Use of protein chip mass spectrometry to monitor biotinylation reactions.

Surface-enhanced laser desorption/ionization time-of-flight analysis was used to monitor both the kinetics and heterogeneity of product formation during the biotinylation of a number of model proteins and peptide targets. The selected molecules were the IgG-binding protein, protein A, human serum albumin, and a synthetic peptide corresponding to the N terminus of a streptococcal M1 protein. The extent of biotinylation was determined by kinetic analysis of the shift in molecular mass from the native material. Each residue modified by reaction with N-hydroxysuccinimide biotin resulted in an addition of approximately 341 amu to the native protein or polypeptide. The novelty of the method was in the ability to determine the molecular mass shift, without first separating the targeted molecule from the biotinylating reagent. The analysis was rapid, simple, and provided information on the average number of biotin molecules added and the homogeneity of the resulting product.

Self-actuated, thermo-responsive hydrogel valves for lab on a chip.

An easy to fabricate, thermally-actuated, self-regulated hydrogel valve for flow control in pneumatically driven, microfluidic systems is described. This microvalve takes advantage of the properties of the hydrogel, poly(N-isopropylacrylamide), as well as the aqueous fluid itself to realize flow control. The valve was designed for use in a diagnostic system fabricated with polycarbonate and aimed at the detection of pathogens in oral fluids at the location of the sample collection. The paper describes the construction and characterization of the hydrogel valves and their application for flow control, sample and reagent metering, sample distribution into multiple analysis paths, and the sealing of a polymerase chain reaction (PCR) reactor to suppress bubble formation. The hydrogel-based flow control is electronically addressable, does not require any moving parts, introduces minimal dead volume, is leakage and contaminant free, and is biocompatible.

Electrokinetic measurements of dielectric properties of membrane for apoptotic HL-60 cells on chip-based device.

The specific membrane capacitance and conductance of mammalian cells reflect the surface morphological complexities and barrier functions of cell membrane, respectively, and could potentially respond to cell physiological and pathological changes in a measurable manner. In this study, an electrokinetic system was developed by using negative dielectrophoretic force (nDEP force) assisted positioning and electroroation (ROT) measurement. Numerical simulations regarding the geometric model of the electrode were performed primarily for the electric field analysis. The dielectric responses of membrane for apoptotic HL-60 cells induced by bufalin were detected. The membrane capacitance of the cells was found to fall from an initial value of 15.6 +/- 0.9 mF/cm(2) to 6.4 +/- 0.6 mF/cm(2) after a 48 h treatment with 10 nM bufalin. However, the membrane conductance remained almost constant at (2.25 +/- 1.1) x 10(3) S/m(2) during the first 12 h of bufalin treatment and then increased distinctly to (4.2 +/- 1.3) x 10(3) S/m(2) thereafter. Scan electron microscopy (SEM) studies of the cells revealed a decreased complexity in cell membrane morphology following bufalin treatments, suggesting that the observed changes in the membrane capacitance was dominated by the alterations of cell surface structures. The results demonstrate that the ROT technique gives a quantitative analysis of the toxic damage by chemicals to cells and can be exploited in the testing and development of new pharmaceuticals and active cell agents.

Electrochemical detection of pathogen infection using cell chip.

The immobilized cell using self-assembled synthetic oligopeptide was applied to the electrochemical detection of pathogen infection. Thin films based on cysteine-terminated synthetic oligopeptides were fabricated for the immobilization of HeLa cell on gold (Au) substrate. Layer formation and immobilization of the cell were investigated with surface plasmon resonance (SPR) and electrochemical impedance spectroscopy (EIS). Experimental results showed that the thin film of cysteine-terminated synthetic oligopeptide was successfully fabricated and it could be applied for the immobilization of HeLa cells. The adhered living cell was exposed to E. coli O157:H7, which induced the change of SPR angle and electrochemical impedance signal. The proposed cell immobilization method using self-assembly technique can be applied to construct the cell microarray for on-site pathogen monitoring.

Characterization of trypsin immobilized on the functionable alkylthiolate self-assembled monolayers: a preliminary application for trypsin digestion chip on protein identification using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

Self-assembled monolayers (SAMs) on coinage metal provide versatile modeling systems for studies of interfacial electron transfer, biological interactions, molecular recognition and other interfacial phenomena. Recently the bonding of enzyme to SAMs of alkanethiols onto Au electrode surfaces was exploited to produce a bio-sensing system. In this work, the attachment of trypsin to a SAMs surface of 11-mercaptoundecanoic acid was achieved using water soluble N-ethyl-N '-(3-dimethylaminopropyl)carbodiimide hydrochloride and N-hydroxysuccinimide as coupling agent. The thickness of SAMs was determined by optical ellipsometer; contact angles of the modified Au surfaces were measured in air using a goniometer. The Second Harmony Generation data displays the last few percents of the alkylthiol molecules adsorbed and produced the complete monolayer by inducing the transition from a high number of gauche defects to an all-trans conformation. Using X-ray Photoelectron Spectroscopy (XPS) and Fourier-Transformed Infrared Reflection-Absorption and Attenuated Total Reflection Spectroscopes (FTIR-RAS and ATR), we examined the chemical structures of samples with different treatments. By matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS), we demonstrated the digestion of bovine serum albumin (BSA) on the trypsin-immobilized SAMs surface. Experimental results have revealed that the XPS C1s core levels at 286.3 and 286.5 eV (Amine bond), 288.1 eV (Amide bond) and 289.3 eV (Carboxylic acid) illustrate the immobilization of trypsin. These data were also in good agreement with FTIR-ATR spectra for the peaks valued at 1659.4 cm(-1) (Amide I) and 1546.6 cm(-1) (Amide II). Using MALDI-TOF MS observations, analytical results have demonstrated the BSA digestion of the immobilized trypsin on the functionalized SAMs surface. For such surfaces, BSA was digested on the trypsin-immobilized SAMs surface, which shows the enzyme digestion ability of the immobilized trypsin. The terminal groups of the SAMs structure can be further functionalized with biomolecules or antibodies to develop surface-base diagnostics, biosensors, or biomaterials.

The treatment of leukoderma after burns by a combination of dermabrasion and "chip" skin grafting.

The treatment of leukoderma after burns can be difficult. The white patches are often extensive and the adjacent scar tissue itself may need excision and this further increases the size of the final defect. Moreover, when sheet skin grafts are used it is difficult to produce an inconspicuous border between skin graft and surrounding normal skin. We have developed a new technique to treat depigmentation following burns with minimal donor site morbidity and encouraging results.

Tissue reactions following apical plugging of the root canal with infected dentin chips. A histologic study in dogs' teeth.

The object of the present work was to observe whether the condensation of a plug of infected dentin in the apical region, before the filling of a root canal, could show results similar to those found in the absence of contamination. The histologic analysis of the periapical tissues of dogs' teeth containing infected apical plugs showed results that were totally unfavorable, when compared to cases in which the plugs were absent. Such data do not encourage the use of dentin plugs for cases of contaminated root canals, at least until other experiments can better clarify the problem.

Phase II study of iproplatin (CHIP, JM-9) in advanced squamous cell carcinoma of the head and neck.

A phase II study of iproplatin was conducted in advanced squamous cell carcinoma of the head and neck both in patients previously treated with chemotherapy or not. Iproplatin was given intravenously every 4 weeks at the dose of 240-300 mg/m2 without hydratation. A total of 101 4-week courses was given to 50 eligible patients. No antitumor activity was detected. Drug-induced toxicity consisted in moderate myelosuppression, frequent but mild nausea and vomiting, and occasionally diarrhea and stomatitis. This study demonstrates that iproplatin given in this dose and schedule is devoid of activity in advanced squamous cell carcinoma of the head and neck.

Phase II study of iproplatin (CHIP, JM-9) in advanced testicular cancers progressing after prior chemotherapy.

Twenty-two patients with advanced testicular cancer received iproplatin at a dose of 180-240 mg/m2 every 4 weeks. All the patients progressed or recurred after chemotherapy including cisplatin. The most severe toxicity was thrombocytopenia with two toxic deaths after a first cycle of 240 and 180 mg/m2 respectively. Nausea and vomiting were almost universal but mild in intensity. One renal failure occurred 6 weeks after the first cycle while the tumor was progressing. No antitumor activity was observed in this heavily pretreated population of patients.

From neural chip and engineered biomolecules to bioelectronic devices: an overview.

At the first C.E.C. Workshop, in Brussels on 28-29 November 1991, attended by over 70 leading European scientists and industrialists, bioelectronics was defined as 'the use of biological materials and biological architectures for information processing systems and new devices'. At the end of the Frankfurt Workshop, bioelectronics, specifically bio-molecular electronics, was described as 'the research and development of bio-inspired (i.e. self-assembly) inorganic and organic materials and of bio-inspired (i.e. massive parallelism) hardware architectures for the implementation of new information processing systems, sensors and actuators, and for molecular manufacturing down to the atomic scale'. The subject of this overview is to summarize some of the most significant progress in bio-molecular electronics from neural VLSI networks and bio-molecular engineering. As an example of one possible route, emphasis is placed on the results recently obtained within this laboratory.

Using surface plasmon resonance to directly determine binding affinities of combinatorially selected cyclopeptides and their linear analogs to a streptavidin chip.

In our recent report, several HPQ-containing streptavidin ligands were identified from a structurally constrained combinatorial library, and the relative affinities in IC(50) of these tight-binding ligands were revealed by a captured enzyme-linked immunosorbent assay. In the present work, surface plasmon resonance was employed to directly evaluate the binding affinities between immobilized streptavidin and combinatorially selected ligands. The equilibrium dissociation constants and kinetic on/off rates of a previously identified N-to-side chain and newly synthesized N-to-C cyclopeptides were readily deduced using Scatchard analysis and computational simulation. It was found that both cyclopeptides bound streptavidin far more tightly than its linear counterpart ( approximately 1000-fold), while the reversed (QPH) linear and cyclic peptidyl ligands were hardly recognized by streptavidin. Consequently, not only was the binding specificity of synthetic ligands distinguished qualitatively but also the entropic advantage of conformationally constrained cyclopeptides over their linear forms was demonstrated quantitatively by surface plasmon resonance.
