A description of the damaged DNA produced during tissue injury.

Warm ischemia produces DNA damage which is characterized by both single- and double-strand breaks with 5'-PO4 and 3'-OH ends. In contrast, cold ischemia produces mostly single-strand breaks for the first 60 hr and then, abruptly, double-strand damage is produced. Cold ischemia produces both 5'-OH and 5'-PO4 termini, but 5'-OH ends do not appear until after 24 hr of storage. Cold ischemia, also produces 3'-PO4 ends but we have not found any 3'-OH termini. Mechanisms for DNA degradation during warm and cold ischemia are presented to account for these results. It is suggested that damage to the genetic code may be prevented if degradation of the DNA can be confined to the production of the easily repairable 5'-PO4/3'-OH opposed ends.

A new hemoglobin variant altering the alpha 1 beta 2 contact: Hb Chemilly alpha 2 beta 2 99(G1)Asp leads to Val.

Hemoglobin Chemilly (alpha 2 beta 2 99(G1)Asp leads to Val), a high oxygen affinity variant, was uncovered in the red blood cells of a polycythemic patient who reported to the hospital concerning periodic headaches. We describe the molecular abnormality and functional studies of this new abnormal Hb. beta 99(G1)Asp, an invariant residue of hemoglobin, is considered a key amino acid for conformational changes between the R in equilibrium T quaternary structures responsible for the allosteric behavior of hemoglobin. Hb Chemilly exhibits a high O2 affinity, very low cooperativity and reduced Bohr effect. Its functional abnormalities are compared to the 5 other Hb variants at site beta 99(G1) described up to now of the 7 single base substitutions predictable from the genetic code.

Enzymatic synthesis of dihydrosirohydrochlorin (precorrin-2) and of a novel pyrrocorphin by uroporphyrinogen III methylase.

Uroporphyrinogen III methylase was purified from a recombinant hemB-strain of E. coli harbouring a plasmid containing the cysG gene. N-terminal analysis of this purified protein gave an amino acid sequence corresponding to that predicted from the genetic code. From the u.v./visible spectrum of the reaction catalysed by this SAM dependent methylase it was possible to observe the sequential appearance of the chromophores of a dipyrrocorphin and subsequently of a pyrrocorphin. Confirmation of this transformation was obtained from 13C-NMR studies when it was demonstrated, for the first time directly, that uroporphyrinogen is initially converted into dihydrosirohydrochlorin (precorrin-2) and then, by further methylation, into a novel trimethylpyrrocorphin.

Eclampsia and fetal sex.

The study of the possible importance of the sex of the fetus on the frequency and clinical characteristics of 777 singleton pregnancies complicated by antepartum eclampsia showed that; (1) the male to female ratio for the total group was 1.23/1, (2) this figure was 1.27/1 in first pregnancies and 1.32/1 in patients less than 20 years old, (3) the ratio increased to 1.44/1 in women more than 30 years of age, and (4) in contrast, 169 postpartum eclampsia cases had a male to female ratio of 1.07/1. There were no statistically significant differences in the obstetric profiles, the main eclamptic symptoms or the fetomaternal mortality figures attributable to the sex of the fetus. However, since eclampsia is more severe in older women, the clinician may see more complications and maternal deaths associated with male fetuses because of their significantly higher ratio. In addition, these findings suggest that the presence of a male fetus with a hormonal and genetic code more different from the mother than with female offspring may be an accessory to the fundamental causes of this disease.
