimens, concentration spanning from 1.0 to 4.5 mmol/l) statistical analysis gave: BMT (y)/AAS(x): y = 0.0721 + 1.0226x (r = 0.9678); OCFT(y)AAS(x): y = -0.0208 + 1.0720x (r = 0.9836): it is concluded that both methods show a slight positive bias with respect to AAS.

Possible mechanisms of the efflux of glutamate from kidney mitochondria generated by the activity of mitochondrial glutaminase.

The transport of glutamate across the inner membrane of kidney mitochondria and the influx of glutamine into the mitochondria was studied using an oxygen electrode, the swelling technique and by continous recording of the activity of the mitochondrial glutaminase by an NH4+-sensitive electrode. It is well known that the enzyme is activated by inorganic phosphate and strongly inhibited by glutamate. 1. Avenaciolide, Bromocresal purple and Bromothymol blue inhibited the respiration of the mitochondria almost completely in the presence of glutamate as substrate but not in the presence of glutamine. Production of aspartate during the oxidation of glutamine was not significantly inhibited by avenaciolide but it was markedly suppressed by Bomocresol purple and Bromothymol blue. 2. Swelling of kidney mitochondria in an isosmotic solution of glutamine and ammonium phosphate was not inhibted by avenaciolide or Bromocresol purple indicating that these substances do not inhibit the penetration of the mitochondrial membrane by glutamine or phosphate. 3. The activity of the mitochondrial glutaminase was strongly inhibited by avenaciolide or Bromocresol purple in the presence of inhibitos of respiration or an uncoupler but not in ther absence. Experimental data suggest that this was caused by the inhibition of glutamate efflux. The addition of a detergent removed this inhibition. On the basis of these observations it was concluded that two mechanisms exist which enable glutamate to leave the inner space of kidney mitochondria: (a) an electrogenic efflux coupled to the respiration-driven proton translocation and the presence of a membrane potential (positive outside) and (b) an electroneutral glutamate-hydroxyl antiporter which is inhibted by avenaciolide and which operates in both directions. Our observations do not support the existence of the electrogenic glutamine-glutamate antiporter or glutamate-aspartate exchange in the mitochondria studied.

Direct and selective flow-injection method for the simultaneous spectrophotometric determination of calcium and magnesium in red and white wines using online dilution based on "Zone Sampling".

The present work reports a selective and simple flow injection method for the direct and simultaneous determination of calcium and magnesium ions in red, rose, and white wines. Both ions react with methylthymol blue (MTB) at a strongly basic medium to form colored complexes that are monitored spectrophotometrically (lambda(max) = 610 nm). The simultaneous determination is achieved by online masking of magnesium by 8-hydroxyquinoline (8-HQ). Incorporating an online dilution mode based on the "zone sampling" technique in the FI system, the determination of both analytes was achieved without any pretreatment of the samples, in the range 0-350 mg L(-1) and 0-200 mg L(-1) for Ca(II) and Mg(II), respectively. The 3 sigma detection limits were quite satisfactory (2.1 and 1.8 mg L(-1) for Ca(II) and Mg(II) respectively), and the precision was 1.2% (at a mixture of 100.0 mg L(-1) Ca(II) + 100.0 mg L(-1) Mg(II), n = 12). A detailed study of interferences proved that the proposed method is highly selective. The application of the method to the direct analysis of red, rose, and white wines yielded excellent results compared with those obtained by using FAAS as a reference method (e(r) < 2.8%).

[Cryptococcus neoformans chemotyping. Review of the literature. New epidemiologic data on cryptococcosis. Our experience with the use of C.G.B. media in the study of this yeast].

The purpose of this work was to collect the main information from the literature about the biotyping of Cryptococcus neoformans. The more up-to date research concerning the epidemiology of cryptococcosis comprising quite a few articles, mainly after the advent of AIDS, was also reviewed. The Cryptococcus neoformans varieties neoformans and gattii are well defined biochemically nowadays chiefly through the C.G.B. medium, according to Kwon-Chung et al. (1982). The isolation of C. neoformans var. gattii from flowers and leaves of Eucalyptus camaldulensis and Eucalyptus tereticornis, specially in Australia, through the works of Ellis & Pfeiffer (1990) and Pfeiffer & Ellis (1992) permitted very interesting epidemiological investigations on C. neoformans, a capsulated yeast by which Sanfelice, in Italy (1894; 1895) attracted attention of medical class. Busse, in 1894, described the first human case of cryptococcosis under the presentation of a bone lesion simulating sarcoma. In this paper, the Brazilian researchers focused on this subject were pointed out, followed by the Author's experience with the C.G.B. medium (L-canavanine, glycine and bromothymol blue) proposed by Kwon-Chung et al. (1982) with very good results. It was possible with such medium the study of 50 C.N.S. liquor samples, being 39 from AIDS patients (78%) and 11 from non-AIDS ones (22%). Thirty-seven out of the 39 HIV-positive patients (74%) were identified as C. neoformans var. gattii. From the negative HIV, 8 (16%) were classified as C. neoformans var. neoformans and 3 (6%) as C. neoformans var. gattii. We could not perform the serotyping of the above referred samples. It is evident anyway that in Brazil there exist both varieties gattii and neoformans, agents of neurocryptococcosis, including AIDS patients. The importance of neurocryptococcosis, mainly among AIDS patients, is stressed here, showing once more the value of C.G.B. medium in the typing of C. neoformans in its two varieties. Also, it is of relevant importance the demonstration that some species of eucalyptus may act as "host-trees" of C. neoformans var. gattii.

Characterization of discontinuous buffer junctions using pH indicators in capillary electrophoresis for protein preconcentration.

An effective sample preconcentration technique for proteins and peptides was recently developed using capillary electrophoresis (CE) with discontinuous buffers [C.A. Nesbitt, J.T.-M. Lo, K.K.-C. Yeung, J. Chromatogr. A 1073 (2005) 175]. Two buffers of different pH created a junction to trap the sample molecules at their isoelectric points and resulted in over 1000-fold preconcentration for myoglobin within 30 min. To study the formation of pH junctions in CE, a pH indicator, bromothymol blue, is used in this work to reveal the pH changes at the discontinuous buffer boundary. Bromothymol blue (BTB) exhibits a drastic change in its visible absorption spectrum (300-600 nm) going from the acidic to basic pH conditions, and is therefore ideal for visualizing the changes in pH at the junctions created by various buffer combinations. Preconcentration of myoglobin was performed in discontinuous buffers containing BTB. Major differences in the BTB absorption profiles were identified from buffer systems that differ significantly in preconcentration performance, which in turn, allowed for the identification of ideal buffers for sample preconcentration. Up to 2000-fold preconcentrations of myoglobin were achieved in the buffer systems studied in this work. In addition, the role of the electroosmotic flow (EOF) on the preconcentration performance was investigated. A low EOF was found to be desirable, as the pH junction could stay longer in the capillary for accumulation of proteins. The pH junction also displayed characteristics to resist bandbroadening. Potential laminar flow resulted from the mismatched residual EOFs under the two pH conditions within the discontinuous buffers appeared to have minimal effect on the preconcentration. In fact, external applied pressure can be used to control the migration of the pH junction without compromising the protein preconcentration.

Bromothymol blue broth: improved medium for detection of Ureaplasma urealyticum (T-strain mycoplasma).

Bromothymol blue (B) broth for the cultivation, detection, and identification of Ureaplasma urealyticum is described. In this medium, strains Cook and 960 had shorter generation times (60 min or less) and reached higher populations (over 10(8)) than have yet been reported for this species. Furthermore, the indicator changes color before the end of logarithmic growth, and the cultures retain viability for at least 1 day thereafter, greatly simplifying the handling of the organism. When the populations in cultures of these two strains and seven new isolates were determined, growth was detected earlier and proceeded to higher final titers in B broth than in urease test color medium (U-9 broth). The inclusion of antibiotics in B broth for use in clinical laboratories (B/NL broth) made the medium selective, specific, and more sensitive for the isolation of U. urealyticum. Comparison of B/NL broth with genital mycoplasma (GM) agar and U-9 broth for the primary isolation of U. urealyticum was made with 183 urethral swabs. All 70 isolates were detected on B/NL broth, but only 66 and 63 isolates were detected on GM agar and in U-9 broth, respectively. Moreover, the cultures in B/NL broth were pure and at titers that generally showed good correlation with colony counts on GM agar.

Hydrophilic region of lecithin membranes studied by bromothymol blue and effects of an inhalation anesthetic, enflurane.

A pH-indicator dye, bromothymol blue, was used to probe the hydrophilic surface of dimyristoyl-, dipalmitoyl-, and distearoylphosphatidylcholine bilayer vesicles. The apparent pK of the surface-adsorbed dye was larger than the bulk pK value. The contribution of the choline positive charge on the dissociation constant of the dye adsorbed on the vesicle surface was estimated by screening the charge interaction with 2 M KCl. The effective surface potentials interacting with the dye were thus estimated to be 33.2, 45.6, and 46.8 mV, respectively, for the dimyristoyl-, dipalmitoyl-, and distearoylphosphatidylcholine vesicles. From the differences between the obtained effective potentials and the calculated surface potentials of the charge-determining plane of the choline head, the distances between the prototropic part of the dye and the choline charge-determining plane were estimated to be 10.5, 8.0, and 7.8 A, respectively. These values were obtained at 25 degrees C; the dimyristoylphosphatidylcholine membrane was in the liquid-crystalline phase and the other two were in the solid gel phase. Addition of an inhalation anesthetic, enflurane, decreased the distance in the dimyristoylphosphatidylcholine vesicles and increased the distance in the dipalmitoyl- and distearoylphosphatidylcholine vesicles. The increase of precessional motion of choline head by the inhalation anesthetic is apparently responsible for the changes.

Interference of magnetic resonance imaging contrast agents with the serum calcium measurement technique using colorimetric reagents.

A possible interaction between either linear (Gd-DTPA-BMA and Gd-DTPA) or macrocyclic (Gd-DOTA) gadolinium complexes used as magnetic resonance imaging (MRI) contrast agents and colorimetric technique reagents for the measurement of serum calcium was evaluated on human serum pools, and its mechanism was investigated by means of UV spectrometry and electro-spray ionization mass spectrometry (ESI-MS). The highest concentration tested was 2.5 mM (corresponding to a putative strictly intravascular distribution of the compound) and the lowest dose was 0.2 mM (i.e. about two elimination half lives). Serum calcium was dosed in duplicate by conventional colorimetric techniques involving o-cresol-phthalein complexone (OCP) or methylthymol blue (MTB) as reagents. No interference was detected when mixing Gd DOTA with serum, whatever the concentration. Gd DTPA (2.5 mM) did not interfere with the colorimetric technique either. Conversely, the Gd DTPA-BMA solution induced a concentration-related variation in apparent calcium levels. In the UV experiments, solutions of 2.5 mM MRI contrast media were mixed with OCP or MTB and UV absorption spectra were recorded between 400 and 800 nm. For Gd-DOTA/OCP and Gd-DOTA/MTB, no significant variations in the absorbance were detected. However, in the presence of Gd DTPA BMA, the absorbance of OCP and MTB showed substantial and immediate variations over time. The ESI-MS studies showed a complete displacement of Gd3+ ion in the case of Gd-DTPA BMA. In the presence of OCP, we observed the disappearance of Gd-DTPA BMA and the formation of the free ligand DTPA BMA and a new complex Gd OCP with an original stoichiometry of 2/2. Such a phenomenon did not occur in the case of Gd DOTA and Gd DTPA. The decomplexation of Gd-DTPA BMA in the presence of OCP can probably be explained by the weaker thermodynamic stability of Gd-DTPA BMA compared to that of Gd-DOTA and Gd DTPA.

Anesthetic-protein interaction: surface potential of bovine serum albumin estimated by a pH-sensitive dye.

It is often contended that inhalation anesthetics act on proteins via perturbation of lipid membranes. However, direct interaction between anesthetics and water-soluble proteins also has been demonstrated. We postulate that the anesthetic action is directed to the interface between water and macromolecules, irrespective of lipid membranes or proteins. The present study deals with anesthetic effects upon interfacial properties of a water-soluble, crystalline delipidated bovine serum albumin. A pH-indicator dye, bromothymol blue, was used to probe the surface potential of the protein. When a pH-indicator dye binds to a macromolecule, the pH, indicated by the color of the dye, differs from the bulk pH measured by a pH meter. This is because the pH of the microscopic area, where the dye is adsorbed, differs from the bulk due to the surface electrostatic potential that interacts with hydrogen ions (electrostatic terms), and the physical property that affects the color of the dye at the bound region is different from the bulk (nonelectrostatic terms). The mismatch between the bulk pH and the color of the bound pH indicator can be used to probe the property of the dye binding site. By screening the electrostatic effects with high ionic strength, the anesthetic effects upon the nonelectrostatic term were shown to be negligible under the present experimental conditions; the pH-color mismatch was mainly caused by the anesthetic effect upon the electrostatic potential of the macromolecular surface interacting with the dye. Accordingly, the surface potential of the dye binding site was estimated from the mismatch. It was found that inhalation anesthetics decreased the surface potential. The partial pressures of diethylether, enflurane, and methoxyflurane that decreased the surface potential by 10 mV were 2.1 X 10(-2), 1.7 X 10(-2), and 0.17 X 10(-2) bar, respectively, which were in agreement with the minimal alveolar concentrations of these anesthetics to achieve surgical anesthesia.

Spectral analysis of biochemical reactions used for identification of bacteria.

To define parameters for optimizing automated discrimination of bacterial biochemical reactions certain theoretical considerations of spectrophotometric analysis were explored. One-hundred and one recent clinical isolates of gram-negative bacilli (21 species) were inoculated into AP1 20 E strips and read manually after 24 hours. With spectrophotometric scanning, the AP1 reactions could be classified into three analytical categories: pH change, production of new products, and darkening of the medium. Whereas single wavelength analysis gave 2.9% disagreement from the visual, multiple wavelength analyses were uniformly more accurate. The best results for pH change reactions were obtained by calculating a ratio of two wavelengths. New color reactions were best interpreted by demonstration of the new peak, whereas darkening reactions required quantitation of the area under the entire curve. With these methods, a 99.3% overall agreement of individual reactions and a 97% agreement of identification were achieved. Multiple-point analysis of spectra coupled with computerized interpretation of the data should help resolve the problem of equivocal reactions in bacterial identification schemes optimized for spectral analysis.
