Demonstration of beta 1H globulin together with C3 in the dermal-epidermal junction of patients with systemic lupus erythematosus.

Skin lesions and clinically normal skin from 10 patients with systemic lupus erythematosus (SLE) were examined by immunofluorescence for the presence of C3 and its control protein, beta 1H globulin. beta 1Hwas always found in association with deposited C3; in the instance where C3 deposits were not found beta 1H was also not found. Granular deposits of C3 and beta 1H were found in the dermal-epidermal junction (DEJ) in all of the 5 nonlesional skins studied. In the lesional skin, C3 was found in 4 of 5; in 2 of these 4, beta 1H was also found. As previously demonstrated for in vitro systems, beta 1H also binds in vivo to fragments of C3, presumably C3b, generated during activation of the complement system.

In vitro synthesis of factor B of the alternative pathway of complement activation by mouse peritoneal macrophages.

Factor B of the alternative pathway of complement activation was shown to be synthesized and secreted by unstimulated mouse peritoneal macrophages. The activity of B in the culture supernatants from macrophage monolayers was detected by consumption of C3 in reaction mixtures containing supernatant and guinea pig factors C3, D and insoluble C3b. Using a monospecific antiserum, factor B in concentrated culture supernatants was shown by immunodiffusion and immunoelectrophoresis to be identical to factor B in mouse plasma and to form a characteristic complex with cobra venom factor in the presence of D. A steady rate of factor B secretion was observed for 4 days providing the medium was changed every 24 h. Cycloheximide (0.5 mug/ml), an inhibitor of protein synthesis, caused inhibition (90%) of factor B production. Incubation of culture medium containing 14C-labeled amino acids with the macrophage monolayer resulted in incorporation of radioactivity into factor B as detected by autoradiography of precipitation lines formed with anti-B antiserum; This indicated that synthesis of factor B had occurred. In the same culture supernatants the presence of newly synthesized C3 was also demonstrated.

Haplotype analysis of the linkage group HLA-A:HAL-B:Bf and its bearing on the interpretation of the linkage disequilibrium.

The analysis of 650 HLA-A:HAL-B:Bf three-factor haplotypes revealed significant associations only between alleles of the very closely linked genes HLA-A and HLA-B, and Bf, respectively. Most striking is the highly significnat association of the rare Bf variant F1 with HLA-B18 and of S1 with HLA-B13, HLA-B14, and HLA-Bw21. Only random allele distributions were observed when considering the somewhat more distant genes HLA-A and Bf or the higher order interaction at all three genes. From these findings it seems likely that the linkage disequilibrium within the MHC is not due to selective forces, but rather due to a short evolutionary period.

Bf polymorphism: study of a new variant (F0.55).

A variant band in the Bf polymorphism has been found in the serum of a healthy women, her mother, her sister, and two brothers. By direct comparison the band was found to migrate faster than F but slower than F1. It is highly probable that the band represents a new allele at the Bf locus, Bf 0.55. The new allele was transmitted in this family together with the HLA haplotype A11, B27.

Genetic loci of components of the classical and alternate pathway of complement activation: a new dimension of the immunogenetic linkage group (HLA) on chromosome 6 in man.

In this review, a working hypothesis is put forward that functional cooperation of various types of cells and proteins in immune recognition, mediation and response is maintained by a common chromosomal region which evolved over millions of years from a common ancestor by gene duplication. In brief, the known functions of the H-2 complex are discussed (susceptibility and resistance to viral infection, immune response genes, T-B cell interaction as non-self recognition and response). The addition of loci of the classical and alternate pathway of complement activation to the HLA region (i.e., C2, C4 and the Bf system) is reviewed with respect to functional relationship to immune recognition and mediation mechanisms. As expected according to this hypothesis, genes for late-acting components (C3, C5, C7 and C8 in man, C5 in mice) have so far not proved to be linked to HLA.

Localization of the Bf locus within the HLA region. Report on an informative family and critical evaluation of available data on Bf mapping.

Since the estimation of recombination fractions is only an arbitrary means to map genetic loci on chromosomal regions, family studies of cases with informative cross-overs within the region in question are of decisive importance. This paper reports the study of a family with a HLA-B to HLA-D cross-over which is informative in the Bf system. The possible reasons for conflicting published data on Bf gene mapping are discussed.

Bf polymorphism: another variant (S0.8).

A "new" variant band in the Bf system has been found in the serum of three individuals belonging to a tribe of Brazilian Indians (Karaja--Bananal--Goias) and in the serum of a Caucasian individual from the area of Strasbourg. It is highly probable that the band represents another allele at the Bf locus BfS0.8.

Genetic polymorphism of C3 and serum levels of immunoglobulins, C3, C4 components of complement and C3-proactivator in four different populations of Afghanistan.

The C3 phenotype distribution was studied in 4 different populations from Afghanistan. The gene frequencies of C3S allele were: Tajiks (0,8547), Pushtoons (0.8812), Hazaras (0.9036) and Osbeks (0.8530). These values were significantly higher than in European populations studied previously. No significant differences were found between the mean serum levels of C3, C4 and C3-proactivator among 4 population groups. A higher concentration of IgG, IgA and IgM was observed in Afghanistan sera than reported for Europeans.
