Subcellular localization of gold in suprarenal testicle and thyroid glands after injection of allochrysine in rats.

Gold injected under the form of anti-rheumatoid polyarthritis soluble solutions provokes, as observed by electron microscope, a deposit of crystalline micro-needles in different tissues, like in cells of adrenal and suprarenal glands, in Leydig cells of the testicles, in isolated thyrocytes and in thyroid endothelial cells. They are present as bundles, often of incurvated type, of high electron-density, present in lysosomes, which contain moreover a hyaline, emptied vesicle. These structures are named "aurosomes". The microanalysis, using the MS 46 (Cameca) and the Camebax (MBX) demonstrates that these crystalline structures are composed by gold associated with sulphur, and not with phosphate.

[Spontaneous course of allergic bronchopulmonary aspergillosis].

This case report illustrates the spontaneous course of allergic bronchopulmonary aspergillosis (ABPA). The x-ray images cover a time-frame of nearly 20 years. All of the main criteria of ABPA are fulfilled. Despite intermittent oral corticosteroid treatment, recurrent eosinophilic infiltrates of the lungs occurred. New strategies in diagnosis and treatment of ABPA are discussed.

Chelation therapy in workers with lead exposure.

Occupational lead overexposure remains a major problem. To evaluate the settings in which physicians appropriately prescribe chelation therapy for lead exposure, 7 cases were identified from physician phone calls and mandatory laboratory reporting of elevated blood lead levels to the California Department of Health Services. In the 2 workers with the highest blood lead levels (both of whom had severe symptoms), treatment was indicated. Physicians inappropriately prescribed chelating agents to workers with ongoing lead exposure as prophylaxis against rising blood lead levels and to treat atherosclerotic heart disease. Workers' personal physicians identified lead overexposure in 5 of the 7 cases. Workplace lead medical surveillance programs mandated by the federal Occupational Safety and Health Administration were inadequate in all 5 of the workplaces where information was available.

[The effect of aurotherapy on the level of unsaturated fatty acids during the treatment of rheumatoid arthritis patients].

Patients with rheumatic arthritis displayed alterations in blood serum fatty acid spectrum such that polyunsaturated fatty acids tended to be on the decrease, arachidonic acid in particular, monounsaturated ones on the increase. Long-term aurotherapy (for up to 1.5 yr) made for normalization of serum content of unsaturated fatty acids, which observation was accompanied by clinical-and-laboratory remission of rheumatic arthritis. Crisanole effect got higher with incrementing dosage of metallic gold. Results of studies made pathogenetically validate the expediency of long-term aurotherapy in patients with rheumatic arthritis.

The effect of succimer therapy in lead intoxication using postural balance as a measure: a case study in a nine year old child.

Postural balance testing was used as a measure of the effect of therapy on a 9 year old boy with high lead levels. Following therapy with CaEDTA and succimer, the patient's postural sway responses were comparable to a low-lead (< 10 micrograms/dL) comparison group for 3 out of 4 tests which rely relatively less on the higher centers for balance. This improvement in postural balance may be attributable to the combined influence of pharmacologic and age associated maturational effects. This case study provides suggestive evidence that while chelation therapy can reduce PbB levels quickly, it can also modify gross neuromotor function manifested by postural balance characteristics.

Management of idiopathic pulmonary fibrosis: prognostic indicators.

There now exists a number of approaches that can be used to predict likely responses to therapy and disease progression in idiopathic pulmonary fibrosis. These tools should be used in management decisions. It must be remembered that destroyed, fixed fibrotic lung will not improve and that the computed tomography pattern and extent of the "fibrotic" pattern should be used to determine if the best response to treatment that can be expected is a prevention of deterioration rather than improvement.

Antioxidant action via p53-mediated apoptosis.

The biological effects of antioxidants are often considered in terms of their effects on oxygen or lipid radicals. However, antioxidants can also exert their effects through altering the cellular redox potential. Herein, we report that sulfur-containing antioxidants such as N-acetylcysteine and dimercaptopropanol induced apoptosis in several transformed cell lines and transformed primary cultures but not in normal cells. In contrast, chain-breaking antioxidants such as vitamin E lacked this activity. An increased glutathione level was not required for apoptosis; however, all apoptosis-inducing antioxidants elevated the total cellular thiol levels. Antioxidant-induced apoptosis required the p53 tumor suppressor gene. N-Acetylcysteine elevated p53 expression posttranscriptionally by increasing the rate of p53 mRNA translation rather than by altering the protein stability. The p53 induction occurred in normal cells. These observations indicate a redox sensor for p53 induction in vivo, with additional transformation-specific information being required for apoptosis. Manipulating p53-dependent apoptosis with nontoxic antioxidants may have a direct clinical application.

Interference by 2, 3-dimercapto-1-propanol (BAL) in angiotensin I radioimmunoassay.

The angiotensinase inhibitor 2,3-dimercaptopropanol (BAL) interferes with peptide-antibody binding when certain sensitive antisera are used in angiotensin I radioimmunoassay systems. Three of nine antisera tested showed sufficient interference to produce serious errors in data obtained using these antisera together with BAL. For PRA determinations in human plasma, at both pH 5.7 and pH 7.3, relationships between different PRA'S are altered, and results of renin stimulation tests are changed in unpredictable ways. Determination of renin concentration in rat plasma does not require use of BAL as inhibitor, and it is best avoided. For human plasma renin determinations, use of BAL-sensitive antisera should be avoided, since there is no satisfactory way to correct data for the resulting error. BAL itself, rather than its oxidation products, is probably the interfering substance. The interference appears to be due to an interaction between BAL and the BAL-sensitive antiserum. It is not related to the known actions of BAL as chelating or reducing agent.
