Polymorphism of clarithromycin.

It is well recognized that physicochemical properties of drugs are affected by the type of polymorphic crystalline form of drugs. Clarithromycin is known to exist in at least three polymorphic crystalline forms. Since conventional means to obtain the most thermodynamically stable form (Form II) for the antibiotics is known to be associated with a low purity of the stable form, we developed a novel method to improve the purity of the crystalline form by a modification of the preparation process. The new method involved a simple recrystallization of clarithromycin in solvents having 5-12 carbon atoms (e.g., hexane and heptane) or ethers with 4-10 carbon atoms (e.g., isopropyl ether) and, thus, less likely to be associated with the problem in purity of resulting crystal. Differential scanning calorimetry and powder X-ray diffraction were used to compare the crystalline form of the resultant powder with Form II crystal prepared by the conventional method. The crystal prepared by the new method was identical to Form II crystal of the conventional method as evidenced by the lack of the exothermic peak near 110 degrees C in differential calorimetry scan, indicating that Form II crystal could be readily prepared by the new process. Therefore, these data indicated that the improvement in the purity of the Form II crystal for clarithromycin as well as a significant cost reduction is likely by the novel method.

[Rheological phase synthesis and characterization of lanthanum salicylate and luminescence properties of Tb3+ -doped lanthanum salicylate].

Lanthanum salicylate and Tb3+ -doped lanthanum salicylate were synthesized with the rheological phase reaction method. Elemental analysis, IR, TG, DTA and powder X-ray diffraction were investigated to determine the composition, crystal structure and coordination manner between the COO- and ion La3+ of lanthanum salicylate. The emission and excitation spectra of Tb3+ -doped lanthanum salicylate were also discussed. Powder X-ray diffraction suggests that the compound has a layered monoclinic structure, and the lattice parameters are a = 21.6010 A, b = 13.8015 A , c = 3.8103 A, beta = 97.11 degrees, V = 1127.2 A3, Z = 2, rhocal = 1.621 g x cm(-3) and rhoexp = 1.653 g x cm(-3). The Tb3+ -doped lanthanum salicylate exhibits very strong green luminescence of Tb3+ under the excitation of UV light. And the transition from 5D4 to 7F5 is the strongest one.

Sobrerol enantiomers and racemates: solid-state spectroscopy, thermal behavior, and phase diagrams.

The characterization of the solid state of sobrerol enantiomers and racemates has been accomplished by a number of techniques on solid phase such as thermal analysis (DSC) and spectroscopy (IR, 13C NMR, and X-ray diffraction both on powders and on single crystal). Experimental and theoretical binary phase diagrams of cis- and trans-sobrerol enantiomers and their mixtures have been drawn and are discussed. Thermal analysis allowed, moreover, the detection of cis racemate polymorphism. Finally, the quantitative analysis of the cis racemate as an impurity of the trans racemate by means of microcalorimetric determinations is reported.

Organoapatite growth on an orthopedic alloy surface.

We report here a method to coat orthopedic metals with the artificial bone material organoapatite. The growth of organoapatite on titanium alloy surfaces of foils and porous cylinders involves sequential preadsorption of poly(L-lysine) and poly(L-glutamic acid) on metal, followed by exposure to organoapatite-precipitating solutions. The organoapatite characterization of the coating was carried out by transmission electron microscopy, electron diffraction, scanning electron microscopy, energy-dispersive X-ray scattering, powder X-ray diffraction, FT-IR, and elemental analysis. The preadsorbed poly(amino acids) in the form of a self-assembled bilayer of oppositely charged macromolecules can lead to a surface coverage of titanium alloy in the range of 70-90%. The deposition mechanisms could involve the surface capture of embryonic crystals and the nucleation of apatite on the bilayer. Bioabsorbable organoapatite could serve as a tissue-engineering scaffold for bone regeneration into porous implants.

Cation mobility and kinetics of ion exchange in zirconium hydrogen monothiophosphate hydrate, Zr(HPO(3)S)(2)x1.5H(2)O.

The ion conductivity of zirconium hydrogen monothiophosphate (Zr(HPO(3)S)(2)x1.5H(2)O) has been measured by impedance spectroscopy. The measured value of proton conductivity is 3 x 10(-5) S/cm at 298 K. Conductivity was shown to decrease with increasing temperature due to a dehydration process. Above 450 K, the conductivity is likely governed by proton transport in the anhydrous phase Zr(HPO(3)S)(2). The activation energies of proton conductivity were measured to be 18 +/- 2 kJ/mol for Zr(HPO(3)S)(2)x1.5H(2)O and 60 +/- 3 kJ/mol for the anhydrous compound. The kinetics of ion exchange was studied with the use of potentiometric titration for several ion pairs, H(+)/Na(+), H(+)/Zn(2+), and Na(+)/Zn(2+) in Zr(HPO(3)S)(2)x1.5H(2)O. The diffusion coefficient values for H(+)/Na(+) ion exchange in Zr(HPO(3)S)(2)x1.5H(2)O are lower than those reported in alpha-zirconium phosphate. At the same time, the mobility of zinc ions in Zr(HPO(3)S)(2)x1.5H(2)O is higher than sodium ion mobility. The ion exchange H(+)/Zn(2+) is accompanied by the slow hydrolysis of the initial compound. In all cases, the powdered solids were evaluated by powder X-ray diffraction, and particle sizes were controlled by grinding and sieving the powders.

[Identification on Chinese materia medica Radix Rehmanniae preparata by X-ray diffraction Fourier pattern method].

OBJECTIVE: To develop a new identification and analysis method of Chinese medicinal materia Radix Rehmanniae Preparata. METHODS: Powder X-ray Diffraction Fourier Pattern. RESULTS: Experiments and analysis were carried out on five samples. The standard X-ray diffraction Fourier Pattern and characteristic diffraction peaks of Radix Rehmanniae Preparata were obtained. CONCLUSION: This method can be used for identification on Chinese medicinal materia Radix Rehmanniae Preparata.

Cross-beta order and diversity in nanocrystals of an amyloid-forming peptide.

The seven-residue peptide GNNQQNY from the N-terminal region of the yeast prion protein Sup35, which forms amyloid fibers, colloidal aggregates and highly ordered nanocrystals, provides a model system for characterizing the elusively protean cross-beta conformation. Depending on preparative conditions, orthorhombic and monoclinic crystals with similar lath-shaped morphology have been obtained. Ultra high-resolution (<0.5A spacing) electron diffraction patterns from single nanocrystals show that the peptide chains pack in parallel cross-beta columns with approximately 4.86A axial spacing. Mosaic striations 20-50 nm wide observed by electron microscopy indicate lateral size-limiting crystal growth related to amyloid fiber formation. Frequently obtained orthorhombic forms, with apparent space group symmetry P2(1)2(1)2(1), have cell dimensions ranging from /a/=22.7-21.2A, /b/=39.9-39.3A, /c/=4.89-4.86A for wet to dried states. Electron diffraction data from single nanocrystals, recorded in tilt series of still frames, have been mapped in reciprocal space. However, reliable integrated intensities cannot be obtained from these series, and dynamical electron diffraction effects present problems in data analysis. The diversity of ordered structures formed under similar conditions has made it difficult to obtain reproducible X-ray diffraction data from powder specimens; and overlapping Bragg reflections in the powder patterns preclude separated structure factor measurements for these data. Model protofilaments, consisting of tightly paired, half-staggered beta strands related by a screw axis, can be fit in the crystal lattices, but model refinement will require accurate structure factor measurements. Nearly anhydrous packing of this hydrophilic peptide can account for the insolubility of the crystals, since the activation energy for rehydration may be extremely high. Water-excluding packing of paired cross-beta peptide segments in thin protofilaments may be characteristic of the wide variety of anomalously stable amyloid aggregates.

Identification of a crystalline cyanide-containing compound in blast furnace sludge deposits.

During blast furnace operation, a cyanide-containing muddy waste referred to as blast furnace sludge is generated in large amounts. In Germany it was and is still common practice to pump this sludge into surface deposits. Depending on species, cyanide has very different toxicity. To this day there is no information about the type of cyanide occurring in blast furnace sludge deposits. In order to identify the type of cyanide we investigated by means of wet chemical and powder X-ray diffraction analyses 37 samples of three blast furnace deposits. Wet chemical results indicate that both the extremely toxic free cyanide (HCN and CN ) and toxic weak metal-cyanide complexes, for example [Zn(CN)4]2-, are not present in the sludge. By powder X-ray diffraction we identified the crystalline cyanide-containing compound potassium zinc hexacyanoferrate(II) nonahydrate, K2Zn3[Fe(CN)6]2 x 9H2O, as the cyanide-bearing compound. Our study is the first that identifies potassium zinc hexacyanoferrate(II) nonahydrate in the environment. As the iron-cyanide complex [Fe(CN)6] is not acutely toxic, any direct hazard comes from cyanide occurring in the investigated wastes. Under the predominant pH milieu of the sludge (pH about 8) the solubility of potassium zinc hexacyanoferrate(II) nonahydrate is low, thus minimizing the mobility of cyanide.

Evolution of the structural chemistry of vanadium organodiphosphonate networks and frameworks: structural consequences of fluoride incorporation in the development of stable phases with void channels.

Hydrothermal reactions of solutions containing a vanadate source, an organodiphosphonate, an organonitrogen component, and HF (V/P/O/F) yield a series of oxyfluorovanadium-diphosphonates with charge-compensation provided by organoammonium cations or hydronium cations. While V/P/O/F networks provide the recurrent structural motif, the linkage between the layers and the details of the polyhedral connectivities within the layers are quite distinct for the five structures of this study. [H2pip][V4F4O2(H2O)2{O3P(CH2)3PO3}2] (1) (pip = piperazine) is a conventional three-dimensional (3D) "pillared" layer structure, whose V/P/O/F networks are buttressed by the propylene chains of the diphosphonate ligands. In contrast, [H2en][V2O2F2(H2O)2{O3P(CH2)4PO3}] (2) and [H2en]2[V6F12(H2O)2{O3P(CH2)5PO3}2 {HO3P(CH2)5PO3H}] (3) are two-dimensional (2D) slablike structures constructed of pairs of V/P/O/F networks sandwiching the pillaring organic tethers of the diphosphonate ligands. Despite the common overall topology, the layer substructures are quite different: isolated {VO5F} octahedra in 2 and chains of corner-sharing {VO(3)F(3)} octahedra in 3. The 3D structure of [H2en]2[V7O6F4(H2O)2{O3P(CH2)2PO3}4].7H2O (4.7H2O) exhibits a layer substructure that contains the ethylene bridges of the diphosphonate ligands and are linked through corner-sharing octahedral {VO6} sites. The connectivity requirements provide large channels that enclose readily removed water of crystallization. The structure of [H3O][V3F2(H2O)2{O3P(CH2)2PO3}2].3.5H2O (5.3.5H2O) is also 3D. Because of the similiarity with 4.7H2O, it exhibits V/P/O/F layers that include the organic tethers of the diphosphonates and are linked through corner-sharing {VO6} octahedra. In contrast to the network substructure of 4.7H2O, which contains binuclear and trinuclear vanadium clusters, the layers of 5.3.5 H2O are constructed from chains of corner-sharing {VO4F2} octahedra. Thermal studies of the open framework materials 4 and 5 reveal that incorporation of fluoride into the inorganic substructures provides robust scaffoldings that retain their crystallinity to 450 degrees C and above. In the case of 4, dehydration does not change the powder X-ray diffraction pattern of the material, which remains substantially unchanged to 450 degrees C. In the case of 5, there are two dehydration steps, that is, the higher temperature process associated with loss of coordinated water. This second dehydration results in structural changes as monitored by powder X-ray diffraction, but this new phase is retained to ca. 450 degrees C. The materials of this study exhibit a range of reduced oxidation states: 1 is mixed valence V(IV)/V(III) while 2 and 4.7H(2)O are exclusively V(IV) and 3 and 5.3.5H2O are exclusively V(III). These oxidation states are reflected in the magnetic properties of the materials. The paramagnetism of 1 arises from the presence of V(III) and V(IV) sites and conforms to the Curie-Weiss law with C = 2.38 em K/(Oe mol) and = -66 K with mu(eff) (300 K) = 4.33 mu(B). Compounds 3-5 exhibit Curie-Weiss law dependence of magnetism on temperature with mu(eff) (300 K) = 5.45 mu(B) for 3 (six V(III) sites), mu(eff) = 4.60 mu(B) for 4 (seven V(IV) sites) and mu(eff) = 4.13 mu(B) for 5 (two V(III) sites). Compound 2 exhibits antiferromagnetic interactions, and the magnetism may be described in terms of the Heisenberg linear antiferromagnetic chain model for V(IV). The effective magnetic moment at 300 K is 2.77 mu(B) (two V(IV) sites).

Water vapor adsorption behavior of sodium deoxycholate anhydrous forms.

Pseudopolymorphism of sodium deoxycholate (NaDC) was investigated. Intact NaDC (dihydrate) was dried at 60 degrees C under reduced pressure resulting an anhydrous amorphous phase. On the other hand, intact NaDC was altered to an anhydrous crystalline form by heating at 200 degrees C. The water vapor adsorption and desorption isotherms of dehydrated NaDCs were determined using an automatic gravimetric water vapor adsorption analyzer. In the case of NaDC dehydrated at 60 degrees C, the weight was increased in rising relative humidity and it was transformed into the NaDC tetrahydrate above 60% RH, which was identified by TG/DTA and powder X-ray diffraction. During the water vapor adsorption process of the sample dehydrated at 200 degrees C, the NaDC dihydrate was obtained in the range of 50 to 70% RH and then transformed into the NaDC octahydrate above 85% RH. The NaDC octahydrate was characterized by TG/DTA and powder X-ray diffraction for the first time. During the desorption process, the octahydrate was changed to the tetrahydrate between 80 and 40% RH.

Two-dimensional [TIn2] polyanions in BaTIn2 (T=Rh, Pd, Ir, Pt)--the collapse of the three-dimensional indium polyanion of BaIn2.

The title compounds were prepared from the elements by reactions in sealed tantalum tubes in a high-frequency furnace. The four compounds were investigated by X-ray diffraction both on powders and single crystals, and the structures of the rhodium and platinum compounds were refined from single-crystal data: Cmcm, a = 447.68(8), b = 1131.1(2), c = 805.6(2) pm, wR2 = 0.0561, 354 F2 values for BaRhIn2; a = 452.06(8), b = 1162.4(2). c = 801.5(1) pm, wR2 = 0.1427, 362 F2 values, for BaPtIn2: with 16 variables for each refinement. The structures are isopointal to MgCuAl2 and can be considered to be a transition metal (T) filled CaIn2 type, in which the indium atoms form a distorted network like hexagonal diamond (lonsdaleite). The indium substructure is cut apart in BaTIn2 and resembles together with the transition metal atoms a two-dimensional polyanion rather than a three-dimensional polyanion as found in the compounds CaTIn2, CaTSn2, and SrTIn2. Semiempirical band structure calculations support the assumption of a two-dimensional polyanion in which the strongest interactions are found for the T-In contacts.

Hexacyanometalate molecular chemistry: heptanuclear heterobimetallic complexes; control of the ground spin state.

Following a bottom-up approach to nanomaterials, we present a rational synthetic route from hexacyanometalates [M(CN)(6)](3-) (M=Cr(III), Co(III)) cores to well-defined heptanuclear complexes. By changing the nature of the metallic cations and using a localised orbital model it is possible to control and to tune the ground state spin value. Thus, with M=Cr(III), d(3), S=3/2, three heptanuclear species were built and characterised by mass spectrometry in solution, by single-crystal X-ray diffraction and by powder magnetic susceptibility measurements, [Cr(III)(CNbondM'L(n))(6)](9+) (M'=Cu(II), Ni(II), Mn(II), L(n)=polydentate ligand), showing spin ground states S(G)=9/2 [Cu(II)], with ferromagnetic interactions J(Cr,Cu)=+45 cm(-1), S(G)=15/2 [Ni(II)] and J(Cr,Ni)=+17.3 cm(-1), S(G)=27/2 [Mn(II)], with an antiferromagnetic interaction J(Cr,Mn)=-9 cm(-1), (interaction Hamiltonian H=-J(Cr,M) [S(Cr)Sigma(i)S(M)(i)], i=1-6). With M=Co(III), d(6), S=0, the heptanuclear analogues [Co(III)(CN-M'L(n))(6)](9+) (M'=Cu(II), Ni(II), Mn(II)) were similarly synthesised and studied. They present a singlet ground state and allow us to evaluate the weak antiferromagnetic coupling constant between two next-nearest neighbours M'-Co-M'.

Aluminum orthovanadate (AlVO4): synthesis and characterization by (27)Al and 51V MAS and MQMAS NMR spectroscopy.

Polycrystalline samples of AlVO(4) have been prepared by two methods of synthesis and characterized by (27)Al and (51)V MAS NMR spectroscopy at 14.1 T. The MAS NMR spectra clearly reveal that essentially pure samples with minor impurities of V(2)O(5) and alumina have been obtained. From these samples, (27)Al quadrupole coupling parameters and isotropic chemical shifts as well as the magnitudes and relative orientations of the (51)V quadrupole coupling and chemical shift tensors have been determined with high precision for AlVO(4). These data have been obtained from a combined analysis of multiple-quantum (MQ) MAS NMR spectra and MAS NMR spectra of the central and satellite transitions. The (27)Al and (51)V NMR data show that the asymmetric unit for AlVO(4) contains three isolated VO(4) tetrahedra, one pentacoordinated Al site, and two AlO(6) octahedra. This is in agreement with the supposition that AlVO(4) is isostructural with FeVO(4) and with a recent structure refinement for AlVO(4) based on powder X-ray diffraction (XRD) data. The favorable agreement between the refined crystal structure from powder XRD and the NMR parameters is apparent from a convincing correlation between experimental (51)V quadrupole tensor elements and calculated (51)V electric field gradient tensor elements obtained by the point-monopole approach. An assignment of the (27)Al NMR data is obtained from similar calculations of the (27)Al electric field gradients and by estimation of the distortion of the AlO(6) octahedra.

Disordered zinc in Zn4Sb3 with phonon-glass and electron-crystal thermoelectric properties.

By converting waste heat into electricity, thermoelectric generators could be an important part of the solution to today's energy challenges. The compound Zn(4)Sb(3) is one of the most efficient thermoelectric materials known. Its high efficiency results from an extraordinarily low thermal conductivity in conjunction with the electronic structure of a heavily doped semiconductor. Previous structural studies have been unable to explain this unusual combination of properties. Here, we show through a comprehensive structural analysis using single-crystal X-ray and powder-synchrotron-radiation diffraction methods, that both the electronic and thermal properties of Zn(4)Sb(3) can be understood in terms of unique structural features that have been previously overlooked. The identification of Sb(3-) ions and Sb(2)(4-) dimers reveals that Zn(4)Sb(3) is a valence semiconductor with the ideal stoichiometry Zn(13)Sb(10). In addition, the structure contains significant disorder, with zinc atoms distributed over multiple positions. The discovery of glass-like interstitial sites uncovers a highly effective mechanism for reducing thermal conductivity. Thus Zn(4)Sb(3) is in many ways an ideal 'phonon glass, electron crystal' thermoelectric material.

X-ray diffractometer combining synchrotron radiation and pulsed magnetic fields up to 40 T.

A synchrotron X-ray diffractometer incorporating a pulsed field magnet for high fields up to 40 T has been developed and a detailed description of this instrument is reported. The pulsed field magnet is composed of two coaxial coils with a gap of 3 mm at the mid-plane for passage of the X-rays. The pixel detector PILATUS 100K is used to store the diffracted X-rays. As a test of this instrument, X-ray diffraction by a powder sample of the antiferromagnet CoO is measured below the Néel temperature. A field-dependent lattice distortion of CoO due to magnetostriction is observed up to 38 T.

Pharmaceutical properties of freeze-dried formulations of egg albumin, several drugs and olive oil.

The freeze-dried ternary formulations of meclizine (MZ, an anti-motion sickness drug), prednisolone (PRED, an anti-inflammatory drug) and norfloxacin (NFLX, an anti-microbial drug) which are poorly water-soluble and are low bioavailability drugs, were prepared using egg albumin and olive oil. The powder X-ray diffractions, the dissolution rate and the bioavailabilities in vivo of these formulations were studied in comparison with each drug alone. By forming ternary formulations of these drugs, the dissolution rates of the drugs from the formulations were significantly improved compared with each drug alone. The results of their powder X-ray diffraction measurements showed that these drugs in the ternary formulations presented in an amorphous form, indicating increased dissolution rates. On the other hand, the plasma concentrations of these drugs increased significantly after oral administration in formulations to rats, except for the NFLX formulation, and the areas under the concentration-time curves (AUC) of the ternary formulations of MZ, PRED and NFLX were 2.1, 1.6 and 1.3 times those of the drugs alone, respectively. From these results, it was proven that formulations consisting of egg albumin, olive oil and poorly water-soluble drugs were useful preparations for improving the drug's disadvantageous pharmaceutical properties.

Preparation, characterization and in vitro dissolution studies of solid dispersion of meloxicam with PEG 6000.

The poor solubility and wettability of meloxicam leads to poor dissolution and hence showing variations in bioavailability. The present study is aimed to increase solubility and dissolution of the drug using solid dispersion techniques. The solid binary systems were prepared at various drug concentrations (5-40%) with polyethylene glycol 6000 by different techniques (physical mixing, solvent evaporation). The formulations were characterized by solubility studies, differential scanning calorimetry, fourier transform infrared spectroscopy and in vitro dissolution rate studies. The solubility of drug increased linearly with increase in polymer concentration showing A(L) type solubility diagrams. Infrared spectroscopy studies indicated the possibility of hydrogen bonding with polymer. The differential scanning calorimetry and powder X ray diffraction demonstrated the presence of polymer as eutectica or monotectica in solid dispersion along with the physical characteristics of the drug (crystalline, amorphous or a mixture of both). The solid dispersions of the drug demonstrated higher drug dissolution rates than physical mixtures and pure meloxicam, as a result of increased wettability and dispersibility of drug in a solid dispersion system.

Crystal structure prediction for eniluracil.

State-of-the-art molecular modeling tools were used to predict the crystal structure of eniluracil, a compound for which it has not been possible to grow a single crystal. Two methods were used, one that incorporates molecular structure and powder X-ray diffraction data and another that employs molecular structure and lattice energy calculations into the search algorithm. Two structures were identified, one with P2(1)/c and the other with P2(1) symmetry, both of which are consistent with the infrared and Raman spectra. A detailed analysis of the simulated and experimental powder X-ray diffraction patterns indicates that the P2(1)/c structure is the best representation of the crystal structure.
