Intracerebroventricular vs. subcutaneous drug administration: apples and oranges?

For numerous reasons, the icv route is commonly used to administer endogenous opioid peptides and other newly discovered or synthesized substances and may often indicate whether the drug has actions on the brain. Effects are then routinely compared to the actions of prototypic agents given by some systemic route, such as sc or ip. Unfortunately, there is little appreciation that qualitative as well as quantitative differences can be seen when the same drug is administered by central as opposed to peripheral routes. Numerous factors undoubtedly contribute to the final effect. Whatever the explanation, failure to recognize that such differences may occur could result in incorrect conclusions as to the involvement of specific receptor types or subtypes.

Buoyant density and molecular weight calculations on an Apple II microcomputer from equilibrium banding experiment on analytical ultracentrifuge.

Buoyant density is one of the non-destructive measurements of viruses, nucleic acids etc. which help in characterizing these entities in terms of their chemical composition and physical conformation. In recent years equilibrium sedimentation in CsCl buoyant density gradient has been proved to be a powerful tool for genome characterization. The method enables one to obtain information such as the absolute and the relative buoyant density of the material and its molecular weight. This communication describes a program in BASIC which calculates these parameters from the raw data obtained directly from the experiment. The program is fully interactive, user-oriented, and written keeping in view biologists and biochemists as the main users. The program has a built-in option to calculate either the absolute or the relative buoyant density and the molecular weight.

On-line data acquisition system using an Apple computer: ISI and PST histograms.

A combined hardware/software system for the study of the temporal distributions of spike data is described. Through extensive use of assembly language subroutines the program is able to simultaneously acquire, analyze and display the incoming data. Production of a "Temporal Memory Map" makes possible the examination of acquired data at a wide variety of different bin widths. Interaction with the user is made extremely simple. Provisions are included for the labelling and printing of the output histograms and for the compact storage of data onto floppy disks. Maximum temporal resolution is 0.1 millisecond and maximum total acquisition width is 12.8 minutes. Hardware requirements beyond the basic computer consist of an external signal conditioning circuit and a slot-independent peripheral card containing two integrated circuits. The system has been successfully used in the analysis of neural and EMG evoked responses and should be widely useable in other applications.

Antioxidant defense system in the apple snail eggs, the role of ovorubin.

A novel role of ovorubin as a protection system against oxidative damage in eggs from Pomacea canaliculata was investigated. Carotenoid composition, and their antioxidant capacity, as well as the carotenoid-apoprotein interaction, were studied for this lipoglycocarotenoprotein. Carotenoid extracts from ovorubin were analysed by TLC and spectrophotometry. The major carotenoid was astaxanthin in its free (40%), monoester (24%), and diester (35%) forms, mainly esterified with 16:0 fatty acid. The antioxidant capacity of ovorubin carotenoids was studied by the inhibition of microsomal oxidation in a non-enzymatic system, showing strong protection against oxidative damage (IC50=3.9 nmol/mg protein). The carotenoid-apoprotein interaction was studied by spectrophotometry and electrophoresis using reconstituted ovorubin. Astaxanthin does not seem to affect the structural characteristics of ovorubin, however the carotenoid-protein association significantly protected astaxanthin against oxidation. Ovorubin therefore, besides its role in providing energy and structural precursors during embryogenesis, would be an antioxidant carrier, protecting at the same time this pigment from oxidation in the perivitellin fluid environment of the egg.

An apple a day keeps the doctor away: children's evaluative categories of food.

This study explores how children evaluatively categorize foods based on their nutritional value. Three-year-olds, four-year-olds, seven-year-olds, and adults completed a task in which they categorized a list of 70 foods as healthy or junky. The results showed important developmental differences in participants' ability to accurately classify foods as healthy/junky and to provide relevant justifications for these classifications. These results suggest that a large amount of category learning occurs with development, especially as children incorporate different types of information about food nutrition into their evaluative category representations.

Flies and their golden apples: the effect of dietary restriction on Drosophila aging and age-dependent gene expression.

Reduced nutrient availability (dietary restriction) extends lifespan in species as diverse as yeast, nematode worms, Daphnia, Drosophila, and mammals. Recent demographic experiments have shown that moderate nutrient manipulation in adult Drosophila affects current mortality rate in a completely reversible manner, which suggests that dietary restriction in Drosophila increases lifespan through a reduction of the current risk of death rather than a slowing of aging-related damage. When examined in the light of the new demographic data, age-dependent changes in gene expression in normal and diet-restricted flies can provide unique insight into the biological processes affected by aging and may help identify molecular pathways that regulate it.
